We investigated the contributions of platelet microRNAs (miRNAs) to the rate of growth and regulation of gene expression in primary ectopic tumors using mouse models. We previously identified an inhibitory role for platelets in solid tumor growth, mediated by tumor infiltration of platelet microvesicles (microparticles) which are enriched in platelet-derived miRNAs. To investigate the specific roles of platelet miRNAs in tumor growth models, we implanted pancreatic ductal adenocarcinoma cells as a bolus into mice with megakaryocyte-/platelet-specific depletion of mature miRNAs. Humpback whales are known for the plastic and variable nature of their acoustic communication, social interactions, and foraging behaviors. Distinct foraging strategies are used to capture different prey types at the surface and on the seafloor. Sounds produced during foraging are thought to facilitate intraspecific social communication. Previous studies have identified specific sound types produced by humpbacks associated exclusively with benthic foraging strategies, suggesting functions related to foraging behavior. Climate is highly variable at scales that are not captured by most weather and climate models due to local effects of topography and living systems. This variation in microclimate has been noted for many decades but has been difficult to incorporate into quantitative understanding of ecological systems and processes due to the large amount of data and complex models required to adequately describe fine-scale patterns across complex landscapes. Hemlock forests in eastern North America face a deadly threat: the invasive insect, hemlock woolly adelgid (HWA), Adelges tsugae. Early detection of this pest remains a key focus for management groups to ensure rapid response to control and stop the spread of HWA. In Chapter II, our goals were to develop an affordable, easy-to-use trap that is compatible with airborne eDNA sampling techniques and assess its efficiency as a monitoring tool for HWA. Communities worldwide are facing environmental crises such as air pollution, water shortages, climate change, and other forms of environmental change and degradation. While technical solutions for environmental change are essential, so too are solutions that consider social acceptability, value cultural relevance, and prioritize equity and social justice. Social work has a critical and urgent role in creating and implementing macrolevel social responses to environmental change. Experimental pain trials have been unable to determine whether cannabinoid analgesia is attributable to intoxication, analgesic expectancies, and/or pharmacological action. One approach to resolving this issue involves testing the effects of analgesic expectancies and cannabidiol (CBD), a non-psychoactive cannabinoid, on human pain reactivity using a balanced placebo design. Despite its frequent use for pain relief, no experimental research has tested the analgesic effects of CBD in humans. Pyrroloindolines and related systems are present in a large number of complex natural products. These core structures have generated considerable synthetic interest, as many of the compounds possess challenging, elaborate structures and interesting biological properties. Recently we have focused on using trichloroacetimidates for the synthesis of these fascinating molecules. Trichloroacetimidates can be used an electrophilic source of an alkyl group to form the pyrroloindoline directly from tryptamine derivatives. In this manner trichloroacetimidates provide a flexible solution to accessing highly functionalized pyrroloindoline core structures, needing only a catalytic amount of a Lewis acid to effect the requisite transformations.

Ketone hydrogenation:A model system of carbonyl (ketone) hydrogenation, acetone hydrogenation over supported Pt catalysts in the gas-phase fixed bed reactor was investigated to understand both the mechanism and rate controlling steps in a fundamental level. The experiments and outcomes include the detailed macro- and micro-kinetic analysis of hydrogenation of acetone. The apparent reaction orders of hydrogen and acetone, apparent activation energies, and observed kinetic isotope effects over a large range of temperatures and concentrations (compound partial pressures) are reported in this document. Seamounts are important marine habitats that are hotspots of species diversity. Relatively shallow peaks, increased productivity and offshore locations make seamounts vulnerable to human impact and difficult to protect. Present estimates of seamount numbers vary from anywhere between 10,000 to more than 60,000. Seamount locations can be estimated by extracting large, cone-like features from bathymetry grids (based on criteria of size and shape). These predicted seamounts are a useful reference for marine researchers and can help direct exploratory surveys. The experiment was conducted at the Experimental Station of Paranavaí/IAPAR-PR-Brazil, from October 1998 to May 1999, to evaluate the effects of N fertilization on Panicum maximum cv. IPR 86 pastures. Four doses of N (0; 150; 300; and 450 kg.ha-1.year-1) were applied to pastures under rotational grazing with grazing cycles (GCs) of 40 days (5 days-grazing and 35 days-rest). The effects of grazing management on seed production, seed bank size and seedling emergence patterns of Trifolium repens cv. Zapicán (WC) and Lotus corniculatus cv. San Gabriel (BFT) oversown swards were evaluated. A complete randomized block design with 4 replicate blocks was used, in which 4 grazing strategies (grazing all year (SI), summer spelling for seed production (S2), winter rest plus summer spelling (S3) and autumn rest plus summer spelling (S4)), were combined with two defoliation severalties (4 and 10 cm height post-grazing residuals). This perspective on Penn State Extension Services and plain people is based on my personal experiences as a plain person, in which I interacted with Extension Services first as a farmer, and then (while working on a doctorate) as a part of the Extension system. Penn State Extension started over a century ago and was deliberate in reaching out to plain (conservative Anabaptist) farmers since the beginning, which led to a history of trust-based cooperation. Certain aspects of Amish agriculture have been studied extensively but much is still unknown about Amish environmental attitudes and beliefs. This is due, in part, to the difficulty of directly soliciting responses from adherents. This article—part of a larger study of the Kishacoquillas Valley Amish settlement in Mifflin County, Pennsylvania—reflects on methodological challenges that arose during a study on Amish environmental attitudes and behaviors. Farmers from two Amish groups, as well as two non-Amish groups used for comparison, were interviewed about their environmental attitudes. Anabaptists have a strong history of agricultural innovation and care for the land. Their innovative spirit was forged out of persecution, migration, and the need to survive in challenging circumstances. This article examines the agricultural practices of seventeenth- and eighteenth-century Swiss and South German Anabaptist farmers and those of eighteenth-century Anabaptist immigrants to Pennsylvania. European Anabaptist tenant farmers distinguished themselves by their family-centered mixed agriculture and their investment in both the land (using manure, lime, gypsum, and crop rotation to improve the soil) and livestock (improving natural meadows and planting pastures for fodder, maintaining clean barns, practicing confinement feeding, and …

Purpose: The purpose of this qualitative multi-case study was to describe and analyze how schools that implement a STEAM program include the arts in their integrated program. The study focused on 3 K-12 schools in Southern California that are located in the Antelope Valley high desert region of the state. This multi-case study analyzed an elementary school, a middle school, and a high school that are in different districts in the area.

Cytokine production is a critical component of cell-extrinsic responses to DNA damage and cellular senescence. Here, we demonstrated that expression of the gene encoding interleukin-19 (IL-19) was enhanced by DNA damage through pathways mediated by c-Jun amino-terminal kinase (JNK) and cGAS-STING and that IL19 expression was required for the subsequent production of the cytokines IL-1, IL-6, and IL-8. IL19 expression was stimulated by diverse cellular stresses, including inhibition of the DNA replication checkpoint kinase ATR (ataxia telangiectasia and Rad3-related protein), oncogene expression, replicative exhaustion, oxidative stress, and DNA double-strand breaks. The heart is a major organ whose function is to transport nutrients and waste throughout the body. This organ can become infected by pathogens, such as viruses, bacteria, or parasites. Infection of the middle heart layer, or myocardium, is often caused by a viral agent. This disease has three stages: viral infiltration, adaptive immune system activation, and finally either viral clearance or cardiac cell remodeling. Background: African Americans (AAs) suffer a higher stroke burden due to hypertension. Identifying genetic contributors to stroke among AAs with hypertension is critical to understanding the genetic basis of the disease, as well as detecting at-risk individuals.

Methods: In a population comprising over 10,700 AAs treated for hypertension from the Genetics of Hypertension Associated Treatments (GenHAT) and Reasons for Geographic and Racial Differences in Stroke (REGARDS) studies, we performed an inverse variance-weighted meta-analysis of incident stroke. In this paper, we outline the need for a coordinated international effort toward the building of an open-access Global Ocean Oxygen Database and ATlas (GO2DAT) complying with the FAIR principles (Findable, Accessible, Interoperable, and Reusable). GO2DAT will combine data from the coastal and open ocean, as measured by the chemical Winkler titration method or by sensors (e.g., optodes, electrodes) from Eulerian and Lagrangian platforms (e.g., ships, moorings, profiling floats, gliders, ships of opportunities, marine mammals, cabled observatories). Codling moth (CM) (Cydia pomonella L.), a devastating pest, creates a serious issue for apple production and marketing in apple-producing countries. Therefore, effective nondestructive early detection of external and internal defects in CM-infested apples could remarkably prevent postharvest losses and improve the quality of the final product. In this study, near-infrared (NIR) hyperspectral reflectance imaging in the wavelength range of 900–1700 nm was applied to detect CM infestation at the pixel level for three organic apple cultivars, namely Gala, Fuji and Granny Smith. ACEs are adverse childhood experiences experienced in the first 18 years of life. They are present in over a third of the population and lead to adverse health outcomes (Karatekin, 2017). The goal of this study was to examine whether ACEs could be used to identify stress levels of college students during the COVID-19 pandemic. Data on ACEs and stress levels were collected from students actively enrolled at a college or university (N= 183). Findings indicated that high ACEs were predictive of high stress, specifically considering stressful factors experienced during the COVID-19 pandemic. The results of this study suggest the …

Introduction: Mutations in the KCNN2 gene have been linked to phenotypes of movement disorders and autism. The goal of this experiment was to use the CIUSPR-Cas9 system to create a deletion within the KCNN2 gene and to observe the effect on the expression of movement in zebra fish embryos.

There are functional benefits to exercise in muscle, even when performed late in life, but the contributions of epigenetic factors to late-life exercise adaptation are poorly defined. Using reduced representation bisulfite sequencing (RRBS), ribosomal DNA (rDNA) and mitochondrial-specific examination of methylation, targeted high-resolution methylation analysis, and DNAge™ epigenetic aging clock analysis with a translatable model of voluntary murine endurance/resistance exercise training (progressive weighted wheel running, PoWeR), we provide evidence that exercise may mitigate epigenetic aging in skeletal muscle. Hevea brasiliensis (rubber tree) is a large tree species of the Euphorbiaceae family with inestimable economic importance. Rubber tree breeding programs currently aim to improve growth and production, and the use of early genotype selection technologies can accelerate such processes, mainly with the incorporation of genomic tools, such as marker-assisted selection (MAS). However, few quantitative trait loci (QTLs) have been used successfully in MAS for complex characteristics. Recent research shows the efficiency of genome-wide association studies (GWAS) for locating QTL regions in different populations. Objectives

To compare phagocytic activities of monocytes in peripheral blood mononuclear cells (PBMCs) from acute gout patients and normal subjects, examine monosodium urate monohydrate (MSU) crystal-induced IL-1β secretion ± recombinant human proteoglycan 4 (rhPRG4) or interleukin-1 receptor antagonist (IL-1RA), and study the anti-inflammatory mechanism of rhPRG4 in MSU stimulated monocytes.

Methods

Acute gout PBMCs were collected from patients in the Emergency Department and normal PBMCs were obtained from a commercial source. Monocytes in PBMCs were identified by flow cytometry. Cognitive load refers to the amount of used working memory resources, which is limited in both capacity and duration. Predicting cognitive load from raw electroencephalogram (EEG) recordings remains a challenge because of the high degree of noise due to technical variations in the recording process and the multi-factorial nature of the mapping between the EEG data and cognitive load. We present parameter-optimized deep convolutional neural network (CNN) models to predict four levels of cognitive load.
