Viruses and their hosts have coevolved for a long time. This coevolution places both the pathogen and the human immune system under selective pressure; on the one hand, the immune system has evolved to combat viruses and virally infected cells, while viruses have developed sophisticated mechanisms to escape recognition and destruction by the immune system. SARS-CoV-2, the pathogen that is causing the current COVID-19 pandemic, has shown a remarkable ability to escape antibody neutralization, putting vaccine efficacy at risk. One of the virus’s immune evasion strategies is mitochondrial sabotage: by causing reactive oxygen species (ROS) production, mitochondrial physiology is impaired, …

S100 proteins are multifunctional calcium-binding proteins of vertebrates that act intracellularly, extracellularly, or both, and are engaged in the progression of many socially significant diseases. Their extracellular action is typically mediated by the recognition of specific receptor proteins. Recent studies indicate the ability of some S100 proteins to affect cytokine signaling through direct interaction with cytokines. S100P was shown to be the S100 protein most actively involved in interactions with some four-helical cytokines. To assess the selectivity of the S100P protein binding to four-helical cytokines, we have probed the interaction of Ca2+-bound recombinant human S100P with a panel of 32 …

In this study, we examined the interplay between protein intrinsic disorder, hepatitis C virus (HCV) infection, and signaling pathways induced by Toll-like receptors (TLRs). To this end, 10 HCV proteins, 10 human TLRs, and 41 proteins from the TLR-induced downstream pathways were considered from the prevalence of intrinsic disorder. Mapping of the intrinsic disorder to the HCV-TLR interactome and to the TLR-based pathways of human innate immune response to the HCV infection demonstrates that substantial levels of intrinsic disorder are characteristic for proteins involved in the regulation and execution of these innate immunity pathways and in HCV-TLR interaction. Disordered regions, …

Proteomic analysis revealed the preservation of many proteins in the Heslington brain (which is at least 2600-year-old brain tissue uncovered within the skull excavated in 2008 from a pit in Heslington, Yorkshire, England). Five of these proteins—“main proteins”: heavy, medium, and light neurofilament proteins (NFH, NFM, and NFL), glial fibrillary acidic protein (GFAP), and myelin basic (MBP) protein—are engaged in the formation of non-amyloid protein aggregates, such as intermediate filaments and myelin sheath. We used a wide spectrum of bioinformatics tools to evaluate the prevalence of functional disorder in several related sets of proteins, such as the main proteins and …

Complications related to atherosclerosis account for approximately 1 in 4 deaths in the United States and treatment has focused on lowering serum LDL-cholesterol levels with statins. However, approximately 50% of those diagnosed with atherosclerosis have blood cholesterol levels within normal parameters. Human fortilin is an anti-apoptotic protein and a factor in macrophage-mediated atherosclerosis and is hypothesized to protect inflammatory macrophages from apoptosis, leading to subsequent cardiac pathogenesis. Fortilin is unique because it provides a novel drug target for atherosclerosis that goes beyond lowering cholesterol and utilization of a solution nuclear magnetic resonance (NMR) spectroscopy, structure-based drug discovery approach requires milligram …
Being Black In Sports, Vongai Kwenda
Being Black In Sports, Vongai Kwenda
Emerging Writers
This essay examines how black sportspeople aren’t sheltered from racism, though they are famous and loved. In the past, black players faced segregation and more overt, aggressive forms of racism. But today, the sports world has come a long way with integration, stricter policies, and protests going global. Racism is perpetuated by racist fans online and in the stands, and sports commentators who use racial tropes rooted in biological racism. More awareness and debunking are needed. White players can take a stand in the heat of an incident to send a stronger message too. It worked in the past, so …

Journal of Mathematics and Science: Collaborative Explorations
Using a mixed-methods design, this body of work from the SUMMIT-P consortium explores possible effective conditions for the sustainable reform of STEM teaching and learning at the collegiate level. A model of catalysts for successful and sustainable change is proposed, based on five years of data collection and observations. These catalysts include institutional support, intrinsic and extrinsic motivation of faculty involved, measures of student success, institution size, prior faculty experience, faculty buy-in, and institutional culture. The discussion ends with a delve into the potential broader impacts of this work. For example, this model may help institutions better understand how to …

While the public-school classroom has increasingly become more diverse, the teaching population has not. Research studies on race and its role in the classroom have been necessary considering the predominantly white teaching force and the opportunity gap that exists in the education outcomes of students of color and that of their white peers; furthermore, a gap in rural and urban studies on white teachers working with students of color provided the additional impetus for the study. This study seeks to add to the emerging field of second-wave white teacher identity studies. The purpose of this case study was to discover …
Benchmarking Modeling Methods For G Protein Coupled Receptor Ligand Discovery And Application To Orphan Receptors Bb3, Gpr88 And Gpr52, Paige Potts
Electronic Theses and Dissertations Archive
The diversity of the known and presumed physiological roles of the members of the GPCR superfamily motivate studies to expand both pharmacological and general knowledge. Computational GPCR ligand discovery/interaction studies require either a protein structure or a large number of known ligands. There are 120 unique GPCR with structures deposited in the Protein Data Bank (August 2021), leaving many GPCR without experimentally-determined structures. Additionally, many GPCR are orphan receptors lacking known endogenous ligands (potentially lacking other known synthetic small molecule ligands). Chapter I provides a brief overview of two common GPCR ligand discovery methods—homology modeling and pharmacophore modeling. Each of …

There is a great clinical need for effective methods to prevent infection following musculoskeletal trauma caused by severe injuries or implantation surgeries. Up to 80% of infections are due to the formation of microbial biofilms, which are difficult to eradicate as they possess mechanisms to evade immune cell clearance and withstand antibiotic treatment. The persistence of biofilms as well as the rise of multi-drug resistant bacterial strains further exemplify the need for non-antibiotic therapeutics to hinder bacterial growth. By incorporating anti-biofilm agents into biomaterials, the incidence of infection will decrease, as will patient trauma and hospitalization costs. The overall research …

Programming shape changes in soft materials require precise control in directionality and magnitude of their mechanical response. Among ordered soft materials, liquid crystal elastomers (LCEs) exhibit remarkable and programmable shape-shifting when their molecular order changes. In this dissertation, we proposed a new method to fabricate complex spatially-varying ordered LCEs by a maskless projection display system. By programming the synchronization of the rotated polarizer and projected segments with different shapes, various configurations of topological defects ranging from single to two-dimensional lattice of arbitrary patterns are created in a deterministic manner. We synthesized, remotely programmed, and modeled the reversible and complex morphing …

Anger is an important aspect of the negative emotion associated with posttraumatic stress disorder (PTSD), and difficulties with anger regulation are implicated in more severe PTSD trajectories as well as associated problems such as aggression perpetration. Anger is also known to erode social relationships and is implicated in poorer psychosocial functioning and having fewer close relationships. From a diagnostic conceptualization, anger is included in both the hyperarousal symptom cluster (e.g. irritability and angry reactions) and the negative alterations in cognitions and mood symptom cluster (e.g. negative emotions like anger, fear, shame, etc). Although anger is included in the diagnostic symptoms …

Black youth with high body weights (BYHW; BMI ≥ 95th percentile) endure unique stressors that may contribute to psychopathology, but more research is needed on factors associated with fewer stress-related reactions among these youth. Utilizing linear regression modeling and controlling for youth BMIz scores, the current study examined how resilience and weight-related quality of life were associated with stress-related reactions as reported by 32 treatment-seeking BYHW (Mage = 14.19; MBMIZ = 2.59) and one of their primary caregivers (N = 64). Findings showed that dyadic differences exist in what BYHW and their caregivers perceive as associated with youth stress-related reactions. …

Vascular complications result in significant morbidity and mortality in individuals with type 1 diabetes. Certain measures such as the urinary albumin-to-creatinine ratio can assess risk for later diabetes complications by determining the amount of protein leaking from the kidneys (an early sign of kidney damage). Based on a conceptual framework, this study used growth curve and latent variable mixture modeling to evaluate individual and family characteristics, psychosocial responses, individual and family responses, and metabolic variables (hemoglobin A1c and DKA hospitalizations) that predict urinary albumin-to-creatinine ratio (ACR) trajectories over the span of three years. Adolescents (N = 142) 12-18 years old …

Evolution works on the variation present in a population and that variation is the individual. But, what makes an individual? Part of this is the physical and genetic individuality, but for animals a large part of their individuality comes from the consistent behaviors that they express that are different from others. These behaviors can be developed innately, such as those that are considered ‘personality,’ or learned through their experiences. These effects compound each other and will impact the fitness of each animal. Additionally, the individuality of other organisms can be vital information. If individuals vary in behavior, then the appropriate …
Voices From The Classroom: Perspectives Of Elementary Teachers On Teacher Absenteeism, And The Related Factors Of Leadership Behaviors And Teacher Preparation, Amanda Brabham

Employee absence can arise in any organization or institution; however, in education, teacher absenteeism place burdens on schools and hinders the ability of schools to provide quality and consistent instruction to students. Teacher absenteeism has been a topic of discussion in the West Tennessee study school district for many years. School and district leaders have pondered over what can be done to improve the attendance behaviors of teachers. Research asserts that teachers are the most important in-school determinant of student success, and for this reason, school and district leaders in the West Tennessee study district must gain an understanding of …
Mate Preference And Parental Investment In Wild Songbirds, Kelly Danielle Miller
Mate Preference And Parental Investment In Wild Songbirds, Kelly Danielle Miller
Electronic Theses and Dissertations Archive
In sexually reproducing species, fitness maximization requires obtaining a mate whose optimal reproductive strategy may not be compatible with one’s own optimum. Thus, the classic tradeoff between investment in current offspring vs. surviving to produce future offspring is shaped by various decisions, including one’s choice of mate. Because males and females face fundamentally different selective pressures, the “perfect match” seldom exists, requiring that individuals shift investment strategies according to social and environmental conditions. I examined these tenets of life-history evolution in two species of wild songbird. First, I manipulated incubation effort of female Carolina wrens to assess the role of …

Transgender athletes face discrimination based on negative societal attitudes in many life arenas; they particularly confront prejudice in the arena of sport. This study examined the attitudes of some of the most influential people in an athlete’s life, coaches. The study examined coach gender, conformity to masculinity norms (particularly hegemonic masculinity norms of power over women and heterosexual self-presentation), and level of physical contact in sport as related to negative attitudes toward transgender athletes. In light of the recent spate of anti-transgender legislation focusing on transwomen athletes, attitudes toward directionality of transition within transgender athletes were also investigated. Data were …

The capital femoral physis is a growth plate located between the head of the femur and the femoral neck and forms a temporary joint where growth cartilage differentiates into bone by endochondral ossification during development. A slip occurs when a shear stress across the physis overcomes the mechanical integrity of the bone-cartilage-bone interface, known as slipped capital femoral epiphysis. Though this disorder is widely studied the etiology is not completely understood. Joint morphology may play a critical role in stability, until the joint closes in early adulthood. During development morphological changes in the joint emerge and provide natural mechanical mechanisms …

It is reported that 7.5 million African Americans are diagnosed with mental illness, and up to 7.5 million more may be affected without formal diagnosis (Davis, 2005). Acknowledging a 2:1 gender ratio of depression, women may be over-represented in this population (Immerman & Mackey, 2003). African American women’s lower rates of utilization of mental health services, which has been suggested to be impacted by stigma surrounding mental health, serves as an additional barrier for the attainment of optimal mental health. Thompson-Sanders et al. (2004) suggested that Black women who believe that living with psychological pain is part of her role …

James Baldwin describes the Black male in the White gaze as a “fixed star,” meaning that there is only one way that Black males can be, and that way is deviant. However, we often do not examine what happens in Black-male psychology because of the social ascription of criminality, via tropes like the “Myth of the Black-Male Rapist” and “thug.” In this project, I investigate what this does to Black-male freedom and self-recognition. I make the argument that narratives influence one’s concept of self and that the political negotiation one must do in return impedes their freedom. To help solve …

This research is situated at the nexus of race, racism, and care in education law, policy, and practice. State takeovers of schools and school districts in the United States have been rising for over thirty years, significantly increasing in the past decade. These takeovers raise questions of equity, accountability, integrity, and capability in many areas, specifically in terms of educating the nation's youth living in urban settings. State-led reconstitutions of schools and school districts have been and continue to be linked to racialized policy agendas, most notably those that seek to disenfranchise, dispossess, and confine Black and Brown students, families, …

The neuropeptide oxytocin alters the salience of rewards and is being investigated as a treatment for substance use disorder; however, the effects of oxytocin on reward-related neural circuits are not understood. The detection of rewarding stimuli is regulated in part by the mesolimbic dopamine system, with dopaminergic cell bodies in the ventral tegmental area (VTA) that project to the nucleus accumbens (NAc). The current study aimed to determine the effect of oxytocin receptor activation in the NAc on mesolimbic dopamine release. Fixed potential amperometry was used to measure stimulation-evoked dopamine release in the NAc of anesthetized C57BL/6J mice before and …

Animals must be able to collect and utilize information from their environment, such as olfactory cues, to make decisions that impact their fitness. Meadow voles make excellent experimental subjects for studying the functional aspects of olfactory communication. While several body sources of meadow voles were previously observed to communicate the vole’s sex, I aimed to determine whether each source also communicates information distinct from other sources. This was accomplished by utilizing a habituation-test method. I found that scent marks from three body sources of a female meadow vole communicated information that was both redundant and distinct from one another. In …

Proton-coupled electron transfer (PCET) occurs in many different types of chemical reactions. PCET involves the transfer of protons and electrons from one location to another. It is involved in both organic and inorganic chemistry in systems such as those of photosynthesis, metabolism, and respiration. The theoretical formulations of PCET reactions are founded on the theoretical treatment of electron transfer. These reactions lie on a spectrum from hydrogen atom transfer (HAT) to multiple-site couple proton-electron transfer (MS-CPET). The properties that any given reaction has can be quantified with the use of a potential energy surface (PES). The specific geometry of the …
