Azobenzene is a photoswitchable molecule known for its high conversion yield and repeatability. The inclusion of an azobenzene moiety with a known inhibitor of your target of interest allows for greater study of function as well as potential therapies. Three derivatives were designed from azobenzene and known inhibitor of EAATs, TBOA, to synthesize photoswitchable inhibitors of EAATs. Further investigation into the function of EAATs and development of a specific inhibitor for EAAT3, an important glutamate transporter in the brain required for proper function, can then be conducted. This dissertation contains two parts of study on the Friedan states and on the conformal bootstrap with symmetries.

The Friedan states are potential physical boundary states of the free boson. However, they exhibited a continuous spectrum in the open string sector, in contrast to more standard examples. The explicit expressions for the density of states of the Friedan states is obtained. Some pathologies and possible contradictions of these states are explored. The $g$ functions of the Friedan states are shown to be infinite, suggesting an infinite number of degrees of freedom in the theory.

Image manipulation detection (IMD) aims to determine whether an image has been tampered with and to identify the manipulated regions. These capabilities have become increasingly important with the rapid advancement of media editing and generation technologies, such as Photoshop and generative AI methods, which underscore the need for robust tools for media authentication. Although current state-of-the-art (SoTA) methods achieve strong results on common manipulation types, such as splicing, copy-move, and removal, they often struggle to generalize to manipulation types not represented in the training data. An eddy covariance (EC) flux system was integrated onto a Navy Oceanographic Meteorological Automatic Device (NOMAD) floating lidar buoy fitted for measuring the offshore wind resource. The EC flux system consisted of two sonic anemometers and an inertial measurement unit that together provided high-frequency (10 Hz) motion-corrected wind measurements. To assess possible impacts of flow distortion, EC momentum fluxes (10 min) were calculated in gravity-aligned, natural, and planar fit (PF) coordinate frames. Scholars, policymakers, changemakers, and social workers have attempted to define, understand, and eradicate poverty. Its endurance raises the prospect that these efforts may be incomplete or faulty. This thesis will probe this concern by examining poverty and poverty alleviation programs through the lens of a holistic intervention-based framework known as the Sustainable Livelihoods Framework (SLF). Variability in the high-latitude wintertime stratospheric flow, often referred to as the stratospheric polar vortex, can precede anomalous conditions in tropospheric flow on subseasonal-to-seasonal (S2S) timescales and impact surface weather. The range of stratospheric impacts on tropospheric conditions range from shifts in the North Atlantic storm tracks, formation of cold spells, and development of extreme precipitation. Impacts of stratospheric variability are known to persist for up to 60 days, providing ample windows of opportunity in forecasting on S2S timescales. Insulin has widespread actions within the brain, including energy homeostasis, modulation of feeding, and as a key component of memory processes. Further, brain insulin dysfunction correlates with Alzheimer's disease and other neurodegenerative disorders. To this point, measurement of hippocampal insulin has been limited. Latin America and the Caribbean (LAC) represent 8 percent of the global population but about one-third of global homicides, yet interpersonal violence in the region receives limited attention in the English-language literature. This dissertation contains three distinct studies of the causes and consequences of crime and violence in LAC. The first study examines the predictive effects of inflation on robbery, burglary, and homicide rates in Argentina, which possesses one of the highest robbery rates worldwide and has endured high inflation – and, at times, hyperinflation – for years. The purpose of this Dissertation was to establish the theoretical and practical limits of Kolmogorov-Zurbenko periodograms with dynamic smoothing in contrast to standard periodograms with static smoothing, along with a practical application of Kolmogorov-Zurbenko time series analysis methods. It was determined that Kolmogorov-Zurbenko periodograms with dynamic smoothing were superior to standard periodograms with static smoothing in estimating signal frequencies with respect to sensitivity, accuracy, and resolution, and that they are robust in the face of missing data. The University at Albany designed and manufactured 59 low-cost air quality sensor packages to continuously measure PM₂.₅, CO, O₃, NO₂, and NO at 38 New York State Mesonet (NYSM) sites located in the New York City Metropolitan Area. Prior to use for monitoring, low-cost sensors require calibration to correct for environmental sensitivities. Calibration models can be developed using data collected from co-location periods in which low-cost sensors are installed at sites with Federal Reference Methods and/or Federal Equivalent Methods instruments. MicroRNAs (miRNA) are a class of small non-coding RNAs that play a key role in gene silencing. miRNAs form an RNA-induced silencing complex (RISC) with the argonaute 2 (AGO2) protein in order to control gene expression through silencing messenger RNAs (mRNA). The ability for a miRNA-mRNA complex to fit into AGO2 and form the RISC is based on the location and characteristics of a central bulge, but the mechanism and interactions that lead to a functional RISC remain unknown. Accurate wind and gust forecasts are crucial in operational meteorology, yet there is a distinct gap in the literature regarding the verification of operational gust forecasts. In the present work, we assess the performance of the High-Resolution Rapid Refresh's (HRRR) gust potential, verifying it against the Automated Surface Observing System's (ASOS) observed gusts across the Continental United States (CONUS). The HRRR gust's diurnal cycle reveals a significant underprediction of the gust during daylight hours and a slight overprediction of the gust during hours of darkness. Conventional computed tomography produces a 2D slice or 3D image from multiple conventional images to avoid overlapping structures and increase visualization of the internal features of the object. A set up, macro controls and processing for computed tomography was created for the Center for X-ray Optics for the first time, and demonstrated in this work. Adding differential phase contrast to conventional x-ray images highlights edges. This qualitative case study examines how peer interactions among six seventh-grade English Learners (ELs) and non-ELs serve as both a context for and a method of developing adolescents' literacy and disciplinary literacy practices. Data sources include two semi-structured interviews with each student and their four classroom teachers, student and teacher ethnographic interviews, 300 hours of classroom observations (in English Language Arts, Social Studies, Math and Science, English as a New Language Tutorial), and student-generated artifacts (e.g., worksheets).

CRISPR-Cas12a is a rapidly evolving technology for improved gene editing in cells, as well as in vivo. The system, consisting of Cas12a nuclease and approximately 40-nt long guide RNA (gRNA), can efficiently recognize and cut genomic double-stranded DNA (dsDNA). Chemical modifications to the gRNA can potentially improve its dsDNA binding and enhance Cas12a-mediated nuclease activity. In this work, 5-methyl cytidine (m5C) and N6-methyl adenosine (m6A) were explored to improve the binding between gRNA and target DNA.
