Myeloid-derived suppressor cells (MDSCs) represent an innate immune cell population comprised of immature myeloid cells and myeloid progenitors with very potent immunosuppressive potential. MDSCs are reported to be abundant in the lungs of active tuberculosis (TB) patients. We sought to perform an in-depth study of MDSCs during latent TB infection (LTBI) and active TB (ATB) using the nonhuman primate (NHP) model of pulmonary TB. We found a higher proportion of granulocytic, polymorphonuclear myeloid-derived suppressor cells (PMN-MDSCs) in the lungs of ATB animals compared to those with LTBI or naive control animals. Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused a historic pandemic of respiratory disease (coronavirus disease 2019 [COVID-19]), and current evidence suggests that severe disease is associated with dysregulated immunity within the respiratory tract. However, the innate immune mechanisms that mediate protection during COVID-19 are not well defined. Here, we characterize a mouse model of SARS-CoV-2 infection and find that early CCR2 signaling restricts the viral burden in the lung. We find that a recently developed mouse-adapted SARS-CoV-2 (MA-SARS-CoV-2) strain as well as the emerging B.1.351 variant trigger an inflammatory response in the lung characterized by the expression of …

G protein-coupled receptors (GPCRs) are the largest family of human proteins. They have a common structure and, signaling through a much smaller set of G proteins, arrestins, and effectors, activate downstream pathways that often modulate hallmark mechanisms of cancer. Because there are many more GPCRs than effectors, mutations in different receptors could perturb signaling similarly so as to favor a tumor. We hypothesized that somatic mutations in tumor samples may not be enriched within a single gene but rather that cognate mutations with similar effects on GPCR function are distributed across many receptors. Autism spectrum disorder (ASD) is a neurodevelopmental disorder with strong genetic influences. There is an increasing demand for ASD genetic testing beyond the traditionally recommended microarray and syndromic autism testing; however, the current whole genome sequencing (WGS) and whole exome sequencing (WES) methods are lacking an academic standard for WGS variant annotation, reporting, and interpretation, tailored towards patients with ASD and offer very limited interpretation for clinical significance. Using WGS data from six family trios, we demonstrate the clinical feasibility and technical implementation of an evidence-based, fully transparent bioinformatics pipeline and report framework for an ASD-focused WGS genetic report. The objective of this study was to assess the effect of Italian ryegrass (Lolium multiflorum Lam.) sward height on lamb performance. The experiment was carried out at the Research Station of UFRGS, Eldorado do Sul, Brazil. Yearling no-castrated male lambs were assessed in terms of individual animal performance and gain per hectare. The experiment was carried out at the Experimental Station of the IAPAR in Paranavaí– PR, Brazil, from October 1998 to May 1999 to evaluate the response of crossbred steers grazing pastures of Panicum maximum cv. IPR 86 fertilized with four doses of N (0; 150; 300 and 450 kg.ha-1.year). Interest in grazing systems is growing among farmers in the USA as a means of reducing feed costs for lactating dairy cows. An experiment was conducted near Gainesville, FL to compare milk production and composition and milk income minus feed costs from two pasture-based systems with those of a conventional confinement housing system over a 276-d period. - The recent popularity of sex reassignment surgery is logically untenable and immoral when understood in the light of Kantian philosophy. From a Kantian perspective of synthetic a priori judgments, I argue that a biological male cannot rationally claim to “feel like a woman inside.” As a male, any female is part of the noumenal world and cannot be known apart from perception. The statement “I feel like a woman inside” assumes all women feel the same on the inside. Kant’s explanation of the noumenal and phenomenal excludes the possibility of knowing that all women or men feel the same inside …

The characterization of the most common photosynthetic biofilms in the Nerja Cave by the continuous monitoring of the in vivo chlorophyll a (Chl a) fluorescence and the incorporation of the irradiance as a new environmental variable related to previous studies in the cave, have allowed us to improve our knowledge about the photosynthetic pattern of the biofilms of the cave. Effective quantum yield (ΔF/Fm′) and relative electron transport rate (rETR) were determined during periods of the light, whereas the maximal quantum yield (Fv /Fm) was determined during dark periods. Purpose: The purpose of this exploratory phenomenological study was to identify and describe the strategies used by exemplary rural superintendents to maintain personal resiliency based on Everly, Strouse, and McCormack’s (2015) five factors of personal resilience (active optimism, decisive action, moral compass, relentless tenacity, and interpersonal support).
Methodology: This exploratory phenomenological study used purposeful criterion sampling and recommendations from the county superintendents in the identified geographic area. Heavy-duty trucks contribute approximately 20% of fuel consumption in the United States of America (USA). The fuel economy of heavy-duty vehicles (HDV) is affected by several real-world parameters like road parameters, driver behavior, weather conditions, and vehicle parameters, etc. Although modern vehicles comply with emissions regulations, potential malfunction of the engine, regular wear and tear, or other factors could affect vehicle performance. Predicting fuel consumption per trip based on dynamic on-road data can help the automotive industry to reduce the cost and time for on-road testing. A modular and scalable strategy, which remodels 3-methylglutaric anhydride to 2-oxopiperidines bearing at least three contiguous stereocenters is described. The approach relies on the chemoselective and stereocontrolled annulation of 1,3-azadienes with the anhydride component. The resulting acid-tethered allylic 2-oxopiperidines are then engaged in several selective fragment growth processes, including catalytic denitrative alkenylation, halolactonization, and Vilsmeier–Haack functionalization.

Fisheries Research Reports
In July 2021, the Department of Primary Industries and Regional Development convened an ecological risk assessment (ERA) of the fisheries that access the State-wide Small Pelagic Scalefish Resource. The ERA considered the potential ecological impacts of the West Coast Purse Seine Fishery, South Coast Purse Seine Fishery, Purse Seine Development Zones and the recreational fishers who catch small pelagic scalefish. The assessment evaluated the impact of each fishing sector/method on all relevant retained and bycatch species, endangered, threatened and protected species, habitats and the broader environment.

More Alike Than Different: Student Perceptions Of Academic Librarians By Genders And Subject Areas, Elizabeth Price, Jody C. Fagan

This study explored differences in how undergraduate students of different genders and academic disciplines perceived academic librarians at a U.S. public university. No evidence indicated differences between students identifying as male or female, and few statistically significant differences were found among four Subject Areas (Arts, Humanities + Social Sciences, Business, Health + Education, and STEM). Our results have implications for local practice though they are not generalizable to other institutions. Despite a lack of significant findings, librarians should continue to explore ways to measure how students of different disciplines perceive academic libraries and/or librarians as valuable to their academic success.

The goals of this research project were to collect repeat samples at designated reaches and under different flow conditions along the Grand River, sample the entire Grand River over a short period of time, collaborate with wastewater treatment plant labs and technicians to analyze water samples, and to calculate a Water Quality Index (WQI) for samples collected.

Background: About 1 in 4 women and 1 in 10 men have experienced intimate partner violence (IPV; CDC, 2019). Although there have been numerous studies on IPV, our knowledge pertains to IPV in heterosexual couples. Although IPV occurs at similar rates in both opposite and same sex couples, studies suggest that individuals are more likely to evaluate IPV in opposite-sex couples as more severe compared to same-sex couples. Melanin is a major virulence factor in pathogenic fungi that enhances the ability of fungal cells to resist immune clearance. Cryptococcus neoformans is an important human pathogenic fungus that synthesizes melanin from exogenous tissue catecholamine precursors during infection, but the type of melanin made in cryptococcal meningoencephalitis is unknown. We analyzed the efficacy of various catecholamines found in brain tissue in supporting melanization using animal brain tissue and synthetic catecholamine mixtures reflecting brain tissue proportions. Solid-state NMR spectra of the melanin pigment produced from such mixtures yielded more melanin than expected if only the preferred constituent dopamine had been incorporated, …

The Gardner-Webb University community is remembering the contributions and legacy of Dr. Gerald Keown, who passed away on Dec. 14. Keown was professor emeritus of Biblical Studies and the former associate dean of the Gardner-Webb University M. Christopher White School of Divinity. He began his tenure at GWU in 1996 as a professor of Old Testament Interpretation and served as associate dean from 2006 to 2019.

Chemical investigation of the ethyl acetate extract from the marine-derived Streptomyces sp. isolate B1848 resulted in three new isoquinolinequinone derivatives, the mansouramycins E–G (1a–3a), in addition to the previously reported mansouramycins A (5) and D (6). Their structures were elucidated by computer-assisted interpretation of 1D and 2D NMR spectra, high-resolution mass spectrometry, and by comparison with related compounds. Cytotoxicity profiling of the mansouramycins in a panel of up to 36 tumor cell lines indicated a significant cytotoxicity and good tumor selectivity for mansouramycin F (2a), while the activity profile of E ( …

However, training the machine learning models, especially deep learning models require thousands to millions of samples in order to reach an acceptable accuracy level. Calcium ion plays a significant role in plant signal transduction pathways, including regulation of stress-induced gene expression. Sensor proteins such as proteins with calcium-binding C2 domains can perceive changes in cellular calcium level. Functional analysis of such sensor proteins will give us a better understanding of plant stress signaling. Synaptotagmins are a group of proteins containing C2 domains that are capable of binding calcium ions and regulate gene expression in response to external abiotic stress stimuli. This study investigated mathematics teachers’ beliefs about the nature of mathematics and the contributory factors shaping them. A survey of 200 trained and untrained mathematics teachers from public and private schools was carried out in Karachi Pakistan, a context with a strong influence of religion on the socio-cultural practices of the people. The survey was followed by a focus-group discussion with selected teachers and is the focus of this paper. This is a ground-breaking study in a hitherto under-researched socio-religious context.
