In vitro derivation of pancreatic β-cells from human pluripotent stem cells holds promise as diabetes treatment. Despite recent progress, efforts to generate physiologically competent β-cells are still hindered by incomplete understanding of the microenvironment's role in β-cell development and maturation. Here, we analyze the human mesenchymal and endothelial primary cells from weeks 9-20 fetal pancreas and identify a time point-specific microenvironment that permits β-cell differentiation. Further, we uncover unique factors that guide in vitro development of endocrine progenitors, with WNT5A markedly improving human β-cell differentiation. WNT5A initially acts through the non-canonical (JNK/c-JUN) WNT signaling and cooperates with Gremlin1 to inhibit …

Background & Aims: Biliary atresia (BA) is the commonest single etiology indication for liver replacement in children. As timely access to liver transplantation (LT) remains challenging for small BA children (with prolonged waiting time being associated with clinical deterioration leading to both preventable pre- and post-transplant morbidity and mortality), the care pathway of BA children in need of LT was analyzed—from diagnosis to LT—with particular attention to referral patterns, timing of referral, waiting list dynamics and need for medical assistance before LT. Methods: International multicentric retrospective study. Intent-to-transplant study analyzing BA children who had indication for LT early in life …
Implementation Of A Genome-Wide Survey Of Induced Mutations To Identify Agronomically Valuable Variants In Chenopodium Quinoa, Andrew Alarcon Parker
Implementation Of A Genome-Wide Survey Of Induced Mutations To Identify Agronomically Valuable Variants In Chenopodium Quinoa, Andrew Alarcon Parker
Theses and Dissertations
Quinoa has been utilized for millennia in the Andes region of South America as a nutritious and hardy food crop. In recent years interest in quinoa has grown as need increases for an alternative to traditional cereal crops that can tolerate marginal environments while offering superior nutrition. Growers outside the Andes have experienced several complications adopting quinoa, including undesirable secondary metabolites, poor yield, lodging, and height inconsistency. Unfortunately, access to native ecotypes for crop improvement is limited, and desirable traits are difficult to introduce into available quinoa cultivars because of its allotetraploid genome and tendency to self-pollinate. A genome-wide survey …

Hydrogen sulfide (H2S) serves as an important gaseous signaling molecule that is involved in intra- and intercellular signal transduction in plant–environment interactions. In plants, H2S is formed in sulfate/cysteine reduction pathways. The activation of endogenous H2S and its exogenous application has been found to be highly effective in ameliorating a wide variety of stress conditions in plants. The H2S interferes with the cellular redox regulatory network and prevents the degradation of proteins from oxidative stress via post-translational modifications (PTMs). H2S-mediated persulfidation allows the rapid response of proteins in signaling networks …
The Connection Between Body Modification And Personality, Taylor Shoemaker
The Connection Between Body Modification And Personality, Taylor Shoemaker
Steeplechase: An ORCA Student Journal
Previous research has been inconsistent in its findings regarding the associations between body modifications (e.g., piercings, tattoos, augmentation, scarification, split tongue) and the Big Five personality traits (i.e., openness to experience, conscientiousness, extraversion, agreeableness, neuroticism). All traits have been found to be significantly correlated with body modification in at least one study, but their significance differed from study to study. The purpose of the current study was to examine the associations between body modification and each domain of personality concurrently to add to the literature surrounding differences between modified and unmodified individuals. To participate in this study, participants were asked …

Mycotoxins are secondary metabolic products of fungi. They are poisonous, carcinogenic, and mutagenic in nature and pose a serious health threat to both humans and animals, causing severe illnesses and even death. Rapid, simple and low-cost methods of detection of mycotoxins are of immense importance and in great demand in the food and beverage industry, as well as in agriculture and environmental monitoring, and, for this purpose, lateral flow immunochromatographic strips (ICSTs) have been widely used in food safety and environmental monitoring. The literature to date describing the development of ICSTs for the detection of different types of mycotoxins using …

The Academic Work-Life Balance Of Entry-Level Athletic Training Students And Its Implications, Grant Pheil
School of Health and Human Performance Theses
Athletic training students (ATS) have been documented to experience stressors and perceive burnout up to this point in their assimilation into the profession. This is understood through recent work-life balance (WLB) and burnout research within the student population, stemming off research recommendations surrounding the professional setting. However, following current educational shifts and worldwide events, prior research is limited surrounding specific graduate level ATS WLB, stressors, coping responses, and comparisons across ATS demographics. The purpose of this study is to identify this balance in graduate level students and highlight stressors or management techniques that will benefit future students pursuing athletic training …

Nutrient Transporter Gene Expression In The Early Conceptus—Implications From Two Mouse Models Of Diabetic Pregnancy, Claudia Kappen, Claudia Kruger, Sydney Jones, J. Michael Salbaum
Division of Basic Science Faculty Publications
Maternal diabetes in early pregnancy increases the risk for birth defects in the offspring, particularly heart, and neural tube defects. While elevated glucose levels are characteristic for diabetic pregnancies, these are also accompanied by hyperlipidemia, indicating altered nutrient availability. We therefore investigated whether changes in the expression of nutrient transporters at the conception site or in the early post-implantation embryo could account for increased birth defect incidence at later developmental stages. Focusing on glucose and fatty acid transporters, we measured their expression by RT-PCR in the spontaneously diabetic non-obese mouse strain NOD, and in pregnant FVB/N mouse strain dams with …

Pharmacy
Topical conveyance of antifungal agents like itraconazole ITZ has been giving good grounds for expecting felicitous antifungal medicines. The defiance of topical delivery of this poorly water soluble and high-molecular-weight drug, however, mightily entail an adequate vehiculation. ITZ aspasomes, newer antioxidant generation of liposomes, have been designed and enclosed in a cream to ameliorate skin deposition. The proposed creams containing non-formulated ITZ or encapsulated in aspasomes (0.1% or 0.5%) were topically applied in patients with diagnosed diaper dermatitis complicated by candidiasis, tinea corporis (TC), and tinea versicolor (TVC). Placebos (void aspasomal cream and cream base) were also utilized. The obtained …

Background: Sex-specific differences in fetal lung maturation have been well described; however, little is known about the sex-specific differences in microRNA (miRNA) expression during human fetal lung development. Interestingly, many adult chronic lung diseases also demonstrate sex-specific differences in prevalence. The developmental origins of health and disease hypothesis suggests that these sex-specific differences in fetal lung development may influence disease susceptibility later in life. In this study, we performed miRNA sequencing on human fetal lung tissue samples to investigate differential expression of miRNAs between males and females in the pseudoglandular stage of lung development. We hypothesized that differences in miRNA …
Diel Gene Expression Improves Software Prediction Of Cyanobacterial Operons, Philip Heller
Diel Gene Expression Improves Software Prediction Of Cyanobacterial Operons, Philip Heller
Faculty Research, Scholarly, and Creative Activity
Cyanobacteria are important participants in global biogeochemical process, but their metabolic processes and genomic functions are incompletely understood. In particular, operon structure, which can provide valuable metabolic and genomic insight, is difficult to determine experimentally, and algorithmic operon predictions probably underestimate actual operon extent. A software method is presented for enhancing current operon predictions by incorporating information from whole-genome time-series expression studies, using a Machine Learning classifier. Results are presented for the marine cyanobacterium Crocosphaera watsonii. A total of 15 operon enhancements are proposed. The source code is publicly available.

Sex Ratios And The City: Is Parental Corticosterone A Mediator Of The Relationship Between Urbanization And Offspring Sex Allocation?, Denyelle Kilgour
Master of Science in Integrative Biology Theses
The Trivers-Willard hypothesis states that mothers can adjust their offspring sex ratios according to their own condition and the environmental conditions they face during breeding. Past tests of this hypothesis in avian species have focused on how natural variation shapes sex allocation trade-offs. However, urbanization also influences individual condition and presents animals with novel challenges in optimizing their brood sex ratio. One likely mediator of the relationship between environmental conditions, parental condition, and sex ratios is corticosterone (CORT), the primary glucocorticoid in birds. However, no research has examined the links between urbanization, individual condition, CORT, and adaptive sex allocation within …

The Tibetan Plateau harbors the largest number of glaciers outside the polar regions, which are the source of several major rivers in Asia. These glaciers are also major sources of nutrients for downstream ecosystems, while there is a little amount of data available on the nutrient transformation processes on the glacier surface. Here, we monitored the carbon and nitrogen concentration changes in a snowpit following a snowfall in the Dunde Glacier of the Tibetan Plateau. The association of carbon and nitrogen changes with bacterial community dynamics was investigated in the surface and subsurface snow (depth at 0–15 and 15–30 cm, …

Infant feeding practices can greatly impact a child's life with far reaching implications based upon decisions a mother makes for her newborn. Breastfeeding is the preferred method of infant feeding because of its association with health benefits to both the mother and child. The prevalence of breastfeeding among rural, low income, disadvantaged groups to include Native American (NA) and African American (AA) adolescent mothers is low. NAs and AAs bear a disproportionate burden of illness and health risks compared to other races and ethnic groups in the United States. Breastfeeding is a targeted public health strategy to improve the health …

Percent soil organic matter (SOM), pH and crop yield are among the biophysicochemical process-driven soil health indicators (SHIs). However, identifying sustainable soil health conditions using these SHIs is limited due to the lack of Integrated Productivity Efficiency (IPE) models. We define IPE as a concept that identifies best-to-worst-case soil health outcomes by assessing the effect of agronomic practices on weighted abundance of functional guilds (WAFG) of beneficial soil organisms and SHIs simultaneously. Expressing WAFG of all beneficial nematodes (x-axis) and SHIs (y-axis) as a percent of untreated control and regression of x and y reveals four quadrants describing worst-to-best-case outcomes …

Recently, a new ontological way of exploring sport and physical activity (PA) has been proposed: the type of PA performed is classified using the four ontological dimensions. This phenomenon has classified PA as individual (I-me), encounter (I-You), team (I-Society), and nature (I-Nature). There has only been one study that has examined how participating in these various types of PA influences moods; however, that study asked individuals to recall what types of PA were performed and how they felt more than 60 days before the commencement of the study. PURPOSE: To identify whether daily moods differ based on the four …

Educational-based psychological skills training (PST) is effective in terms of Positive Psychology outcomes. Mental toughness (MT), a Positive Psychology construct, is positively associated with sports performance via mostly correlational research. Sports training emphasizes working on the weaknesses of the athlete. Positive Psychology is rooted in strength-based interventions. In Applied Sports Positive Psychology, where females are underrepresented, the two approaches appear contradictory. PURPOSE: To examine the effects of deficit- versus strength-based interventions on MT levels of female collegiate athletes. METHODS: Out of 161 female SUNY Plattsburgh athletes, 95 participated. MT scores were collected via the eight-item, Mental Toughness Index …

International Journal of Exercise Science: Conference Proceedings
Recently, moderate-to-vigorous intensity physical activity (MVPA) has attracted additional scientific interest (e.g., ACSM recommendations) not only thanks to a) the physical and psychological benefits that can be experienced by the general population following just a single bout, but also b) since high levels of MVPA seem to mitigate the mortality risk associated with high levels of sedentary behavior, such as sitting. Non-pharmaceutical interventions against the COVID-19 (e.g., stay-at-home orders) have altered people’s lifestyles (e.g., physical activity, sitting time). Grit, a higher-order personality trait based on two lower-order components (i.e., perseverance and consistency), is highly predictive of both success and performance. …
