Cyberbiosecurity and workforce development in agriculture and the life sciences (ALS) is a growing area of need in the curriculum in higher education. Students that pursue majors related to ALS often do not include training in cyber-related concepts or expose the ‘hidden curriculum’ of seeking internships and jobs. Exposing students through workforce development training and hands-on engagement with industry professionals can provide learning opportunities to bridge the two and is an area of growth and demand as the workforce evolves. The objectives of this work were 1) to learn key concepts in cybersecurity, including data security, visualization, and analysis, to …

The field of Alzheimer’s disease (AD) has witnessed recent breakthroughs in the development of disease-modifying biologics and diagnostic markers. While immunotherapeutic interventions have provided much-awaited solutions, nucleic acid-based tools represent other avenues of intervention; however, these approaches are costly and invasive, and they have serious side effects. Previously, we have shown in AD animal models that tolfenamic acid (TA) can lower the expression of AD-related genes and their products and subsequently reduce pathological burden and improve cognition. Using TA as a scaffold and the zinc finger domain of SP1 as a pharmacophore, we developed safer and more potent brain-penetrating analogs …

We utilize a coupled economy–agroecology–hydrology modeling framework to capture the cascading impacts of climate change mitigation policy on agriculture and the resulting water quality cobenefits. We analyze a policy that assigns a range of United States government’s social cost of carbon estimates ($51, $76, and $152/ton of CO2-equivalents) to fossil fuel–based CO2 emissions. This policy raises energy costs and, importantly for agriculture, boosts the price of nitrogen fertilizer production. At the highest carbon price, US carbon emissions are reduced by about 50%, and nitrogen fertilizer prices rise by about 90%, leading to an approximate 15% reduction in fertilizer applications for …

Chronic hypoxia may have a huge impact on the cardiovascular and renal systems. Advancements in microscopy, metabolomics, and bioinformatics provide opportunities to identify new biomarkers. In this study, we aimed at elucidating the metabolic alterations in kidney tissues induced by chronic hypoxia using untargeted metabolomic analyses. Reverse phase ultrahigh performance liquid chromatography-mass spectroscopy/mass spectroscopy (RP–UPLC–MS/MS) and hydrophilic interaction liquid chromatography (HILIC)–UPLC–MS/MS methods with positive and negative ion mode electrospray ionization were used for metabolic profiling. The metabolomic profiling revealed an increase in metabolites related to carnitine synthesis and purine metabolism. Additionally, there was a notable increase in bilirubin. Heme, N-acetyl-L-aspartic …

The Importance Of Caregivers, Family, And Context For Child Welfare Involved Youth And A National Child Sample: A Structural Equation Modeling Perspective, Nathan Michael Lutz
Dissertations
This bundled, three-study dissertation examines how the relationships that children have with adult family members impact mental health outcomes, including internalizing symptoms and externalizing behaviors. The first study examines the moderating effect of relationship satisfaction on the relationship between kin involvement and child psychopathology. The second study uses data from the 2017 National Survey of Children's Health (NSCH), examining the mediating effect of the parent-child relationship on the associations across various systems in which children are embedded and mental health diagnoses. The third paper examined the interactions between both family strengths and caregiver involvement with the parent child relationship and …

Introduction: Though the nature of breastfeeding is critical, scant information is available on how the action of the milk transfer from mother to infant is regulated in humans, where the points of dysfunction are, and what can be done to optimize breastfeeding outcomes. While better therapeutic strategies are needed, before they can be devised, a basic scientific understanding of the biomechanical mechanisms that regulate human milk transfer from breast to stomach must first be identified, defined, and understood. Methods: Combining systems biology and systems medicine into a conceptual framework, using engineering design principles, this work investigates the use of biosensors …

Fear Reduced Through Unconditional Stimulus Deflation Is Behaviorally Distinct From Extinction And Differentially Engages The Amygdala, Gabrielle R. Bonanno, Erisa Met Hoxha, Payton K. Robinson, Nicole C. Ferrara, Sydney Trask
Purdue University Libraries Open Access Publishing Fund
Background
Context fear memory can be reliably reduced by subsequent pairings of that context with a weaker shock. This procedure shares similarities with extinction learning: both involve extended time in the conditioning chamber following training and reduce context-elicited fear. Unlike extinction, this weak-shock exposure has been hypothesized to engage reconsolidation-like processes that weaken the original memory.
Methods
We directly compared the weak-shock procedure with extinction using male and female Long Evans rats.
Results
Both repeated weak-shock exposure and extinction resulted in decreased context freezing relative to animals that received context fear conditioning but no subsequent context exposure. Conditioning with …
Examining The Bidirectional Associations Between Parenting And Adolescent Coping In The Context Of Exposure To Community Violence, Jenny Phan
Dissertations
Coping refers to all the strategies and processes that an individual uses to eliminate, reduce or tolerate stress (Lazarus & Folkman, 1984; Rosario et al., 2003). Within the general coping literature, research has observed that use of approach-oriented coping strategies is associated with positive psychosocial outcomes, and avoidant-oriented coping strategies is associated with negative psychosocial outcomes (e.g., Compas et al., 2001; Wadsworth & Compas, 2002). However, most of these studies have focused exclusively or predominantly on middle-class White participants (e.g., Compas et al., 2001; Wadsworth & Compas, 2002) which may limit the generalizability of these findings to other groups. Further, …
Interactions Of Lewis Acids And Carbonyls In The Presence Of Ligands Or Additives: How Solution Behavior Differs And The Implication On Catalytic Systems, Sophi Rose Todtz
Interactions Of Lewis Acids And Carbonyls In The Presence Of Ligands Or Additives: How Solution Behavior Differs And The Implication On Catalytic Systems, Sophi Rose Todtz
Dissertations
Lewis acids have proven extremely valuable to the field of organic synthesis. Their diverse application in reactions of carbonyl-containing substrates has caused a multitude of efforts towards characterization of solution interactions between Lewis bases and Lewis acids. Though Lewis initially described a 1:1 interaction between one acid and one base, our lab has previously employed in-situ IR spectroscopy to characterize 2:1, 3:1, and 4:1 solution structures under catalytic conditions. These insights have played a critical role in our understanding of catalyst behavior in carbonyl-containing systems, allowing us to describe byproduct inhibition in the carbonyl-olefin metathesis mechanism. The research presented in …
A Descriptive Exploration Of Quality And Opportunities In Outdoor Urban Play Spaces: The Case Of Chicago Public Schools, Katherine Anne Varey
A Descriptive Exploration Of Quality And Opportunities In Outdoor Urban Play Spaces: The Case Of Chicago Public Schools, Katherine Anne Varey
Dissertations
This study investigated the quality of children’s play spaces, both green and built, in and surrounding urban public schools in Chicago. Given children’s declining access to nature and the outdoors, especially in urban areas, the aim of the study was to both assess the current potential for high-quality outdoor experiences, as well as to uncover additional, low-cost, and low-burden opportunities that may exist for enhancing outdoor experiences. Based on a selected sample of 60 Chicago Public Schools, this mixed methods study employed quantitative ratings of play space and neighborhood quality, geographic information systems (GIS) mapping techniques to show distribution of …

Environments Of War: The Pacific Northwest And The Waging Of World War Ii, Katherine L. Macica
Dissertations
The Pacific Northwest served an essential role in America’s “Arsenal of Democracy.” Weapons and material produced in the Northwest proved essential to the Allied victory in World War II. These weapons served as agents of destruction abroad, but became agents of transformation for the places in which they were produced. This dissertation examines four of the most important resources and industries in Oregon and Washington to emerge or expand as a result of the war: hydroelectric power, forest products, shipbuilding, and aircraft manufacture. The wartime development of these industries profoundly reshaped the physical and economic landscape of the Pacific Northwest, …

Parenting And Psychosocial Functioning In Youth With Spina Bifida: Making Connections With Other Cns-Related Chronic Health Conditions, Adrien M. Winning
Dissertations
Parenting practices have been linked to psychosocial outcomes in children who have chronic health conditions that affect the central nervous system (CNS), such as spina bifida (SB). However, there remains a need for (1) more evidence-based assessments that capture parenting in SB, (2) additional research focused on parenting in early childhood in SB, as this is a critical developmental period that has been understudied, and (3) systematic reviews that synthesize parenting research across CNS-related conditions to guide future work. This dissertation includes three papers that aim to address each of these gaps in the current literature. Specifically, the first study …

“But You Don't Look Sick…:” Shedding Light On The Socially Constructed Problems Of Invisible Chronic Illness And The Benefits Of Shifting To A Dynamic Illness Framework, Samantha Susan Ropski
Master's Theses
Invisible chronic illness is a common, complex phenomenon that applies to any illness, disease, or collection of symptoms that is not always or ever outwardly apparent or discernable to onlookers, even in biomedical contexts. Interestingly, despite 96% of those with chronic illness fitting under the “invisible” umbrella, patients are frequently misdiagnosed, invalidated, dismissed and marginalized because they do not “look sick” enough within the context of the disease-focused medical model. This thesis aims to argue that many of the most significant and damaging problems associated with chronic illness are not due to the actual illnesses themselves but rather, are socially …

The loss of contact inhibition is a key step during carcinogenesis. The Hippo-Yes-associated protein (Hippo/YAP) pathway is an important regulator of cell growth in a cell density-dependent manner. However, how Hippo signaling senses cell density in this context remains elusive. Here, we report that high cell density induced the phosphorylation of spectrin α chain, nonerythrocytic 1 (SPTAN1), a plasma membrane-stabilizing protein, to recruit NUMB endocytic adaptor protein isoforms 1 and 2 (NUMB1/2), which further sequestered microtubule affinity-regulating kinases (MARKs) in the plasma membrane and rendered them inaccessible for phosphorylation and inhibition of the Hippo kinases sterile 20-like kinases MST1 and …

The interphase genome is dynamically organized in the nucleus and decorated with chromatin-associated RNA (caRNA). It remains unclear whether the genome architecture modulates the spatial distribution of caRNA and vice versa. Here, we generate a resource of genome-wide RNA-DNA and DNA-DNA contact maps in human cells. These maps reveal the chromosomal domains demarcated by locally transcribed RNA, hereafter termed RNA-defined chromosomal domains. Further, the spreading of caRNA is constrained by the boundaries of topologically associating domains (TADs), demonstrating the role of the 3D genome structure in modulating the spatial distribution of RNA. Conversely, stopping transcription or acute depletion of RNA …
