Acinetobacter baumannii is an opportunistic pathogen of growing concern, as isolates are commonly multidrug resistant. While A. baumannii is most frequently associated with pulmonary infections, a significant proportion of clinical isolates come from urinary sources, highlighting its uropathogenic potential. The type II secretion system (T2SS) of commonly used model Acinetobacter strains is important for virulence in various animal models, but the potential role of the T2SS in urinary tract infection (UTI) remains unknown. Here, we used a catheter-associated UTI (CAUTI) model to demonstrate that a modern urinary isolate, UPAB1, requires the T2SS for full virulence. A proteomic screen to identify …

Nanodiamonds are at the heart of a plethora of emerging applications in areas ranging from nanocomposites and tribology to nanomedicine and quantum sensing. The development of alternative synthesis methods, a better understanding, and the availability of ultrasmall nanodiamonds of less than 3 nm size with a precisely engineered composition, including the particle surface and atomic defects in the diamond crystal lattice, would mark a leap forward for many existing and future applications. Yet today, we are unable to accurately control nanodiamond composition at the atomic scale, nor can we reliably create and isolate particles in this size range. In this …

Impacts Of Population Level Environmental Contaminants, Sex Hormones, And Fibroblast Cell Subsets On Systemic Sclerosis (Ssc), Noelle N. Kosarek
Dartmouth College Ph.D Dissertations
Systemic sclerosis (SSc) is a rare autoimmune disease characterized by fibrosis of the skin and internal organs, vascular abnormalities, and autoantibody formation. The etiology of SSc is unknown, though the disease is thought to arise in genetically predisposed individuals after exposure to an environmental factor. There are few FDA approved disease modifying medications available to treat SSc.
The express aims of this dissertation are tripart. First, we aimed to validate a 3D tissue model known as the self-assembled skin equivalent (saSE) model. Second, we sought to describe the geographic distribution of SSc in a US Medicare population to better understand …

Technology Use By Registered Dietitians For Patient Care In An Outpatient Setting, Donika Emerllahu
Theses
Background Technology use in digital health tools has increased significantly since the COVID-19 pandemic disrupted many in-office patient care services. Although healthcare practices have adopted several technologies, they remain surrounded by a saturated increase in consumer-based wearables and smartphone applications. As a result, attitudes and acceptability toward integrating these technologies for patient care are evolving. Digital health tools is an umbrella term including several technologies utilized among health care providers for the purpose of patient care. The inclusion of digital health tools by Registered Dietitian Nutritionists’ (RDNs) in practice provides a more robust and personalized approach to patients and their …

Neuroblastoma (NB) is a childhood cancer arising from sympatho-adrenal neural crest cells. MYCN amplification is found in half of high-risk NB patients; however, no available therapies directly target MYCN. Using multi-dimensional metabolic profiling in MYCN expression systems and primary patient tumors, we comprehensively characterized the metabolic landscape driven by MYCN in NB. MYCN amplification leads to glycerolipid accumulation by promoting fatty acid (FA) uptake and biosynthesis. We found that cells expressing amplified MYCN depend highly on FA uptake for survival. Mechanistically, MYCN directly upregulates FA transport protein 2 (FATP2), encoded by SLC27A2. Genetic depletion of SLC27A2 impairs NB survival, and …

BACKGROUND: Lymphoid neoplasms, including multiple myeloma (MM), non-Hodgkin lymphoma (NHL), and NK/T cell neoplasms, are a major cause of blood cancer morbidity and mortality. CD38 (cyclic ADP ribose hydrolase) is a transmembrane glycoprotein expressed on the surface of plasma cells and MM cells. The high expression of CD38 across MM and other lymphoid malignancies and its restricted expression in normal tissues make CD38 an attractive target for immunotherapy. CD38-targeting antibodies, like daratumumab, have been approved for the treatment of MM and tested against lymphoma and leukemia in multiple clinical trials.
METHODS: We generated chimeric antigen receptor (CAR) T cells targeting …

Human metapneumovirus (HMPV) inclusion bodies (IBs) are dynamic structures required for efficient viral replication and transcription. The minimum components needed to form IB-like structures in cells are the nucleoprotein (N) and the tetrameric phosphoprotein (P). HMPV P binds to the following two versions of the N protein in infected cells: N-terminal P residues interact with monomeric N (N

Detecting ocular conditions and initiating treatment at the earliest is essential for preserving the remaining visual health in various ocular diseases such as glaucoma, diabetic retinopathy, and inner retinal diseases such as retinoblastoma. Ocular treatment strategies include topical administration of drugs (e.g., Timolol for glaucoma), ocular surgeries (e.g., trabeculoplasty), gene therapy (e.g., for treating age related macular degeneration), light-based (e.g., laser) therapies and direct injection (e.g., intravitreal injections) of drugs depending on the target site of the ocular therapy. Topical administration of drugs to the anterior segment and photothermal therapy of the posterior segment of the eye are among the …

With the rise of deep neural networks, the challenge of explaining the predictions of these networks has become increasingly recognized. While many methods for explaining the decisions of deep neural networks exist, there is currently no consensus on how to evaluate them. On the other hand, robustness is a popular topic for deep learning research; however, it is hardly talked about in explainability until very recently. In this tutorial paper, we start by presenting gradient-based interpretability methods. These techniques use gradient signals to assign the burden of the decision on the input features. Later, we discuss how gradient-based methods can …

The main protease, Mpro, of SARS-CoV-2 is required to cleave the viral polyprotein into precise functional units for virus replication and pathogenesis. Here, we report quantitative reporters for Mpro function in living cells in which protease inhibition by genetic or chemical methods results in robust signal readouts by fluorescence (enhanced green fluorescent protein [eGFP]) or bioluminescence (firefly luciferase). These gain-of-signal systems are scalable to high-throughput platforms for quantitative discrimination between Mpro mutants and/or inhibitor potencies as evidenced by validation of several reported inhibitors. Additional utility is shown by single Mpro amino acid variants and structural information combining to demonstrate that …

Investigating The Role Of Amyloid Beta Peptides In Inflammation And Alzheimer’S Disease Pathogenesis, Nguyen Le
Theses and Dissertations
Alzheimer’s Disease (AD) is a neurodegenerative disease characterized by severe memory decline and cognitive impairments. In AD patients’ brains, aggregates of amyloid beta (Aβ) peptides, called amyloid plaques, are hypothesized to trigger innate immune responses that contribute to AD pathogenesis. Interestingly, recent studies demonstrated that an increased level of Aβ protected the host against pathogen infections. Using Drosophila as a model organism, our preliminary data showed that the loss of the Drosophila APP homolog, APPL, led to immune deficits against parasite infection, and that overexpression of Drosophila Aβ produced an inflammatory phenotype. These findings suggested that Aβ might be required …

Nonsmall-cell lung cancer (NSCLC) is the most common type of lung cancer, with a dismal prognosis. NSCLC is a highly vascularized tumor, and chemotherapy is often hampered by the development of angiogenesis. Therefore, suppression of angiogenesis is considered a potential treatment approach. Tannic acid (TA), a natural polyphenol, has been demonstrated to have anticancer properties in a variety of cancers; however, its angiogenic properties have yet to be studied. Hence, in the current study, we investigated the antiproliferative and antiangiogenic effects of TA on NSCLC cells. The (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) (MTS) assay revealed that TA induced a dose- and time-dependent …

Epigenetic abnormalities in DNA hydroxymethylation (5hmC) have been detected in patients with myeloid neoplasms, suggesting that 5hmC might act as a valuable epigenetic mark to reflect the disease status of myeloid neoplasms. Here, we report systematic genome-wide mapping of the DNA hydroxymethylomes in over 70 patients with myeloid neoplasms. Our integrative analysis leads to the identification of distinct 5hmC signatures that can sensitively discriminate patients from healthy individuals. At the molecular level, we unveiled dynamic 5hmC changes within key transcription factor (e.g., the CEBP family) binding motifs that are essential for hematopoiesis and myeloid lineage specification. 5hmC redistribution was found …

A vehicle (e.g., automobile, motorcycle, a bus, a recreational vehicle (RV), a semi-trailer truck, a tractor or other type of farm equipment, a train, a plane, a helicopter, etc.) often has proximity sensors to detect adjacent objects. A head unit of the vehicle may use sensor data such as proximity sensor data to detect an empty parking space. The head unit may then report the detected empty parking space to a cloud computing system, such as a mapping service. Data from multiple vehicles may be used to crowd source a relatively detailed understanding of the amount of empty parking spaces …
