Clonal hematopoiesis (CH) is an independent risk factor for atherosclerotic cardiovascular disease. Murine models of CH suggest a central role of inflammasomes and IL-1β in accelerated atherosclerosis and plaque destabilization. Here we show using single-cell RNA sequencing in human carotid plaques that inflammasome components are enriched in macrophages, while the receptor for IL-1β is enriched in fibroblasts and smooth muscle cells (SMCs). To address the role of inflammatory crosstalk in features of plaque destabilization, we conducted SMC fate mapping in Ldlr-/- mice modeling Jak2VF or Tet2 CH treated with IL-1β antibodies. Pediatric cancer consists of a diverse group of rare diseases. Due to limited patient populations, standard randomized and controlled trials are often infeasible. As a result, single-arm trials are common in pediatric oncology and the use of external controls is often desirable or necessary to help generate actionable evidence and contextualize trial results. In this paper, we illustrate unique features in pediatric oncology clinical trials and describe their impact on the use of external controls. Background: POLARIS (phase 2 [ph2]; NCT03911869) evaluated encorafenib (BRAF inhibitor) in combination with binimetinib (MEK1/2 inhibitor) in BRAF/MEK inhibitor-naïve patients with BRAF V600-mutant melanoma with asymptomatic brain metastases.
Methods: The safety lead-in (SLI) assessed tolerability for high-dose encorafenib 300 mg twice daily (BID) plus binimetinib 45 mg BID. If the high dose was tolerable in ph2, patients would be randomized to receive high or standard dose (encorafenib 450 mg once daily [QD] plus binimetinib 45 mg BID). The aim of this study is to evaluate the baseline F18-FDG PET/CT findings of individuals diagnosed with giant cellarteritis (GCA) and to explore its association with clinical findings and classification criteria.Materials and methods: We analysed data from patients who underwent F18-FDG PET/CT scans to investigate large vessel (LV)involvement between 2010 and 2019. Only patients with a clinical diagnosis of GCA and at least 6 months of follow-up were included.We compared initial clinical features and laboratory findings based on the presence of LV vasculitis on PET/CT and the maximumstandard uptake value (SUVmax) of vascular territories.Results: Twenty-nine patients (median age at diagnosis: …

Background: Bioprostheses with RESILIA tissue demonstrate a reduction in calcification and improve health outcomes in pre-clinical and clinical studies. Prior economic analyses which relied on 5 years of evidence from the COMMENCE trial demonstrate financial savings for RESILIA tissue valves relative to mechanical valves after surgical aortic valve replacement (SAVR). Given the recent release of 7-year COMMENCE data, this economic evaluation updates the estimate for long-run savings of bioprosthetic valves with RESILIA.
Methods: Simulation models estimated disease progression across two hypothetical SAVR cohorts (tissue vs. mechanical) of 10,000 patients each in the US. The primary comparison calculated the SAVR-related expenditures …

Anne Frank is linked to her contemporaries in about 80% of 18 English-language published and produced plays. The remaining plays pair Frank and African American icons Harriet Tubman, Emmett Till, and Martin Luther King, Jr. Research on dramatic literature with Frank as a character, the writing of plays linking her with African American personages, or history, analysis, or comparison of the process of multiple plays about Frank does not exist. Pedophilia is a complex, multifaceted disorder that has existed, in some form, throughout all of human history. It was not until 1952, with the inception of the Diagnostic and Statistical Manual of Mental Disorders (DSM-1), that it was officially characterized as a psychiatric disorder. Treatment options have included surgeries, medications, cognitive behavior therapy, combined treatments (antiandrogenic drug and psychotherapy), and preventative programs. Pedophilic offenders released to the community must complete approved treatment. The community treatment may be manualized, evidence-based and likely to be effective. The applied dissertation compares the lifestyle and behavioral characteristics of mass, serial, and spree murderers. Historically, studies have focused on serial murder according to the perpetrator's typology or indicators at the crime scene. The current topics of research lack a comparison of these classifications. While the classification and described typologies are essential for academic and investigative purposes, understanding the impactful lifestyle factors of these perpetrators may also serve as an essential factor in predictive analysis, victimology, and the motive behind their heinous crimes.

Legal responses to litigation funding could take many forms—consumer protection, regulation of interest rates and fees, perhaps even prohibition (as under the old doctrines of champerty and maintenance). In the U.S., procedural responses, often emphasizing disclosure of funding, have featured prominently. From the perspective of one directly involved in the U.S. rulemaking effort, this Article explores the challenges of designing such a rule. Among other things, it notes that litigation funding actually could support important law-implementation features of American litigation. The Monster Mash is a course proposal for an upper-division undergraduate literature course focused on exploring monsters in literature and building connections between classic and more contemporary texts using high-impact practices (HIPs) with student success in mind. I build on previous work in the field of Monster Studies and introduce my own original monster pattern that prompts students to interpret monsters as they trek through Origin, Separation, Power, Threat, and Diminishment. The inspiration for this dissertation began as a practitioner. As a Wall Street equity analyst, I calculated the valuation of various types of stocks using relative and intrinsic valuation methods on a daily basis. I noticed an interesting phenomenon while performing these calculations. The estimated stock prices were often largely disconnected from the actual stock market price leading me to wonder what the role of estimated valuations was in the overall investment process.

Background: Antiangiogenic therapy had been tested in urothelial cancer (UC) without reaching the clinic.
Objective: We provide a systematic review and meta-analysis of trials to assess efficacy of immune checkpoint inhibitors (ICI) combined with antiangiogenic agents in UC.
Methods: Following PRISMA guidelines, we searched for trials with at least one arm of patients with UC treated with ICI plus antiangiogenics. Data were analyzed with the "meta" package from R using a one-staged frequentist meta-analysis.
Results: After screening 13,708 titles and abstracts, 9 studies were selected for analysis with 14 identified cohorts comprising 621 patients: 448 were ICI-naïve (ICI-N) and 173 …

Mechanisms of voluntary motor control are complex and require inputs via cortical centers, peripheral sensory systems, and spinal inputs such as central pattern generators. It is hypothesized that control is simplified via utilization of motor primitives, which may rely on structures within the spinal cord, however this remains unknown in both human and non-human primates. The goal of this dissertation is to characterize muscle activity in the upper extremity in relation to the corresponding anatomical organization of spinal motoneurons to provide insight into how primitives are both organized and activated to produce movement.

The phenomenon of maternity has been repeatedly described as an event that shakes the very foundations of social and physical identity. As the flesh of the pregnant person literally divides to produce new life, one subject becomes enclosed within another, dramatically affecting the pregnant person’s sense of self and causing a confluence of intense, and often conflicting, feelings. Market humiliation is a corrosive relational process to which the law repeatedly fails to respond due to the law’s heavy reliance on the discrimination paradigm. In this process, providers of market resources, from housing and work to goods and services, use their powers to reject or mistreat other market users due to their identities. They thus cause users severe harm and deprive them of dignified participation in the marketplace. The problem has recently reached a peak. The discussion in 303 Creative v. This Article uncovers a critical yet unexplored dimension of policing: the strategic oscillation of police officers between their roles as state actors and private individuals, and its significant implications for police accountability frameworks. As officers toggle between these two roles to their legal advantage, they exploit a deep, systemic flaw in the structural design of policing. Tracing the trajectory of policing from its vigilante origins to its institutionalized form today, this Article argues that contemporary policing merges state-sanctioned power with vestiges of vigilantism to blur the public-private divide. Medieval Feminist Forum: A Journal of Gender and Sexuality
This essay shines a light on the previously ignored queer representations of women on Gothic secular ivories, taking as its case studies three popular iconographical themes: the Castle of Love, the game of chess, and the courtly hunt. Refusing to yield to compulsory heterosexuality when looking at the indeterminately gendered bodies that populate these objects reveals both the ivories’ inherent queerness and their potential to interrogate their own normativity. Like the romance literature that inspired the images on these objects, they represent and enforce idealized gender norms with remarkable reluctance. The study explored the readiness and challenges of social media adoption at the Public Records and Archives Administration Department (PRAAD) in Ghana and was guided by the Iacovou, Charalambos L., Izak Benbasat, and Albert S. Dexter model of Electronic Data Interchange adoption. Using the qualitative approach, 13 participants drawn from the Archives Division were interviewed to explore their perspectives. The results showed that besides the participants high understanding and awareness of the benefits of social media adoption, clients were interested to engage PRAAD on social media. In 1895 the Nebraska Legislature passed a bill officially adopting the nickname the “Tree Planter’s State” on account of the founding of Arbor Day, the Timber Culture Act of 1873, and the “millions of trees planted by early settlers as windbreaks, woodlots, and orchards.” Since that time, Arbor Day and the Timber Culture Act have been the subject of countless historical studies, while the subject of settler tree planting remains largely unexplored. Pirates during the late seventeenth and early eighteenth centuries experienced the height of Atlantic piracy. The Golden Age of Piracy caused major political problems for the British overseas colonies and constituted revisions in colonial governance. Piracy during this period began as buccaneers who worked for the state during wartime as privateers and rogues during peacetime. This group changed however, by the end of the seventeenth century had transitioned into pirates, attacking any and all shipping around the colonies regarless of origin. On the 400th anniversary of American enslavement the New York Times (NYT) 1619 project launched an interactive digital experience including a popular podcast centering the contributions and narratives of Black Americans. This study sought to understand how HBCU students responded to learning Black music history through what we term a “pop culture podcast.” This study explored the ways in which this particular podcast could support the development of Critical Race Media Literacy (CRML) based on a media discourse at a Historically Black College/University (HBCU). Since the 18th National Congress of the Communist Party of China, the construction of Ecological Civilization (EC) in China has undergone historic, transitional, and comprehensive changes from theory to practice. The construction of system is the core content and the basic guarantee for the success of EC construction. Based on the government documents of EC construction and related research literatures, this study analyzes the formation and evolution characteristics of China’s EC construction system, parses the development process of representative innovation policies, and then predicts the trend of the further deepening reform of EC system.
