Objectives: This study aimed to assess the prevalence of aberrant opioid-related behaviours (AORB) among patients with sickle cell disease (SCD) in Oman and identify associated risk factors. Methods: This cross-sectional analytical study was conducted at Sultan Qaboos University Hospital, Muscat, Oman from June to December 2022. Patients with SCD aging 18–65 years old were included and clinically observed for more than 3 months. The Current Opioid Misuse Measure (COMM) was used to evaluate AORB with scores of ≥9 indicating high risk. Demographic and clinical data were collected. The association between AORB and clinical-demographic variables was examined using the Chi-square test. …

In this nonpayment case, the court awarded the tenants a substantial rent abatement of over $76, 000 for the landlord's breach of the warranty of habitability. The tenants successfully proved the existence of numerous conditions, including leaks, mold, and defective surfaces, using HPD violations and trial testimony. While the landlord was granted a final money judgment for the remaining arrears, the court significantly reduced the amount owed. The decision underscores the importance of a fact-based defense in abatement cases to meet the preponderance of the evidence standard.

Acute myeloid leukemia (AML) that evolves from myeloproliferative neoplasm (MPN) is known as post-MPN AML. Current treatments do not significantly extend survival beyond 12 months. B-cell lymphoma-extra large (BCL-xL) has been found to be overexpressed in leucocytes from patients with MPN, making it a potential therapeutic target. We investigated the role of BCL-xL in post-MPN AML and tested the efficacy of DT2216, a platelet-sparing BCL-xL proteolysis-targeting chimera, in preclinical models of post-MPN AML. We found that BCL2L1, the gene encoding BCL-xL, is expressed at higher levels in patients with post-MPN AML than in those with de novo AML. Single-cell multiomics …

Clitellata is a major clade of Annelida that includes almost all freshwater and terrestrial annelids as well as many marine species. Aquatic oligochaetes include the union of the two multispecies families: Naididae and Tubificidae. There has been much debate regarding the phylogeny and classification of the cosmopolitan and diverse Naididae. In this study, the total mitogenome sequences of Potamothrix hammoniensis (Naididae: Tubificinae), Stylaria fossularis, Stylaria lacustris, Chaetogaster diaphanus, Chaetogaster diaphanus sp. B, and Slavina appendiculata (Naididae: Naidinae) were sequenced and characterized and can be accessed from NCBI under accession numbers OQ654101, PP909790, PP909791, PP909792, PP909793, and PP893275, respectively. Functional neurologic symptom disorder (FNSD) is a multifaceted condition that can present with neurologic symptoms inconsistent with recognized neurological disease, often in the context of psychosocial stressors, though these are not required for diagnosis. We report the case of a 12-year-old Hispanic female who developed episodes of limb paralysis, motor unresponsiveness, and self-injurious behaviors following a history of sexual abuse by a family member. Despite the initial absence of depressive symptoms, the patient later met criteria for major depressive disorder (MDD) and post-traumatic stress disorder (PTSD). Her neurological symptoms lacked identifiable medical findings, following unremarkable results from comprehensive blood work, …

The Response Of Mitochondrial Position To Glucose Stimulation In The Min6 Model System, Luis Perez
The Response Of Mitochondrial Position To Glucose Stimulation In The Min6 Model System, Luis Perez
Arts & Sciences Graduate Student Theses and Dissertations
The compartmentalization of eukaryotic cells into membrane-bound organelles with specific subcellular positioning enables precise spatial and temporal control of cellular functions. While functionally significant mitochondrial localization has been demonstrated in cells such as neurons, it remains unclear how general these cell principles are. Here, we examine the spatial organization of mitochondria within MIN6 pancreatic beta cells under variable glucose conditions. We observe glucose-dependent redistributions of mitochondria, favoring peripheral localization at elevated glucose levels when insulin secretion is also elevated. Our results suggest that active mitochondrial transport along microtubules and calcium activity, but not ATP synthesis, are critical regulators of this …

Wildfires are a natural part of the western U.S. landscape and provide numerous benefits, including enhanced nutrient cycling and habitat rejuvenation. Utah’s fish populations evolved with wildfire as a common disturbance. However, the increasing size and severity of wildfires, combined with Utah’s heavily engineered and fragmented stream networks, pose a contemporary risk to fish populations. This fact sheet reviews wildfire impacts on erosion and Utah's watersheds and fisheries. It outlines measures we can take to prevent post-wildfire fish die-offs.

Racial disparities in the clinical outcomes of triple-negative breast cancer (TNBC) have been well-documented, but the underlying biological mechanisms remain poorly understood. To investigate these disparities, we employed a multi-omic approach integrating imaging mass cytometry and spatial transcriptomics to characterize the tumor microenvironment (TME) in self-identified Black American (BA) and White American (WA) TNBC patients. Our analysis revealed that the TME in BA patients is marked by a network of endothelial cells, macrophages, and mesenchymal-like cells, which correlates with reduced patient survival. In contrast, the WA TNBC microenvironment is enriched in T-cells and neutrophils, indicative of T-cell exhaustion and suppressed …

In this perspective, we highlight the relevance of the FA-Hippo signaling pathway and its regulation of the Yes-associated protein (YAP) and the transcriptional coactivator with a PDZ-binding domain (TAZ) as main players in the process of implants integration. The modulation and responses of YAP/TAZ triggered by substrate and ECM stiffness are of particular interest in the construction of materials used for medical implants. YAP/TAZ nuclear localization and activity respond to the substrate stiffness by several mechanisms that involve the canonical and non-canonical Hippo signaling and independently of the Hippo cascade. YAP/TAZ regulate the expression of genes involved in several mechanisms …

Background: Limited ancestral diversity has impaired our ability to detect risk variants more prevalent in ancestry groups of predominantly non-European ancestral background in genome-wide association studies (GWAS). We construct and analyze a multi-ancestry GWAS dataset in the Alzheimer's Disease Genetics Consortium (ADGC) to test for novel shared and population-specific late-onset Alzheimer's disease (LOAD) susceptibility loci and evaluate underlying genetic architecture in 37,382 non-Hispanic White (NHW), 6728 African American, 8899 Hispanic (HIS), and 3232 East Asian individuals, performing within ancestry fixed-effects meta-analysis followed by a cross-ancestry random-effects meta-analysis.
Results: We identify 13 loci with cross-population associations including known loci at/near CR1, …

Primary testicular (PT) diffuse large B-cell lymphoma (DLBCL) is a rare and aggressive lymphoma with distinct clinical and molecular characteristics. To identify prognostic biomarkers in PT-DLBCL, in this study we analyzed DNA and RNA samples of PT-DLBCL tumors from 206 patients using next-generation sequencing platforms and assays. Genetic alteration analysis found that multiple chromosomal copy number variations (CNVs), TP53 transcript mutations with high variant allele frequency, and MCD subtype had significantly adverse prognostic effects, whereas elevated microsatellite instability had a significantly favorable prognostic effect in PT-DLBCL. Targeted RNA-seq analysis identified a PTL gene expression signature by comparing PT-DLBCL with systemic …

Neuroactive steroids (NAS) have long been recognized for their hypnotic and anesthetic properties in both clinical and preclinical settings. While sex differences in NAS sensitivity are acknowledged, the underlying mechanisms remain poorly understood. Here, we examined sex-specific responses to an endogenous NAS epipregnanolone (EpiP) in wild-type mice using behavioral assessment of hypnosis (loss of righting reflex, LORR) and in vivo electrophysiological recordings. Specifically, local field potentials (LFPs) were recorded from the central medial thalamus (CMT) and electroencephalogram (EEG) signals were recorded from the barrel cortex. We found that EpiP-induced LORR exhibited clear sex differences, with females showing increased sensitivity. Spectral …

Small extracellular vesicles (sEVs) have emerged as powerful vectors for liquid biopsy, offering a noninvasive window into the dynamic physiological and pathological states of the body. However, to fully leverage the clinical potential of sEV biomarkers, it is imperative to develop robust and efficient technologies for their isolation and analysis. In this study, we introduce a novel sharp-edge acoustofluidic platform designed for rapid and effective sample preparation, coupled with sensitive detection of specific sEV populations based on their surface markers. Our approach utilizes acoustically activated sharp-edge microstructures to concentrate bead-bound sEVs within the microfluidic device, facilitating immediate visualization by fluorescence …

Research Outputs: 2025-Present
Microbial nanocellulose (MC) draws special attention from the research community due to its unique nano-bio fabrication. This nanofiber-constructed biomaterial is biosynthesized by certain microbes in the purest form (devoid of lignin, pectin, and hemicellulose). Due to its striking similarities to animal collagen fibers, MC nanofibers have been used to develop osteoconductive biomaterial for soft tissue regeneration. High surface area, notable rheological properties, and biocompatibility make this material attractive for tissue regeneration. Concurrently, their bio-friendly characteristics are due to various bio-ceramic materials like calcium phosphate (CP) employed in conjunction with MC (for the development of scaffold/bio-composite). The significant interaction between MC's …

Machine Learning Modeling Approaches For Predicting The Extent Of Fatigue Damage In Polycrystalline Alloy Structures, Hassan Alqahtani
Yanbu Journal of Engineering and Science
Using predictions of crack-tip-opening-displacement (CTOD) to measure the extent of fatigue damage has provided the opportunity to prepare an efficient strategy for protecting mechanical structures from damage and developing a structural health monitoring system. The objective is to forecast non-measurable CTOD by using machine learning methods. In this paper, an optical metrology device, which is built by Alicona on a confocal microscope and hereafter referred to as Confocal Microscope, has been used to measure CTOD. However, two factors limit the usage of Alicona Apparatus: (i) the size of optical images, where a CTOD over 400 micrometers cannot be measured; and …
