Heart failure (HF) and chronic kidney disease (CKD) frequently coexist, which elevates the risks of hospitalization, disease progression, and death. Despite advances in treating each condition independently, many challenges remain in diagnosing and managing them in combination. In March 2024, Kidney Disease: Improving Global Outcomes (KDIGO) held the Controversies Conference on Kidney Disease and Heart Failure: Recent Advances and Current Challenges. Discussions highlighted the complex, bidirectional relationship between HF and CKD, including shared risk factors and overlapping pathophysiology as well as nuances in interpreting biomarkers such as natriuretic peptides and serum creatinine. Background:

Menopause associated asthma impacts a subset of women and is less responsive to current treatments. Mechanisms driving this late onset asthma are unknown. We recently developed a mouse model of menopause associated asthma using a combination of 4-Vinylcyclohexene Diepoxide (VCD) and House Dust Mite (HDM) exposures. The goal of this study was to determine how hormone replacement therapy during perimenopause impacts lung function and inflammation.

Obsessive-compulsive disorder (OCD) is a chronic, serious psychiatric disorder that affects 2%-3% of the population and is associated with high personal and societal costs. Genetic factors are estimated to explain roughly half the risk of developing OCD, and genomic studies are just beginning to identify common and rare genetic variants mediating this risk. A major goal of genomic studies is to yield insights into the etiology of OCD and identify molecular targets for the development of novel therapeutics. ΔFOSB, an unusually stable member of the AP-1 family of transcription factors, mediates long-term maladaptations that play a key role in the pathogenesis of drug addiction, cognitive decline, dyskinesia, and several other chronic neurological and psychiatric conditions. We have recently identified that 2-phenoxybenzenesulfonic acid-containing compounds disrupt the binding of ΔFOSB to DNA in vitro in cell-based assays, and one such compound, JPC0661, disrupts ΔFOSB binding to genomic DNA in vivo in the mouse brain with partial efficiency. When a marriage goes wrong, a court may use a non-disparagement order to prevent a party from speaking negatively about another in some way, either permanently or temporarily. Family courts often use non-disparagement orders to protect parties or their children from psychological harm during divorce or child-custody proceedings. In such cases, non-disparagement orders forbid parents from denigrating one another in front of their children and/or third parties. To protect the child, non-disparagement orders are often imposed upon both parents by the court. But courts do not always protect children and other parties from disparagement in this way. For centuries, American criminal courts have overwhelmingly favored “general” verdicts over “special” verdicts, without giving the latter a fair trial. A general verdict requires criminal jurors to deliver only an ultimate judgment of “guilty” or “not guilty” on a charged offense. In contrast, a special verdict would require jurors to answer a sequence of questions about whether each legal element of the charged offense has been proven beyond a reasonable doubt before they deliver a final judgment. One of the most important issues for product-liability lawsuits against drug manufacturers today is federal preemption. Plaintiffs injured by prescription drugs regularly turn to state tort law for a remedy. Defendants in these cases—often brand-name drug manufacturers—frequently invoke preemption doctrine to argue that the Food and Drug Administration’s (“FDA”) rigorous approval process for prescription drugs has displaced state causes of action. Transcription factor EB (TFEB) is a master regulator of lysosomal biogenesis and cellular clearance pathways. TFEB activity is tightly controlled by multiple post-translational mechanisms, but the exact molecular mechanism controlling its stability has remained elusive. Here, we identify the IκB kinase (IKK) complex as a key regulator of TFEB protein stability through a phosphorylation-ubiquitination cascade. A high-content kinase inhibitor screen reveals that IKK inhibition increases TFEB protein levels, and genetic ablation of IKK components increases TFEB stability, upregulates lysosomal genes, and enhances lysosomal biogenesis and degradative capacity. INTRODUCTION: Recent advances in osteoimmunology have illuminated the significant role of neuroimmune interactions in bone and joint health. The neuroimmune system critically regulates bone cell function through a complex network of molecular mediators - including signals from the sympathetic and parasympathetic nervous systems, pro- and anti-inflammatory cytokines, cortisol and other hormones, and growth factors. These mediators interact within a nonlinear network where each component can influence others reciprocally, with the brain serving as a central coordinator.

Pelvic organ prolapse remains a common condition requiring durable surgical correction, with sacrocolpopexy considered the gold standard. Transvaginal natural orifice transluminal endoscopic surgery (vNOTES) has emerged as a scarless minimally invasive approach, while the integration of robotic single-port technology may further enhance surgical precision and ergonomics. However, data on robotic-assisted single-port vNOTES (RSP-vNOTES) sacrocolpopexy remains limited. We report a case of a 58-year-old Asian woman with symptomatic apical pelvic organ prolapse. The pelvic organ prolapse quantification (POP-Q) assessment demonstrated stage II uterine prolapse. Purpose: To generate a comprehensive profile of microRNAs (miRNAs) present in human tear fluid using next-generation sequencing (NGS), establish a reference miRNome for healthy human tear fluid, and investigate whether miRNA expression varies by sex, race, or age.

Methods: Tear samples were collected from 32 adults using Schirmer strips. RNA was isolated using the miRNeasy Serum/Plasma Kit, and cDNA libraries were prepared using the QIAseq miRNA Library Kit. Barcoded libraries were sequenced on the NovaSeq 6000 platform. Background: Siblings and household contacts of physically abused children are at increased risk for physical abuse and may have injuries that are not clinically apparent. However, injury testing recommendations for contact children remain debated.

Objective: Our objective was to describe testing practices and yield for contacts of physically abused children.

Participants and settings: This is a cross-sectional study of children < 10 years old evaluated for physical abuse at 10 US pediatric referral centers in the CAPNET research network.

Methods: We report the proportion of contact children who had a skeletal survey (SS) or neuroimaging and describe the proportion of tests that identified injuries.

The purpose of this project was to understand public perceptions of brands using influencers. The brands Dunkin’ and Tarte Cosmetics served as case studies for this study. A mixed-methods approach was undertaken. Specifically, social media listening in the form of a content analysis was conducted. That was then followed by an online survey of students from a medium-sized university in the northeast. A number of factors were measured related to participants’ stated brand and influencer effectiveness on social media in general, as well as specific to Dunkin’ and Tarte. However, most existing models operate under the unrealistic closed-world assumption, where training and test data are drawn from the same distribution. This paper presents Rosetta-XAI, a comprehensive software framework for evaluating and explaining Large Language Model (LLM) behavior in cross-language code conversion tasks. The system implements a four-stage automated pipeline: (1) code generation by LLMs accessed through the Ollama API inference service, (2) regex-based extraction of code blocks from markdown responses, (3) language-specific syntax and compilation validation with temporary artifact management, and (4) execution with timeout protections and CSV-based checkpoint recovery. Importance: Left bundle-branch pacing (LBBP) has been proposed as an alternative to biventricular pacing (BiVP) for patients with heart failure with left bundle-branch block (LBBB). However, robust clinical evidence from randomized clinical trials is lacking.

Background:

Sorting Nexin 27 (SNX27), a key regulator of synaptic receptor trafficking and endosomal recycling, has been implicated in maintaining synaptic homeostasis and cognitive function. To date, variants in SNX27 have been reported in a small number of patients across three publications with severe neurodevelopmental phenotypes. However, the genetic and functional landscape of SNX27-related disorders remains poorly understood, and further evidence is needed to confirm its association with disease and to better delineate the associated phenotype.

Web caching offers several advantages, such as increasing cache hit rates, lowering the workload on origin servers, and minimizing network traffic. Nevertheless, limited cache capacity poses a major challenge in web caching systems. Moreover, repeatedly fetching same media objects from origin servers leads to unnecessary bandwidth consumption. Furthermore, traditional caching policies, including Least Recently Used (LRU), are vulnerable to cache pollution. This study introduces a collaborative caching policy based on the Naïve Bayes (NB) Machine Learning (ML) algorithm. This study explores how digital technology is integrated into mathematics education, focusing on the views of secondary school mathematics teachers and using a quantitative research method through a survey. This study aimed to examine the effectiveness of a story-based counseling program in developing spiritual intelligence skills among first-grade students in the Sultanate of Oman. The researchers adopted a quasi-experimental approach and applied tools including a spiritual intelligence scale and a specially designed counseling program to achieve the study’s objectives. Digital Citizenship Among Middle School Students In Oman: A Study Considering Various Variables, Suad Saleh Al Wahaibi, Khalfan Zahran Al-Hijji, Raja Maznah Raja Husain

This study examined the level of digital citizenship (DC) among grade 10 students in Oman with respect to gender, internet usage duration, and exposure to online threats. A total of 1,022 students participated during the 2022/2023 academic year. The study employed a questionnaire constructed on Ribble’s (2015) Respect, Educate, and Protect (REP) model and applied a random cluster sampling technique. Results revealed that students’ DC level was moderate overall. This study examines the impact of voluntary tipping compared to mandatory service charges on consumer satisfaction. An analysis was conducted which contained data from restaurants based in the coastal region of southern California. The dataset utilizes a citizen-curated list compiled of restaurants that impose mandatory service charges alongside a list of restaurants. This second list is compiled of restaurants that offer comparable cuisine and pricing but contrasting tipping practices. Restaurants that impose mandatory service charges of 15% or 20% were compared to restaurants that use traditional voluntary tipping.
