Modification of antibodies to chemically couple labels or immobilization reagents is essential for developing biosensors. Typically, conjugation occurs through chemical methods that leverage reactive amines and thiols on native antibodies; however, this nonspecific approach can interfere with antibody function. Microbial transglutaminase (mTG) is an enzyme that has been used for site-specific conjugation of chemical modifiers to the Fc region of native antibodies, but thus far mTG-mediated conjugation has been limited to production of antibody-drug conjugates with human IgGs. Towards Novel Porphyrinoid Architectures: Synthesis Of Porphyrin Analogues With Two Six-Membered Exocyclic Rings, Amy L. Le, Timothy D. Lash

Tripyrranes with terminal ester protective groups were prepared by reacting 7-acetoxy-4,5,6,7-tetrahydroindoles with 3,4-diethylpyrrole in refluxing acetic acid-ethanol. Dibenzyl esters were deprotected by hydrogenolysis over 10% Pd/C and the resulting dicarboxylic acids were condensed with a series of dialdehydes in the presence of trifluoroacetic acid, followed by oxidation with FeCl3 or DDQ, to give porphyrinoid products with two six-membered exocyclic rings. Reactions with a pyrrole dialdehyde gave porphyrins in 27-30% yield and good yields were also obtained using 2,5-furandicarbaldehyde and 4-hydroxy-2,6-pyridinedicarbaldehyde to prepare oxaporphyrins and oxypyriporphyrins. However, poorer results were noted for thiaporphyrins, benzocarbaporphyrins and oxybenziporphyrins due to their poor …

Advances in detector physics, particularly in neutrino experiments and rare-event searches, have expanded our ability to probe phenomena that challenge how we describe the universe. People who inject drugs (PWID) account for the majority of hepatitis C virus (HCV) infections in the United States. The injection-equipment-sharing network likely plays an important role in shaping the dynamics of HCV transmission. Recognizing the emerging HCV epidemic in rural communities, we developed an agent-based network simulation model of HCV transmission via injection equipment sharing and used data on rural PWID networks to inform model parameterization and calibration. We then simulated an array of networks that varied key network properties to understand their impact on the magnitude and distribution of HCV incidence. Early-stage governance is often hindered by limited understanding of technological implications, rapid innovation cycles, and resistance from powerful industry actors who favor minimal oversight. Yet, timely and effective governance is essential, as new technologies are most malleable in their formative stages. Teachers who are culturally responsive use classroom materials that reflect their students' ethnicities, traditions, and values. Western teachers have access to diverse books and classroom tools. They have the agency to choose materials that support the curriculum. However, when Western educators work abroad, they must adapt their teaching methods and tools to local norms. Working abroad may require censoring or omitting information and content they believe in and support. As a result, teachers may feel dissonance or conflict when censoring. Teacher attrition, particularly among experienced educators, significantly impacts school stability and student outcomes. This quantitative study examined the relationship between mid-to-late-career elementary teachers’ perceptions of their principals’ servant leadership behaviors and their own retention decisions within Minnesota public schools. Grounded in servant leadership theory, the research also explored whether perceptions of leadership varied significantly based on teacher gender or teaching position. Utilizing a non-experimental, correlational design, data were collected via an online survey administering the Servant Leadership Scale and the Turnover Intention Scale (TIS-6). This quantitative study examined the relationship between mentoring and teacher professional development among public senior high school teachers in Ghana’s Central Region. Although mentoring is widely promoted as a strategy for strengthening instructional quality, limited empirical research has investigated how specific dimensions of mentoring influence in-service secondary teachers within Sub-Saharan African contexts. Grounded in social cognitive theory and adult learning theory, this study addressed that gap by differentiating among structural participation in mentoring, duration of mentoring involvement, and mentorship participation intensity. We find all non-negative integers n such that congruence modulo n is stable under exponentiation by positive integers, i.e. we show, that n = 1, 2, 6, 42, 1806 are the only numbers for which the condition a ≡n b and c ≡n d imply that a c ≡n b d , a, b, c, d ∈ N is satisfied. The goal of my project, Predicting the Paycheck: What Factors Truly Influence Graduate Earnings?, was to see if we could actually predict a college graduate's starting salary using data instead of just guessing. I used a dataset of 172 majors from the American Community Survey to look at how a student's field of study, the gender balance of their major, and the current job market all impact their first paycheck. I wanted to create a tool that helps students understand the financial reality of their degree before they even graduate.

Statically-annotated types have been shown to aid developers in a number of programming tasks, and this benefit holds true even when static type checking is not used. It is hypothesized that this is because developers use type annotations as in-code documentation. In this study, we aim to provide evidence that developers use type annotations as in-code documentation. Dimensionality reduction techniques are developed from the assumption that high-dimensional data often arises from low-dimensional structures embedded into the high-dimensional ambient space. Classical dimensionality reduction methods rely on the Euclidean distance, which may fail to capture the geometric structures of the datasets. This dissertation includes alternative metrics for dimensionality reduction techniques and challenges in applying these techniques.

Understanding the impact of specific biomedical interventions is crucial for optimizing HIV prevention strategies. In this study, we estimate of the prevalence of voluntary medical male circumcision (VMMC) and lay out a framework for estimating preexposure prophylaxis (PrEP) in the three high-burden sub-Saharan African countries of Malawi, South Africa, and Zimbabwe. For VMMC, we use a cohort-based simulation framework to model circumcision prevalence in all males over time, allocating VMMC count data to cohorts over time. Epidermal differentiation disorders (EDDs) are a heterogeneous group of inherited skin diseases characterized by impaired epidermal barrier function, scaling, and erythema. Although substantial progress has been made in identifying the genetic causes of EDD, long-standing clinical observations—including impaired growth, ectropion, and increased rates of preterm birth—remain poorly characterized across genotypes. The prevalence, severity, and clinical significance of these manifestations have not been systematically evaluated in large, genotype-stratified cohorts, limiting the ability to provide anticipatory guidance and tailored clinical care. Legal scholars have paid little attention to these national-security laws, and have not recognized that the government uses them specifically to assert a right to suppress information held in patent applications and trade secrets. In this Article, I present a new history of the relationship between export controls and intellectual property to show how the government regulates IP transfers, and how the state uses IP laws to national-security ends. Coastal wetlands are typically considered greenhouse sinks, specifically playing an important role in the global carbon cycle (Chmura et al., 2003), but new studies have pointed to anthropogenic interference leading to the understanding that altered tidal salt marshes may also be producers of methane to the atmosphere, contributing to the greenhouse effect. This thesis investigates whether the selection of thermodynamic ensemble introduces quantifiable differences in the stress relaxation modulus G(t), storage modulus G”(w), loss modulus G”(w), and phase angle d(w) computed from equilibrium molecular dynamics simulations of the AAA-1 asphalt model. A 72-molecule, 12-component representation of AAA-1 bitumen was simulated under three ensembles (NPT, NVT, and NVE) at three temperatures (400.15 K, 443.15 K, and 533.15 K) with three independent replications at each condition, yielding 27 total simulations. Stress autocorrelation functions were computed from each production trajectory and Fourier-transformed to obtain frequency-dependent rheological properties.

The Ocean State Ocean Model (OSOM), funded and developed through the National Science Foundation’s (NSF) Established Program to Stimulate Competitive Research (EPSCoR), was designed to simulate the physical dynamics of Rhode Island’s estuaries and shelf waters. However, the model struggled to capture key features of summer circulation in Rhode Island Sound (RIS), most notably the westward flowing coastal current and vertical stratification profile found in May through September. Significant errors in modeled lateral and vertical density gradients indicated that the thermohaline structure, physical mixing parameters, and forcing inputs required comprehensive revision. Self-supervised learning (SSL) has emerged as a principled approach to visual representation learning that derives supervisory signal directly from unlabeled data, enabling foundation models to be trained at scale without manual annotation. Deployments in medical imaging and biometric recognition have demonstrated the potential of this paradigm, yet the assumptions that make SSL effective on natural image benchmarks fail systematically in specialized domains. Circadian rhythms are approximately 24-hour cycles of physiology and behavior. Circadian rhythms of eating, or meal timing, are linked to cardiometabolic dysfunction in humans. Mistimed eating, such as breakfast skipping and eating late at night, increases the risk of obesity, diabetes, and cardiovascular disease in humans. Likewise, disrupted eating rhythms cause obesity in mice. This dissertation investigates how estrogens regulate eating behavior rhythms and tissue clock timing in female mice. Marine sediments serve as an important geochemical record used to study how the Earth’s climate and oceans have changed throughout geologic time. As such, it is critical to understand how these sediments can be altered following deposition on the seafloor. This dissertation targeted two key geochemical archives, carbonate and pyrite, to link changing burial conditions with the resulting marine sediment chemistry. Wetlands are both critical habitats and bellwethers of the health of the environment. Coal, being the remains of wetlands, offers the possibility of understanding the evolution of wetlands over large climatic shifts. The paleoenvironment of coals can be determined from palynology, petrology, and geochemistry. Stable isotope ratios within coal, including carbon (δ13C), nitrogen (δ15N), and sulfur (δ34S), are established tools for paleoenvironmental reconstructions. These ratios can be correlated to flora, geochemistry, macerals, and bacterial activity.
