This work addresses three objectives related to the design and modeling of an instrumented knee sleeve and its high deflection strain gauges for at-home knee rehabilitation. First, a multi-objective Bayesian approach was developed to economically optimize a high deflection strain gauge's mechanical and electrical behavior. Four objective functions were simultaneously optimized with two design variables in seven iterations; 90% faster than a grid search approach. The presented algorithm adjusts variables in the manufacturing process of high deflection strain gauges and produces a sensor with optimized properties for a predetermined purpose. In recent years, with the severe safety problems of organic electrolytes, all-solid-state electrolytes (ASSEs) have been considered as an appealing choice to replace organic electrolytes due to their high reliability and safety. Allometric scaling describes the relationship of trait size to body size within and among taxa. The slope of the population-level regression of trait size against body size (i.e. static allometry) is typically invariant among closely related populations and species. Such invariance is commonly interpreted to reflect a combination of developmental and selective constraints that delimit a phenotypic space into which evolution could proceed most easily. Thus, understanding how allometric relationships do eventually evolve is important to understanding phenotypic diversification. The famed ancient herb, known to the Romans as silphium (Greeksilphion), is widely regarded as the first recorded instance of human-induced species extinction. Modern scholars have largely credited direct exploitation (e.g., over-harvesting; over-grazing) as the primary cause of silphium's extinction, due to an overwhelming demand for the plant in ancient times. Recent research has revealed strict cold-stratification requirements for the germination of silphium's closest living relatives, revealing the likelihood that silphium shared these same germination requirements. Winters in northeastern North America have warmed faster than summers, with impacts on ecosystems and society. Global climate models (GCMs) indicate that winters will continue to warm and lose snow in the future, but uncertainty remains regarding the magnitude of warming. Here, we project future trends in winter indicators under lower and higher climate-warming scenarios based on emission levels across northeastern North America at a fine spatial scale (1/16°) relevant to climate-related decision making. Under both climate scenarios, winters continue to warm with coincident increases in days above freezing, decreases in days with snow cover, and fewer nights below freezing. …

Multiple Myeloma (MM) is a deadly blood malignancy, characterized by the uncontrolled proliferation of aberrantly differentiated plasma cells. MM is challenging to diagnose and treat, accounting for approximately 12% of hematologic malignancies. The overexpression of anti-apoptotic group of Bcl-2 family proteins, particularly Myeloid cell leukemia 1 (Mcl-1), play a critical role in the pathogenesis of MM. The overexpression of Mcl-1 is associated with drug resistance and overall poor prognosis. Thus, inhibition of the Mcl-1 protein is an attractive therapeutic strategy against myeloma cells. Strains of Dirofilaria immitis suspected of lack of efficacy (LOE) to macrocyclic lactone (ML) preventive drugs have been increasingly reported in dogs by practicing veterinarians since 2005 in the Lower Mississippi Delta region. If proven, and not controlled in the early stages, the emergence of ML drug resistance threatens to become a widespread problem in the US that may limit the effectiveness of current preventive drug treatment methods. Thousand cankers disease (TCD) is hypothesized to have a greater impact on eastern black walnut (Juglans nigra) in urban forests and plantations compared with natural forest stands. Along with other factors such as resource availability, the phytobiome could partly account for observed differences in disease severity across management regimes. We investigated the extent to which J. nigra-associated soil microbiomes from plantations and natural forests modulate (i) the amount of necrosis caused by Geosmithia morbida in 1-year-old seedlings, and (ii) relative abundance of rhizosphere endophytes and opportunistic pathogens Fusarium and Rhizoctonia in response to aboveground inoculation with G. morbida. We report a radially and azimuthally polarized (raPol) microscope for high detection and estimation performance in single-molecule orientation-localization microscopy (SMOLM). With 5000 photons detected from Nile red (NR) transiently bound within supported lipid bilayers (SLBs), raPol SMOLM achieves 2.9 nm localization precision, 1.5° orientation precision, and 0.17 sr precision in estimating rotational wobble. Within DPPC SLBs, SMOLM imaging reveals the existence of randomly oriented binding pockets that prevent NR from freely exploring all orientations. Treating the SLBs with cholesterol-loaded methyl-β-cyclodextrin (MβCD-chol) causes NR’s orientational diffusion to be dramatically reduced, but curiously NR’s median lateral displacements drastically increase from 20.8 to …

Nucleoside modified mRNA combined with Acuitas Therapeutics' lipid nanoparticles (LNPs) has been shown to support robust humoral immune responses in many preclinical animal vaccine studies and later in humans with the SARS-CoV-2 vaccination. We recently showed that this platform is highly inflammatory due to the LNPs' ionizable lipid component. The inflammatory property is key to support the development of potent humoral immune responses. A majority of the largest, single-site outbreaks of COVID-19 infections in the United States have been in prisons and jails since the beginning of the pandemic. These outbreaks threaten the lives and well-being of incarcerated people, correctional staff, and people who live in the communities to which incarcerated people return. This study employs both linear and logistic multivariate regression models to examine data from the UCLA’s COVID Prison Data Project, IPUMS CPS, the National Center for Health Statistics, and the Prison Policy Initiative to better understand the facility, county, and state-level predictors of COVID-19 infections and deaths in correctional facilities. …

The Rubber Hand Illusion (RHI) creates distortions of body ownership through multimodal integration of somatosensory and visual inputs. This illusion largely rests on bottom-up (automatic multisensory and perceptual integration) mechanisms. However, the relative contribution from top-down factors, such as controlled processes involving attentional regulation, remains unclear. Following previous work that highlights the putative influence of higher-order cognition in the RHI, we aimed to further examine how modulations of working memory load and task instructions—two conditions engaging top-down cognitive processes—influence the experience of the RHI, as indexed by a number of psychometric dimensions. Tumor growth is associated with metabolic reprogramming of various organs including the liver. This metabolic reprogramming is responsible for the development of behavioral fatigue represented by decreased voluntary wheel running in a murine model of lung cancer. To determine whether interleukin (IL-)6 induced by the tumor is responsible for the metabolic reprogramming, mice injected with Lewis lung carcinoma cells in the flank were treated with an anti-mouse IL-6 monoclonal neutralizing antibody using a 2 × 2 factorial design (+/- tumor and +/- anti-IL-6 antibody). Toxoplasma gondii commonly infects humans and while most infections are controlled by the immune response, currently approved drugs are not capable of clearing chronic infection in humans. Hence, approximately one third of the world's human population is at risk of reactivation, potentially leading to severe sequelae. To identify new candidates for treating chronic infection, we investigated a series of compounds derived from diversity-oriented synthesis. Bicyclic azetidines are potent low nanomolar inhibitors of phenylalanine tRNA synthetase (PheRS) in T. gondii, with excellent selectivity. BACKGROUND: One of the challenges in human neuroscience is to uncover associations between brain organization and psychopathology in order to better understand the biological underpinnings of mental disorders. Here, we aimed to characterize the neural correlates of psychopathology dimensions obtained using two conceptually different data-driven approaches.

Learning engages a high-dimensional neuronal population space spanning multiple brain regions. However, it remains unknown whether it is possible to identify a low-dimensional signature associated with operant conditioning, a ubiquitous form of learning in which animals learn from the consequences of behavior. Using single-neuron resolution voltage imaging, here we identify two low-dimensional motor modules in the neuronal population underlying Aplysia feeding. Our findings point to a temporal shift in module recruitment as the primary signature of operant learning. The respiratory syncytial virus (RSV) fusion (F) protein undergoes two furin-cleavage events to become fusion competent, resulting in the release of a twenty-seven amino acid peptide (p27). Recent studies indicate that the p27 region of the F protein was an immunodominant antigen in young children. In this study, we evaluated the kinetics of the serum antibody response to the p27 peptide following natural RSV reinfection in adults. Abstract Marine fungi exist as three major cell types: unicellular yeasts, filamentous hyphae and zoosporic early-diverging forms, such as the Chytridiomycota (chytrids). To begin to understand the ecological and biogeochemical influence of these cell types within the wider context of other plankton groups, cell size and macromolecular composition must be assessed across all three cell types. Using a mass–balance approach to culture, we describe quantitative differences in substrate uptake and subsequent macromolecular distribution in three model marine fungi: the yeast Metschnikowia zobellii, the filamentous Epicoccum nigrum and chytrid Rhizophydium littoreum. Importance For Black US residents, experiences of racial discrimination are still pervasive and frequent. Recent empirical work has amplified the lived experiences and narratives of Black people and further documented the detrimental effects of racial discrimination on both mental and physical health; however, there is still a need for further research to uncover the mechanisms connecting experiences of racial discrimination with adverse health outcomes.

Objective To examine neurobiological mechanisms that may offer novel insight into the association of racial discrimination with adverse health outcomes.

Extension materials that are sensitive to changing demographics and culture increase relevance and compliance with food safety practices. Produce safety extension materials were developed for U.S. Virgin Islands (USVI) produce growers to help with compliance with a new food safety rule. We developed employee training materials based on a needs assessment and behavioral change was evaluated six months after dissemination. The original materials were not seen as culturally appropriate but after modifications, improvements in food safety practices and behavior changes were observed. These results suggest that extension educators should seek feedback from target populations about potential interventions before implementation.

Feed and forages take the bulk of inputs required in livestock production. Inadequate forages in quality and quantity in sub-Saharan Africa is responsible for low livestock productivity. Addressing livestock feed requirements presents quick wins in livestock productivity compared to breeding and health. In a Climate Smart Dairy project implemented in Tanzania, we demontrated several forage options to farmers including, two cultivars of Pennisetum purpureum, two Urochloa hybrids, Chrloris gayana and Tripsacum andersonii (control). Where applicable, the grasses were intercropped with either one of the following forage legume, Desmodium intortum, Lablab purpureus or Stylosanthes guiyanensis. Fourteen forage treaments …

Production of sheep, goat, and cattle are major agricultural enterprises on West Texas rangelands, especially in the Edwards Plateau. In this region, the use of fire as a management tool was suppressed until recently. Although previous studies have been conducted to evaluate cattle use of burned versus unburned patches, there has been a lack of studies where combinations of livestock species grazing together on patch burned areas have been evaluated. This experiment, conducted at the Federal University of Agriculture, Abeokuta, Nigeria evaluated the morphogenesis of three varieties of Pennisetum purpureum used in this study, which were the local, purple, and FI variety at different stages of growth. The plants were established through vegetative plant parts (stems) sourced from a previously established plot and planted using 1 m × 1 m spacing on plots measuring 4 m × 5 m. An experiment was conducted in Jos, Nigeria to evaluate the effects of stages of growth on dry matter yield and nutrient composition of Red clover (Trifolium pratense var. AberClaret) in the year of establishment. The treatments were five (5) stages of growth (5, 9, 13, 17 and 21) weeks after sowing (WAS). The five stages of growth were replicated five times in a Latin Square Design. The land was divided into twenty five plots of 3 m X 3 m each.
