Introduction/aims: Prior studies have emphasized the role of inflammation in the response to injury and muscle regeneration, but little emphasis has been placed on characterizing the relationship between innate inflammation, pain, and functional impairment. The aim of our study was to determine the contribution of innate immunity to prolonged pain following muscle contusion.

Methods: We developed a closed-impact mouse model of muscle contusion and a macrophage-targeted near-infrared fluorescent nanoemulsion. Closed-impact contusions were delivered to the lower left limb. Pain sensitivity, gait dysfunction, and inflammation were assessed in the days and weeks post-contusion. Macrophage accumulation was imaged in vivo by injecting …

The Ubiquitin Specific Peptidase 22 (USP22), a component of the Spt-Ada-Gcn5 Acetyltransferase (SAGA) histone modifying complex, is overexpressed in multiple human cancers, but how USP22 impacts tumorigenesis is not clear. We reported previously that Usp22 loss in mice impacts execution of several signaling pathways driven by growth factor receptors such as erythroblastic oncogene B b2 (ERBB2). To determine whether changes in USP22 expression affects ERBB2-driven tumorigenesis, we introduced conditional overexpression or deletion alleles of Usp22 into mice bearing the Mouse mammary tumor virus-Neu-Ires-Cre (MMTV-NIC) transgene, which drives both rat ERBB2/NEU expression and Cre recombinase activity from the MMTV promoter resulting …

AMG 256 is a bi-specific, heteroimmunoglobulin molecule with an anti-PD-1 antibody domain and a single IL-21 mutein domain on the C-terminus. Nonclinical studies in cynomolgus monkeys revealed that AMG 256 administration led to the development of immunogenicity-mediated responses and indicated that the IL-21 mutein domain of AMG 256 could enhance the anti-drug antibody response directed toward the monoclonal antibody domain. Anti-AMG 256 IgE were also observed in cynomolgus monkeys. A first-in-human (FIH) study in patients with advanced solid tumors was designed with these risks in mind. AMG 256 elicited ADA in 28 of 33 subjects (84.8%). However, ADA responses were …

Science can provide accurate information to society to inform decision-making and behavior. One contemporary topic in which the science is very clear, yet behavioral change has lagged, is climate change mitigation. Climate change scientists use evidence-based research to advocate to the public to adopt emission-reducing behaviors in various sectors such as transportation and food. However, scientists themselves often do not change their own behaviors according to the scientific consensus. We present a case study of a group of natural sciences PhD students, who, when presented with evidence and an opportunity for a behavioral change with implications for climate change mitigation, …

A Second Look At Active Shooter Events: Expanding Our Understanding Of Key Factors, Kyle Gamache, Matt Zaitchik, Judith Platania, Cecelia Coslovi, Willem Pontbriand

Arts & Sciences Faculty Publications

In the current study, we investigate active shooter events (ASEs) with emphasis on expanding our understanding of key factors involved in these events. The impetus for this study is a 2015 exploration (Gamache et al., 2015) that highlighted common perpetrator and event characteristics in ASEs occurring in the US prior to 2014. Since the 2015 study, mass-shooting events have moved from tragic, isolated incidents, to a phenomenon that is widely discussed in the media and highly politicized. Our goal was to investigate continued trends, if any exist, and to compare our current sample of ASEs to the sample utilized in …

Phosphorylation Regulates Viral Biomolecular Condensates To Promote Infectious Progeny Production, Nicholas Grams, Matthew Charman, Edwin Halko, Richard Lauman, Benjamin A Garcia, Matthew D Weitzman

2020-Current year OA Pubs

Biomolecular condensates (BMCs) play important roles in diverse biological processes. Many viruses form BMCs which have been implicated in various functions critical for the productive infection of host cells. The adenovirus L1-52/55 kilodalton protein (52K) was recently shown to form viral BMCs that coordinate viral genome packaging and capsid assembly. Although critical for packaging, we do not know how viral condensates are regulated during adenovirus infection. Here we show that phosphorylation of serine residues 28 and 75 within the N-terminal intrinsically disordered region of 52K modulates viral condensates in vitro and in cells, promoting liquid-like properties. Furthermore, we demonstrate that …

The double-stranded DNA (dsDNA) sensor STING has been increasingly implicated in responses to "sterile" endogenous threats and pathogens without nominal DNA or cyclic di-nucleotide stimuli. Previous work showed an endoplasmic reticulum (ER) stress response, known as the unfolded protein response (UPR), activates STING. Herein, we sought to determine if ER stress generated a STING ligand, and to identify the UPR pathways involved. Induction of IFN-β expression following stimulation with the UPR inducer thapsigargin (TPG) or oxygen glucose deprivation required both STING and the dsDNA-sensing cyclic GMP-AMP synthase (cGAS). Furthermore, TPG increased cytosolic mitochondrial DNA, and immunofluorescence visualized dsDNA punctae in …

Development And Initial Validation Of A Hospital Stress Questionnaire, Daniel M Ford, Rebecca Lawton, Elizabeth Travis, Elizabeth A Teale, Daryl B O'Connor

Faculty, Staff and Student Publications

BACKGROUND: Hospitalisation can be a traumatic experience, where inpatients are exposed to an abundance of physical and psychological stressors. Evidence suggests that these hospital-related stressors negatively impact health: a phenomenon known as post-hospital syndrome. The current study aimed to identify hospital-related stressors, and to develop and provide initial validation for a new measure of in-hospital stress.

METHODS: Measure development occurred in three stages: (i) semi-structured interviews, (ii) item generation, and (iii) pilot testing. Twenty-one patients were interviewed regarding their recent hospital experiences, and a list of hospital-related stressors was produced. These stressors were compiled into a questionnaire and piloted on …

Introduction: Nadofaragene firadenovec (Ad-IFNα/Syn3) is now approved for BCG-unresponsive bladder cancer (BLCA). IFNα is a pleiotropic cytokine that causes direct tumor cell killing via TRAIL-mediated apoptosis, angiogenesis inhibition, and activation of the innate and adaptive immune system. We established an immunocompetent murine BLCA model to study the effects of murine adenoviral IFNα (muAd-Ifnα) gene therapy on cancer cells and the tumor microenvironment using a novel murine equivalent of Nadofaragene firadenovec (muAd-Ifnα).

Methods: Tumors were induced by instilling MB49 cells into the bladders of mice; luciferase imaging confirmed tumor development. Mice were treated with adenovirus control (Ad-Ctrl; empty vector), or muAd-Ifnα …

An Empirical Study Of The Dmca's Anti-Circumvention Provisions, Clark Asay Jan 2024

An Empirical Study Of The Dmca's Anti-Circumvention Provisions, Clark Asay

Faculty Scholarship

The DMCA has been a flashpoint during most of its twenty-five-year existence. One of the most controversial parts of the DMCA is Section 1201. Among other things, Section 1201 prohibits third parties from circumventing certain controls to copyrighted content or trafficking in tools that enable circumvention of technological controls. However, despite its nearly quarter-of-a-century lifespan, we know very little about Section 1201 empirically. While certain aspects of the broader DMCA have received empirical assessments, Section 1201 has not. Our understanding of Section 1201 is largely based on anecdotal evidence, in the form of leading opinions from historically prominent copyright circuits. …

Extra-Legal Uses Of Tm, Clark Asay, Lareina Hingson, Stephanie Plamondon Jan 2024

Extra-Legal Uses Of Tm, Clark Asay, Lareina Hingson, Stephanie Plamondon

Faculty Scholarship

Theoretical accounts of trademark law suggest that trademarks serve multiple marketplace functions, including protecting consumers and producers from unfair dealing, reducing consumer search costs, and incentivizing higher quality goods and services. Producers of goods and services often use the ‘TM’ symbol to signal their assertion of legal rights in a mark, arguably helping advance these goals. But recently, an interesting phenomenon has arisen in which individuals not engaged in typical commercial activities have been using the TM signifier in ways that don’t necessarily correspond to the symbol’s legal function. These so-called “extra-legal” uses of the TM symbol can offer insights …

Prevalence Of Food Insecurity Among Caregivers Of Young Children During The Covid-19 Pandemic In Ireland, Elizabeth J. O'Sullivan, Caoimhe Daly, Aileen Kennedy Jan 2024

Prevalence Of Food Insecurity Among Caregivers Of Young Children During The Covid-19 Pandemic In Ireland, Elizabeth J. O'Sullivan, Caoimhe Daly, Aileen Kennedy

Articles

Our objectives were to explore the prevalence of food insecurity in primary caregivers of young children in Ireland and to compare two tools for measuring food insecurity during the COVID-19 crisis. A cross-sectional online survey was conducted among caregivers of children aged <2 years in Ireland in May/June 2020. Relevant survey questions were closed-ended using two established tools for measuring food poverty/insecurity; the Irish Food Poverty Indicator and the Food Insecurity Experience Scale, developed by the Food and Agriculture Organization. Descriptive statistics were used to calculate the prevalence of food poverty/insecurity. To explore agreement in the classification of food insecurity by the Food Poverty Indicator and the Food Insecurity Experience Scale, Cohen's κ was used. Analyses are based on 716 participants; most had a 3rd-level education and were married or in a partnership. Per the food poverty index, Ireland's national measure of food poverty, 3.9% (n = 28) of our sample were experiencing food poverty. This rose to 10.5% (n = 75) experiencing food insecurity when using the Food Insecurity Experience Scale, which also measures worry/anxiety around access to food. There was low agreement between the tools, with 11.3% of the sample classified as food secure by one tool and food insecure by the other. Our current measure of food poverty in Ireland may not be sufficient to describe the food-access struggles or worry/anxiety about food access, experienced by the population, particularly during an emergency like COVID-19.

Strain Engineering is a widely adopted approach to modulate the opto-electronic performance of 2-Dimensional (2D) materials. Recently, anisotropic Van der Waals (vdW) based 2D As2S3 monolayer has gained significant attention within the scientific community due to its stability in ambient conditions. Similar compounds like As2Te3 have also been theoretically explored. However, its indirect bandgap nature limits its application in optical devices. In this study, a systematic study of compressive and tensile strain on three profiles–Uniaxial along a-axis, Uniaxial along b-axis and biaxial strain from −10% to +10%, is performed for As2S3 …

College students experiencing psychological distress have significantly greater negative emotions than students who practice compassionate thinking. We have developed Eight Steps to Great Compassion (ESGC), an innovative brief and no-cost online video training program about how to increase compassion among busy and young adult university students. To examine the effectiveness and benefits of the ESGC, a single-group pre-test–post-test quantitative design with undergraduate university students (N = 92; Mage = 20.39) evaluated its effects. The results from the post-test showed that the ESGC had a significant positive impact on increased feelings of compassion towards oneself, compassion for others, and the …

Covid, Elite Rhetoric, And Americans’ Attitudes Toward U.S. Trade Practices With China, Dmitriy Nurullayev, Ping Xu, James Garand Jan 2024

Covid, Elite Rhetoric, And Americans’ Attitudes Toward U.S. Trade Practices With China, Dmitriy Nurullayev, Ping Xu, James Garand

Political Science Faculty Publications

Objectives: In this article, we consider how the COVID-19 pandemic and evaluations of Donald Trump combined to influence Americans' attitudes toward U.S. trade practices with China. We suggest that psychological factors—such as confirmation bias and transfer of grievances—influence how individuals evaluate the costs and benefits of trade. Specifically, we argue that the severity of the pandemic (reflected by both cumulative deaths and news coverage of the pandemic) and the rhetoric of (then) President Trump came together to influence Americans’ attitudes toward trade with China.

Methods: To assess the COVID effect, the Trump effect, and their interaction effect on …

Determining The Impact Of The U.S. Foreign Military Sales Program On The Proliferation Of Democracy, Matteo Garofalo Jan 2024

Determining The Impact Of The U.S. Foreign Military Sales Program On The Proliferation Of Democracy, Matteo Garofalo

Journal of Strategic Security

The Foreign Military Sales (FMS) program is one of two systems by which foreign nations can purchase U.S. military technologies, and it is additionally the largest component of U.S. foreign security cooperation assistance. As such, it functions as a key tool in the arsenal of U.S. foreign policy, and is therefore tasked with supporting foreign policy objectives, such as the promotion of democracy abroad. This study analyzes all nations to have received FMS sales (for which data has been made available) to determine if either the receipt of FMS contracts impacts their level of democracy, or, alternatively, if changes in …

Asymmetry In The Digital Age: Cyber Deterrence Strategies For Small States, Tony Yuan Li

Journal of Strategic Security

In an era where cyberwarfare poses an invisible but potent threat to national security, traditional deterrence theories fall short in providing nations with a viable strategy to defend themselves. This is especially the case for small states, where limited national resources and cyber capabilities prevent them from effectively deterring larger powers. This paper investigates whether small states can achieve cyber deterrence, and the means to do so. This research challenges the preconception that small states are at a disadvantage in cyber contestation, and instead proposes that their small size offers unique defensive benefits. A smaller attack surface, faster incident response …

The Security Dilemma And Conflict In Space: Impossible Or Inevitable?, Mackenzie Enholm

Journal of Strategic Security

Space has been identified as the next warfighting domain. With tensions simmering between the great spacefaring nations amid a space arms race, or at the very least a race for superior space technology, this article will examine such tensions through the lens of the security dilemma. Do the technology escalations between the great space powers and dual-use space technologies make space conflict inevitable? The United States, China, and Russia continue to develop advanced counterspace capabilities with the ability to target other space systems and satellites. Given the nature of states to maximize their own security amid uncertainty in space, physical …

Teacher Retention And Quality In The Four-Day School Week, Andrew Camp

Education Reform Faculty and Graduate Students Publications

The four-day school week is a school calendar that has become increasingly common following the COVID-19 pandemic. Proponents of the calendar often claim that offering teachers a regular 3-day weekend will help schools better retain existing teachers and recruit new teachers to their district without incurring additional costs due to higher salaries or other pecuniary benefits. However, there is scant empirical evidence assessing these claims. I use difference-in-differences and synthetic difference-in-differences models to assess the impact of four-day school week calendars on teacher retention and teacher quality in Arkansas. I find evidence that the calendar may help retain teachers who …

Innovation Misunderstood, Maurice E. Stucke, Ariel Ezrachi Jan 2024

Innovation Misunderstood, Maurice E. Stucke, Ariel Ezrachi

American University Law Review
