Lysosomal membrane permeabilization (LMP) and cathepsin release typifies lysosome-dependent cell death (LDCD). However, LMP occurs in most regulated cell death programs suggesting LDCD is not an independent cell death pathway, but is conscripted to facilitate the final cellular demise by other cell death routines. Previously, we demonstrated that Caenorhabditis elegans (C. elegans) null for a cysteine protease inhibitor, srp-6, undergo a specific LDCD pathway characterized by LMP and cathepsin-dependent cytoplasmic proteolysis. We designated this cell death routine, lysoptosis, to distinguish it from other pathways employing LMP. In this study, mouse and human epithelial cells lacking srp-6 homologues, mSerpinb3a and SERPINB3, …

A graphene oxide (GO)-based cortisol biosensor was developed to accurately detect corti-sol concentrations from sweat samples at point-of-care (POC) sites. A reference electrode, counter electrode, and working electrode make up the biosensor, and the working electrode was functional-ized using multiple layers consisting of GO and antibodies, including Protein A, IgG, and anti-Cab. Sweat samples contact the anti-Cab antibodies to transport electrons to the electrode, resulting in an electrochemical current response. The sensor was tested at each additional functionalization layer and at cortisol concentrations between 0.1 and 150 ng/mL to determine how the current response differed. A potentiostat galvanostat device was …

Background: Irreversible electroporation (IRE) has emerged as a viable consolidative therapy after induction chemotherapy, in which this combination has improved overall survival of locally advanced pancreatic cancer (LAPC). Optimal timing and patient selection for irreversible electroporation remains a clinically unmet need. The aim of this study was to investigate preoperative factors that may assist in predicting progression-free and overall survival following IRE.
Methods: A multi-institutional, prospectively maintained database was reviewed for patients with LAPC treated with induction chemotherapy followed by open-technique irreversible electroporation from 7/2015-5/2019. RECIST 1.1 criteria were used to assess tumor response and radiological progression. Overall survival (OS) …

Extracellular signal-regulated kinase 3 (ERK3) is an atypical member of the mitogen-activated protein kinase (MAPK) family, members of which play essential roles in diverse cellular processes during carcinogenesis, including cell proliferation, differentiation, migration, and invasion. Unlike other MAPKs, ERK3 is an unstable protein with a short half-life. Although deubiquitination of ERK3 has been suggested to regulate the activity, its ubiquitination has not been described in the literature. Here, we report that FBXW7 (F-box and WD repeat domain-containing 7) acts as a ubiquitination E3 ligase for ERK3. Mammalian two-hybrid assay and immunoprecipitation results demonstrated that ERK3 is a novel binding partner …

Grasslands Classification In Yobe State Nigeria Using Integrated Orderly Classification System Of Grassland (Iocsg), S. Usman, M. G. Bala, Y. Shen
IGC Proceedings (1977-2023)
Integrated Orderly Classification System of Grassland (IOCSG) was initiated by Professor Ji-Zhou Ren in the 1960s and it has satisfactorily served to direct theory and practice of grassland classification for the last several decades in China. In this study IOCSG is adopted to classify the grasslands in Yobe state Nigeria, using 30 years (1984-2013) data on precipitation and temperature from 12 satellite stations across the state. Humidity (moisture index) was estimated as the ratio of precipitation per annum to annual temperature. The results revealed a trend of decline in precipitation and moisture index with latitude; from 772.63 mm and 0.72 …

Forb Counts In Grassland Are Sensitive To Analytical Method, Gene A. Fults
IGC Proceedings (1977-2023)
An analysis of forbs from 9 years of Plant Census protocol (NRI 2019) is compared to data from the Line Point Intercept (LPI) protocol. Both protocols were conducted on the same geospatially separate macroplots (N=1230). The macroplots locations were stratified by NRI based on a land hierarchy which has had long-term use in the United States (Salley etal 2015). This ties vegetation and land together in the data. The analysis showed that the 15-minute plant census method increased forb species data by 29 percent over the LPI method which takes 40-60 minutes. Both methods captured similar total ratios and rank …

IGC Proceedings (1977-2023)
Grasslands are known to host crop pests and may act as the reservoir of pests affecting maize production. This research aimed at determining the effect of grassland composition on abundance of stemborers on grasses sorrouding maize farms in western Kenya. The experimental set up followed a complete random block design (CRBD); with four elevations at Lambwe, Homabay, Luanda and Mt. Elgon assigned as blocks based on different altitudes. Sampling was done in two seasons, during the short rain season of 2019 and the long rain season of 2020. Grasslands surrounding four pairs of push-pull and non-push-pull maize farms were assessed. …

Degradation Issue Of Fescue-Forbs Rangeland In The Mountain Steppe Of Mongolia, Otgontuya Lkhagvajav, Lkhagvajav Namdag, Munkhbat Chimed
IGC Proceedings (1977-2023)
The total area of Fescue-forbs rangeland in Mongolia is 10 million hectares, accounting for 8.5% of total rangeland area. The Fescue-forbs rangeland is the main type of rangeland in the mountain steppe. The Mongolian rangeland has degraded in the last two decades mainly due to overgrazing and climate change. The main objective of this study was to determine the change of plant community and reduction of frequency and productivity of the dominant species Festuca lenensis in different degradation states of rangeland. The dominant species and main plant functional groups have completely changed in Fescue-forbs rangeland in all three states of …

An Opisthacanthus asper (Peters, 1861) (Hormuridae) shows a relatively rare example of pedipalp regeneration in which the lost tibia and tarsus was replaced by a smaller, curved element of uncertain homology to either the fixed or free finger. A comparable abnormal palp described in the literature hints that pedipalps can only regenerate a structure of this form, regardless of the site of amputation. An Olivierus caucasicus (Nordmann, 1840) (Buthidae) is described in which claws (pretarsus) of leg III regenerated directly at the distal end of the tibia, while in leg IV the claws regenerated at the end of a truncated …

The idea that there are 2 distinct processing modes is seen throughout social and cognitive psychology research. One mode is generally fast, automatic, and relatively effortless, while the other is slow, systematic, and effortful. One mechanism of effortful processing is cognitive reflection which is one’s ability to reflect on their intuition. The Cognitive Reflection Test (CRT) is a 3-item measure that quantifies a person’s ability to recognize an intuitive answer as incorrect and identify the correct response. Additionally, a yea-yeaing score was collected to measure how often a person agrees with a statement and its opposite (i.e., endorses contradictory beliefs). …

There are many different ways to convert a carboxylic text acid to an aldehyde. A common way to accomplish this transformation is to use multiple steps (e.g., reduction of the carboxylic acid to the corresponding alcohol followed by selective oxidation to the aldehyde). Our success in reducing the diacid muconic acid (1) to muconaldehyde (4) in a relatively mild, one-pot process has led us to examine whether the method can be expanded to other carboxylic acids. We report here our findings using benzoic acid as a representative substrate.

SARS-CoV-2, the virus responsible for the COVID-19 of which several variants have emerged, such as the B.1.351 SARS-CoV-2 variant. The Receptor Binding Domain (RBD), located within the Spike protein is an immunogenic epitope for potent neutralizing antibodies. Current mRNA vaccines encode for the Spike protein, allowing the body to build antigen-specific antibodies. Assays measuring protective antibodies are essential to manage the COVID-19 pandemic and can be used as a platform for variant screening. RBD-foldon 2.2 is a novel antigen produced by fusing RBD with the trimerization domain Fibritin from Bacteriophage T4. Its amino acid sequence is based on the original …

Decomposition Rates Of Organic Material Across Herbivore Treatments In A Nutrient-Rich Semi-Arid Sodic Savanna, L. L. Erasmus, H. Van Coller, S. Claassens, F. Siebert
IGC Proceedings (1977-2023)
Decomposition is a major determinant of terrestrial nutrient cycling and therefore an important regulator of ecosystem structure and function. It has been widely documented that large mammalian herbivores (LMH) act as a significant driver of changes to aboveground structure and modifications to edaphic properties. Little is known about the role of herbivory, and particularly the loss thereof, in mediating essential ecological processes in a herbivore-adapted system. The Nkuhlu exclosures, a large-scale, long-term exclusion experiment in the Kruger National Park, South Africa, provided an opportunity to explore the effects of herbivory and/or its long-term exclusion on decomposition and stabilisation of detrital …

Comparative Anatomical Study Of The Grasses In The Range Lands Of Kovilpatti, Tamil Nadu Of India, B. Makesh Kumar, J. Stephan, P. Kumar
IGC Proceedings (1977-2023)
Grasses are the crucial component of the rural rangeland ecology. Besides the morphological studies, micro-morphological characters such as stem anatomy were also investigated in this study. So as to get detailed understanding about variations during the summer and monsoon within species and also differentiating the species with the help of supplementary characters such as number of cortical layers and vascular bundles, xylem vessel length etc. In grass anatomy the characters are rather constant for some species and highly variable for others probably often due to the climatic variation. Still the characters are very helpful to differentiate and determine the genus. …

Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has become a serious threat to global public health, underscoring the urgency of developing effective therapies. Therapeutics and, more specifically, direct-acting antiviral development are still very much in their infancy. Here, we report that two hepatitis C virus (HCV) fusion inhibitors identified in our previous study, dichlorcyclizine and fluoxazolevir, broadly block human coronavirus entry into various cell types. Both compounds were effective against various human-pathogenic CoVs in multiple assays based on vesicular stomatitis virus (VSV) pseudotyped with the spike protein and spike-mediated syncytium formation. The antiviral effects …

Identifying the composition of avian diets is a critical step in characterizing the roles of birds within ecosystems. However, because birds are a diverse taxonomic group with equally diverse dietary habits, gaining an accurate and thorough understanding of avian diet can be difficult. In addition to overcoming the inherent difficulties of studying birds, the field is advancing rapidly, and researchers are challenged with a myriad of methods to study avian diet, a task that has only become more difficult with the introduction of laboratory techniques to dietary studies. Because methodology drives inference, it is important that researchers are aware of …

These could potentially alter regeneration trajectories following severe fire or through cumulative impacts of climate-mediated drying, fire, and/or anthropogenic disturbance. We used lidar-derived point cloud metrics, and site-specific locational attributes to assess trajectories of post-disturbance vegetation regeneration in boreal peatlands south of Fort McMurray, Alberta, Canada using a space-for-time-chronosequence. The objectives were to (a) develop methods to identify conifer trees vs. deciduous shrubs and trees using multi-spectral lidar data, (b) quantify the proportional coverage of shrubs and trees to determine environmental conditions driving …

The objective of this case-control study was to determine if feeding behavior data collected from an automated milk feeder (AMF) could be used to predict neonatal calf diarrhea (NCD) in the days surrounding diagnosis in pre-weaned group housed dairy calves. Data were collected from two research farms in Ontario between 2017 and 2020 where calves fed using an AMF were health scored daily and feeding behavior data (milk intake (mL/d), drinking speed (mL/min), number of rewarded or unrewarded visits) was collected. Calves with NCD were pair matched to healthy controls (31 pairs) by farm, gender, and age at case diagnosis …
Heterologous Production Of Plant Secondary Metabolites Resveratrol And Caffeic Acid In E.Coli And Synechocystis Sp. Pcc 6803, Yong Xue
Theses and Dissertations
Caffeic acid and resveratrol are polyphenol compounds produced by plants as secondary metabolites to protect against biotic and abiotic stresses. In the last decades, numerous studies have demonstrated the beneficial effects of these compounds on human health as anticancer, antioxidant, anti-virus and anti-inflammatory agents. In plants, both caffeic acid and resveratrol are synthesized either from phenylalanine or tyrosine. Recently, heterologous production of phenylpropanoids using genetically engineered bacteria is gaining more and more attentions from industry because it can dramatically improve the yield with reduced costs. The major aim of the work is to explore the possibility of using Synechocystis or …
Massively Parallel Assessment Of The Functional Effects Of De Novo Autism Spectrum Disorder Patient-Derived Genetic Variants In 3’ Untranslated Regions, Tomas Lagunas Jr
Massively Parallel Assessment Of The Functional Effects Of De Novo Autism Spectrum Disorder Patient-Derived Genetic Variants In 3’ Untranslated Regions, Tomas Lagunas Jr
Arts & Sciences Graduate Student Theses and Dissertations
Human genetic studies have identified a large number of patient-derived de novo variants in presumptive regulatory regions of the genome that pose a challenge for interpretation of their effects since the impact of regulatory variants is highly dependent on the cellular context. In particular, current investigations have made significant investments in sequencing the genomes of autism spectrum disorder (ASD) families as a diagnostic method and to further understand the genetic architecture of this complex neurodevelopmental disease. These studies have revealed an enrichment of mutations in the untranslated regions (UTRs) of genes, which are noncoding regulatory regions, in ASD genomes. Assessment …
