The reproductive health of women is influenced by microorganisms and their metabolites, namely those representing the vaginal microbiota and those producing toxins that are ingested. To manipulate the vaginal microbiota toward a health-associated, Lactobacillusdominant state, an approach adopting prebiotic lactulose was taken. Using batch culture, lactulose supported Lactobacillus maintenance and positively altered metabolites, while not disrupting indigenous L. crispatus epithelial adherence. The vagina also harbours abundant glycogen, but initial assumptions that lactobacilli utilize it were incorrect. I have now shown that glycogen selectively stimulates organisms associated with dysbiosis and L. iners in a self-limiting manner. Other compounds such as environmental …

Developmental exposure to bisphenol A (BPA), a ubiquitous endocrine disrupting chemical, is associated with organ dysfunction and diseases in adulthood. However, little is known about its effects on the adrenal glands. Therefore, this thesis addresses this important question using both in vivo and in vitro approaches. BPA at environmentally relevant doses was administrated via diet to pregnant mice from embryonic day 7.5 to birth, following which mice were switched to a standard chow. At two months postnatally, adrenal glands and blood samples were collected from adult mouse offspring for structural and functional analysis. I found that: (a) BPA increased adrenal …

Metastasis is the primary cause of mortality in cancer patients. Inhibition of proteins that are involved in the regulation of metastasis are expected to suppress metastasis and represent treatment targets. Our focus is on prostate cancer metastasis and we have developed a novel high-throughput means of performing in vitro screens for regulators of prostate cancer metastasis. We propose to use a focused CRISPR library screen that will “knock out” all human kinases to determine which ones are responsible for prostate cancer metastasis. Transitions in Care are challenging for people with dementia due to impairments in communication and disorientation from being in a foreign environment. Transitions in care have lead to incidents of unnecessary physical and chemical restraints of this vulnerable population. To investigate how to optimize care transitions, qualitative descriptive information was collected through three personal interviews to gain a broader understanding of mechanisms of care transitions in London, ON. Objectives: The aim of this study was to investigate the effect of different orthodontic materials on the formation of the protein pellicle on the bracket surface.
Materials and Methods: We used X-ray Photoelectron Spectroscopy (XPS) to analyze the atomic composition of orthodontic metallic and ceramic brackets. Following XPS, the brackets were immersed in human whole saliva supernatant for 2-hour incubation at 37°C. Hydroxyapatite (HA) discs were used as control. Acquired pellicle was harvested with sonication for each group, and liquid chromatography electrospray ionisation mass spectrometry was used for protein identification.

BACKGROUND: Previous studies showed that synaptic transmission is affected by general anesthetics, but an anesthetic dose response in freely moving animals has not been done. The hippocampus provides a neural network for the evaluation of isoflurane and pentobarbital on multisynaptic transmission that is relevant to memory function.
METHODS: Male Long-Evans rats were implanted with multichannel and single electrodes in the hippocampus. Spontaneous local field potentials and evoked field potentials were recorded in freely behaving rats before (baseline) and after various doses of isoflurane (0.25 to 1.5%) and sodium pentobarbital (10 mg/kg intraperitoneal).

The RND superfamily of efflux pumps plays vital roles in the intrinsic defence mechanisms of bacterial pathogens. The staphylococci have two genes encoding RND transporters; farE, which promotes efflux of antimicrobial fatty acids, and an uncharacterized gene we have named femT. Although RND pumps are known to contribute to maintenance of cellular homeostasis and antimicrobial resistance, the function of FemT, and the relationship between FemT and FarE in staphylococci have not been identified. Hockey Fans in Training (Hockey FIT) is a gender-sensitized weight loss and healthy lifestyle program for overweight and obese male hockey fans, consisting of a 12-week active phase followed by a 40-week maintenance phase. Following program completion, interviews and focus groups were conducted to explore the social support given and received by the men involved in Hockey FIT. © 2017 American Society for Surgery of the Hand Purpose To investigate the effect of implant positioning on ulnohumeral contact using patient-specific distal humeral (DH) implants. Methods Seven reverse-engineered DH implants were manufactured based on computed tomography scans of their osseous geometry. Native ulnae were paired with corresponding native humeri and custom DH implants in a loading apparatus. The ulna was set at 90° of flexion and the humeral component (either native bone or reverse-engineered implant) was positioned from 5° varus to 5° valgus in 2.5° increments under a 100-N compressive load. Introduction: Individuals with head and neck cancer (HNCa) face myriad biopsychosocial challenges. Even after treatment completion, these challenges may continue to cause diminished quality of life (QoL). Resilience may serve to minimize the impact of HNCa and, thus, maximize QoL. The purpose of this study was to identify resilience in HNCa survivors and explore its potential relationship with QoL.
Methods: Thirty-one HNCa survivors completed three validated, self-report questionnaires pertaining to the collection of resilience and QoL data. Descriptive, correlational, and observational analyses were performed.

With 50% of patients in North America not taking their medications as they should (Brown & Bussell, 2011), a better understanding of medication adherence among older patients could be helpful to health professionals and service providers. The purpose of this study was to examine whether the perceived pharmacist-patient quality of communication is associated with diabetes medication adherence. Eighty-four older adults from the Primary Care Diabetes Support Program were recruited. Diabetes medication adherence and pharmacist-patient quality of communication were measured using self-report questionnaires. High-dose-rate brachytherapy (HDR-BT) is an interstitial technique for the treatment of intermediate and high-risk localized prostate cancer that involves placement of a radiation source directly inside the prostate using needles. Dose-escalated whole-gland treatments have led to improvements in survival, and tumour-targeted treatments may offer future improvements in therapeutic ratio. The efficacy of tumour-targeted HDR-BT depends on imaging tools to enable accurate dose delivery to prostate sub-volumes. This thesis is focused on implementing ultrasound tools to improve HDR-BT needle localization accuracy and efficiency, and evaluating dynamic contrast enhanced magnetic resonance imaging (DCE-MRI) for tumour localization.

The applications of PET-MRI in radiotherapy are only starting to be realised. However, quantitative accuracy of PET relies on accurate attenuation correction (AC) of, not only the patient anatomy but also MRI hardware and current methods, which are prone to artefacts caused by dense materials. PURPOSE: To develop an evidence-based clinical practice guideline for the prevention of oral mucositis in children (0-18 years) receiving treatment for cancer or undergoing haematopoietic stem cell transplantation (HSCT).
METHODS: The Mucositis Prevention Guideline Development Group was interdisciplinary and included internationally recognised experts in paediatric mucositis. For the evidence review, we included randomised controlled trials (RCTs) conducted in either children or adults evaluating the following interventions selected according to prespecified criteria: cryotherapy, low level light therapy (LLLT) and keratinocyte growth factor (KGF). Integrin β1 is essential for pancreatic beta-cell development and maintenance throughout life in rodents and human fetal islets. However, the effects of a postnatal β1 integrin knockout (β1KO) specific to pancreatic beta-cells of mice is undetermined. We generated mice with CreERT recombinase specific to the mouse insulin promoter (MIP), allowing us to induce a β1KO upon injection of tamoxifen (MIPβ1KO model).
At 3-4 weeks of age tamoxifen was injected and mice were sacrificed at 8 (male) and 16 weeks (female) post-tamoxifen. MIPβ1KO mice had impaired glucose tolerance, reduced beta-cell mass and islet density. The impairment in glucose tolerance remained in …

OBJECTIVES: To evaluate the extent of association between systemic inflammation and periodontal disease in American adults, and to assess whether socio-economic position mediated this relationship. METHODS: We used data from the National Health and Nutrition Examination Survey (NHANES IV) (2001-2010). Systemic inflammation was defined by individual and aggregate (cumulative inflammatory load) biomarkers (C-reactive protein, white blood cell counts, neutrophil counts, and neutrophil: lymphocyte ratio). Loss of attachment and bleeding on probing were used to define periodontal disease. Poverty:income ratio and education were indicators of socio-economic position. Drawing on interview and focus group data, this article explores research undertaken as part of a larger research project exploring precarity in the nonprofit employment services sector in a mid-sized Canadian city. We critically survey major legislative changes to Canadian employment and income security policies and programs, including the restructuring of work and labor relations, growth of performance-based contracting-out, erosion of intergovernmental transfers, worker stress, and emotional tolls. Our study’s results demonstrate how employment precarity in the nonprofit employment services sector is amplified by top-down and centralized relationships with funding partners and policymaking divorced from the employment experiences of frontline …
