This qualitative study employed a hybrid hermeneutic phenomenological design to explore how technical high school teachers experienced and interpreted literacy instruction for Black and Brown neurodivergent students. Grounded in Critical Race Theory (CRT) and Disability Critical Race Theory (DisCrit), the research examined how educators’ perceptions, instructional practices, and collaborative relationships shaped access, equity, and empowerment in career and technical education contexts. Data was collected from seven academic and trade teachers through semi-structured interviews, focus groups, lesson artifacts, and reflective journals. Human macrophage isolation was successful with a 10% yield. Isolations are being further refined using an autoMACS cell separator, providing students instructional training on industry-level equipment that is commonly used. Through the primary human myoblast and murine macrophage co-culture, a significant shift in phenotype was demonstrated. Macrophage morphology varies with polarization and was used to evaluate the impact of factors and myoblasts at polarizing macrophages. Macrophage morphology was measured using an Image J analysis that quantified the axial ratio of macrophages. Axial ratio was calculated using the major axis divided by minor axis. Aim: The rising prevalence of opioid use during pregnancy poses serious public health concerns. The placenta is a critical organ during gestation, and opioid exposure can disrupt its function and fetal development. However, the molecular mechanisms through which opioids such as oxycodone affect feto-placental development remain poorly understood. This study aimed to investigate the effects of chronic in-utero oxycodone exposure on the composition and signaling functions of placenta-derived small extracellular vesicles (PSEVs) using a rat model.

This dissertation explores how Shakespeare’s plays and their film adaptations reflect and respond to the conditions of the Anthropocene, arguing that both forms of art can foster an environmental ethic attuned to the interconnectedness of human and nonhuman worlds. It proposes that the Anthropocene did not begin with industrialization but rather intensified a much older pattern of human ecological intervention, a process that becomes increasingly visible from the early modern period to the twentieth century. This phenomenological qualitative study explored how youth extracurricular involvement shapes the home environments of Millennial African American families. Guided primarily by Ecological Systems Theory, with Critical Race Theory as a complementary lens, the study examined how parents navigated interconnected systems of home, school, peers, and community while responding to racialized barriers embedded in youth programs. Data were collected through semi-structured interviews, observations, and document analysis with six African American mothers. Seven themes emerged: extracurricular activities, family dynamics, parental involvement, academic achievement, social development, cultural identity, and time management. My dissertation includes two projects, centered on transcription regulation by checkpoint kinases of the DNA damage response (DDR). The first project focused on the role of checkpoint kinase Mec1p in transcription termination and regulation of the torpedo exonuclease Rat1p. Termination, closely linked with pre-mRNA 3’ processing, dissociates RNA polymerase II (RNAPII) from DNA and releases the nascent RNA transcript. Efficient termination is required for maintaining a pool of RNAPII that is available for re-entry into new transcription cycle. Previous results showed that inactivation of Mec1p in the absence of exogenous genotoxic stress downregulates the efficiency of transcription termination. This study …

Staphylococcus aureus is a leading cause of prosthetic joint infection (PJI) typified by biofilm formation. Anti-inflammatory granulocytic myeloid-derived suppressor cells (G-MDSCs) represent the main leukocyte population in a mouse model of S. aureus PJI, followed by neutrophils (PMNs), and macrophages (Mφs), which is also seen during human PJI. Defining how each leukocyte population responds to S. aureus biofilm vs planktonic bacteria could have important implications for how S. aureus evades immune detection to facilitate biofilm persistence. Sudangrass can provide high-quality summer forage, supporting cow/calf pairs and growing cattle. In this study, sudangrass or a sudangrass–sunnhemp mix was grazed over two years by cow/calf pairs and heavy stockers (~880 lb). There was no difference in carrying capacity between forage types. Sunnhemp inclusion had no effect on cow body condition or nursing calf gain. Cows maintained condition through peak lactation, and nursing calves gained 2.27 lb/day. Stockers gained 1.56 lb/day on sudangrass alone and 1.73 lb/day on the mix. However, the added seed cost of sunnhemp increased the cost of gain in year 1 and …

Brominated and chlorinated benzenes are common industrial chemicals linked to kidney injury. This study evaluated the nephrotoxicity of several mono- and dihalogenated derivatives using isolated kidney cells from male Fischer 344 rats. LDH release and trypan blue exclusion were measured after 30- and 60-minute exposures to 0.25–1.0 mM to test compounds. Toxicity increased with increased concentration and longer times. Mixed-halogen bromobenzenes, particularly 1-bromo-4-chlorobenzene, showed one of the highest toxicity levels, while 1,4dichlorobenzene was the most toxic chlorobenzene. Antioxidants (glutathione, ascorbate, and αtocopherol) markedly reduced injury, indicating oxidative mechanisms play a role in toxicity. cytochromes P450 (CYP) inhibitors (piperonyl butoxide and …

Dichlorodiphenyltrichloroethane, commonly known as DDT, was an organochlorine pesticide widely used in the mid-20th century. It is highly stable and resistant to degradation, leading to its long-term presence in the environment. This chemical bioaccumulates in ecosystems and in human tissue, particularly adipose tissue, raising concerns about chronic exposure. Emerging research links DDT exposure to adverse neurological outcomes, including increased risk for neurodegenerative disorders such as Parkinson’s Disease. Mechanistically, DDT and its metabolites may disrupt dopaminergic neurons, induce oxidative stress, and interfere with mitochondrial function, contributing to neurotoxicity. Amyloids, insoluble protein aggregates of cross ß-sheet structure, are commonly associated with amyloidosis diseases. However, more recent studies support that some amyloids serve essential functions evolutionarily conserved across several taxa, particularly in reproduction. The nematode Caenorhabditis elegans serves as an excellent model organism in reproductive research. Prior to this study, no research has investigated the presence of functional amyloid in the C. elegans reproductive system. Stroke, a major cause of long-term disability and cognitive problems in the U.S., significantly affects people’s quality of life. Exposure to particulate matter (PM2.5) may result in systemic inflammation and oxidative stress, which can worsen the cognitive function of post-thrombectomy stroke patients. This project thus assessed the relationship between PM2.5 exposure and its effect on the cognitive function of post-thrombectomy stroke patients. Fifteen stroke patients (9 non-Appalachian and 6 Appalachian) undergone thrombectomy were recruited. Personal PM2.5 exposure was measured using wearable sensors for a week during the study period. A small to midsized university supported science, technology, engineering, and mathematics (STEM) education by partnering with a reputable online learning company to provide students and faculty access to artificial intelligence powered textbook problem sets and chatbots. An in-house formative assessment study was conducted in Fall 2024 and Spring 2025 to understand how traditional undergraduate students engaged with the platform. Quantitative results suggests that the general population did not utilize it as much as initially expected. The Black church has historically served as a cornerstone for African American spiritual and communal life yet faces significant challenges in addressing mental health needs rooted in generational trauma and systemic oppression. This dissertation explores the crisis of silence surrounding mental health in Black church contexts, where stigma, pastoral limitations, and historical mistrust of institutional systems create barriers to healing. Coastal soil salinization from rising seawater levels has adverse impacts on soil function, seed germination, and plant growth. Root exudates play a key role in supporting microbial activity, nutrient cycling, and plant health, yet little is known about the combined effects of salinization and the addition of artificial root exudates (AREs) on soils. We investigated the impacts of sodium chloride (NaCl) and AREs, alone and in combination, on soil microbial function and the growth and survival of Chamaecyparis thyoides saplings. Congenital birth deformities often arise during epithelial morphogenesis, displaying the need to understand the mechanisms underlying this process. Cell shape changes that mediate morphogenesis often require cell surface expansion to accommodate shifts in 3D tissue geometry. Our lab has identified Four Wheel Drive (Fwd), a Golgi-localized phosphatidylinositol 4-kinase (PI4K) IIIβ ortholog, as a regulator for cell surface expansion during Drosophila ventral furrow formation, a well-characterized model for epithelial folding. The development of high-performance, rare-earth-free hard magnetic materials is critical for next-generation energy, information, and spin-based technologies. This dissertation investigates structure-property relationships in iron-based magnetic systems, including iron carbides, Fe-Co-B boride alloys, hexaferrites, and FeCo nanostructures, synthesized via chemical and metallurgical routes. Through controlled synthesis and comprehensive structural and magnetic characterization, this work demonstrates that magnetic anisotropy, coercivity, and thermal stability can be systematically controlled through morphology design, compositional tuning, microstructural refinement, and interparticle interaction modulation.
