Microbiota Dynamics, Metabolic And Immune Interactions In The Cervicovaginal Environment And Their Role In Spontaneous Preterm Birth, Stanely Onyango, Jia Dai Mi, Anjela Koech, Patricia Okiro, Marleen Temmerman, Peter Von Dadelszen, Rachel Marie Tribe, Geoffrey Omuse
Pathology, East Africa
Differences in the cervicovaginal microbiota are associated with spontaneous preterm birth (sPTB), a significant cause of infant morbidity and mortality. Although establishing a direct causal link between cervicovaginal microbiota and sPTB remains challenging, recent advancements in sequencing technologies have facilitated the identification of microbial markers potentially linked to sPTB. Despite variations in findings, a recurring observation suggests that sPTB is associated with a more diverse and less stable vaginal microbiota across pregnancy trimesters. It is hypothesized that sPTB risk is likely to be modified via an intricate host-microbe interactions rather than due to the presence of a single microbial taxon …

This paper investigates fairness and bias in Canonical Correlation Analysis (CCA), a widely used statistical technique for examining the relationship between two sets of variables. We present a framework that alleviates unfairness by minimizing the correlation disparity error associated with protected attributes. Our approach enables CCA to learn global projection matrices from all data points while ensuring that these matrices yield comparable correlation levels to group-specific projection matrices. Experimental evaluation on both synthetic and real-world datasets demonstrates the efficacy of our method in reducing correlation disparity error without compromising CCA accuracy.

Background: Zebrafish larvae are translucent, allowing in vivo analysis of gut development and physiology, including gut motility. While recent progress has been made in measuring gut motility in larvae, challenges remain which can influence results, such as how data are interpreted, opportunities for technical user error, and inconsistencies in methods.
Methods: To overcome these challenges, we noninvasively introduced Nile Red fluorescent dye to fill the intraluminal gut space in zebrafish larvae and collected serial confocal microscopic images of gut fluorescence. We automated the detection of fluorescent-contrasted contraction events against the median-subtracted signal and compared it to manually annotated gut contraction …

Boron is an essential microelement for plant growth. Tomato is one of the most cultivated fruits and vegetables in the world, and boron deficiency severely inhibits its yield and quality. However, the mechanism of tomato in response to boron deficiency remains largely unclear. Here, we investigated the physiological and molecular bases of the boron deficiency response in hydroponically grown tomato seedlings. Boron deficiency repressed the expression of genes associated with nitrogen metabolism, while it induced the expression of genes related to the pentose phosphate pathway, thereby altering carbon flow to provide energy for plants to cope with stress. Boron deficiency …

Most uveal melanoma cases harbor activating mutations in either GNAQ or GNA11. Despite activation of the mitogen-activated protein kinase (MAPK) signaling pathway downstream of Gαq/11, there are no effective targeted kinase therapies for metastatic uveal melanoma. The human genome encodes numerous understudied kinases, also called the "dark kinome". Identifying additional kinases regulated by Gαq/11 may uncover novel therapeutic targets for uveal melanoma. In this study, we treated GNAQ-mutant uveal melanoma cell lines with a Gαq/11 inhibitor, YM-254890, and conducted a kinase signaling proteomic screen using multiplexed-kinase inhibitors followed by mass spectrometry. We observed downregulated expression and/or activity of 22 kinases. …

Atherosclerotic cardiovascular disease is the leading cause of mortality worldwide due to being the predominant factor in heart attack and stroke deaths. Atherosclerotic cardiovascular disease is caused by cholesterol accumulating in arteries. It is historically thought that the cell type most responsible for causing atherosclerosis via excessive cholesterol accumulation is intimal monocyte-derived macrophages. However, macrophage-like cells (MLC) of vascular smooth muscle cell (VSMC) origin have now been proposed as another cell type to trigger atherosclerosis, since VSMC cholesterol accumulation results in these cells to trans-differentiate into a MLC phenotype. Three major genes involved in cholesterol metabolism are ABCA1, ABCG1, and …

The Foundation Review
With place-based philanthropy, a foundation provides extensive, long-term support for a comprehensive mix of programs within specific communities, with the expectation that this will produce benefits at a communitywide level. One of the key questions in designing a place-based initiative is how much the foundation will control local decision-making.
In some initiatives, the foundation dictates the issues that community groups must address and/or the nature of the planning process that will be used to develop solutions. This sometimes produces ineffective or irrelevant solutions. In contrast, other initiatives allow local groups considerable discretion in naming the issues and choosing the solutions, …

The genetic potentials of rice cultivars will need to be expressed to their fullest if global rice production is to be expanded enough by 2050 to meet the increased demand of the expanding population while the availability of land and water per capita dwindles. New and ‘improved’ rice varieties have contributed greatly to increased production over the past 50 years, but the rate of rice yield increase based on genetic changes has declined in recent decades compared with the early years of the Green Revolution. In fact, many rice consumers continue to prefer to consume ‘traditional’ rice varieties (referred to …
Projecting, Persuading, & Protecting: A Mixed Methods Analysis Of Adult Children’S Predictions Of Parents’ Quality-Of-Life Valuations In Serious Illness States, Jessica Hahne
Arts & Sciences Graduate Student Theses and Dissertations
Older adults often face complex serious illness decisions and involve their adult children as shared or surrogate decision makers. However, previous research asking family members to predict older adults’ treatment preferences in serious illness situations typically shows poor concordance with older adults’ actual preferences. In the current mixed methods analysis, we surveyed older adults (n = 38) on their health state valuations in five serious illness scenarios. Two of their adult children (n = 76) predicted their parent’s preferences. Families also participated in a semi-structured conversation to discuss their preferences and predictions. Quantitative analyses showed that parents’ average valuations were …

High Stakes, Bad Odds: Health Laws And The Revived Federalism Revolution, Nicole Huberfeld
Faculty Scholarship
The Supreme Court’s 2021 term produced a remarkable number of blockbuster decisions, nearly hiding an underlying federalism agenda that surfaced in health care, reproductive rights, administrative law, and public health related domains. Health law has been a vehicle for constitutional change before, but the stakes for older laws, most of which rely on states to accomplish national goals, have been raised. The Court has doubled down on interpretive methods that limit governmental power, using formalist tools like clear statement rules that demand specificity and offer little deference to lawmakers or regulators. These rules have constitutional dimensions, including separation of powers …

Placental Co-Culture Model Reveals Listeria Monocytogenes Utilization Of Monocytes For Vertical Transmission, Breanna Mackenzie Amelunke
Online Theses and Dissertations
Listeria monocytogenes is an opportunistic pathogen that can cross the protective placental barrier and infect the fetus, leading to congenital disabilities or death. Extracellular L. monocytogenes infection of the placenta has been well studied; however, the bacteria have not been observed translocating this barrier intracellularly. As an intracellular pathogen, L. monocytogenes could infect phagocytic cells that can be transported across the placental barrier. A novel cell co-culture system comprised of BeWo and HPVEC cell lines was established to model the human placental barrier. Barrier integrity was confirmed using immunostaining of tight junctions and a FITC-dextran permeability assay. The model was …

Coral-reef development is modulated by growth processes that deposit calcium carbonate onto the reef, such as calcification, and destructive processes that remove calcium carbonate from the reef, such as bioerosion. Wherever calcification rates exceed bioerosion rates, there is a net accretion of the reef through time, but where bioerosion rates exceed calcification rates, there is a net destruction of the reef through time. A reef’s framework is the legacy of the interaction of these processes throughout a reef’s history. Studying the fossil frameworks of reefs can shed light on the environmental factors that have controlled reef development in the past, …

The Therapeutic Role Of Lm11a-31 (A P75ntr Modulator) In The Outcome Of Ischemic Stroke And Hiv-1 Pathogenesis, Golnoush Mirzahosseini
Theses and Dissertations (ETD)
The objective of this research project is to explore the potential therapeutic role of LM11A-31 in both ischemic stroke and human immunodeficiency virus-1 (HIV-1) infection. There are limited treatment options for ischemic stroke, with tissue plasminogen activator (tPA) being the only effective medication and a narrow administration window, which is also accompanied with the high incidence of ischemic stroke in HIV-1 infected individuals. Therefore, it is critical to explore an effective therapeutic agent to address the urgent medical need. LM11A-31 is a p75 neurotrophin receptor (p75NTR) modulator and is orally bioavailable, and it can penetrate the intact blood-brain barrier (BBB). …
