Protein-folding chaperone heat shock protein 90 (HSP90) buffers genetic variation in diverse organisms, but the clinical significance of HSP90 buffering in human disease remains unclear. Here, we show that HSP90 buffers mutations in the BRCT domain of BRCA1. HSP90-buffered BRCA1 mutations result in protein variants that retain interactions with partner proteins and strongly rely on HSP90 for protein stability and function in cell survival. Moreover, HSP90-buffered BRCA1 variants confer poly (ADP-ribose) polymerase (PARP) inhibitor resistance in cancer cells. Low-level HSP90 inhibition overcomes this resistance, revealing a cryptic and mutant-specific HSP90-contingent synthetic lethality. The bankruptcy of 23andMe was an inflection point for the direct-to-consumer genetics market. Although the privacy of consumer data has been highlighted by many as a concern, we discuss another key tension in this case: the corporate enclosure of scientific data that has considerable potential value for biomedical research and public health.

This Doctor of Ministry thesis project examined how structured spiritual formation practices can cultivate attentiveness to God and strengthen ministry readiness among leaders within a congregational context. The project addressed a critical need in the ministry field: many leaders serve faithfully but lack sustained rhythms of reflection, Scripture meditation, and contemplative rest that foster long-term spiritual vitality. The purpose of this quantitative, predictive, correlational study was to examine how well course clarity and utility value predict student engagement among online learners at a Canadian post-secondary institution. This study is important because it can provide educators and curriculum developers with insights into the factors that positively impact student engagement. Most foster parents feel they do not receive adequate emotional support, funding, and preparation for their foster children in their care, which can lead to turnover, placement disruptions, and additional unnecessary caregiver changes for foster children. Attachment Theory explains children need consistent, warm, and loving caregiver relationships for healthy emotional, cognitive, and social development (Bowlby, 2018). This qualitative historical study investigates how music functions as a unifying force within global social and political movements, transcending cultural, linguistic, and national boundaries to foster collective identity and sustain long-term activism. Drawing on Social Identity Theory, Cultural Diffusion Theory, and Framing Theory, the research analyzes archival documents, historical records, media sources, and case studies from movements such as the U.S. Civil Rights Movement, the Anti-Vietnam War Movement, anti-apartheid struggles, Black Lives Matter, the Arab Spring, and contemporary climate justice activism. Church revitalization is a complex process. The stress it brings can challenge even the most experienced pastor’s spiritual and emotional resilience. Revitalization experts note that revitalization anxiety is especially true for pastors with certain personality traits. One such trait is the DISC personality category of steadiness. Leadership specialists argue that relief from this stress is found through gaining emotional intelligence. The purpose of this DMIN action research project was to teach emotional intelligence to eight to ten pastors with the steadiness personality. The relationship between horses and humans has evolved and now horses are a part of therapy, leisure activities, competitions, and companionship with humans, and this evolution beckons further research into aspects that contribute to positivity in this relationship. This research examined the themes of personality among professional horse trainers by utilizing the Riso-Hudson Enneagram Type Indicator (RHETI), the Ten-Item Personality Inventory (TIPI), and semi-structured interviews. Exploring The Relationship Between Loneliness And Real-Time Measurements Of Gsr And Hrv During Concept Mapping, And Cortisol And Oxytocin Levels, Jangwon Seo

This study attempts to explore loneliness among college students from a bio-physio-psycho-social perspective, and it utilizes real-time physiological measurements (GSR and HRV) and biomarkers (cortisol and oxytocin) in conjunction with psychosocial assessments. The study employs both qualitative and quantitative methods, which involve concept mapping exercises, with the aim of comprehending the full nature of how loneliness interacts with physiological and psychological processes. At Time 1 (T1) we found significant positive correlations between loneliness and elevated GSR levels, which is indicative of increased sympathetic arousal (r = .35, p < .01). Additionally, there were significant negative correlations between loneliness and mean RR intervals in HRV that indicated autonomic dysregulation featured by increased sympathetic activities as well as reduced parasympathetic functions (r = -.28, p < .05). However, at Time 2 (T2), these correlations were not significant indicating possible temporal fluctuations due to adaptive responses or extraneous variables such social support systems or coping mechanisms. The qualitative data from concept mapping enriched the analysis by providing deeper insights into the cognitive and emotional processes underlying loneliness. Despite limitations like demographic skewness, funding constraints, logistical challenges, etc., this, therefore, highlights the need for multidimensional investigations of psychological phenomena. This has implications for future research as well as clinical practice, which could include using these integrative measures to develop holistic interventions for mental health disorders.

Cesarean sections remain the most common surgical procedures in the United States, but they come with significant risks, including increased pain, infection, longer recovery, and higher health care costs. Enhanced Recovery After Surgery (ERAS) guidelines outline an evidence- based, multidisciplinary approach that optimizes surgical recovery and improves outcomes. This Doctor of Ministry thesis project addresses worship fragmentation at Beach Church in Virginia Beach, Virginia, where sacramental, evangelical, and charismatic elements have historically functioned independently, leading to theological imbalance and diminished congregational unity. Many healthy and unhealthy adult behaviors are transmitted from parent to child as generational behavior cycles. Conflict avoidance, codependency, and people-pleasing are all unhealthy relationship dynamics that are generationally passed down in such a way. Breaking these generational cycles of unhealthy relationship dynamics is often difficult. The goal of this phenomenological qualitative study was to explore the common themes among individuals who have broken the generational cycles of conflict avoidance, co-dependency, and people-pleasing. This qualitative phenomenological research project will explore the lived experiences of Black women pastoring at least three (3) who are not genetically related, weekly attending male congregants in the historically patriarchal culture of the Christian Methodist Episcopal (CME) Church with a minimum of at least one hundred (100) weekly attendees or more, and actively ordained. The CME Church has traditionally believed that the Bible prohibits women from serving as pastors or in any position of control over men. This qualitative phenomenological study explored the lived experiences of East African immigrants with accented English in Michigan mental health counseling context. The purpose was to understand how accented speech shaped communication, emotional safety, and therapeutic connection between clients and counselors. Grounded in B.F. Skinner’s theory of language learning, along with the language transfer and critical period hypothesis frameworks, the study employed semi-structured interviews with ten East African immigrant adults who had received mental health counseling in Michigan. This quantitative archival study examined the relationship between adverse childhood experiences (ACEs) and intimate partner violence (IPV) among participants in a specialized Domestic Violence Treatment Court (DVTC) in New Mexico. Between January 2019 and December 2024, 69 individuals volunteered for the DVTC, with 49 graduating successfully, reflecting a graduation rate of approximately 78%. The study’s purpose was to determine whether a sample of offenders (N=22) reported higher levels of ACEs compared to the U.S. general population. The purpose of this quantitative experimental pretest -posttest design using an experimental and control group was to determine if gaming escape room simulation will decrease BSN nursing students' stress in high fidelity simulation (HFS). The sample size of 64 students had 32 students randomly assigned to each group, the control and the experimental group. The purpose of this qualitative phenomenological study was to describe the shared lived experiences of women affected by marital infidelity in North Carolina. Physical, emotional, and spiritual issues arise due to betrayal and brokenness of secure attachments. Findings from this study revealed that some shared emotional issues women faced following infidelity included depression, guilt, self-blame, and low self-esteem. Some issues took longer to recover from than others, such as loss of trust, low self-esteem, and varying physical health-related issues.
