Malaria is a vector-borne disease and is a major global public health threat caused by the genus Plasmodium. Of the five species that infected humans, Plasmodium vivax and Plasmodium falciparum contribute the highest rates of morbidity and mortality. Research efforts to develop antimalarial drugs and effective vaccines is imperative to the prevention of future cases. Plasmodium has three reproductive stages: sporogonic, liver, and blood. Sporozoites are injected into the human host from an infected female Anopheles mosquito, which travel to the liver to initiate liver stage malaria. Background: Long coronavirus disease (COVID), which affects an estimated 44.69-48.04 million people in the United States, is an ongoing public health concern that will persist as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continues to spread.
Methods: We developed a computational simulation model representing the clinical course, health effects, and associated costs of a person with long COVID.
Results: Simulations show that the average total cost of a long COVID case can range from $5084-$11 646 (assuming symptoms only last 1 year) with 92.5%-95.2% of these costs being productivity losses. Therefore, the current number of long COVID cases could cost …

Reverse genetics approaches in mice are widely used to understand gene functions and their aberrations in diseases. However, limitations exist in translating findings from animal models to human physiology. Humanized mice provide a powerful bridge to understanding human physiology and mechanisms of disease pathogenesis while maintaining the feasibility of working with small animals. BACKGROUND: Chronic limb-threatening ischemia (CLTI) presents a major clinical challenge in patients with Type 2 Diabetes Mellitus (T2DM), requiring lower extremity revascularization (LER) to mitigate adverse cardiovascular and limb outcomes. Lipoprotein(a) (Lp(a)) has been implicated in cardiovascular risk, but its role in patients with T2DM and CLTI undergoing revascularization remains unclear. Thus, this study aimed to investigate the prognostic value of Lp(a) levels in diabetic CLTI patients for major adverse cardiovascular events (MACE), major adverse limb events (MALE), or both after LER.

The BCG vaccine has been used against tuberculosis (TB) for over a hundred years; however, it does not protect adults from pulmonary TB. To develop alternative vaccines against TB, we generated Mycobacterium tuberculosis H37Rv (Mtb)-derived vaccine strains by rationally deleting key virulent genes, resulting in single (SKO; ΔfbpA), double (DKO; ΔfbpA-ΔsapM), triple (TKO-D; ΔfbpA-ΔsapM-ΔdosR and TKO-Z; ΔfbpA-ΔsapM-Δzmp1), and quadruple (QKO; ΔfbpA-ΔsapM-Δzmp1-dosR) strains. Soil-transmitted helminths (STHs) are intestinal parasites that affect over a billion people worldwide. STH control relies on microscopy-based diagnostics to monitor parasite prevalence and enable post-treatment surveillance; however, molecular diagnostics are rapidly being developed due to increased sensitivity, particularly in low-STH-prevalence settings. The genetic diversity of helminths and its potential impact on molecular diagnostics remain unclear. Using low-coverage genome sequencing, we assess the genetics of STHs within worm, faecal, and purified egg samples from 27 countries, identifying differences in the genetic connectivity and diversity of STH-positive samples across regions and cryptic diversity between closely related human- and pig-infective species. Zoonoses emergence and endemicity are serious global public health problems that disproportionately affect low-resource settings. One Health (OH) serves as a framework to understand the interconnection of zoonoses risk factors and incentivizes collaboration, coordination, and communication among sectors. Background: Sexual minority women (SMW) experience higher rates of substance use relative to heterosexual women in the U.S. but remain an under-studied population. SMW who inject drugs (SMWWID) navigate an unpredictable drug market and may experience complex relationships within their social spheres. We explore cisgender SMWWID’s strategies to maintain safety while injecting drugs, and the influence of their social support systems on those strategies. Methods: We conducted N = 16 phone-based, semi-structured, in-depth interviews with SMWWID in Baltimore, Maryland between June-October 2021. Enhancers play critical roles in gene expression, but a full understanding of their complex functions has yet to be defined. The cellular response to excess zinc levels in Caenorhabditis elegans requires the HIZR-1 transcription factor, which binds the high-zinc activation (HZA) enhancer in the promoters of multiple target genes. Cadmium hijacks the excess zinc response by binding and activating HIZR-1. By analyzing the genome-wide transcriptional response to excess zinc and cadmium, we identified two positions in the genome where head-to-head oriented genes are both induced by metals. BACKGROUND: Mental illnesses constitute a large and escalating portion of the global burden of disease, particularly in low- and middle-income countries like Uganda. Understanding community perceptions towards mental illness is crucial for developing effective interventions.
AIMS: To explore beliefs about the perceived causes and treatment of common mental illnesses (depression, anxiety, alcohol use disorder) and suicidality in rural eastern Uganda.

BACKGROUNDSymptoms of early-onset neonatal sepsis (EOS) in preterm infants are nonspecific and overlap with normal postnatal physiological adaptations and noninfectious pathologies. This clinical uncertainty and the lack of reliable EOS diagnostics results in liberal use of antibiotics in the first days to weeks of life, leading to increased risk of antibiotic-related morbidities in infants who do not have an invasive infection. METHODSTo identify potential biomarkers for EOS in newborn infants, we used unlabeled tandem mass spectrometry proteomics to identify differentially abundant proteins in the umbilical cord blood of infants with and without culture-confirmed EOS. CD16A is an activating Fc receptor on NK cells that mediates antibody-dependent cellular cytotoxicity (ADCC), a key mechanism in antiviral immunity. However, the role of NK cell-mediated ADCC in SARS-CoV-2 infection remains unclear, particularly whether it limits viral spread and disease severity or contributes to the immunopathogenesis of COVID-19. We hypothesized that the high-affinity CD16AV176 polymorphism influences these outcomes. Background: Hypoxaemia predicts mortality at all levels of care, and appropriate management can reduce preventable deaths. However, pulse oximetry and oxygen therapy remain inaccessible in many primary care health facilities. We aimed to develop and validate a simple risk score comprising commonly evaluated clinical features to predict hypoxaemia in 2-59-month-old children with pneumonia.
Methods: Data from seven studies conducted in five countries from the Pneumonia Research Partnership to Assess WHO Recommendations (PREPARE) dataset were included. Readily available clinical features and demographic variables were used to develop a multivariable logistic regression model to predict hypoxemia (oxygen saturation < 90%) at presentation to care. The adjusted log coefficients were transformed to derive the PREPARE hypoxemia risk score and its diagnostic value was assessed in a held-out, temporal validation dataset. The model and risk score were analysed by evaluating the area under the receiver operating characteristic curve (AUC), sensitivity and specificity.

African horse sickness (AHS) is caused by infection with the African horse sickness virus (AHSV). In March 2020, AHS was first reported in Thailand due to the importation of subclinically infected host animals, such as zebras. In this study, we conducted a serological survey for AHSV-specific antibodies across 21 horse farms in the northeastern provinces (n=3) of Thailand. BACKGROUND: Takotsubo syndrome (TTS) is considered as a transient acute heart failure syndrome predominant in females. However, data on Chinese TTS patients are limited. Therefore, this study aimed to investigate the clinical characteristics of Chinese TTS patients, identify prognostic markers.
METHODS: A total of 258 TTS patients were enrolled. Data on clinical characteristics and in-hospital major adverse cardiovascular events (MACE) were collected and analyzed. The latter included severe heart failure, shock, malignant arrhythmia, and death.
RESULTS: The average age of TTS patients was 59.3 ± 19.2 years. Female patients accounted for 68.9%. MACE occurred in 41.4% of patients during hospitalization. …

This study investigates the efficacy and fundamental mechanisms of varying doses of racecadotril in combination with bifidobacterium triple viable for treating this condition in children. Methods: Employing a randomized controlled trial design, participants diagnosed with pediatric persistent diarrhea were randomly allocated to a high-dose group (observation) and a low-dose group (control), both treated with racecadotril combined with bifidobacterium triple viable. Objective: To determine the expression, as well as the underlying mechanism, of lncRNA MZF1 antisense RNA 1 (MZF1-AS1) in Mycoplasma pneumoniae pneumonia (MPP). Methods: GSE179051 was analyzed for differentially expressed lncRNAs in MP infection. The levels of MZF1-AS1 were determined by quantitative Real-time PCR in children with MPP and healthy subjects. Objective: This study analyzed the relationship between clinical parameters and prognosis in children with Mycoplasma pneumoniae pneumonia positive lobar pneumonia and developed an early identification model. Methods: Relevant clinical parameters were collected. Patients were then categorized into two groups based on their length of hospital stay: 116 cases in the refractory group (≥10 days) and 94 cases in the non-refractory group (<10 >days). Objective: Early-onset neonatal sepsis (EOS) is a common cause of mortality in the neonatal period. The purpose of the study was to determine the value of the neutrophil-to-lymphocyte ratio (NLR) in the early diagnosis of EOS. Methods: This was a prospective cross-sectional study. A total of 102 newborns with early-onset infection (EOI) were enrolled within the first 72 hours of life in the neonatal intensive care unit of a tertiary referral hospital, the largest pediatric center in central Vietnam. Objective: A decline in healthcare utilisation and hospital admissions resulted from the COVID-19 pandemic, severely affecting many aspects of life. In this study, we sought to evaluate the pandemic impact on general paediatric admissions by comparing the one-year pre-pandemic period. Methods: From the hospital records, all paediatric admissions from the beginning of March 2020 to the end of February 2021 and the corresponding period in 2019–2020 were included in this descriptive study. We aimed to evaluate the effectiveness and outcomes of medical, percutaneous, and surgical treatments in pediatric HD cases. Methods: A retrospective review was conducted on pediatric patients diagnosed with HD between January 2013 and January 2023. Data on patients were analyzed. This review highlights the biosynthesis of nanoparticles using plant extracts as a sustainable, economical and non-toxic alternative. Plant-derived phytochemicals such as flavonoids, phenolics, and alkaloids serve dual roles as natural reducing and stabilizing agents, enabling the production of nanoparticles typically ranging between 10 and 100 nm. Antibiotic resistant bacteria are becoming increasingly difficult-to-treat resulting in increased morbidity and mortality, especially for common infections such as urinary tract infections (UTIs). Globally, UTIs afflict over 400 million people a year. Numerous different gram-negative and gram-positive bacterial species cause UTIs, including uropathogenic Escherichia coli (UPEC), Klebsiella pneumoniae, and Acinetobacter baumannii.
