Physiology Faculty Publications

The fusion of skeletal muscle stem cell (MuSC) to myofibers during hypertrophy has exclusively focused on the transfer of the MuSC nucleus, leaving the fate of other MuSC organelles, such as mitochondria, largely unexplored. The objective of this study was to determine whether MuSCs transfer their mitochondria upon myofiber fusion in response to a hypertrophic stimulus. To achieve this goal, we specifically labeled MuSC mitochondria with Dendra2 fluorescence by crossing the MuSC-specific CreER (Pax7CreER/CreER) mouse with the Rosa26-Dendra2 mouse to generate the Pax7-Dendra2 mouse. To induce the fusion of MuSC to myofibers, Pax7-Dendra2 mice underwent synergist ablation surgery …

Hormonal Decay After Death: Evaluating Dhea-S As A Potential Molecular Marker For Postmortem Estimation, Marisa Lydia Novak Jan 2025

Hormonal Decay After Death: Evaluating Dhea-S As A Potential Molecular Marker For Postmortem Estimation, Marisa Lydia Novak

Graduate Student Theses, Dissertations, & Professional Papers

Post-mortem interval (PMI) estimation is a central component of forensic anthropological investigation, but traditional methods for estimating time since death lose accuracy after ~72 hours. Recent research indicates that some steroid hormones may undergo predictable degradation patterns after death, offering potential use as biochemical clocks for PMI estimation beyond this threshold. Dehydroepiandrosterone (DHEA), and its sulfate form (DHEA-S) are of particular interest due to their metabolic stability, systemic influence, and association with trauma. This study investigates whether DHEA-S hormonal degradation patterns correlate with time elapsed since death and body temperature, evaluating their potential as biochemical PMI markers. 160 mL of …

Estrogen Receptor-Mediated Effects Of Resveratrol On Muscle Function, David Ayi-Bonte

Open Access Master's Theses

Artemis missions are set to send the next man and the first woman back to the moon and enable long-term missions on the lunar surface. However, exposure to lunar gravity has significant physiological effects characterized by loss of muscle mass, weakness and decreased bone density. Nutraceutical countermeasures such as Resveratrol (RSV), a naturally occurring polyphenol, have been identified as promising strategies to preserve musculoskeletal health; however, its pleiotropic effect hinders our ability to identify the primary cellular pathways through which it delivers its benefits. Furthermore, its efficacy in simulated lunar gravity is unknown. Based on the benefits observed with RSV …

“Democracy,” Usa, Marc Edelman Jan 2025

“Democracy,” Usa, Marc Edelman

Publications and Research

What does it mean to be an effective citizen in a democracy? Answering this question requires unpacking “democracy” and an analysis of institutions that systematically disempower and alienate citizens. This paper briefly examines the contributions of psychoanalysis to understanding democracy and authoritarianism. It scrutinizes U.S. governance institutions and points to democratic deficits and backsliding present even before Trump 1.0 and 2.0. These range from the Electoral College, the Senate, and the Supreme Court to the Federal Communications Commission. All contributed to institutionalizing minority rule and elite capture. Governance institutions failed to deliver what the American people tell opinion pollsters they …

A Systematic Review Of The Outcomes And Efficacy Of Complementary And Alternative Non-Pharmacological Interventions In Youth With Attention Deficit Hyperactivity Disorder, Kelly M. Kohler Jan 2025

A Systematic Review Of The Outcomes And Efficacy Of Complementary And Alternative Non-Pharmacological Interventions In Youth With Attention Deficit Hyperactivity Disorder, Kelly M. Kohler

Theses and Dissertations

A number of behavioral and pharmacological treatments have been widely accepted as evidenced-based approaches to treating symptoms of attention-deficit hyperactivity disorder (ADHD) in children and adolescents. While pharmacological approaches are safe and effective at managing symptoms, a large subset of caregivers do not have access to medication, refuse it, or find it ineffective, necessitating the need for non-pharmacological treatments. Such behavioral interventions are also generally effective approaches to symptom management and reducing the functional impairment of ADHD and may even demonstrate better parent satisfaction than medication alone. However, these interventions are often difficult to access and burdensome to implement, especially …

The Role Of Fruiting Bodies In Protecting A Community Of Myxococcus Xanthus Spores From The Environment, Dave Lall Jan 2025

The Role Of Fruiting Bodies In Protecting A Community Of Myxococcus Xanthus Spores From The Environment, Dave Lall

Wayne State University Dissertations

Environmental microorganisms have evolved a variety of strategies to survive fluctuations in environmental conditions, including the production of biofilms and differentiation into spores. Myxococcus xanthus are ubiquitous soil bacteria that produce starvation-induced multicellular fruiting bodies filled with environmentally resistant spores (a specialized biofilm). Isolated spores have been shown to be more resistant than vegetative cells to heat, ultraviolet radiation, and desiccation. The evolutionary advantage of producing spores inside of fruiting bodies is not clear. Here, we examine a hypothesis that the fruiting body provides additional protection from environmental insults. We developed a high-throughput method to compare the recovery (outgrowth) of …

Environmental Pollution And Early-Life Metabolic Programming, Lisa Koshko Jan 2025

Environmental Pollution And Early-Life Metabolic Programming, Lisa Koshko

Wayne State University Dissertations

Substantial evidence demonstrates that parental pollutant exposures can induce harmful health effects in the offspring, including the predisposition to Type II Diabetes. As rates of Type II Diabetes continue to rise in younger populations, a crucial gap lies in understanding the role of parental influence in the etiology of later-life metabolic disease. Therefore, the central goal of this work was to determine how early life exposure to smoking level benzene, an endocrine-disrupting air pollutant, affects the metabolic health of offspring, and to identify potential developmental mechanisms involved in disease susceptibility. This goal was achieved using two models of early life …

Legal Barriers To Maternal Healthcare Equity, Ashley Keith Jan 2025

Legal Barriers To Maternal Healthcare Equity, Ashley Keith

Akron Law Faculty Publications

In examining contemporary maternal healthcare inequity in clinical care delivery, we must study the critical role played by the legal institutions in creating and perpetuating this problem. Comprehensive systems within the United States require legal advocacy and judicial enforcement to take shape and maintain their structural integrity. Healthcare in this country is no different. Legal institutions helped form and further, both intentionally and inadvertently, the racial disparities that we currently battle. Legal history plays an integral role in understanding why racial disparities in maternal mortality persist. One way for legal professionals to contribute to the ongoing efforts to achieve maternal …

Department of Biomedical and Translational Sciences Faculty Publications

Troponin C (TnC) is the Ca²⁺-sensing subunit of troponin that is responsible for activating thin filaments in striated muscle, and, in turn, for regulating the systolic and diastolic contractile function of cardiac muscle. The secondary structure of vertebrate TnC is mainly composed of α-helices, with nine helices named sequentially, starting from the amino terminus, from N to A–H. The N-helix is a 12-residue-long α-helix located at the extreme amino terminus of the protein and is the only helical structure that does not participate in forming Ca²⁺-binding EF-hands. Evolutionarily, the N-helix is found only in TnC from mammalian species and most …

[Summary] Cardiomyocytes endure physical stress from the myocardium environment while generating their own mechanical strains. The force generated by sarcomeres is transmitted both longitudinally to adjacent sarcomeres and laterally to the cytoskeleton via intermediate filaments. This mechanical stimulus impacts other organelles, including the nucleus, thus playing a vital role in sensing and signaling nuclear adaptations. However, there is limited understanding of how changes in cardiac contractility affect nuclear mechanics. Here, we sought to investigate the effects of hyper- and hypo-contractility in nuclei of human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) bearing TNNT2 pathogenic variants associated with hypertrophic (HCM) or dilated …

The Hidden Cost Of Venture Capital, Emilie Aguirre Jan 2025

The Hidden Cost Of Venture Capital, Emilie Aguirre

Faculty Scholarship

Founders, employees, consumers, and even funders increasingly expect businesses to pursue social goals alongside financial performance. Yet even the most committed firms have found it difficult to maintain social performance over time. Scholars in economics, management, and law have put forth several explanations for this “mission drift,” including inappropriate governance, poor management, lack of genuine commitment, and threat of takeovers. Puzzlingly, research to date primarily focuses on later-stage firms, even though the events and decisions that take place in a firm’s early stages can critically impact retention of its social performance.

Drawing from over five years of qualitative field research …
