Population Health, East Africa

Background: When confronted with diagnostic uncertainty and a decision on whether to start treatment or not, clinicians consider the potential harm and benefit of offering versus withholding treatment. Treatment can be offered if the probability of tuberculosis (TB) in the patient is above the “therapeutic threshold” (ThT): the probability of disease at which the expected utility of treating and not treating is the same. We estimated ThT for TB in clinical and community settings in Southern Africa using two methods: an adapted nominal group technique (aNGT), and decisions made based on clinical vignettes (CVs).

Methods: We enrolled health …

Effects Of Cell-Free Supernatants Of Commercial Yogurt Products On Salmonella Survival, Juliet Durant, Akayla King

Student Posters

Salmonellosis remains a global public health challenge and Salmonella enterica serovar Enteritidis and Typhimurium are the primary causes. Salmonella virulence mechanisms involve attachment and invasion of intestinal epithelial cells. Gut microbiota and probiotics that contain Lactobacillus species have been shown to prevent Salmonella infection. Yogurt is a commonly consumed probiotic product containing Lactobacillus species. Yogurts made under laboratory conditions and the cell-free supernatants have been shown to inhibit Salmonella growth. The many commercial yogurt products on the market may vary in antimicrobial components based on production methods. Therefore, the present study aimed to evaluate the survival of 5 log CFU/mL …

Background: Merkel cell carcinoma (MCC) is a rare, aggressive cutaneous malignancy with neuroendocrine differentiation. Several molecular pathways have been implicated in MCC development and multiple cell-of-origin candidates have been proposed, including neural crest cells, which express acetylcholine receptors (AChRs). The role of nicotinic acetylcholine receptors (nAChRs) in MCC has not been explored. In this study, we investigated if MCC expresses nAChRs and if nAChR expression correlates with patient characteristics.

Methods: The study included 71 MCC cases diagnosed with sufficient tissue available to perform immunohistochemical analysis. The median follow-up was 29.8 months (range, 2.7-234.1). We performed immunohistochemistry using antibodies against the …

Host effector CD4+ T cells emerge as critical mediators for tumor regression but whether they can be activated by adoptively transferred CD8+ T cells remains unknown. We previously reported that adoptive transfer of interleukin 9 (IL-9)-producing cytotoxic CD8+ T (Tc9) cells achieved long-term control of tumor growth. Here, we demonstrate that murine tumor-specific Tc9 cells control the outgrowth of antigen-loss relapsed tumors by recruiting and activating host effector CD4+ T cells. Tc9 cells secreted IL-24 and recruited CCR7-expressing conventional type 2 dendritic cells (cDC2 cells) into tumor-draining lymph nodes to prime host CD4+ T cells against relapsed tumors. Host CD4+ …

Leukocytes migrate through the blood and extravasate into organs to surveil the host for infection or cancer. Recently, we demonstrated that intravenous (IV) anti-CD45.2 antibody labeling allowed for precise tracking of leukocyte migration. However, the narrow labeling window can make this approach challenging for tracking rare migration events. Here, we show that altering antibody administration route and fluorophore can significantly extend the antibody active labeling time. We found that while both IV and intraperitoneal (IP) anti-CD45.2 antibody labeled circulating leukocytes after injection, they had different kinetic properties that impacted labeling time and intensity. Quantification of circulating antibody revealed that while …

Pulsesight: Ai-Powered Smartphone Solution For Non-Invasive Oxygen Saturation, Respiration Monitoring & Emr Integration, Kazi Zawad Arefin

Dissertations (1934 -)

The utilization of non-invasive, contactless methods to detect physiological parameters such as oxygen saturation (SpO2) and respiration rate has the potential to significantly improve healthcare delivery. This dissertation suggests a new system that utilizes photoplethysmography (PPG) signals extracted from facial and fingertip video recordings. These video recordings are captured using a standard smartphone. The system accomplishes real-time, contactless health monitoring without the necessity of specialized medical equipment by employing advanced image processing and signal analysis techniques. This method addresses critical health challenges, particularly for vulnerable populations, by facilitating continuous monitoring in resource-constrained environments. The development of a context-aware mobile application …

Prediction Of Grade 4 Radiation-Induced Lymphopenia During Chemoradiation Therapy For Lung Cancer Patients: Insights From Two Past Trials, Serena Monti, Giuseppe Palma, Ting Xu, Radhe Mohan, Zhongxing Liao, Laura Cella

Faculty, Staff and Student Publications

Background and purpose: Radiation-induced lymphopenia (RIL) is a significant side effect associated with radiation therapy (RT) with important prognostic implications. We developed and tested a normal tissue complication probability (NTCP) model for Grade 4 (G4) RIL in patients with locally advanced Non-Small-Cell Lung Cancer (NSCLC) who underwent concurrent chemotherapy and RT, analyzing data from patients enrolled in two clinical trials.

Materials and methods: We retrospectively analyzed the data from NCT00915005 (MDA-cohort) and NCT00533949 (RTOG0617-cohort) trials. After finding the candidate predictors of G4-RIL, defined as absolute lymphocyte count (ALC) at nadir < 0.2*109 cells/l during RT, we trained an NTCP model on the MDA-cohort and tested it on the RTOG-cohort, based on common available variables in the two cohorts. Model performance was assessed in terms of discrimination and calibration.

Results: In the MDA-cohort, 55 out of 161 (34%) patients …

Giving Voice To Student Grief: Using Community-Based Participatory Research To Develop And Pilot A Translational Storytelling Intervention For Bereaved College Students, Cassidy Rae Taladay-Carter

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

The purpose of this dissertation was to employ community-based participatory research (CBPR, Wallerstein et al., 2018) methods to attend to a pressing issue in the community. In partnership with one local grief center, we identified the need for enhanced grief support for current emerging adult undergraduate students in our area. Nearly 50% of U.S. emerging adults enrolled in four-year residential universities experience the death of an important figure (Cox et al., 2015; Cupit et al., 2021), at a time when they are developing important facets of their narrative identity. Without the coherence and meaning structures provided by narrative identity, students …

Chief Justice Roberts of the United States Supreme Court has said that "the way to stop discrimination on the basis of race is to stop discriminating on the basis of race. "I In this Article, I examine what it means to discriminate on the basis of race-or what it means to stop discriminating on the basis of race. I consider interventions designed to enhance social welfare in an economy with racially discriminatory games operating at its base. One set of interventions is color-blind, in the sense that it treats all actors alike regardless of race. The other set of interventions …
