Objective: Ultrasound (US) has been proposed as a potential tool for assessing skin fibrosis in systemic sclerosis (SSc). However, a large-scale comparison of US-based assessment with histologic markers of skin fibrosis has not been reported. We evaluated US-based skin assessments for their face validity (differentiation between involved SSc and healthy control [HC] skin), construct validity (comparison to modified Rodnan skin score [mRSS]), and criterion validity (comparison to histologic and gene expression fibrosis markers).

Methods: Twenty HCs and 52 patients with SSc underwent clinical and US assessment followed by a forearm skin biopsy. Predefined areas were assessed on the finger, hand, …

Introduction: Past studies of hepatitis B vaccines in people with HIV (PWH) have shown mixed results about improving responses with increases in dose or dose frequency, leading to inconsistent guidelines and practice. We examined vaccine series completion and revaccinations in PWH lacking a seroprotective response.

Methods: Hepatitis B vaccination histories were evaluated as a retrospective study in 561 participants of A5379 trial (NCT04193189). Individuals in 41 sites in Africa, Asia, South America, and the United States qualified for the trial with antibody titers below the seroprotective level (< 10 mIU/mL) despite past hepatitis B vaccinations. The primary measure of interest was the number of vaccines in the past vaccination records.

Results: Of the 1098 trial candidates, 561 (51%) did not …

Faculty, Staff and Student Publications

Background: In cases of massive transfusion, intravenous access can be limited. Additionally, delivering different products through separate lines and infusion methods adds unnecessary steps. RBC, plasma, and whole blood (WB) are delivered via rapid infusers; cryoprecipitate (CRYO) is infused through separate access and is not approved with these devices.

Study design and methods: We examined the impact of different transfusion methods on hemostatic performance and factor activity using two available CRYO products. Ten bags of CRYO, ten bags pathogen-reduced CRYO (Intercept® Fibrinogen Complex, IFC), and 20 units of WB were obtained. Each CRYO bag was infused with one unit of …

Are Drains After Trauma Splenectomy Associated With Organ Space Surgical Site Infections?, Stephanie Martinez Ugarte, Mokunfayo O Fajemisin, Seokhun Kim, William D Rieger, Renee W Green, Gabrielle E Hatton, Lillian S Kao

Faculty, Staff and Student Publications

Introduction: Left upper quadrant (LUQ) drain use after trauma splenectomy varies, with minimal existing data to guide practice. We aimed to investigate the association of LUQ drains with organ space surgical site infection (OS-SSI) after splenectomy. We hypothesized that LUQ drains after trauma splenectomy are associated with increased odds of OS-SSI.

Patients and methods: A retrospective study of adult blunt trauma patients who underwent splenectomy from 7/2018 to 6/2024 was performed. We excluded patients who survived < 5 days after operation, and those with an intraoperatively identified pancreatic injury. Demographics, surgical details, and drain information were abstracted from the medical record. Inverse probability of treatment weighting (IPTW) was used to balance variables between patients who did and did not receive an LUQ drain after splenectomy. Multivariable logistic regression with IPTW was performed to estimate the association of LUQ drains with OS-SSI.

Results: Of 179 patients, 68% (n = 121) were male, the median age was 38 years (interquartile range 28, 55), and 27% (n = …

Research outputs 2022 to 2026

SummaryBackground and AimsDietary nitrate and nitrite may have source-dependent effects on liver health: plant-sourced nitrate may confer hepatoprotective benefits via its conversion to nitric oxide, whereas nitrate and nitrite from animal foods, processed meats, or contaminated drinking water may promote liver injury via their conversion to N-nitrosamines. We aimed to examine associations between source-specific nitrate and nitrite intakes and incident metabolic dysfunction-associated steatotic liver disease (MASLD) in two large cohorts. Secondary aims were to assess associations with liver MRI biomarkers and to evaluate whether nitric oxide-pathway gene variants modify associations with plant-sourced nitrate/nitrite.MethodsWe analysed 53,854 participants from the Danish Diet, …

Research outputs 2022 to 2026

This study compared five solvents - urea, carbonate-bicarbonate, citrate, phosphate, and water - for their ability to extract proteins from beech (BM) (Hypsizygus marmoreus) and Swiss brown (SB) (Agaricus bisporus) mushrooms. Using mass spectrometry-based proteomics and bioinformatics, we found that urea produced the highest total protein identifications by data-dependent acquisition at 1% false discovery rate (FDR): BM: 1739, SB: 1907; and by data-independent acquisition: BM: 1969, SB: 2172 proteins, especially enriching cytosolic and ribosomal proteins. Carbonate-bicarbonate favoured the extraction of metabolic and translational proteins, while citrate targeted extracellular and membrane-associated proteins related to cell wall dynamics. Phosphate-enriched catalytically active proteins, …

Beyond The First Bout: Adaptations To Repeated Injuries Across Physiological And Pathological Conditions, Cory W. Baumann, Christopher P. Ingalls, Kazunori Nosaka, Jarrod A. Call

Research outputs 2022 to 2026

Skeletal muscle exhibits remarkable plasticity following injury, yet most research has focused on responses to a single bout of eccentric contractions. This review addresses adaptations to repeated eccentric contraction–induced injuries across physiological and pathological conditions, with emphasis on insights from preclinical rodent models. In healthy muscle, the repeated bout effect (RBE) reduces strength loss and accelerates recovery after subsequent bouts. However, these adaptations are highly condition dependent. Aging can attenuate the RBE, while dystrophic muscle remains vulnerable to repeated injury despite compensatory remodeling. Other factors, including but not limited to, chronic alcohol intake and malignant hyperthermia can influence these responses, …

Research outputs 2022 to 2026

Background: Health service transitions of child, adolescent, and young adult cancer survivors between paediatric and adult cancer care services are pervasively fragmented, resulting in poor coordination, reduced satisfaction, and adverse outcomes for cancer survivors and significant others (e.g., caregivers and family members). Numerous trials and clinical guidelines have been developed to support care transitions, but their convergence, divergence, and methodological quality remain unclear. To advance clinical practice and quality research about care transitions, a synthesis of trials and guidelines on child, adolescent, and young adult care transition across age-appropriate cancer services is required to inform future research, practice, guideline, and …

Research outputs 2022 to 2026

NRAS mutations occur in 10%–30% of cutaneous melanomas and are associated with high tumor mutational burden. Mutant NRAS signaling drives aberrant cell growth and proliferation, in part, through activation of the RAF–MEK-ERK1/2 kinase pathway; however, targeted therapies to this pathway have limited effectiveness in patients with NRAS mutant melanoma. The role of other targetable signaling pathways in NRAS mutant melanoma is poorly characterized. Here, we demonstrated that one isoform of diacylglycerol kinase, diacylglycerol kinase eta (DGKη), a lipid signaling regulator, was highly expressed in NRAS mutant melanoma patient samples. Knockdown of DGKH in NRAS mutant melanoma cell lines resulted in …

Doecke, Pratishtha Chatterjee, Steve Pedrini, Hamid R. Sohrabi, Nicholas J. Ashton, Henrik Zetterberg, Manohar L. Garg, Kaj Blennow, Ralph N. Martins

Research outputs 2022 to 2026

Alterations in plasma sphingomyelin (SM) levels have been reported in Alzheimer's disease (AD), pointing to disturbances in lipid metabolism that may contribute to disease pathogenesis. Neuronal damage in early AD triggers tau release into central and peripheral systems. Despite influence from peripheral contributions, alterations in plasma total-tau (T-tau) remain valuable in indicating AD-related neurodegeneration. Investigating relationships between SM metabolism and tau release during preclinical AD may uncover important biochemical processes and support advancing early non-invasive detection and treatment approaches. This cross-sectional study investigated cognitively unimpaired (CU) older adults from the KARVIAH cohort, grouped by cortical amyloid-β (Aβ) status through positron …

Research outputs 2022 to 2026

Amyloidogenic proteins, such as amyloid-β (Aβ), self-assemble into cross-β fibrils whose accumulation is central to Alzheimer's disease (AD). Measuring Aβ aggregation and clearance in living cells remains challenging using current cell-based assays, which are often low-throughput or not suited for real-time monitoring. This study aimed to (1) develop a robust, quantitative, and scalable fluorescence-based assay using Amytracker to monitor Aβ accumulation and clearance in an Aβ-producing neuronal cell model, and (2) validate its utility for mechanistic studies and therapeutic screening. We established a plate-based fluorescence assay using Amytracker in MC65 neuronal AD model expressing the Amyloid precursor protein C-terminal fragment …

Anthropogenic stresses in dryland regions can alter hydrology, salinization, and biogeochemical processes. How these pressures influence methane (CH4) cycling and emissions from aquatic ecosystems remains poorly understood. Over the course of 1 year, we measured diffusive and ebullitive CH4 emissions from resacas—shallow, hydrologically managed distributary channels prevalent throughout the Lower Rio Grande Valley along the US–Mexico border. Despite warm, nutrient-rich waters, CH4 emissions from resacas were low, and more comparable to rates observed in estuarine systems. CH4 fluxes were positively related to temperature but negatively related to salinity and rainfall. Elevated salinity, driven by evaporation and source water from the …

Research outputs 2022 to 2026

Proton exchange membrane fuel cell (PEMFC) systems’ reliable operation under variable load conditions is a significant challenge because of voltage fluctuations, transient variations, and the high computational requirements of sophisticated control techniques like intelligent and model predictive controllers. Even while these state-of-the-art methods provide great precision, complexity and cost often limit their real-time applicability, underscoring the need for a simpler but efficient control solution. To achieve effective voltage regulation in PEMFC systems, this study suggests a DC–DC buck converter in conjunction with a comparator-based proportional–integral–derivative (PID) control technique. In order to provide stable operation under both constant and changing load …

The Rise And Fall Of The World's Greatest Marine Biodiversity Hotspot, Alexandre C. Siqueira, Kiessling, Nussaïbah Raja, David R. Bellwood

Research outputs 2022 to 2026

Habitat condition and area shape global species distributions, with shallow-water reefs hosting a disproportional share of marine biodiversity. Although reef area is a well-established predictor of marine species richness, its historical context is less well understood. Here, we show that the rise of tropical marine biodiversity is closely tied to reef expansion in space and time. During the Early/Mid Miocene (23 to 11.6 million years ago), Indo-Pacific reefs reached unprecedented size and thickness, surpassing any reef systems in the past 66 million years. These massive reefs, likely driven by unique environmental, biotic, and tectonic conditions, fostered the expanding diversity and …

Between-Session Reliability And Sensitivity Of Force Production Characteristics Across Various Force-Time Points During The Isometric Single-Leg Long-Lever Bridge, Adam E. Sundh, Nicholas J. Ripley, A. J. Lamb, Conor J. Cantwell, Paul Comfort

Research outputs 2022 to 2026

The purpose of this study was to assess the between-session reliability of peak force and early force-time variables in a unilateral isometric hamstring assessment (isometric long-lever bridge). The sample consisted of 30 collegiate male athletes (age: 19.4±1.3 years; height: 1.79±0.1 m; body mass: 80.4±10.3 kg) with no history of recent hamstring injury (<6 months) who volunteered to participate in the study. All participants performed 3 unilateral maximal voluntary isometric contractions (MVIC) with their heels on force plates and shoulders elevated 15.24 cm, while their hips were secured with a loaded barbell. Data was analyzed to assess the reliability of force values at 50, 100, 150, 200, 250 ms, and peak force (N) across three repeated sessions. Moderate-good relative and absolute reliabilities were observed across all early force-time variables with lower bound ICC values≥0.51 (0.51–0.71) and point estimate CV%≤12.65% (7.63–12.65), with moderate relative (ICC≥0.74; range 0.74–0.92) and absolute (CV%≤6.50%; range 6.48–6.50) reliability for peak force. Trivial to small effect sizes (g≤0.4) were observed between sessions. These findings suggest that the isometric single-leg long-lever bridge (SL-LLB) can be used to reliably monitor peak and rapid force generating capacities to track performance changes over time due to training adaptations as well as acute or chronic fatigue.

Research outputs 2022 to 2026

Growth anomalies (GAs) are coral diseases characterised by tumour-like skeletal lesions reported globally, yet their causes remain poorly understood. Microorganisms are integral to coral health, but the role of bacterial communities in GAs remains unclear. We investigated an outbreak of GAs in Isopora palifera at the Cocos (Keeling) Islands using 16S rRNA amplicon sequencing to compare bacterial communities of GA-affected and asymptomatic corals, surrounding water and potential pollution sources. Significant differences in bacterial beta diversity were observed across sites, with an interaction between location and coral health status. Coral and water samples hosted distinct microbial communities, but there was no …
