Evaluation Of Performance Characteristics And Groundwater Contamination Risks Associated With On-Farm Swine Carcass Disposal Via Composting And Shallow Burial With Carbon, Gustavo Castro Garcia
Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–
This dissertation evaluates the performance of three on-farm swine carcass disposal methods: whole carcass composting (WCC), ground carcass composting (GCC), and shallow burial with carbon (SBC), focusing on biosecurity factors and groundwater contamination risks. Foreign animal diseases (FADs), such as African swine fever and classical swine fever, pose severe economic and animal well-being risks if introduced to the United States. With confirmation of an FAD, swine movement will be halted, creating an urgent need for practical, biosecure, and environmentally responsible on-farm disposal strategies for swine carcasses. Nebraska, as a leading swine-producing state, exemplifies the vulnerability of high-density livestock regions to …

Intention To Take Pre-Exposure Prophylaxis (Prep) As An Hiv Preventive Medication: An Application Of The Health Belief Model And Theory Planned Behavior, Besuthu Nakekelwa Nkambule
Graduate Theses and Dissertations
South Africa (SA) has 7.7 million citizens (12.6% of its 2023 population) living with human immunodeficiency virus (HIV). The use of pre-exposure prophylaxis (PrEP) as an antiretroviral medication for HIV-negative individuals is a promising HIV prevention strategy in SA that can help the attainment of an HIV free generation. PrEP offers greater than 90% protection against HIV acquisition. Despite SA’s efforts in increasing PrEP coverage for its citizens, primarily to those at substantial risk, its adoption remains relatively low. Structural barriers related to access, costs, lack of awareness, and, more recently, US funding cuts exacerbate the challenges of PrEP’s potential …
Social And Economic Gradients Of Resource Transfer Behavior In Gorongosa, Mozambique, Karl J. Mertens
Social And Economic Gradients Of Resource Transfer Behavior In Gorongosa, Mozambique, Karl J. Mertens
Boise State University Theses and Dissertations
Why do people who experience the same environmental and social conditions vary so much in their resource transfer behavior? This dissertation addresses that question through examination of behavioral variation in subsistence horticulturalists in the dry tropical forest bufer conservation area of Gorongosa National Park in Mozambique. In this region of moderate and acute food shortages at both household and regional levels, individuals face similar challenges of addressing social and resource uncertainty yet respond with marked behavioral variation that cannot be explained solely by diferences in wealth, kinship, or reciprocity patterns.

Medical Surveillance In Custodial Jsas: Preventing Bloodborne Pathogen Exposure, Kiara Gadea
Electronic Theses, Projects, and Dissertations
Custodians play an important role in maintaining our surroundings safe and sanitary. However, in doing so, they are their exposure to health hazards that put them at risk, yet these exposures are often overlooked. One major risk they face is contact with bloodborne pathogens, which put custodial staff at risk for infections such as hepatitis B. While Job Safety Analyses (JSA’s) outline various protective measures, many fail to incorporate medical surveillance as part of their risk management program. Overlooking this important detail leaves custodians vulnerable to health risks that can be prevented.
Adding a medical surveillance section to custodial JSA’s, …

The Role Of The Jnk Gene In Ovarian Cancer Inflammatory Pathways, Tashoy D. Campbell
The Role Of The Jnk Gene In Ovarian Cancer Inflammatory Pathways, Tashoy D. Campbell
Graduate Student Dissertations, Theses, Capstones, and Portfolios
Ovarian cancer, one of the leading causes of cancer-related deaths in women globally, presents a significant challenge due to its common late-stage diagnosis and chemoresistance (inability of the tumor to respond to drugs). The c-Jun N-terminal kinase (JNK) signaling pathway plays a crucial role in ovarian cancer progression, contributing to inflammation, tumor survival, metastasis, and chemotherapy resistance. This literature review examines the role of the JNK signaling pathway in ovarian cancer, with a focus on its role in inflammation, metastasis, and chemoresistance. JNK regulates key inflammatory cytokines, such as TNF-α and IL-6, which promote tumor growth and immune evasion. Moreover, …
Computational Frameworks To Unravel The Immune Landscape, Shan He
Computational Frameworks To Unravel The Immune Landscape, Shan He
Dissertations and Theses (Open Access)
Recent advances in immunotherapy, including immune checkpoint blockade (ICB) and adoptive cell therapy, face challenges such as resistance and immune-related adverse events, partly due to our limited understanding of the immune signaling pathways. While high- throughput genomic data provide unprecedented resolution into these immune pathways, their full potential is limited by the lack of well-annotated, context-specific immune gene sets. To address this need, I developed a workflow to construct immune gene sets by integrating RNA- seq datasets and performing decomposition. Using this approach, I constructed 28 immune- specific gene sets from 83 bulk RNA-seq datasets and 12 Natural Killer (NK) …

Dissertations and Theses (Open Access)
Prion diseases are fatal neurodegenerative disorders caused by the misfolding of the normal prion protein (PrPC) into its infectious form (PrPSc). While the zoonotic potential of chronic wasting disease (CWD) remains uncertain, the presence of prions in food products raises public health concerns. Additionally, therapeutic strategies for prion diseases remain limited. This thesis presents a methodological exploration of the Protein Misfolding Cyclic Amplification (PMCA) technique to address these challenges in two key areas: (1) detecting CWD prions in processed meats subjected to common cooking procedures, and (2) identifying potential anti-prion compounds through a high-throughput PMCA adaptation. …

Studies Of The Mechanisms Of Reverse Signaling By Cd40l, Barbara M. Nassif Rausseo
Dissertations and Theses (Open Access)
CD40 ligand (CD40L) is a transmembrane protein expressed on activated CD4 T cells, traditionally known for its role in forward signaling by engaging CD40 on antigen- presenting cells to promote immune activation. While this pathway is well characterized, there is evidence that CD40L also mediates reverse signaling, acting as a signaling receptor on T cells which enhances T cell function. However, the extent, consequences, and mechanism of CD40L-mediated reverse signaling in CD4 T cells remain poorly defined. This dissertation investigates the mechanisms and effects of CD40L reverse signaling on T cell function using both primary human T cells and the …
Accurate Temporal Integration Schemes For Nonlinear Adsorption Problems, Evan D. Butterworth
Accurate Temporal Integration Schemes For Nonlinear Adsorption Problems, Evan D. Butterworth
All Dissertations
We consider a nonlinear transport problem to model the chromatography process of high-capacity multimodal membranes. Robust and efficient algorithms that simulate these bioseparation processes are critical to developing therapeutics for various chronic illnesses and infectious diseases. However, much of the current methodology focuses on stabilization and linearization techniques, often implementing low-order time-discretizations and linearized adsorption, resulting in inefficiencies and inaccuracies in the numerical solution. Utilizing Rothe's method, we develop various time-discretization schemes coupled with the finite element method to solve the fully implicit problems. Stability and solvability results are presented for several methods. Through multiple high-level software implementations paired with …

Orchestrated Architecture Dismantling: Borrelia Burgdorferi’S Coordinated Destruction Of Adaptive Immune Responses, Craig A. Land
Electronic Theses and Dissertations
Borrelia burgdorferi represents the most prevalent vector-borne bacteria in North America, affecting ~476,000 Americans annually yet persisting despite robust immune responses. This project reveals spirochetes target adaptive immunity through coordinated lymphatic colonization and precision immune dysregulation.
Using comparative host models (C57BL/6 vs. reservoir Peromyscus leucopus), we demonstrate lymph node colonization within 48-hours, with spirochetes exploiting LYVE-1-positive lymphatic structures as dissemination highways while avoiding the interior of follicular regions. Critically, we identify targeted CD4+ T-cell elimination in germinal center border zones at 21-DPI, precisely when these cells should be coordinating protective immunity. Concurrent upregulation of inhibitory receptor CD32b creates molecular …

Complement anaphylatoxins C3a and C5a are potent immunomodulators whose impact extends well beyond their traditional roles in innate immunity. Acting through G protein-coupled receptors C3aR, C5aR1, and C5aR2, these peptides take part in coordinating immune cell recruitment, vascular tone, and tissue remodeling. Yet their functions are deeply context-dependent: while they play essential roles in microbial clearance and immune coordination, their overactivation contributes to immunopathology in a wide range of diseases. Although both short and long sleep duration are associated with elevated hypertension risk, our understanding of their interplay with biological pathways governing blood pressure remains limited. To address this, we carried out genome-wide cross-population gene-by-short-sleep and long-sleep duration interaction analyses for three blood pressure traits (systolic, diastolic, and pulse pressure) in 811,405 individuals from diverse population groups. We discovered 22 novel gene-sleep duration interaction loci for blood pressure, mapped to 23 genes. Investigating these genes' functional implications shed light on neurological, thyroidal, bone metabolism, and hematopoietic pathways that necessitate future investigation for blood pressure management that caters to sleep health …

Objectives: This study aimed to evaluate the antimicrobial efficacy of three commercially available denture cleaning tablets Efferdent, Polident, and Val-clean against Staphylococcus aureus biofilms formed on two denture base materials: milled polymethyl methacrylate (PMMA) and 3D printed denture resin.
Material and methods: Eighty samples were fabricated, with 40 specimens per denture base material: CAD/CAM milled PMMA (Lucitone Digital Fit LT; Dentsply Sirona, Charlotte, NC, USA) and 3D printed resin (Denture Base Resin; Formlabs Inc., Somerville, MA, USA). Each group was divided into four subgroups (n = 10), Efferdent, Polident, Val-clean, and deionized water (control). Surface roughness was measured using a …
