Importance Of Specific Nk Cell Subsets For Antitumor Immunity In Hpv+ Cancers, Madison O'Hara
Importance Of Specific Nk Cell Subsets For Antitumor Immunity In Hpv+ Cancers, Madison O'Hara
Dissertations and Theses (Open Access)
High-risk type human papillomaviruses (HPV) are associated with genital and oral cancers, and the incidence of HPV+ head and neck squamous cell cancers is fast increasing worldwide. Survival rates for patients with locally advanced disease are poor and variable after standard of care (SOC) treatment. Identifying the antitumor host immune mediators important for treatment response and designing strategies to promote them are essential for improving clinical outcome. The natural killer (NK) cells are a critical component for antitumor innate effector immunity. Among the multitude of activation and inhibitory receptors on immune cells, HLA-DR is recognized as an important activation marker …
Molecular Basis Of Cell Membrane Adaption Against Daptomycin In Enterococcus Faecalis, April Nguyen
Molecular Basis Of Cell Membrane Adaption Against Daptomycin In Enterococcus Faecalis, April Nguyen
Dissertations and Theses (Open Access)
Daptomycin is a last-resort lipopeptide antibiotic that disrupts cell membrane (CM) and peptidoglycan homeostasis (Grein et al., 2020a; Müller et al., 2016a). Enterococcus faecalis has developed a sophisticated mechanism to avoid daptomycin killing by redistributing CM anionic phospholipids away from the septum. The CM changes are orchestrated by a three-component regulatory system, designated LiaFSR, with a possible contribution of cardiolipin synthase (Cls) (Cesar A Arias et al., 2011a; Khan et al., 2019b; Palmer et al., 2011a; Panesso et al., 2015a; Reyes et al., 2015a). However, the mechanism by which LiaFSR controls the cell membrane response and the role of Cls …
Regulation Of Cd4 T Cell Functions By Ncrna-Mediated Signaling Pathways During Chronic Viral Infections, Lam Nguyen
Regulation Of Cd4 T Cell Functions By Ncrna-Mediated Signaling Pathways During Chronic Viral Infections, Lam Nguyen
Electronic Theses and Dissertations
CD4 T cell homeostasis and competency are critical for the effectiveness of antiviral immunity. However, CD4 T cells derived from people living with HIV (PLWH) and individuals with chronic HCV infection often exhibit an inflammaging phenotype, evidenced by persistent inflammation, immune activation, exhaustion, senescence, and cellular apoptosis. Despite intensive investigations, the molecular mechanisms underlying CD4 T cell dysfunction in antiretroviral therapy (ART)-controlled PLWH and HCV-infected patients remain poorly understood. By investigating the roles of non-coding (nc)RNA transcripts in regulating the functions of CD4 T cells derived from PLWH and HCV-infected patients, we demonstrated that long non-coding (lnc)RNA - growth arrest-specific …

Hierarchal Single-Cell Lineage Tracing Reveals Differential Fate Commitment Of Cd8 T-Cell Clones In Response To Acute Infection., Leena Abdullah
Hierarchal Single-Cell Lineage Tracing Reveals Differential Fate Commitment Of Cd8 T-Cell Clones In Response To Acute Infection., Leena Abdullah
Dartmouth College Ph.D Dissertations
Generating balanced populations of CD8 effector and memory T cells is necessary for immediate and durable immunity to infections and cancer. Yet, a definitive understanding of CD8 differentiation remains unclear. Here we used CARLIN, a processive lineage recording mouse model with single-cell RNA-seq and TCR-seq to track endogenous antigen-specific CD8 T cells during acute viral infection.
Transcriptional profiling of antigen-specific T cells at the peak of the effector response to Vesicular stomatitis virus uncovered a diverse repertoire of expanded antigen-specific T-cell clones represented by seven transcriptional states. TCR enrichment analysis identified TCR clones with differential memory- or effector-fate biases during …
Exploring Suicide Loss Survivorship And Postvention Support During The Coronavirus 2019 Pandemic, Sean Robert Kershaw
Exploring Suicide Loss Survivorship And Postvention Support During The Coronavirus 2019 Pandemic, Sean Robert Kershaw
Dissertations
Suicide is both a severe and pervasive issue in our society (Centers for Disease Control and Prevention [CDC], 2023b). For every suicide, the loved ones of the deceased who are left behind face a long, arduous journey of grieving. Suicide loss survivors who emerged throughout the COVID-19 pandemic arguably encountered more unique challenges during this global crisis than in times past. However, research regarding suicide loss survivorship experiences during the pandemic remains largely overlooked. Therefore, this novel qualitative phenomenological study aimed to capture the lived experiences of those who became suicide loss survivors during the COVID-19 pandemic, in addition to …

One Year Follow Up Of A Cross-Sectional Study Of Hookworm Infection And Response To Deworming In Beposo, Ghana, Taylor Clarkson
Capstone Experience: Master of Public Health
Hookworms are soil-transmitted helminths that infect over 500 million people worldwide, primarily those in low-income countries. Infection with hookworm can cause anemia, malnutrition, and stunted physical and cognitive development. Deworming campaigns have attempted to combat hookworm infections in endemic areas for decades. However, there is growing variability in the effectiveness of deworming drugs, and post-treatment reinfections are abundant. There are significant gaps in our understanding of critical host-parasite factors that mediate hookworm infection, reinfection, and response to deworming treatment in endemic communities. A five-year prospective study was undertaken to better understand drivers of hookworm infection in rural Ghana and thus …

Spatiotemporal Dynamics Of Disease: Social And Environmental Drivers Of Movement, Connectivity, And Disease Transmission In Bighorn Sheep, Lauren E. Ricci
Spatiotemporal Dynamics Of Disease: Social And Environmental Drivers Of Movement, Connectivity, And Disease Transmission In Bighorn Sheep, Lauren E. Ricci
All Graduate Theses and Dissertations, Fall 2023 to Present
Movement is a fundamental component of animal ecology. Animals move in order to access resources and avoid risk. Movement decisions aggregated across time determine how individuals use space, contact dynamics between individuals within a population, and connectivity across a species range. These patterns that emerge from movement decisions have downstream implications for many ecological processes and a mechanistic understanding of movement can help answer broader questions about ecology.
Disease dynamics are intrinsically tied to movement. Understanding the mechanisms that drive movement can elucidate how disease will spread and impact host populations. In this vein, I employed a suite of movement …
Spatial Ecology Of Mule Deer Migrations From Grand Teton National Park And The Teton Range, Justin K. Schwabedissen
Spatial Ecology Of Mule Deer Migrations From Grand Teton National Park And The Teton Range, Justin K. Schwabedissen
All Graduate Theses and Dissertations, Fall 2023 to Present
The Greater Yellowstone Ecosystem hosts several of the longest, fully intact ungulate migrations remaining in the continental United States. However, expanding development and an increasing human footprint continue to truncate migratory routes. While the endpoints are often a seasonal range on protected lands, these migration corridors frequently cross other jurisdictional boundaries, including large tracts of private or multiple-use lands, with varying levels of protection. Thus, it is critical resource managers understand the dynamics of migratory movements to define population-level corridors and prioritize appropriate conservation strategies. Mule deer in Wyoming have been documented traveling long distances between summer and winter ranges; …

Vectors And Vector Borne Disease: Models For The Spread Of Curly Top Disease And Culex Mosquito Abundance, Rachel M. (Frantz) Georges
All Graduate Theses and Dissertations, Fall 2023 to Present
Mathematical models are useful tools in managing infectious disease. When designed appropriately, these models can provide insight into disease incidence patterns and transmission rates. In this work, we present several models that provide information that is useful in monitoring diseases spread by insects.
In the first part of this dissertation, we present two models that predict disease incidence patterns for Curly Top disease (CT) in tomato crops. CT affects a wide variety of plants and is spread through the bite of the Beet Leafhopper. This disease is particularly devastating to tomato crops. When infected, tomato plants present with stunted growth …

Selective Transfection Of A Transferrin Receptor-Expressing Cell Line With Dna-Lipid Nanoparticles And Synthesis Of Parasite-Derived Glycans As Biomarkers For Leishmaniasis, Irodiel Vinales Lozano
Open Access Theses & Dissertations
Despite notable progress in lipid nanoparticle (LNP)-mediated gene delivery, achieving selective transfection of specific cell types, such as cancer cells, remains a significant hurdle, hindering the advancement of innovative gene therapies. In this study, we engineered an LNP formulation encapsulating plasmid DNA (pDNA) encoding the monomeric Green Lantern (mGL) fluorescent reporter protein. The DT7 peptide ligand targeting human transferrin receptor 1 (hTfR1) was also conjugated to the LNP surface for targeted delivery to hTfR1-expressing cells. Optimization of LNP composition yielded favorable particle diameter, ζ-potential, yield, and pDNA encapsulation efficiency. Evaluation of transfection selectivity using a panel of two engineered cell …

Synthesis Of Neoglycoproteins As Potential Biomarkers For Chagas Disease And Cutaneous Leishmaniasis Caused By Leishmania (Viannia) Braziliensis, Eileni Rodriguez Gil
Open Access Theses & Dissertations
Protozoan parasites are responsible for causing numerous diseases in humans and mammals worldwide. Among them are Chagas disease (CD), caused by the parasite Trypanosoma cruzi (T. cruzi), and tegumentary leishmaniasis (TL), caused by the parasite Leishmania (Viannia). Both diseases coexist in many regions of Latin America, and current hurdles to differentially diagnose them and determine chemotherapy success need to be overcome. CD can be diagnosed by traditional serological tests and polymerase chain reaction (PCR), but disadvantages of these methods are that they may produce false negatives because the antigens used in commercial CD kits are not conserved across all T.cruzi …

Institutionalized Stigmas: Hiv Stigma, Racism And Heterosexism Among Medical Students In The Southern United States, Krishen D. Samuel
Graduate Theses and Dissertations
Background: Intersecting and related forms of stigma, including HIV-related stigma, racism, and heterosexism, disproportionately impact both communities vulnerable to acquiring HIV and people living with HIV (PLHIV), ultimately impacting more distal outcomes related to HIV prevention, treatment, and epidemiological patterns in the United States (US). The Southern US is the region most impacted by new HIV diagnoses, mainly among Black men who have sex with men (MSM). However, there is a paucity of research on medical students, specifically those in the Southern US, regarding stigmatizing attitudes they may hold towards PLHIV, and how these associate with racism and heterosexism. This …

Developing Anisotropic Soft Matter As Functional Materials To Interface With Biological Systems, Homa Ghaiedi
Developing Anisotropic Soft Matter As Functional Materials To Interface With Biological Systems, Homa Ghaiedi
Graduate Theses and Dissertations
Liquid crystals (LCs) constitute a subset of soft matter possessing hybrid characteristics from both liquid and crystalline phases. In this state, the center of masses remains uncorrelated similar to an isotropic phase, while the long axis of molecules aligns in parallel, resulting in molecules exhibiting orientational and positional order, either individually or concurrently. The directional alignment inherent in LCs arises from the asymmetrical shape and structure of the constituent molecules, causing a spontaneous self-assembly into ordered phases. This cooperative molecular behavior manifests distinctive optical features when subjected to cross polarizers. The remarkable sensitivity of LCs to external stimuli, including temperature, …

Advancements in comprehending myelodysplastic neoplasms (MDS) have unfolded significantly in recent years, elucidating a myriad of cellular and molecular underpinnings integral to disease progression. While molecular inclusions into prognostic models have substantively advanced risk stratification, recent revelations have emphasized the pivotal role of immune dysregulation within the bone marrow milieu during MDS evolution. Nonetheless, immunotherapy for MDS has not experienced breakthroughs seen in other malignancies, partly attributable to the absence of an immune classification that could stratify patients toward optimally targeted immunotherapeutic approaches. A pivotal obstacle to establishing "immune classes" among MDS patients is the absence of validated accepted immune …

Identifying Temporomandibular Disorder Morphological Risk Factors Via Explainable Deep Learning And Multiscale Biomechanical Modeling, Shuchun Sun
All Dissertations
Clarifying multifactorial musculoskeletal disorder etiologies supports risk analysis and development of targeted prevention and treatment modalities. Deep learning enables comprehensive risk factor identification through systematic analysis of disease datasets but does not provide sufficient context for mechanistic understanding, limiting clinical applicability for etiological investigations. Conversely, multiscale biomechanical modeling can evaluate mechanistic etiology within the relevant biomechanical and physiological context. We propose a hybrid approach combining 3D explainable deep learning and multiscale biomechanical modeling; we applied this approach to investigate temporomandibular joint (TMJ) disorder etiology by systematically identifying risk factors and elucidating mechanistic relationships between risk factors and TMJ biomechanics and …

The survival and health of preterm and critically ill infants have markedly improved over the past 50 years, supported by well-conducted neonatal research. However, newborn research is difficult to undertake for many reasons, and obtaining informed consent for research in this population presents several unique ethical and logistical challenges. In this article, we explore methods to facilitate the consent process, including the role of checklists to support meaningful informed consent for neonatal clinical trials. CONCLUSION: The authors provide practical guidance on the design and implementation of an effective consent checklist tailored for use in neonatal clinical research.

Clostridioides difficile infection (CDI) remains a significant public health threat globally. New interventions to treat CDI rely on an understanding of the evolution and epidemiology of circulating strains. Here we provide longitudinal genomic data on strain diversity, transmission dynamics and antimicrobial resistance (AMR) of C. difficile ribotypes (RTs) 014/020 (n=169), 002 (n=77) and 056 (n=36), the three most prominent C. difficile strains causing CDI in Australia. Genome scrutiny showed that AMR was uncommon in these lineages, with resistance-conferring alleles present in only 15/169 RT014/020 strains (8.9 %), 1/36 RT056 strains (2.78 %) and none of 77 RT002 strains. Notably, ~90 …

Importance: SARS-CoV-2 viral load (VL) in the nasopharynx is difficult to quantify and standardize across settings, but it may inform transmission potential and disease severity.
Objective: To characterize VL at COVID-19 diagnosis among previously uninfected and unvaccinated individuals by evaluating the association of demographic and clinical characteristics, viral variant, and trial with VL, as well as the ability of VL to predict severe disease.
Design, setting, and participants: This secondary cross-protocol analysis used individual-level data from placebo recipients from 4 harmonized, phase 3 COVID-19 vaccine efficacy trials sponsored by Moderna, AstraZeneca, Janssen, and Novavax. Participants were SARS-CoV-2 negative at baseline …

Experiences In Vocal Psychotherapy Of A Music Therapy Intern With Post-Covid Symptoms, Kyungwon Matilda Choi
Theses & Dissertations
The purpose of this study was to explore my experiences in vocal psychotherapy as a music therapy intern with Long COVID symptoms. This study employed a first-person qualitative research design by assessing two personal therapy sessions with heuristic inquiry. I analyzed my experience as a client receiving 60-minute individual vocal psychotherapy sessions. The music therapist was a trained (certified) vocal psychotherapist with at least five years of clinical practice. Data were collected from recorded session audio/video and personal journals. The recorded session audio/video were transcribed so the data could be analyzed. I asked and answered the research questions as this …
