Zika virus (ZIKV) is unique among orthoflaviviruses in its vertical transmission capacity in humans, yet the underlying mechanisms remain incompletely understood. Here, we show that ZIKV induces tunneling nanotubes (TNTs) in placental trophoblasts which facilitate transfer of viral particles, proteins, mitochondria, and RNA to neighboring uninfected cells. TNT formation is driven exclusively via ZIKV non-structural protein 1 (NS1). Zinc oxide Nanowires (ZnO-NWs) are promising biosensor materials and hold the key to overcoming challenges in the field. This chapter provides an introductory overview of biosensing technology, focusing on the fundamental principles and comparing ZnO-NWs with other nanostructures regarding the surface area, reactivity, electrical properties, charge transport behavior, optical, magnetic, and piezoelectric properties, and mechanical flexibility. Providing the synthesis and characterization methods, ZnO-NWs’ biosensing processes are also elaborated on surface modification for selectivity, integration with microfluidic systems, enhancing signal transduction, and connecting with biological elements like enzymes, antibodies, and DNA. BACKGROUND: Chronic alcohol consumption leads to lipid accumulation, oxidative stress, cellular damage, and inflammation in the liver, collectively referred to as alcohol-associated liver disease (ALD). FAF2/UBXD8/ETEA (Fas-associated factor 2) is a ubiquitin ligase adaptor protein that plays a crucial role in the ubiquitin-mediated degradation of misfolded proteins in the endoplasmic reticulum. A recent genome-wide association study indicated an association between FAF2 and ALD; however, the exact contribution of FAF2 to ALD pathogenesis remains unclear. Other conditions such as solid tumor cancers and high blood pressure are less common. Problem: Aging is characterized by a general dysregulation of systemic immune responses that increases susceptibility to infections and malignancies. Immune cells in the female genital tract (FGT) are regulated by sex hormones, but little is known about the impact of aging and menopause on immunology in the FGT. Method of Study: This study conducted an age-focused sub-analysis of cervicovaginal samples collected from 47 women enrolled in the Vaginal Mucosal Systems study in Winnipeg, Canada. Paired cervicovaginal lavage and cervical cytobrush were collected and analyzed by Luminex cytokine array, mass spectrometry based metaproteomics, metabolomics, and high dimensional flow cytometry. Gene editing is one of the most promising tools in science. It enables precise modifications of an organism's genetic material. Metagenomics is considered a powerful tool that unlocks the broad genetic potential found in uncultured microbial communities. Exploring the genetic diversity of uncultured microbial communities helps identify novel functional proteins with unique properties and make the best use of these diverse microbial ecosystems.

Breast cancer is a global health challenge, demanding innovative therapeutic approaches to improve patient outcomes. Nanotechnology has emerged as a promising avenue, facilitating targeted drug delivery to breast cancer cells while minimizing off-target effects. Nanoparticle-based formulations, including liposomes, polymeric nanoparticles and lipid-based nanoparticles offer precise control over drug release kinetics and enhanced bioavailability. Furthermore, gene delivery platforms have garnered attention for their potential to modulate gene expression and overcome drug resistance mechanisms in breast cancer. Moreover, the integration of innovative technologies, like CRISPR-Cas9 gene editing and RNAi, holds promise for targeted gene manipulation and personalized treatment approaches in cancer therapy.

Drug overdose deaths in the United States increasingly include the use of multiple drugs, known as polysubstance use. These include opioids, cocaine, methamphetamine, tranquilizers, and sedatives. Polysubstance use is considered high risk because it can cause dangerous synergistic effects and result in the ingestion of higher amounts of drugs. This brief summarizes findings from a study that used data from the National Survey on Drug Use and Health on U.S. adults from 2002 through 2019 to examine gender and marital status differences in high-risk polysubstance use among U.S. adults ages 18 and older. This research aims to systematically review and map the research topics and themes related to Ḥalāl tourism. A total of 333 articles were reviewed from the Scopus database using the Systematic Literature Review (SLR) method and the VOSviewer application. The results showed that there were 159 Ḥalāl tourism researchers, with 59 research clusters and the highest number of documents held by a researcher being 12. Indonesia emerged as the leading country in Ḥalāl tourism research, with 132 research documents. This is due to the country's focus on developing its Ḥalāl tourism potential, which is supported by its 742 languages and …

PURPOSE: INHALE investigated the impact of seeking pathogens by PCR on antibiotic stewardship and clinical outcomes in hospital-acquired and ventilator-associated pneumonia (HAP and VAP).
METHODS: This pragmatic multicentre, open-label RCT enrolled adults and children with suspected HAP and VAP at 14 ICUs. Patients were randomly allocated to standard of care, or rapid in-ICU syndromic PCR coupled with optional prescribing guidance. Co-primary outcomes were superiority in antibiotic stewardship at 24 h and non-inferiority in clinical cure of pneumonia 14 days post-randomisation. Secondary outcomes included mortality, ICU length of stay and evolution of clinical scores.

Historic relationships with the federal government, converged with the strong cultural heritage, autonomy and self-reliance of the Metlakatla Indian Community (MIC) created foundations for strategic resilience during the COVID-19 pandemic. Sentinel milestone events impacting the MIC during the 19th, 20th, and 21st centuries, initiated by the President of the United States, Congress, and the Federal Courts, are analyzed in the context of their impact and support of evolving tribal-centered development and enaction of protections for the MIC.

Traumatic hemipelvectomy (THP) is a rare, life-threatening injury characterized by the dislocation of the hemipelvis, often resulting from high-energy trauma. It carries a high mortality risk due to severe vascular injuries, with survival contingent on rapid hemorrhage control and a multidisciplinary approach. Limb salvage is rarely achievable due to the extent of the injury and complications such as infection and tissue nonviability. We present a case of incomplete THP in a 29-year-old woman who sustained severe soft tissue, bony, and vascular injuries following a boating accident. A 47-year-old male with a history of immunosuppression and recent intensive care unit admission presented with progressive orbital swelling and pain. Blood cultures grew Trichosporon asahii and Candida guilliermondii, with matching positive cultures from peripheral and central venous samples. Given his fungemia and worsening orbital involvement, induction therapy with amphotericin B and isavuconazole was initiated. Within weeks, clinical improvement was noted, prompting a transition to long-term consolidation therapy with oral posaconazole and isavuconazole. At follow-up, the patient demonstrated sustained clinical stability with no recurrence of infection. Orbital involvement from fungemia due to T. asahii and C. guilliermondii is rare, and …

Medication nonadherence among patients with chronic hepatitis B (CHB) can lead to severe liver disease progression, including liver cirrhosis and hepatocellular carcinoma (HCC). Yet the factors that influence adherence in high-risk groups, like Korean Americans, remain unclear. Thus, this study explored the psychosocial and clinical factors affecting medication adherence in CHB patients. A cohort of 365 Korean American patients with CHB from two clinics in Philadelphia and Los Angeles was studied. The 8-item Morisky Medication Adherence Scale (MMAS-8) gauged their adherence to antiviral medication. Background: Cyclin-dependent kinase 4/6 inhibitors (CDK4/6is) in combination with endocrine therapy are the standard treatment for patients with hormone receptor-positive, HER2-negative metastatic breast cancer (mBC). Despite the efficacy of CDK4/6is, intrinsic resistance occurs in approximately one-third of patients, highlighting the need for reliable predictive biomarkers.
Methods: Single-cell RNA sequencing analyzed metastatic tumors from HR+/HER2- mBC patients pre-CDK4/6i treatment at baseline (BL) and/or at disease progression. BL samples were from CDK4/6i responders (median progression-free survival [mPFS] = 25.5 months), while progressors were categorized as early-progressors (EP, mPFS = 3 months) and late-progressors (LP, mPFS = 11 months). Current Issues in Middle Level Education
The unique needs of high-ability young adolescents should be met just as those students with other diverse needs even when instruction becomes virtual. This paper highlights the work of two teachers who overcame the challenges of remote and hybrid learning to provide quality lessons on creativity, logic, and problem solving. They are accomplishing this for 4th and 5th grade students in their gifted program through project-based learning and other lessons situated around creativity. The strategies presented that encourage students to exercise creativity would be beneficial for all students.

Recent literature has shown an association between social media usage and depressive symptoms, but there remains a gap regarding the perceptions of individuals with clinical depression on their social media usage and their interpersonal relationships. This qualitative study was conducted to uncover how people view social media and their clinical depression as well as the quality of their interpersonal interactions through social media platforms. Semi-structured interviews were conducted with 10 individuals who were diagnosed with clinical depression, used social media for 2 hours daily, and lived with their significant other/partner. Derived from the disease-avoidance model is the hypothesis that people may direct negative cognitive and behavioural responses towards individuals with physical disfiguring conditions, including physical disabilities. According to the behavioral immune system, physical disability—a non-contagious physical disfigurement—may falsely activate cognitive disease-avoidance processes, resulting in prejudicial or negative responses toward individuals with physical disabilities. For the first time this hypothesis is put to the test by investigating whether ratings of attractiveness and comfort for a social interaction vary systematically with physical disability (Studies 1 and 2). Adaptive immunity is critical in eliminating tumors, but cancer-intrinsic factors can subvert this function. Melanoma antigen-A4 (MAGE-A4), a cancer-testis antigen, is expressed in solid tumors and correlates with poor survival, but its role in tumorigenesis and antitumor immunity remains unclear. We found that expression of MAGE-A4 was highly associated with the loss of PTEN, a tumor suppressor, in human non–small cell lung cancers (NSCLC). Here, we show that constitutive expression of human MAGE-A4 with Pten loss in mouse airway epithelia results in metastatic adenocarcinoma. Geothermal hot springs provide unique microbial habitats characterized by extreme temperatures, variable pH, and diverse geochemical conditions. This study investigates the microbial diversity of three hot spring locations in Diamond Fork Canyon, Utah—Diamond Fork Warm Springs, Fifth Water Hot Springs, and Monk’s Hollow Trailhead—using Oxford Nanopore MinION sequencing of the 16S rRNA gene. Samples were collected in triplicate and analyzed to identify the bacterial composition, with a focus on potential opportunistic pathogens. Key Points: Question: What proportion of patients with early syphilis have symptoms of the Jarisch-Herxheimer reaction (JHR) after treatment with benzathine penicillin G, and what are the associated characteristics? Findings: This prespecified secondary analysis of a multisite US randomized clinical trial found that, among 249 adults with early syphilis, JHR symptoms occurred after benzathine penicillin G treatment in 59 participants (23.7%). Onset of JHR was within 12 hours of treatment for 86.0% of participants, and JHR was associated with successful treatment response, secondary syphilis, and absence of HIV. Colorectal precancers in Lynch syndrome (LS) exhibit a distinct immune profile, presenting unique opportunities for developing immune-interception strategies to prevent carcinogenesis. Epigenetic modulation by EZH2 of immune-related genes is implicated in the carcinogenesis of different cancer types, including colorectal cancer. This study utilizes a mouse model of LS and ex vivo colonic organoids to assess the effects of the EZH2 inhibitor GSK503 on immune regulatory pathways, tumorigenesis, and epigenetic reprogramming. Our findings revealed that GSK503 significantly increased CD4+ and CD8+ T cells in both splenocytes and colonic mucosa of treated mice compared with controls.
