Do Auditors Respond To Changes In Clients’ Analyst Coverage? Evidence From A Natural Experiment, Mohammad Alkhamees
Do Auditors Respond To Changes In Clients’ Analyst Coverage? Evidence From A Natural Experiment, Mohammad Alkhamees
USF Tampa Graduate Theses and Dissertations
Prior research has established that analysts serve as an important monitor of managementand improve the quality of firms’ financial reporting. In this study, I utilize the natural experiment resulting from the mergers and closures of brokerage houses, to examine whether this exogenous change in firms’ information environment impacts the audit process. Specifically, I examine whether reductions in analyst coverage have an effect on auditor effort proxied by audit fees and audit report lag, as well as financial reporting quality, measured by future restatements. I find that client firms pay significantly higher audit fees following an exogenous reduction in analyst coverage …

Exertional heat illness (EHI) and malignant hyperthermia (MH) are life threatening conditions associated with muscle breakdown in the setting of triggering factors including volatile anesthetics, exercise, and high environmental temperature. To identify new genetic variants that predispose to EHI and/or MH, we performed genomic sequencing on a cohort with EHI/MH and/or abnormal caffeine-halothane contracture test. In five individuals, we identified rare, pathogenic heterozygous variants in ASPH, a gene encoding junctin, a regulator of excitation-contraction coupling. We validated the pathogenicity of these variants using orthogonal pre-clinical models, CRISPR-edited C2C12 myotubes and transgenic zebrafish. In total, we demonstrate that ASPH variants represent …

John Wick: Keanu Reeves’S Epic Adventure, Ann C. Hall
John Wick: Keanu Reeves’S Epic Adventure, Ann C. Hall
Heroism Science
Three films create the John Wick universe and franchise: John Wick (2014), John Wick: Chapter 2 (2017), and John Wick: Chapter 3, Parabellum (2019). A fourth film is scheduled to be released in March 2023. All are wildly popular, and all are criticized for violence, particularly gun violence. I argue, however, that by examining the visual references that appear in all the films, it becomes clear that the films are defending themselves from such attacks through their allusions to ancient and classical epics from around the world. As Wick battles his way through museums and beautiful cities, the film reminds …

The Extinction Race: Techniques Of The Human In Proust, Via Houellebecq, James Dutton
CLCWeb: Comparative Literature and Culture
In his article, “The Extinction Race: Techniques of the Human in Proust, via Houellebecq” James Dutton “reads” identity and race from the point of view of technics. Namely, he does so through the work of two nominally “Eurocentric” authors, Marcel Proust and Michel Houellebecq, observing how familial and racial resemblance is a living inscription of “lost time.” This inscription comes about through the technical means available to and constitutive of the categories which bind them. Thus, instead of furthering unfinishable racial distinctions which only serve to support discourses of racism, this article follows assertions made in the novels of …
Poetics Of The Crystal-Image: Dreams In Mirror And Ashes Of Time Redux, Yuh-Yi Tan
Poetics Of The Crystal-Image: Dreams In Mirror And Ashes Of Time Redux, Yuh-Yi Tan
CLCWeb: Comparative Literature and Culture
This paper delves into recurrent dreams and related recollections in Andrei Tarkovsky’s Mirror and Wong Kar-wai’s Ashes of Time Redux. Gilles Deleuze has called Mirror a film of “turning crystal” because it reflects the two sides of the protagonist’s relationship with his mother and ex-wife by juxtaposing them in a mysterious labyrinth of past-present-future. The crystal imaging refracts four sides of two couples by showing the bonding of the parents along with the connection to his ex-wife and son. The same structure is apparent in Ashes of Time Redux where another set of two bilateral symmetries is initiated between …
Cooling Down Transmedia Storytelling, Jan Baetens, Domingo Sánchez-Mesa
Cooling Down Transmedia Storytelling, Jan Baetens, Domingo Sánchez-Mesa
CLCWeb: Comparative Literature and Culture
In this article we propose a reading of “Dead End Street”, one of the most successful songs of one of the most popular British pop groups of the 60s, The Kinks. However, we will not discuss the song as such, but its remediation as a music video (a practice that did not have to wait for MTV to make its appearance in mass media culture). The analysis will briefly contextualize the group, the song and the clip, but its major objective is to use the Kinks example to open a new question in the larger debate on intermediality and transmediality …
Precarity In The Times Of Partition: Personal Vs Communal Love In Khushwant Singh’S Train To Pakistan And Saadat Hasan Manto’S “Gurmukh Singh Ki Wasiyat”, Ayesha Perveen
CLCWeb: Comparative Literature and Culture
The paper studies how various shades of love respond to precarity in anarchic times by comparing the narrative representation of the aftermath of the Partition of the British colonized Subcontinent into independent countries of India and Pakistan in 1947 with particular focus on Sikh-Muslim relationships in Punjab as presented in Khushwant Singh’s novel Train to Pakistan and Saadat Hasan Manto’s short story “Gurmukh Singh ki Wasiyat.” Employing Judith Butler’s concept of precarity, the paper analyzes how both the writers sketch precarity in partition times ensuing in post-Partition communal violence and effacement of love. The selection of the texts is significant …

Vaccine Technology VIII
Development of effective vaccines against emerging and re-emerging pathogens is a worldwide public health priority. In response to the current pandemic, global vaccine manufacturers have developed several types of SARS-CoV-2 vaccines, including adenovirus (AdV)-vector based vaccines. Production of viral vector vaccines requires optimization of several cell culture parameters. Using wildtype AdV type 5 infection with suspension HEK293 cells as a model system, we evaluated various conditions (including time of harvest, multiplicity of infection (MOI), cell density, insulin supplementation, and temperature shift) to optimize adenovirus production. In addition, we evaluated cell growth and AdV5 production in different chemically defined media and …

Chicken embryo fibroblasts (CEF) and diploid cells have a long history in vaccine production since their isolation in the 1960s at the Wistar Institute (WI-38 cells) as well as the Medical Research Council (MRC-5 cells). The cells quickly became adopted for a number of vaccines: varicella zoster (VZV), MMR, yellow fever, polio, hepatitis A, rotavirus, rabies, Marek's disease, and dengue virus. Most of these vaccine processes were developed with classical media supplemented with Fetal Bovine Serum (FBS). The Hayflick limit of diploid cells restricted their adaptation to a serum-free process. While some of the vaccines such as polio and rabies …

Influenza virus infects millions of people every year worldwide, with elderly and very young people among the most critically affected. Strains that constitute a pandemic threat are characterized by the severity of the clinical manifestations and mortality rates and tend to require the urgent production of hundreds of millions of vaccine doses in very short periods of time. There is an evident need to develop new generations of influenza vaccines based on robust production systems such as mammalian or insect cell cultures. These systems may allow, in contrast to production in embryonated chicken eggs, a faster response capacity, a superior …

Porcine circovirus associated disease (PCVD) is responsible for a significant economic loss in the livestock industry. A vaccine based on a viral antigen has been developed to prevent this disease, and it is produced industrially in the pig’s kidney cell line PK15. However, both biomass and viral antigen production are limited in the current attachment culture bioprocess set-up.

Vaccines for known and emerging diseases as well as new therapeutic applications of viruses will require multiple doses and high virus input per dose or treatment. Therefore, there is a clear need for innovation in vaccine manufacturing. Yet, process optimization and intensification are cost- and labor-intensive challenges and thus not always considered as economically reasonable. To tackle this problem and compete against outdated cost calculations, this work aims to transfer knowledge and techniques that were acquired within a decade of protein production process development into the field of viral vaccine manufacturing. This comprises the development of monoclonal suspension MDCK cell …

Vaccine Technology VIII
Within 2.5 years since SARS-COV-2 emergence, COVID-19 pandemic has caused more than 6.3 million registered deaths and 530 million registered cases. The quick development of safe and effective vaccines was very important to mitigate the sanitary crisis. However, the continuous emergence of virus variants with increasing transmissibility continues to cause periodic outbreaks worldwide. The original vaccines keep protecting from severe disease and death caused by variants, but not from transmission and mild disease. Thus, new and improved vaccines are necessary, and updatable, multivalent pan-variant vaccines might be one way to control SARS-COV-2.
The spike (S) protein of SARS-COV-2, a highly …

Intensification Of Influenza Virus Production In Fed-Batch And Perfusion Cultures Of Hek293sf Cells, Cristina A.T. Silva, Amine A. Kamen, Olivier Henry
Vaccine Technology VIII
More than half a million people die every year from complications of seasonal influenza, and vaccination stands as the most effective method to prevent and limit outbreaks of the disease. Constant vaccine development based on emerging strains and worldwide distribution of vaccines is a great challenge for public health and vaccine manufacturers, particularly in a potential pandemic scenario. The limited flexibility of the current egg-based production system combined with recent advances in large-scale cell culture techniques have encouraged the development of cell culture processes for influenza vaccine production. While cell culture offers a valuable alternative, productivities are still low when …

Development Of An Automated Microscale Platform For Conjugate Vaccine Production In E. Coli, Jasmin J. Samaras, Martina Micheletti
Vaccine Technology VIII
The current state-of-the-art for the production of glycoconjugate vaccines is a lengthy and complex series of process steps. An exciting alternative is Protein Glycan Coupling Technology (PGCT), where glycoconjugates are produced in vivo 1,2. This offers a significant reduction in the time and cost burden of glycoconjugate production, making this technology much more feasible for providing vaccines in developing countries. Current research seeks to apply this technology for pneumococcal vaccines 2, however much process optimisation must be done before this is achievable. This work seeks to provide an automated screening platform at the microscale, enabling the rapid screening …

Suspension Vero Cell Culture Technology For High Titer Production Of Viral Vaccines, Chun Fang Shen, Rénald Gilbert, Elodie Burney, Seyyed Mehdy Elahi, Stephane Lanthier, Amine Kamen, Martin Loignon
Vaccine Technology VIII
Vero cells are considered as the most widely accepted continuous cell line by the regulatory authorities (such as WHO) for the manufacture of viral vaccines for human use. The continuous Vero cell line has been commercially used, after propagation on microcarriers, for the production of rabies, polio, enterovirus 71, hantaan, more recent COVID19 and other vaccines. Vero cell culture technologies were also explored for productions of many more viral vaccines over the last two decades. The growth of Vero cells is anchorage-dependent, and cells need to be dissociated enzymatically or mechanically for the process of subcultivation. This process is labor …

Newcastle disease (ND) is a highly contagious and often severe global spread syndrome that affects birds including domestic poultry. It is caused by a virus belonging to the paramyxoviridae family.
Indeed, an outbreak of ND can be quite severe, and is considered as a constant threat to the industry and food security worldwide. The disease can be controlled through the administration of effective vaccines.
Immunizations with inactivated or live vaccines, although protective, have some eminent disadvantages.
The aims of this work is the development of a vectored vaccine using a non-replicative human adenovirus vector, expressing the F antigen from Newcastle …

HIV-1-based virus-like particles (VLPs) have high potential as scaffold for the development of chimeric or multivalent vaccines by functionalizing them with specific antigens [1], [2]. The obtention of vaccine formulations independent of cold chain is desirable to facilitate transportation and administration worldwide [3]. Recently, efforts are being made to develop cost-effective scalable processes to obtain these particles. The present study aims to compare some of the most used downstream processing (DSP) technologies for capture and purification of VLPs, taking especial consideration in using technologies enabling operation at large scale [4]. First, suspension adapted HEK 293 cells cultured in chemically defined …

Vero cells are nowadays widely used in the production of vaccines. However, they are anchorage dependent, which greatly complicates upstream processing and process scale-up. Moreover, there is a recognized need to reduce the costs of vaccine manufacturing to develop vaccines affordable worldwide. The use of cell lines adapted to suspension growth contributes to reach this objective. We had previously adapted Vero cells to suspension growth for rabies virus (strain LP2061) production in different serum free media (Rourou et al. 2019).
In this work, we describe the use of a chemically defined medium (CDM) provided by Xell AG to assess the …

Metabolic extremes provide opportunities to understand enzymatic and metabolic plasticity and biotechnological tools for novel biomaterial production. We discovered that seed oils of many Thunbergia species contain up to 92% of the unusual monounsaturated petroselinic acid (18:1Δ6), one of the highest reported levels for a single fatty acid in plants. Supporting the biosynthetic origin of petroselinic acid, we identified a Δ6-stearoyl-acyl carrier protein (18:0-ACP) desaturase from Thunbergia laurifolia, closely related to a previously identified Δ6-palmitoyl-ACP desaturase that produces sapienic acid (16:1Δ6)- rich oils in Thunbergia alata seeds. Guided by a T. laurifolia desaturase crystal structure obtained in this study, …

Hand, Foot and Mouth Disease (HFMD) is an endemic childhood disease in Southeast Asia, with substantial disease burden affecting millions of children each year. Occasionally the central nervous system is involved causing serious and sometimes fatal neurological complications. HFMD outbreaks are also observed outside the Asia-Pacific countries. HFMD can be caused by multiple enteroviruses of which the best known virus is EVA71. However, also other enteroviruses such as CVA6, CVA10 and CVA16 can cause the disease. Inactivated EVA71 vaccines are registered in China, but in order to prevent all HFMD cases, multivalent vaccines are warranted. Intravacc is developing an HFMD …

Lgbtq+ Congregants Navigating Identity In The Context Of "Welcoming But Not Affirming" Evangelical, Pentecostal, And Non-Denominational Religious Institutions: A Queer Narrative Analysis, Sarah E. Rasmussen
University Honors Theses
Welcoming but not affirming Evangelical, Pentecostal, and non-denominational churches invite LGBTQ+ people to attend their church, but do not affirm their identity as a gender and/or sexual minority.
