Library Presentations
In this presentation, the author, the University of Kentucky university archivist, outlines a problem with acquiring currently-created university documents and offers some initial solutions. The problem is that key university records of historical and strategic importance are being distributed or published online and not routinely transferred to the archives the way they were in the past. Unless these documents are proactively acquired by the university archives, they are likely to be lost because of the ephemeral nature of the Web. Yet, crawling or otherwise capturing dynamic and changing web platforms adds technological complexity and thus requires additional resources. Given that …

DROSHA encodes a ribonuclease that is a subunit of the Microprocessor complex and is involved in the first step of microRNA (miRNA) biogenesis. To date, DROSHA has not yet been associated with a Mendelian disease. Here, we describe two individuals with profound intellectual disability, epilepsy, white matter atrophy, microcephaly and dysmorphic features, who carry damaging de novo heterozygous variants in DROSHA. DROSHA is constrained for missense variants and moderately intolerant to loss-of-function (o/e = 0.24). The loss of the fruit fly ortholog drosha causes developmental arrest and death in third instar larvae, a severe reduction in brain size and loss …

Kimmel Cancer Center Faculty Papers
BACKGROUND: Accrual to oncology clinical trials remains a challenge, particularly during the COVID-19 pandemic. For late phase clinical trials funded by the National Cancer Institute, the development of these research protocols is a resource-intensive process; however, mechanisms to optimize patient accrual after trial activation are underdeveloped across the National Clinical Trial Network (NCTN). Low patient accrual can lead to the premature closure of clinical trials and can ultimately delay the availability of new, potentially life-saving therapies in oncology.
OBJECTIVE: The purpose of this study is to formally create an easily implemented tool kit of resources for investigators of oncology clinical …

Department of Biochemistry and Molecular Biology Faculty Papers
Recent studies identified a missense mutation in the gene coding for G protein-coupled receptor kinase 6 (GRK6) that segregates with type 2 diabetes (T2D). To better understand how GRK6 might be involved in T2D, we used pharmacological inhibition and genetic knockdown in the mouse β-cell line, MIN6, to determine whether GRK6 regulates insulin dynamics. We show inhibition of GRK5 and GRK6 increased insulin secretion but reduced insulin processing while GRK6 knockdown revealed these same processing defects with reduced levels of cellular insulin. GRK6 knockdown cells also had attenuated insulin secretion but enhanced proinsulin secretion consistent with decreased processing. In support …

Copyright Enforcement In Content-Sharing Platforms, Kameshwari Shankar
Copyright Enforcement In Content-Sharing Platforms, Kameshwari Shankar
Publications and Research
This paper analyzes the choice between quality improvements and copyright litigation by a proprietary seller who faces a competitive threat from a content-sharing platform. The platform operates like a “public good” with contributors who share content and free-riders who only consume content while adding to congestion on the platform. When the proprietor can identify contributors in the platform with sufficient accuracy, a litigation strategy that targets contributors exacerbates free-riding behavior in the sharing platform and drives down platform quality. In contrast, investing in quality improvements for the copyrighted good does not affect contribution decisions on the platform, leading to a …

Why America’S Army Can’T Win America’S Wars, John A. Nagl
The US Army War College Quarterly: Parameters
Since achieving victory in World War II, the United States military has a less than enviable combat record in irregular warfare. Through a detailed historical analysis, this article provides perspective on where past decisions and doctrines have led to defeat and where they may have succeeded if given more time or executed differently. In doing so, it provides lessons for future Army engagements and argues that until America becomes proficient in irregular warfare, our enemies will continue to fight us at the lower levels of the spectrum of conflict, where they have a good chance of exhausting our will to …

Conference papers
Because of the thermoelectric (TE) effect (or Seebeck effect), a difference of potential is generated as a consequence of a temperature gradient across a sample. The TE effect has been mostly studied and engineered in semiconducting materials and it already finds several commercial applications. Only recently the TE effect in cement-based materials has been demonstrated and there is a growing interest in its potential. For instance, a temperature gradient across the external walls of a building can be used to generate electricity. By the inverse of the TE effect (or Peltier effect), one can also seek to control the indoor …
Arabidopsis Shoot Gravitropism Requires The Mediator Coactivator Complex, Gabriella Newton
Arabidopsis Shoot Gravitropism Requires The Mediator Coactivator Complex, Gabriella Newton
Honors Theses
Gravitropism is a plant response to gravity in directing the growth of its organs, such as the root and the stem. Gravity is sensed by statocytes, unique cells that contain starch-filled amyloplasts, which trigger gravitropic growth response by modulating the distribution of growth hormone auxin in the elongation zone of the stem and root. Despite the fact that multiple factors have been identified as key regulators of shoot negative gravitropism, how these factors are regulated at the transcriptional level is largely unknown. In this study, we explore the role of transcriptional machinery in regulating shoot gravity sensing and responses by …

Free Slave: Artist Statement, Thesis, And Working Novel, Leeland Francis Johnson
Masters Theses
This is a fictional slave narrative about a young man born free and taken into slavery. He wrestles with the concepts of forgiveness and mercy as he undergoes his trials and tribulations. As I am a white man taking on this endeavor, I wrestle with the artist's statement about how much care I have taken to make this a replica of a faithful slave narrative. I hope this work will be viewed as a tributary piece.
