CD4+ T cells are crucial for protective immunity to intracellular pathogens. In addition to secreting cytokines, CD4+ T cells promote control of Mycobacterium tuberculosis infection through cognate interactions with macrophages, but the mechanism has been unclear. Here, we show that SLAMF1/CD150 is highly and uniquely induced in macrophages by antigen-specific interactions with CD4+ T cells. In macrophages, SLAMF1 enhances the generation of reactive oxygen species and restricts Mtb replication. Mtb-infection of mice promotes SLAMF1 expression specifically on infected macrophages, not uninfected bystanders. SLAMF1 expression depends on adaptive immunity and also autophagy. Moreover, Slamf1

CLN3 disease is a neuronopathic lysosomal storage disorder that severely impacts the central nervous system (CNS) while also inducing notable peripheral neuromuscular symptoms. Although considerable attention has been directed towards the neurodegenerative consequences within the CNS, the involvement of peripheral tissues, including skeletal muscles and their innervation, has been largely neglected. Background: Kilquist syndrome (KILQS) is a recently characterized, rare disorder characterized by hearing loss, secretory dysfunction, gastrointestinal system (GIS) abnormalities, and neurological anomalies including hypotonia and global developmental delay. It is caused by variants in the SLC12A2 (Solute Carrier Family 12 Member 2) gene which encodes the Na-K-2Cl cotransporter isoform 1 (NKCC1). Objective: This study aims to contribute to genotype-phenotype correlations for SLC12A2 variants. Non-growth hormone-deficient short stature is a group of growth developmental disorders with complex pathophysiological mechanisms involving multiple pathways, including abnormalities in the growth hormone/insulin-like growth factor 1 signal pathway. We report a 4-year-old girl with severe growth and developmental delay: her height was 91 cm (<–3 standard deviation [SD]), and her growth rate was 3 to 4 cm/year (<–2 SD). Growth hormone stimulation tests indicated a normal peak growth hormone level (11.1 μg/L), while insulin-like growth factor-1 levels were reduced (7.98 nmol/L, <–1SD). Whole exome sequencing and copy number variation analysis revealed a 3.19 Mb deletion at 2q35-q36.1 on chromosome 2, encompassing all coding regions of the Epha4 gene. This study suggests an association between Epha4 gene deletion and non-growth hormone-deficient short stature, highlighting the importance of further research on this gene and its related signaling pathways to understand the molecular mechanisms of the disease and potential therapeutic approaches.

FATCO syndrome (Fibular Aplasia, Tibial Camptomelia, Oligosyndactyly) is characterized by the presence of Fibula Aplasia, Tibial Campomelia and Oligosyndactyly. It is an autosomal dominant congenital genetic disorder, with an infrequent occurrence. Nephronophthisis is a group of autosomal recessive kidney diseases characterized by chronic tubulointerstitial abnormalities leading to kidney failure. The incidence of nephronophthisis varies globally, and its genetic heterogeneity presents significant difficulties in diagnosis. Herein, we present a molecular diagnosis of four unrelated Moroccan families fulfilling the clinical criteria of nephronophthisis. As first diagnosis step, screening for a homozygous NPHP1 gene deletion, the most common mutation, was performed by using multiplex polymerase chain reaction. The aim of this study was to describe sleep-disordered breathing (SDB) and treatment in children with achondroplasia (ACH). A retrospective longitudinal study was conducted at the Sleep Unit, Hospital de Pediatría Garrahan. Children with ACH, aged 0 to 18 years, referred for SDB due to clinical symptoms or foramen magnum stenosis (FMS) with at least one polysomnography (PSG) between 2002 and 2019 were included in the study. The primary outcomes included SDB typification and therapeutic interventions. We included 89 patients; 79.7% had one PSG, 13.4% had two PSGs, and 5.6% had three PSGs. This teacher action research study describes the development and application of the opportunities for growth teaching method to increase learner motivation, persistence, cooperation and positive self-talk among primary school students. The approach was implemented with 30 fourth graders in a Philadelphia public school classroom to foster unity, support attitudes toward academics and participation, redefine failure, and reframe challenges. Before this intervention, most of the students lacked self-regulation, self-awareness, and relationship skills needed to thrive. We aim to expand units coordination and construction assessment by examining the significance of embodiment and teacher noticing, especially in relation to recent developments in equitable mathematics teacher noticing research. The Agency Protocol defines a decentralized trust infrastructure in which autonomous agents cooperate by issuing cryptographically‑signed, domain‑scoped promises and verifiable assessments. Each promise carries a dual‑component stake—an at‑risk credit deposit and a non‑refundable operational‑cost fee—that is locked in content‑addressed storage and returned or slashed according to assessment consensus. Assessments are merit‑weighted; an agent’s merit is a non‑transferable, domain‑specific reputation score updated by matrix‑factorized aggregation of historical outcomes, while credits form a fully transferable internal currency. Merit discounts future stake requirements, creating a positive feedback loop that makes honest behaviour utility‑maximising.

Late-onset sepsis (LOS; sepsis occurring after the first 72 hours of life) increases the morbidity and mortality of premature neonates, especially those with very-low-birth weight (≤1500g). In LOS, Enterobacter cloacae has been rarely described as a cause of secondary brain abscesses. Our case report summarizes important aspects of cerebral abscesses due to an E. cloacae sepsis in a preterm neonate. Our patient received surgical abscess drainage and rational antibiotic treatment employing prolonged meropenem infusions plus fosfomycin and subsequent treatment with oral trimethoprim/sulfamethoxazole. Furthermore, we present a review of the current literature from 2009-2022, comparing cases similar to our case.

This study aimed to retrospectively investigate the respiratory viral agents and epidemiological characteristics, in pediatric patients. Methods: The study included pediatric patients, aged 0-18 years. A retrospective analysis was conducted on 5,253 samples sent to the microbiology laboratory with suspected respiratory tract infection between January 1, 2020 and December 31, 2023. Methods We performed a retrospective study of children with M. pneumoniae pneumonia who were admitted to the Women and Children's Hospital of Ningbo University between January 2022 and December 2023. Objective: The study mainly aimed to explore the impact of Human adenovirus (HAdV) detection by two-child policy and coronavirus disease 2019 (COVID-19) epidemic in China. Methods: Hospitalized children with ARTI were enrolled in Maternal and Child Health Hospital of Hubei Province from January 2014 to June 2022, whose clinical data were collected retrospectively. The effects of two-child policy and COVID-19 pandemic on the detection of HAdV were analyzed. Results: Among 100215 hospitalized children with ARTI, the positive rate of HAdV was 1.93% with the peak in 2019. Objective: This study aims to explore the association between respiratory viruses and febrile seizures (FS), and to analyze the impact of these viruses on the type and management of FS. Methods: We conducted a retrospective, single-center study of 49,445 emergency cases from September 2021 to December 2023. Among these, 266 patients with FS were identified. Nasopharyngeal swabs were performed to detect respiratory viruses. We categorized FS into simple febrile seizures (SFS) and complex febrile seizures (CFS) based on established criteria. Methods: We performed a prospective, observational study of pediatric patients in the general ward from October to December 2016. Objectives: This study aimed to compare short-term outcomes at pediatric intensive care unit (PICU) discharge in critically ill children with and without continuous EEG (cEEG) monitoring. Methods: We retrospectively compared 234 patients who underwent cEEG with 2294 patients without cEEG. Propensity score matching was used to compare patients with seizures and status epilepticus between cEEG cohort and historical cohort. Results: The EEG cohort had higher in-hospital mortality, worse Pediatric Cerebral Performance Category (PCPC) scores, and greater PCPC decline at discharge.
