Kaiso (ZBTB33) is a transcription factor involved in mitotic clonal expansion and tumorigenesis in association with Adenomatous Polyposis Coli (APC) loss of heterozygosity. ENCODE data show strong overlap of the Kaiso promoter-binding site—encode-derived Kaiso-binding site (eKBS) and many other transcription factors, including BRCA1. Here we sought to determine whether BRCA1 is a component of the Kaiso enhanceosome that regulates gene transcription. Using proximal ligation assays, immunoprecipitation followed by mass spectrometry, luciferase assays and ChIP-seq experiments, we evaluated the association between BRCA1 and Kaiso. A proposal in the 2025 tax-reconciliation bill would provide federal Child Development Account–like accounts for all children born between 2024 and 2028, with a provision enabling children born in 2028 or after to open accounts. This Policy Brief is designed to inform public debate about the legislation through a discussion of the evidence underpinning the most fundamental of many lessons learned from SEED for Oklahoma Kids, the Center for Social Development’s long-running, randomized experiment with Child Development Account policy: the need for a centralized savings-plan platform.

California's Imperial Valley, with lithium-rich geothermal brines, extensive flat agricultural fields with abundant desert sunshine, and access to transmission lines, is a leading case to explore interconnected themes around just energy transitions. Despite being the poorest county in California and one of the smallest, Imperial already provides 15% of the state’s solar electricity, and the region as a whole represents on the order of 25% of the state’s electric power capacity. This paper brings to light frictions over solar energy development that have emerged over the history of solar power development in the southern Salton Sea region. It describes the …

Aim: DNA methylation in brain regions represents a potential mechanism linking genetic variation to Alzheimer's disease (ad) risk, yet most studies have focused on blood-derived methylation markers. In this study, we conducted a systematic Mendelian randomization (MR) study to evaluate associations between predicted brain region-specific DNA methylation levels and ad risk, using methylation quantitative trait loci (mQTL) as genetic instruments.
Methods: We analyzed mQTLs from five human brain regions: cerebellum (CRBLM), frontal cortex (FCTX), causal pons (PONS), and temporal cortex (TCTX) from 600 individuals in Gibbs et al's study, as well as mQTLs from dorsolateral prefrontal cortex (DLPFC) of 543 …

Background: Methotrexate is a critical component of pediatric acute lymphoblastic leukemia (ALL) therapy that can result in neurotoxicity which has been associated with an increased risk of relapse. We leveraged machine learning to develop a neurotoxicity risk prediction model in a diverse cohort of children with ALL.
Methods: We included children (age 2-20 years) diagnosed with ALL (2005-2019) and treated in Texas without pre-existing neurologic disease. Clinical information was obtained by medical record review. Neurotoxicity occurring post-induction and prior to maintenance therapy was defined as neurologic episodes occurring within 21 days of methotrexate. Water quality assessments are critical for ensuring timely responses to water-related concerns, particularly in low-resource areas with limited water, sanitation, and hygiene (WASH) infrastructure. In collaboration with the Belize Ministry of Health and Wellness and the Ministry of Education, Culture, Science and Technology, we conducted a survey on WASH infrastructure and resources among 221 schools. We identified 65 schools across all six districts of Belize for water quality testing. Among these 65 schools, 83% had at least one water sample that did not meet the WHO's recommended free chlorine residual level for drinking water. Additionally, coliforms and Escherichia coli were …

Nonstructural Roles Of Junctophilin-2 In The Heart, Daniel F Hutchison, Satadru K Lahiri, Kurt W Prins, Xander H T Wehrens

Junctophilin-2 (JPH2) is an essential structural protein that maintains the integrity of junctional membrane complexes (JMCs) in cardiomyocytes, thereby playing its canonical role in excitation-contraction (E-C) coupling. Recent studies reveal novel, nonstructural functions of JPH2 in cardiomyocytes, including regulation of mitochondrial function, lipid metabolism, nuclear signaling, and innate immune responses. Calpain-mediated cleavage of JPH2 generates nuclear fragments, which differentially modulate stress responses. Moreover, full-length JPH2 also localizes to the nucleus, where it forms phase-separated nuclear droplets during oxidative stress. Collectively, these findings highlight JPH2 as a multifunctional protein with critical roles beyond its canonical structural function in E-C coupling.

Oxygen (O2) is vital for cellular development, function, proliferation, and repair, underscoring its critical role in organogenesis. Both hypoxia (reduced tissue O2) and hyperoxia (excess tissue O2), when prolonged, can trigger inflammation and oxidative stress, contributing to acute and long-term cardiopulmonary and neurodevelopmental morbidity. In sick neonates, immature defense mechanisms and coexisting morbidities demand nuanced and sometimes opposing strategies for O2 saturation targets and therapeutic titration. Most current neonatal O2 targeting guidelines are based on animal models or small clinical studies, resulting in recommendations with limited evidence. While the incidence and survival associated with lymphoma have improved in recent years, outcome disparities related to sociodemographic factors such as age, sex, race, ethnicity, socioeconomic status (SES), and other social determinants of health (SDOH) remain prevalent in the modern era. Clinical trials are crucial for further improving outcomes, yet not all sociodemographic groups are equally represented in contemporary lymphoma trials. We conducted a systematic review of the literature evaluating the impact of SDOH on enrollment into clinical trials in the United States and identified 21 relevant studies. Background: There has been a proliferation of health professions training programmes focused on quality and patient safety, but little information regarding the outcomes of these programmes. The purpose of this study was to examine the impact of an interprofessional quality and patient safety curriculum on career outcomes of interprofessional healthcare learners for the Veterans Affairs Patient Safety Fellowship.
Method: We conducted a survey-based assessment of 117 alumni of the Interprofessional Fellowship in Patient Safety assessing their satisfaction with the training program and career impacts.

The current study explores how and why customer sexual harassment (CSH) experienced by service workers may lead to instigated incivility towards customers in the same week. Specifically, following the tenets of Personality Systems Interaction theory (PSI) and the affective shift model, I hypothesized that week-level affective shifts mediate this relationship, such that upshifts in anxiety and downshifts in self-assurance govern changes in motivational and cognitive functions that impair the employee's ability to provide good customer service. Neuromuscular priming and post-activation potentiation (PAP) are techniques utilized to increase nervous system excitation, muscle force, and rate of force development during subsequent muscle contractions in sport. PAP is a commonly used approach that has the potential to influence neuromuscular status monitoring and flagging, and optimize power training. Specifically, jump height is an important parameter in athletic performance and assessing inter-limb asymmetries. School leadership is often described in terms of vision, strategy, and efficiency. However, the lived experiences of school leaders reveal a far more complex reality—one shaped by relationships, ethical dilemmas, and the daily challenges of guiding a school community. This paper presents insights from a sabbatical study in which the author observed and interviewed leaders at five schools—one public and four faith-based.Four aspects of leadership are illustrated in this essay: the emotionally charged moments, the tensions that arise between competing visions, the fragility of educational institutions, and the role of hospitality in leadership.These narratives illustrate how school leaders navigate conflict, …

Research has documented the positive impact of the nurse practitioner workforce on patient and system outcomes in multiple countries. Achieving these outcomes relies on successfully integrating the workforce into the health system. Research has reported negative effects of integration barriers on the ability for nurse practitioners to improve outcomes. Barriers include scope of practice restrictions, organizational climates inconducive to nurse practitioner care, and lacking mentorship. To design efficacious policies, organizational and national policy decision-makers require knowledge of the factors affecting the integration of this human resource into care settings. This paper applies corpus linguistic discourse analysis to approach the social media discourse of a recent political movement in Iran, known as #WomanLifeFreedom and Mahsa Amini. The movement included vast protests in different parts of Iran and went viral on an international scale. Protesters relied heavily on social media, particularly Twitter, to determine strategies and support each other during the expansive calls. To address the discourse shaped in that platform, a corpus was compiled from the Twitter posts, named Mahsa Amini Tweets Corpus (MATC). The management of the herdsmen-farmers conflict-induced internal displacement in Nigeria was characterised by remote factors that affected the protection and assistance of internally displaced persons (IDPs). This qualitative study was conducted on 12 participants comprising humanitarian workers, IDPs, and lawmakers who were conversant with the persistent herdsmen-farmers conflict-induced internal displacement. The study explored the inter-connected relationship among stakeholders within the humanitarian ecosystem. It found that though the herdsmen-farmers conflict led to mass displacement, remote factors such as unwillingness of government, data challenges, the undiplomatic approach of government in the management of the displacement failed to win support of other stakeholders …

In the last decade we have witnessed rising protests in the United States associated with issues that form part of society’s social fabric that can either facilitate or break down collective behaviour. Rising social inequalities can cause people to no longer share the same values and force individuals to work against each other. This breakdown in social capital can be a key driver for protests as the marginalised attempt to voice their grievances. Unhealthy sexual behaviors among adolescents, such as engaging with multiple partners or neglecting contraceptive use, can lead to significant physical, psychological, and relational consequences. This literature review explores the influence of family communication on adolescent sexual behaviors and attitudes, emphasizing the role of parent-adolescent discussions in promoting healthier sexual practices and reshaping sexual values. Research indicates that open, reciprocal family communication is associated with reduced sexual risk-taking, fewer sexual partners, delayed sexual debut, and increased contraceptive use. Moreover, conversations centered on positive sexual norms, such as love and respect, facilitate adolescents’ adoption of healthier sexual values. Effective family communication also …

The integration of IoT and blockchain enhances security, trust, and data integrity but is hindered by security attacks, scalability, and high latency. In this work, a more efficient method of consensus using Directed Acyclic Graph (DAG)-based Fast Probabilistic Consensus (FPC) and Edwards-Curve Digital Signature Algorithm (EdDSA) is proposed to yield better security, efficiency, and decentralization. By eliminating mining, resource use is optimized, and consensus is hastened. Experimental results show a high throughput of 7228.05 Transactions Per Second (TPS), rapid consensus formation in just 7.62 rounds on average, and high adversary resilience, with the system successfully mitigating 89% of adversarial attacks. …
