African American ancestral lineages are marked by resilience, shaped by centuries of forced migrations, the Transatlantic and domestic slave trades, and subsequent movements within the United States. These historical events have influenced both the cultural and genetic diversity of Legacy African Americans. This study examined mitochondrial DNA (mtDNA) haplogroups among 189 participants—79 Gullah Geechee and 110 African Americans—to explore the Gullah Geechee as potential progenitors of other Legacy African North American lineages. Haplogroup analysis revealed L2 and L3 as the most frequent among both groups, comprising 34.6% and 38.8% in African Americans and 28.0% and 54.7% in Gullah Geechee, respectively. …

Approximately 70% of breast cancer (BC) diagnoses are estrogen receptor positive (ER+) with ∼40% of ER+ BC patients presenting de novo resistance to endocrine therapy (ET). Recent studies identify the tumor microenvironment (TME) as having a key role in endocrine resistance in which adipose-derived stem cells (ASCs) play an essential role in cancer progression. Prior studies have indicated that ASC characteristics such as age and BMI may play a role in cancer progression. Metastasis accounts for over 90% of cancer related deaths and current studies link CD36 to the development of metastasis in several cancer models. CD36 is a scavenger receptor that is involved in fatty acid uptake and oxidized lipoprotein recognition. In both metabolic disorders and cancer, CD36 activity has been associated with lipid accumulation, insulin resistance, and tumor metastasis, making it a promising yet underexplored therapeutic target. Our current aim is to develop a library of small molecules that can be used to inhibit CD36 thereby decreasing production of oxidized low-density lipoprotein (oxLDL).

Lipoxygenases (LOXs) are non-heme iron containing enzymes that catalyze the oxygenation of polyunsaturated fatty acids (PUFAs) such as linoleic acid (LA). These lipoxygenase enzymes are found in both plants and animals. In humans, LOXs have been linked to inflammatory diseases such as asthma, atheroscelerosis, and allergic rhinitis. Plant LOXs have been primarily linked to the growth, development, and stress response of the plant. This paper focuses on Oryza sativa lipoxygenase-1 (OsLOX1), an enzyme found in rice (Oryza sativa). Per- and polyfluoroalkyl substances (PFAS) are a contaminant of emerging concern due to their chemical structure that afford a plethora of issues with regards to degradation on environmental and metabolic fronts. PFAS are amphiphilic and contain a carbon-fluorine bond, which complicates their movement through the environment and ensures their longevity in biological systems. Problem. Provider-patient communication can be sorted into two approaches: patient-centered communication (PCC) and doctor-centered communication (DCC). PCC is characterized by a patient focus, in which patients are encouraged to participate in decisions regarding their care, while DCC is characterized by a doctor focus, in which the physician acts as an informational source for patients (Bientzle et al. 2017). Urbanization affects many aspects of the environment with the potential to alter both ecological and evolutionary processes. However, while examples of urban evolution are accumulating, tests of urban eco-evolutionary dynamics are rare. We carried out a reciprocal transplantation experiment using the terrestrial isopod Oniscus asellus to test how adaptation to the urban heat island influenced ecological processes. As isopods are important decomposers, we asked if leaf-litter decomposition differed among reciprocal transplant treatments and used a separate laboratory experiment to isolate the effects of temperature on rates of isopod leaf consumption. Objective: The distribution of excess white adipose tissue (WAT) in obesity correlates with risk for comorbidities. Thus, understanding depot-specific WAT developmental mechanisms is translationally relevant. SNPs near the gene CEBPA associate with waist to hip ratio, and while C/EBPα is a recognized regulator of adipogenesis, there is no previously known role for C/EBPα in regulating adipose distribution.
Methods: We crossed Cebpa floxed mice to the AdipoQ-Cre transgenic mouse strain, generating mice with adipocyte-specific knockout of Cebpa (Cebpa_ASKO). Mice were phenotyped on a chow diet and after prolonged high-fat diet (HFD) feeding.

Introduction: Sleep disruption is an early feature of Alzheimer's disease (AD), but the cellular mechanisms linking amyloid pathology to sleep loss remain unclear.
Methods: Electroencephalography/electromyography (EEG/EMG) recordings, quantitative EEG analysis, and sleep deprivation were performed in APPswe/PSEN1dE9 (APP/PS1) mice at different stages of pathology relative to normal aging. Amyloid burden and microglial density were quantified with whole-brain light-sheet microscopy. CSF1R-mediated microglial depletion explored effects of microglia on sleep loss.
Results: Amyloid plaques caused non–rapid eye movement (NREM) sleep loss that did not worsen with increased plaque burden. Aging reduced REM sleep and eliminated sleep rebound. Amyloid pathology was associated with …

Intestinal absorption of dietary lipid is essential for systemic lipid homeostasis; however, elevated postprandial plasma lipid levels are associated with obesity and increased risk for atherosclerotic cardiovascular disease. In humans and rodents, biological sex impacts dietary triglyceride absorption, and males tend to have higher postprandial triglyceride levels compared to females, but the physiological basis for this is not well understood. Here, we show that the gene DENND5B is associated with body composition in humans and mice and that genetic deletion of Dennd5b in mice prevents postprandial plasma triglyceride elevations in both sexes. Pediatric cancer survival now exceeds 85% owing, in part, to advances and the use of combination chemotherapy treatments such as CHOP (cyclophosphamide, doxorubicin, vincristine, and prednisone). Despite its efficacy, CHOP may cause off-target effects during critical pediatric development periods such as impairments of skeletal muscle. We evaluated the acute effects of a CHOP administered to C57Bl/6J mice from postnatal day 28 to 48. CHOP slowed body-weight gain and led to a smaller gastrocnemius fiber cross-sectional area by ~25% in both sexes (n ¼ 11 or 12 males; n ¼ 8 or 9 females). Cancer cachexia is a wasting condition, primarily affecting skeletal muscle, impairing patients’ quality of life, prognosis, and survival. The molecular triggers are incompletely defined, but given prior evidence for epigenetic plasticity in muscle, we speculate that dysregulated DNA methylation plays a role in muscle transcriptional alterations mediating cachexia severity. We aimed to describe and integrate the cachexia methylome and transcriptome. Inflammation and endothelial dysfunction are key steps in the pathogenesis of atherosclerosis. Octacosanol (C28H58O, OCT), a very-long-chain saturated aliphatic alcohol (VLCA) with 28 carbons, is the main component of policosanol, a nutraceutical mixture of VLCAs (C20-C34) extracted from plants. Polycosanol in animal models is known to reduce atherosclerosis, but its mechanism of action remains unclear. This study investigates the pathways by which OCT alleviates LPS-induced inflammation in primary human aortic endothelial cells (HAECs). Systematic reviews and meta-analyses have become essential tools for synthesizing evidence to guide agricultural policy and practice, yet whether these syntheses are themselves conducted with sufficient rigor has rarely been systematically assessed. This study provides a field-wide assessment of reporting quality in agricultural evidence synthesis, examining whether existing reviews are conducted with sufficient transparency and rigor to support the policy and practice decisions they are meant to inform. We drew a random sample of 244 systematic reviews and meta-analyses from a bibliometric dataset of 1,709 articles published between 1997 and 2023, evaluating reporting quality using a 12-indicator rubric across four …

Alcohol misuse is a known risk factor for pneumonia, and up to 50% of individuals who misuse alcohol are zinc deficient. Chronic alcohol misuse can lead to alcohol-induced ciliary dysfunction (AICD), preventing effective mucociliary clearance and protection against lung infection. Because zinc salts fail to alter AICD, we hypothesized a form of zinc (Zinpro Zinc LG) with enhanced cell uptake would protect against AICD. Bioceramics are important biomaterials in biomedical engineering because of their biocompatibility, bioactivity, osteoconductivity, and structural resemblance to actual bone tissue. Novel calcium channel blockers (CCBs) with dihydropyridine (DHP) or tetrahydro-4H-chromene moieties are often chiral, so effective enantiomer separation methods are needed for continued drug development. The purpose of this dissertation is to outline a comprehensive method development model for the chiral separations of DHPs and tetrahydro-4H-chromene derivatives. The scope of this dissertation is to address analytical challenges during initial synthesis, chiral separations, and data analysis during post-processing for this class of chiral compounds. Across the span of nearly eight hundred years, from AD 700-1450, the societies of the Casas Grandes region waxed and waned and left an astounding representation in the archaeological record. Best-known through the temporally older site of Convento (occupied during what is known as the Viejo Period) and the later site of Paquimé (occupied during the Medio Period), gradual cultural expansion and florescence across this period are witnessed through a variety of findings across decades of anthropological investigation.
Along with the artifacts and architecture unveiled during initial excavations at Paquimé and Convento, remains of 654 individuals were catalogued as well. …

Identity is a human right, and forensic anthropological methods are often aimed at returning identity to decedents. Three main pathways are used for the identification of human remains: fingerprints, dental records, and DNA analysis. Identification efforts may be hindered when perpetrators attempt to obscure the victim’s identity. In cases where corrosive substances, such as hydrochloric acid (HCl) are used, dental comparison and fingerprinting often fail and genetic analysis becomes the best chance of achieving personal identification.

Background: Timely availability of vaccine packaging is critical to pharmaceutical supply chain reliability, as packaging materials play a key role in maintaining cold chain integrity during distribution. Inadequate availability of packaging materials can disrupt vaccine delivery even when finished vaccines are ready for shipment. Purpose: Childhood trauma has detrimental immediate and lifelong impacts on health. Black and African Americans experience higher rates of potentially traumatic events (PTEs) than the general population. Religiosity, a common coping mechanism for many who experience trauma, could serve to moderate the impacts. This study examines whether the importance of religion buffers against the impact of childhood exposure to PTEs on adult perceptions of health and assesses whether social support and/or coping mediate the relationship between religiosity and perceived health.

Child maltreatment remains a significant public health concern with lifelong consequences for children's health, development, and well-being. As children's caregiving environments become increasingly diverse through split-custody arrangements, blended families, kinship care, and other multi-household contexts, recognizing and assessing child safety concerns may become more complex. Although these family structures are not inherently associated with maltreatment, the distribution of caregiving responsibilities and information across multiple households may complicate child maltreatment risk identification.

Chemotherapy is an effective weapon in the fight against pediatric cancer. However efficient, treatment can lead to side effects that last long after a patient finishes their last round of chemotherapy. This thesis examines current research on chemotherapy-induced cardiotoxicity, a side effect of chemotherapy treatment caused by a class of chemotherapy drugs called Anthracyclines (such as doxorubicin and daunorubicin). While effective chemotherapy agents, these drugs have been known to cause late long-term heart damage when used to treat pediatric cancers such as Acute Lymphoblastic Leukemia (ALL), Acute Myeloid Leukemia (AML) and Pediatric Sarcoma.
