Investigating Factors That Impact Income Generation And Distribution In Western National Park Gateway Communities, Elizabeth Depew Aug 2022

Investigating Factors That Impact Income Generation And Distribution In Western National Park Gateway Communities, Elizabeth Depew

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Many rural towns in the western united states have come to be defined by high levels of tourism, seasonal residents, and migration driven by the desire to be close to natural amenities and the higher quality of life they afford. This shift is especially apparent in communities that are “gateways” to natural amenities such as national parks. Embracing this shift towards the “New West,” many community planners have heralded amenity-led development as a remedy to the waning feasibility of relying on agriculture and extractive industries. However, anecdotal evidence and several case studies indicate the factors which make these gateway communities …

Lentiviral Vector Production At High Cell Density By Transient Transfection Of Suspended Culture Hek Cells, Jacob G. Accordino Aug 2022

Lentiviral Vector Production At High Cell Density By Transient Transfection Of Suspended Culture Hek Cells, Jacob G. Accordino

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Viral vectors are gene carriers that efficiently deliver therapeutic gene constructs to target cells. Viral vectors are frequently produced in suspension-growing cells of mammalian origin, referred to as packaging cells. Lentiviral vectors have become widely used as gene vectors since they were initially developed two and a half decades ago, particularly in research settings. The advantages of lentiviral vectors for treating diseases of genetic origin have driven research into their large-scale manufacture for clinical settings. Currently, three main challenges exist that limit mass production of lentiviral vectors. First, plasmid DNA used in the production process is very expensive. Second, high …

Analysis of the genes retained in the minimized Mycoplasma JCVI-Syn3A genome established that systems that repair or preempt metabolite damage are essential to life. Several genes known to have such functions were identified and experimentally validated, including 5-formyltetrahydrofolate cycloligase, coenzyme A (CoA) disulfide reductase, and certain hydrolases. Furthermore, we discovered that an enigmatic YqeK hydrolase domain fused to NadD has a novel proofreading function in NAD synthesis and could double as a MutT-like sanitizing enzyme for the nucleotide pool. Finally, we combined metabolomics and cheminformatics approaches to extend the core metabolic map of JCVI-Syn3A to include promiscuous enzymatic reactions and …

Reversing The Gaze: Using Indigenous And Western Worldviews To Compare Coverage Of Climate Change And Indigenous Peoples In The News Media, Gina Mccrackin Aug 2022

Reversing The Gaze: Using Indigenous And Western Worldviews To Compare Coverage Of Climate Change And Indigenous Peoples In The News Media, Gina Mccrackin

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

The news media acts as an important conduit for shaping societal views of the socio-politics of climate change. While climate change will indeed affect everyone, it will not affect everyone equally. Indigenous peoples are among the populations whose well-being is threatened the most by climate change. International scholarship finds it is not uncommon for Indigenous cultures, communities, and perspectives to be underrepresented and misrepresented in Western climate change media. Research also indicates that fair Indigenous representation occurs when Indigenous peoples are the authors of news articles themselves. We evaluated the differences in discussions of climate change and environmental issues …

Barriers And Opportunities To Sustainable Urban Agriculture: The Case Of Louisville, Kentucky., Sait Sarr Aug 2022

Barriers And Opportunities To Sustainable Urban Agriculture: The Case Of Louisville, Kentucky., Sait Sarr

Electronic Theses and Dissertations

As the urban populations continue to increase, food insecurity will continue to affect urban residents as well, depriving them of access to abundant and nutritious food. This has captured the attention of many policymakers, urban planners, nonprofits, grassroots movements, and other stakeholders. As a result, there is growing interest in developing new sustainable strategies and policy measures that improve food security through the promotion of sustainable urban agriculture. More than 100,000 Louisville residents, of which 20% are from West Louisville, do not have access to adequate and nutritious foods, and are at a higher risk of developing health issues. West …

The A-Word: Destigmatizing Abortion In American Culture., Kathryn Lafferty Danner Aug 2022

The A-Word: Destigmatizing Abortion In American Culture., Kathryn Lafferty Danner

Electronic Theses and Dissertations

This dissertation uses an interdisciplinary perspective and explores the topic of abortion through legal, medical, philosophical, and cultural perspectives, providing alternative ways to talk about abortion. In some states, especially since the overturning of Roe, abortion has become almost impossible to access, particularly in the South and Midwest. Abortion as a medical procedure is highly stigmatized and although it is a common procedure, it is taboo to discuss in American culture. Exploring how different media work to destigmatize abortion in the United States can lead to a deeper, more nuanced understanding of an overly politicized topic. The introduction is a …

The Evolution Of Aid Protection: A Future Informed By Afghanistan, Tabitha Babcock Aug 2022

The Evolution Of Aid Protection: A Future Informed By Afghanistan, Tabitha Babcock

Capstone Collection

The need for protection during complex crises is unremitting. Studies spanning the past decade prove humanitarian aid workers are at great risk while working within conflict zones and insecure contexts. This influx is due to the proliferation of politicization of humanitarianism, donor involvement, and lack of communication between involved parties. This exploratory study uses longitudinal data collection by way of semi-structured qualitative interviews. The research examined how humanitarian workers’ need for protection, while providing aid to Afghanistan, will change the future of humanitarian response.

It was concluded that although in the past decade, on-site actors felt they had enough physical …

‘Race’ And Academic Performance In International Higher Education: Black Africans In The U.K., Solomon Zewolde Aug 2022

‘Race’ And Academic Performance In International Higher Education: Black Africans In The U.K., Solomon Zewolde

Journal of Comparative & International Higher Education

U.K. higher education research routinely pinpoints the racialized nature of academic performance, but it often fails to even consider if racism contributes to such a pervasive racial or ethnic disparity. While research in the area often focuses on comparing the attainment of home white and ethnic minority students, little attention is given to the experiences of black African international students (BAIS), particularly in U.K. higher education. Using semi-structured qualitative interviews, this study documents how “race” shapes academic performance and achievement by exploring the experiences of 21 BAIS undergraduates studying in ten universities in England. Factors identified, inter alia, include racism …

The versatility of nuclear magnetic resonance (NMR) spectroscopy is apparent when presented with diverse applications to which it can contribute. Here, NMR is used i) as a screening/ validation tool for a drug discovery program targeting the Phosphatase of Regenerating Liver 3 (PRL3), ii) to characterize the conformational heterogeneity of p53 regulator, Murine Double Minute X (MDMX), and iii) to characterize the solution dynamics of guanosine monophosphate kinase (GMPK). Mounting evidence suggesting roles for PRL3 in oncogenesis and metastasis has catapulted it into prominence as a cancer drug target. Yet, despite significant efforts, there are no PRL3 small molecule inhibitors …

Targeting Pathological Changes In The Gut Protects The Cns From Neuropathology., Benjamin Tyler Charpentier Aug 2022

Targeting Pathological Changes In The Gut Protects The Cns From Neuropathology., Benjamin Tyler Charpentier

Electronic Theses and Dissertations

There is growing evidence that the health of the gut microbiome (GM) has an important role on multiple host organ systems. Ethanol-related neuropathologies and Alzheimer’s disease (AD) currently have no effective clinical treatment. Therefore, we developed a novel mouse model of fecal matter transplantation (FMT) utilizing the microbiome from alcoholic hepatitis (AH) patients, characterized by a loss of butyrogenic potential. Minimal neuroinflammation occurred in AH-FMT mice, but significant increases in the endoplasmic reticulum stress response (ERSR) across the majority of neurons in the prefrontal cortex and hippocampus were observed. In the cerebellum, Purkinje cells were specifically affected. Targeting the loss …

The Influence Of Sponsorship Engagement On Brand Loyalty: An Analysis Of The On-Site And Social Media Activational Communications., Achyut Kulkarni

Electronic Theses and Dissertations

The purpose of this study was to investigate the influence of sponsorship engagement (enacted via sponsorship activations) on consumers’ responses to sponsors’ activational communications. Specifically, this study aimed at examining if interaction and engagement with their team’s sponsors’ activational communication on-site, as well as on social media, influenced loyalty towards the sponsors. The study utilized service-dominant (S-D) logic as the theoretical framework. The S-D logic perspective recognizes that consumer behavior is centered on the interactive experiences between a consumer and an object, in this case the sponsor, and that a level of consumer interest and/or personal relevance with respect to …

Rewriting Writing As Transmodal And Translingual: Tranßcribing Japanese., Alex Way Aug 2022

Rewriting Writing As Transmodal And Translingual: Tranßcribing Japanese., Alex Way

Electronic Theses and Dissertations

This dissertation analyzes the use of different scripts in Japanese writing practices to disrupt English-language and Western-centric approaches to multimodal composition. Early chapters establish a brief history of the Japanese writing system (JWS) and explore its functionality. I trace the JWS’s development from borrowed Chinese characters (kanji), which were adapted to the Japanese language through the translation process of kundoku, to the contemporary system which utilizes the supplemental phonetic scripts of hiragana and katakana, in part, to represent Japanese syntax. Building on this historical context, I demonstrate how the use of the Japanese multi-scripts (hiragana, katakana, and kanji) in conventional …

Taiwanese Indigenous Representation, Rhetoric Of Resistance, And Heteroglossia In Warriors Of The Rainbow: Seediq Bale., John Yu-Choh Chang Aug 2022

Taiwanese Indigenous Representation, Rhetoric Of Resistance, And Heteroglossia In Warriors Of The Rainbow: Seediq Bale., John Yu-Choh Chang

Electronic Theses and Dissertations

This dissertation explores the relationship between Taiwanese indigenous narrative and rhetoric, in textual representations of the Seediq people and the 1930 Musha Incident. It explores how the forced colonization of Taiwanese indigenous people affected their identities and cultural representation, and how multi-voiced forms of narrative, storytelling, and meaning-making have rooted in indigenous oral traditions and rituals that counter colonial representations. Across a range of cultural texts, I identify what I call Taiwanese indigenous rhetoric of resistance (TIRR), drawing on Simon J. Ortiz’s theory of indigenous literature and oral traditions as indigenous-nationalist forms of cultural resistance. In addition, I draw on …

Characterization Of Microbotryum Lychnidis-Dioicae Secreted Effector Proteins That Manipulate Its Host Plant, Silene Latifolia., Ming-Chang Tsai Aug 2022

Characterization Of Microbotryum Lychnidis-Dioicae Secreted Effector Proteins That Manipulate Its Host Plant, Silene Latifolia., Ming-Chang Tsai

Electronic Theses and Dissertations

The smut fungal species Microbotryum lychnidis-dioicae is an obligate phytopathogen colonizing the plant host, Silene latifolia. A significant feature of M. lychnidis-dioicae infection is that the fungus can replace pollen on the anthers of susceptible host plants with fungal teliospores, thus earning the fungus the name: anther smut disease of flowers. The fungus synthesizes and secretes effector proteins into the cells of the plant host during infection, and the protein-protein interactions may interfere with and modify metabolism, plant development, and gene expression of the host to allow fungal colonization. Two potential fungal effector proteins, MVLG_06175 and MVLG_05122, were identified …

The Design Of Printed Fanfiction: A Case Study Of Down To Agincourt Fanbinding, Naomi Jacobs, Jsa Lowe Aug 2022

The Design Of Printed Fanfiction: A Case Study Of Down To Agincourt Fanbinding, Naomi Jacobs, Jsa Lowe

Proceedings from the Document Academy

In this paper, we examine the design process of fanbinding, through which physical, printed copies of fanfiction works are created and shared. These are often bespoke, samizdat, singular objects that cannot usually be mass-produced, and include unique hand-bound objects for the designer’s own affective, aesthetic pleasure. Fanbinding suggests durability and preservation, and printed objects can be transformative works in themselves: designed, typeset, and perhaps featuring artwork, maps or other specifically created front/back matter and illustrations. Readers of these born-digital works may produce these in reaction to the fact that texts are published purely in digital, intangible forms, finding themselves craving …
