Cognitive flexibility declines early in Alzheimer's disease, yet the underlying circuit mechanisms remain unknown. Here, we report that young 5xFAD mice exhibit deficits in instrumental reversal learning prior to spatial memory impairment. This behavioral inflexibility is associated with abnormal neuronal reactivation in the medial prefrontal cortex and dorsomedial striatum. Electrophysiological recordings reveal that medial prefrontal cortex neurons are hyperexcitable and receive increased excitatory input. Furthermore, glutamatergic transmission from the medial prefrontal cortex to striatal direct-pathway medium spiny neurons is enhanced and coincides with strengthened inhibitory transmission onto striatal cholinergic interneurons, reduced spontaneous firing, and diminished striatal acetylcholine release. Growth/differentiation factor-15 (GDF15) is a secreted cytokine strongly associated with dementia risk. However, the extent to which GDF15 represents a biomarker and driver of dementia risk remains unclear. Across multiple cohorts, we demonstrated that plasma GDF15 is associated with greater dementia risk over 15- to 25-year follow-up periods when measured in midlife, with stronger associations observed for vascular, compared to Alzheimer's disease (AD), dementia. Two-sample Mendelian randomization supported plasma GDF15's mechanistic role in AD and related dementias, while cohort studies linked it to cerebral small vessel disease, neurodegeneration, phosphorylated tau, and a cerebrospinal fluid proteomic signature indicative of neuroimmune activation. …

Demonstration Of 23-Gene Expression Profile Test Utility Within Prame Immunohistochemistry Results: A Case Series, Gregory A. Hosler, Matthew S. Goldberg, Etan Marks

Histopathological examination using hematoxylin and eosin (H&E)-stained tissue is sufficient for diagnosing most melanocytic neoplasms as benign or malignant; however, diagnostic tools such as PRAME (preferentially expressed antigen in melanoma) immunohistochemistry (IHC) and the 23-gene expression profile (23-GEP) test can aid in the analysis of histopathologically ambiguous lesions, which can be referred to as diagnostically challenging, equivocal, indeterminate, or uncertain. When compared with the gold standard of H&E diagnosis by a dermatopathologist, PRAME IHC showed 83.2% sensitivity and 99.3% specificity in an initial validation cohort and 23-GEP showed 91.5% sensitivity and 92.7% specificity in an independent validation cohort. Karbala International Journal of Modern Science

The current study presents a case of gingivitis caused by Cryptococcus albidus in a 35-year-old male from the city of Karbala in Iraq. This fungal species was identified via morphology, microscopy, additionally to the VITEK system. C. albidus was further confirmed using the polymerase chain reaction (PCR) technique by amplifying the 18S rRNA gene as a universal primer and sequencing. Sequences analysis revealed that this strain of C. albidus had not been previously documented in the NCBI database. Background: In early adolescence, psychological well-being represents an important developmental outcome. This period is marked by significant physical, emotional, and psychosocial transitions. Widely recognized as protective factors, executive function and resilience contribute to positive adolescent adjustment and mental health. However, investigations on the roles of these constructs among early adolescents in Northern Thailand in research remains limited.

Method: This study employed a cross-sectional correlational design, using secondary data from 210 lower secondary school students aged 11 to 13 years in Northern Thailand. There is little doubt as to the mark left by the 1994 Rwandan genocide on historical discourse and perhaps even the American cultural consciousness. U.S. evening news broadcasts carried images of suffering and death into homes across the US, and in the process made Rwanda the site of one of the most recognized traumas in human history. While cultural proximity in mass media coverage played a role, the characterization of media coverage of the genocide, often defined in the press as explicit and constant, requires analysis. In a context marked by an increasing search for meaning and well-being, sanctuaries are re-emerging as significant spaces within contemporary spiritual tourism. This work proposes an approach to sanctuaries located in natural environments as liminal spaces, in the anthropological sense developed by Victor Turner, where processes of passage, transformation, and the reconfiguration of personal experience are facilitated. Amid growing health concerns, increased competition among fitness centers, and declining staff-to-customer ratios, gym operators are seeking effective ways to reduce customer attrition. Drawing on Prospect Theory, this study examines how commitment devices in gym promotions (e.g., deposit contracts and transformation challenges) influence consumers’ intentions to join a gym. Two experiments with 609 participants (Study 1 = 204; Study 2 = 405) investigated the mediating role of perceived attractiveness and the moderating role of contractual flexibility. Results indicate that membership options incorporating a commitment device were perceived as more attractive, leading to stronger registration intentions. However, this effect was observed …
