Adult mammalian hearts exhibit limited regenerative capacity because of the restricted renewal of cardiomyocytes. Recent studies reveal that mammalian hearts exhibit transient regenerative potential within a short time frame after birth, suggesting a regulatory mechanism that prevents adult hearts from initiating a regenerative response to cardiac injury. Here, we discovered that an active form of YAP, named YAP6SA, which is not inhibited by the Hippo signaling pathway and does not interact with TEADs, induces cardiomyocyte cell cycle reentry. In addition, YAP6SA interacts with scaffold protein MPDZ to regulate Rho GTPases and promote cell cycle progression in cardiomyocytes (CMs). Importantly, YAP6SA …

[Objective] To establish a rapid and precise determination method for 9 herbicides (asulam, hexazinone, indaziflam, norflurazon, pyrasulfotole, pyroxasulfone, sulfosulfuron, tebuthiuron, and tepraloxydim) in dairy products that are of international concern but without established maximum residue limits (MRLs) in China. [Methods] The samples are ultrasonically extracted with acetonitrile-acetone mixed reagent (v∶v=8∶2), and the extract is purified by the dispersed solid-phase extraction adsorbents Florisil, N-propylethylenediamine (PSA), and anhydrous MgSO4. A high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method is established for determination. [Results] The nine herbicides are completely separated within 7 min. [Objective] To propose a deep learning-based method for the freshness determination of winter jujube by dividing the fruit into five freshness stages, aiming to improve determination accuracy and reduce the influence of light reflection. [Methods] In this study, a freshness determination method is proposed for winter jujube by combining an efficient ResNet, an attention mechanism, and Faster R-CNN. First, ResNet is used for convolutional processing on the image to extract the global feature map. [Objective] To explore the key non-ribosomal peptide synthetase (NRPS) genes of the active peptide Asp-Leu-Ala-Pro (DLAP) with angiotensin-converting enzyme (ACE) inhibitory activity identified from the mycelia of Ganoderma sinense. [Methods] The effects of key NRPS genes on the synthesis of DLAP are analyzed by PEG-mediated genetic transformation experiment, real-time fluorescence quantitative PCR (qRT-PCR), and high performance liquid chromatography (HPLC). [Results] qRT-PCR results reveal that compared with the wild type, the silenced transformants of GSI_00900 have the reductions of 44.99%, 68.88%, and 66.17% in the expression level of this gene. [Methods] Lueyang black-bone chicken meat was used as the raw material. Two proteases were employed for compound enzymatic hydrolysis, and response surface methodology was applied to optimize the preparation process of Lueyang black-bone chicken oligopeptides and to evaluate their antioxidant activity. [Objective] To investigate the composition and antioxidant properties of flavonoids from the Ampelopsis grossedentata leaf extract. [Methods] Qualitative identification and quantitative determination are conducted on flavonoids from the extract by high-performance liquid chromatography (HPLC). Then, with tea polyphenols and vitamin C as controls, the scavenging rates of the extract are determined against four free radicals, including OH-, DPPH, O-2, and ABTS+, and the antioxidant effect of the extract is studied during potato chip storage. [Objective] This experiment aims to modify extruded whole corn flour through lactic acid bacteria fermentation to investigate its impact on the processing properties of dough. [Methods] Single-factor experiments are conducted using fermentation temperature, fermentation time, and inoculation amount of lactic acid bacteria as key variables. [Methods] The extraction process of mango kernel crude polyphenols is optimized by the orthogonal experiment. Mango kernel polyphenol extract is further separated and purified by the macroporous adsorption resin. Meanwhile, the antioxidant activities of mango kernel polyphenols before and after purification are analyzed with Vc as the control. [Objective] To prevent postharvest fruit decay and extend shelf life, a robust, water-resistant, and self-healing multifunctional edible fresh-keeping film was proposed. [Methods] The edible fresh-keeping film's microstructure, chemical structure, physicochemical properties, mechanical properties, and its effects on the shelf life of bananas were characterized using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), X-ray powder diffractometer (XRD), texture analyzer, and rheometer. [Results] The monomers within the edible film are cross-linked via hydrogen bonds, ester bonds, and electrostatic interactions, forming an irregular network structure on the surface. We investigate the bootstrap methodology and current applications. The bootstrap is a non-parametric resampling method that has universal applicability thanks to the statistical concepts of weak convergence, the law of large numbers, and the central limit theorem. Modern applications of bootstrap methodology have demonstrated time and time again that it is computationally efficient and robust. The earliest days of the bootstrap saw it validated by generating statistics, confidence intervals, and hypothesis tests. This qualitative, narrative research study explored the experiences lived by black female educators with chronic health conditions during and after the Covid-19 pandemic in the Southeastern region of the United States. These educators provided continued instruction in a central Jackson, Mississippi school district and a Georgia school district in the central Atlanta metropolitan area. The investigation during this body of work focused on how black female educators with chronic health conditions were impacted both personally and professionally. The Field-to-Market Fieldprint Calculator is a software tool designed to estimate and compare relative sustainability of differing agricultural practices. This observational research compared conservation techniques to conventional practices in rice and cotton on three separate farms in Northeast Arkansas. Considering management and site conditions, the Fieldprint Calculator was used to compare sustainability metrics to standards at various spatial scales in a spidergraph report. Eight metrics were analyzed to produce reports, including greenhouse gas emissions, water quality and quantity, land and energy use, soil carbon and conservation, and biodiversity. Farmers in Arkansas irrigate more farmland than 47 other states in the U.S. Agricultural production is a vital contributor to the economy of the state; however, it also contributes heavily to water quantity and quality issues within the Mississippi River Basin and the Gulf of Mexico. Row-crop producers have adopted soil and water conservation practices to mitigate these effects, yet many farmers remain hesitant to adopt conservation methods due to uncertainties about their effects on yield and profits. This study evaluates the effectiveness of large language models (LLMs), specifically Claude Sonnet 4.0 and ChatGPT 4.1, for analyzing formative feedback to support student-centered learning (SCL). In large courses, instructors often struggle to promptly review and synthesize student input. We used a low-stakes task: analyzing 63 anonymous student responses to a Muddiest Point prompt in a human physiology class after a lecture on respiratory physiology. Across 20 runs, both LLMs consistently identified "Ventilation and Lung Mechanics" as the most frequent area of confusion, aligning with human analysis. Youth in foster care face disproportionate challenges when it comes to education and personal well-being. Many studies use a deficit lens when evaluating these challenges. In contrast, this study is asset-based, centering first-person accounts from five former foster youth college graduates. Using a counter-narrative framework, we conducted semi-structured interviews with participants to reveal key supports in K-12 schooling participants believed helped them achieve academic and life milestones. These were relationships with teachers and coaches, access to extracurricular activities, and literacy-rich environments. We also reviewed 11 relevant federal educational policies to identify provisions and implementation needs that aligned with participant experiences. …

Accurately measuring street dimensions is essential to evaluating how their design influences both travel behavior and safety. However, gathering street-level information at city-scale with precision is difficult given the quantity and complexity of urban intersections. To address this challenge in the context of pedestrian crossings — a crucial component of walkability — we introduce a scalable and accurate method for automatically measuring crossing distance at both marked and unmarked crosswalks, applied to America’s 100 largest cities. Theoretical accounts typically assume that key features of human socio-cognitive development are universal. This paper reports a large-scale cross-cultural study (17 communities, diverse ethnicities, N = 1,377, 709 female, mean = 5.50 years, -collected March 2022 to January 2024) on gaze following in early childhood. To test for universality, cognitive processing signatures were derived from a computational model treating gaze following as social vector estimation. Results showed substantial variation between communities and individuals. Yet, the processing signature was found in all communities. This study examined the influence of training in SPEAK OUT!® on the perceptual ratings of voice and speech made by communication disorders (CD) graduate students. The purpose of this study was to investigate the influence of SPEAK OUT!® training by comparing those who have undergone SPEAK OUT!® training with those who have not. To address this, 36 participants who attend communication disorders graduate programs within the United States were recruited. Data were collected through a Qualtrics survey distributed via email and analyzed using the Statistical Package for the Social Sciences (SPSS). For the past twenty years, Black women have increasingly ascended to the role of school district superintendent. Black female superintendents navigate education systems that were not built with them in mind creating unique challenges. The purpose of this qualitative phenomenological study was to investigate the challenges that Black female superintendents face while navigating local school boards. This study was informed by glass ceiling theory, gendered organizations theory, and a black feminist framework establishing a research base for this study’s focus. Genetic diseases such as ion channelopathies substantially burden human health. Existing treatments are limited and not genotype specific. Here, we report a 2-step high-throughput approach to rapidly identify drug candidates for repurposing as genotype-specific therapy. We first screened 1,680 medicines using a thallium-flux trafficking assay against Kv11.1 gene variants causing long QT syndrome (LQTS), an ion channelopathy associated with fatal cardiac arrhythmia. We identified evacetrapib as a suitable drug candidate that improves membrane trafficking and activates channels. We then used deep mutational scanning to prospectively identify all Kv11.1 missense variants in an LQTS hotspot region responsive to treatment with evacetrapib. …

[Objective] To optimize the extraction process of black ginseng polysaccharides (BGP) by an enzyme extraction method, and to investigate the antioxidant activity of BGP. [Methods] The optimal dosage of compound enzymes for BGP extraction is obtained using the orthogonal method, and the process conditions for BGP extraction by compound enzymes are optimized using the response surface method. The in vitro antioxidant activity of BGP is evaluated using the DPPH and the ABTS+ radical methods. [Results] The optimal enzyme ratio is 120 mg cellulase, 120 mg pectinase, and 80 mg papain.
