Gram-negative bacteria utilize siderophores to scavenge for environmental iron. Pseudomonas capeferrum produces the siderophores pseudobactin BN7/8, which trigger cell surface signaling (CSS). Iron import CSS pathways consist of an outer membrane TonBdependent transporter, an inner membrane sigma regulator, and a cytoplasmic sigma factor that activates transcription of target genes. The P. capeferrum sigma regulator, PupR, has an N-terminal antisigma domain (ASD). Previously, the PupR ASD was purified as a dimer in the absence of its cognate sigma factor, PupI. Here we use pulldown assays to demonstrate that His6-PupIFL interacts with the PupR ASD as well as with homodimer-disrupting mutants. We …

Effective targeted drug delivery is a critical unmet need in the management of both ulcerative colitis and metastatic colorectal cancer, where conventional systemic therapies may be hindered by suboptimal efficacy and significant side effects. This thesis explores the development and preclinical assessment of polymeric chloroquine (PCQ)-based nanotherapeutics, leveraging stimuli-responsive designs for disease-specific delivery and action.
For ulcerative colitis, a reactive oxygen species (ROS)-responsive nanogel system was engineered to release PCQ, a macromolecular derivative of hydroxychloroquine, specifically in the inflamed areas within the colon. Engineered microorganisms play an increasingly important role in the sustainable production of fuels, bioplastics, and other valuable biomaterials. Successful industrial deployment depends on maximizing product titers and ensuring process robustness at large scale. However, one of the major obstacles is non-genetic cell-to-cell heterogeneity arising from stochastic intracellular processes. Such heterogeneity can result in subpopulations that divert substrates toward growth rather than product biosynthesis, ultimately reducing yield and contributing to batch-to-batch inconsistency. While reinforcement learning has been a vital component in artificial intelligence and machine learning, there exist many open questions about its implementations and how to improve them, in both minds and machines. Among these are i) the contribution of non-neuronal cell types to reinforcement learning, and ii) information-seeking behavior during reinforcement learning. In this thesis, we studied these main topics pertaining to reinforcement learning. Background: Long-term health conditions and multimorbidity are increasing globally, placing an unsustainable pressure on health care systems. Mobile health (mHealth) technologies enable the collection of patient-generated health data outside clinical settings, offering the potential to support personalized care and inform clinical decision-making. However, the ways in which mHealth patient data are being used in clinical practice remain unclear. Farming is a hazardous occupation. Exposures are multifactorial, variable, and shaped by overlapping tasks, environments, and social contexts. This dissertation aimed to illustrate the complexity of agricultural exposures and the barriers to protective practices through three complementary studies.
The first study examined co-exposures to solvents and noise as risk factors for hearing loss (HL) among 7,495 farmers and ranchers in a 7-state region. Results indicated nearly 60% of co-exposed participants reported hearing loss, with an adjusted odds ratio nearly six-fold (OR=6.03, [4.67-7.78]) for moderate/severe HL. Background
Obesity is associated with systemic inflammation, increasing chronic disease risk. Carotenoids possess anti-inflammatory properties, yet the short-term impact of carotenoid-rich juice intake on inflammation in individuals with obesity remains unclear. Therefore, the purpose of this paper was to examine: (1) participant adherence to a 3-week lycopene-rich juice intervention; (2) variance and effect sizes of responses to the dietary intervention; (3) changes in skin and plasma carotenoids, and inflammatory markers among adults with obesity.
Methods

BACKGROUND: Pseudarthrosis remains a significant complication of ACDF, contributing to patient morbidity and increased healthcare costs. Glucagon-like peptide-1 receptor agonists (GLP-1RA), widely prescribed for diabetes and obesity, have been shown to reduced odds of pseudoarthrosis after ACDF. However, it remains unclear whether these reductions meaningfully translate to reduced odds of subsequent cervical surgery.
METHODS: The TriNetX United States Collaborative Network was queried for patients who underwent single- or multi-level ACDF between 2013-2023. Patients with perioperative GLP-1RA use and propensity-score-matched controls without GLP-1RA use were compared for subsequent rates of pseudoarthrosis and cervical reoperation at 6-, 12-, and 24-month intervals. Sporadic early-onset Alzheimer’s disease (sEOAD) represents a substantial but less-studied subtype of Alzheimer’s disease (AD). Here, we generated a single-nucleus multiome atlas derived from the postmortem prefrontal cortex, entorhinal cortex, and hippocampus of nine individuals with or without sEOAD. Comprehensive analyses were conducted to delineate cell type–specific transcriptomic changes and linked candidate cis-regulatory elements (cCREs) across brain regions. We prioritized eight conservative transcription factors in glial cells in multiple brain regions, including RFX4 in astrocytes and IKZF1 in microglia, which are implicated in regulating sEOAD-associated genes. Rhizomelic chondrodysplasia punctata type 1 (RCDP1) is a peroxisomal disorder characterized by skeletal shortening, intellectual disability, seizures, cataracts, and reduced lifespans. RCDP1 is caused by biallelic loss-of-function variants in PEX7, which encodes a protein required for importing select enzymes into the peroxisome matrix, including those essential for ether lipid synthesis (e.g., plasmalogens) and the branched-chain fatty acid catabolism. Plasmalogen deficiency is a hallmark of RCDP1 and other peroxisomal disorders, including RCDP types 2-5 (RCDP2-5) and Zellweger spectrum disorders (ZSD). Adaptive neuromodulation systems and implantable brain-computer interfaces have made notable strides in recent years, translating experimental prototypes into clinical applications and garnering substantial attention from the public. This study investigates how the Danu Kerthi philosophy operates as a framework that integrates Traditional Ecological Knowledge (TEK) with environmental science to support sustainable lake conservation in three upstream lakes in Bali: Beratan, Buyan, and Tamblingan. While previous scholarship has largely focused on the normative and symbolic dimensions of Sad Kerthi, empirical evidence on its ecological effectiveness remains limited. To address this gap, the study adopts a multi-site qualitative design involving participatory observation, document analysis, and semi-structured interviews with 30 participants representing traditional leaders, Hindu priests, tourism managers, village officials, and local residents. The VISIA camera is a device that captures images of the skin, offering a detailed look at skin health by detecting changes that are often missed during physical exams and by the naked eye. It can help identify changes in UV damage, pigmentation, texture, fine lines, and redness. In dermatology, it has become a useful tool to build targeted treatment plans and follow patient progress over time. Background: Survival among children and adolescents and young adults (AYA) with cancer has improved substantially over recent decades; however, dominant survivorship models remain reactive-activated post-treatment and anchored to static exposure- and organ-based screening. This design underuses the anticipatory window at diagnosis and overlooks environmental and social determinants that modulate outcomes across the life course. While Buddhist traditions articulate a parallel moral vision in the early discourses of the Pāli Canon, Western traditions usually locate the roots of ethical reasoning in Socratic inquiry. Especially in their approaches to bodily care, spiritual well - being, and moral responsibility, this article contends that both Socratic and Early Buddhist ideas converge on fundamental principles in medical ethics. Inclusive STEM (science, technology, engineering, and mathematics) education plays a vital role in promoting equitable learning opportunities and fostering sustainable development. However, in Nigeria, many marginalized student groups face barriers that hinder their full participation in STEM fields. This study examines the factors impeding inclusivity in STEM education and their effects on students’ motivation, academic performance, and awareness of sustainable development in Lagos State. This systematic review of literature examines scholarly studies conducted on Black women and Latinas in STEM fields and in STEM higher education. Women are underrepresented in STEM, and on average they comprise 27% of the STEM population (National Science Foundation, 2017). Black women and Latinas are even further underrepresented in STEM by making up only 14.58 % and 3% of the employees in STEM industries (Hispanic Heritage Foundation, 2020; National Science Foundation, 2021). Feminicide and gender violence cases in the last several decades in Puerto Rico have continuously increased, suggesting that this problem becomes worse as time passes. This study, therefore, focuses on identifying actionable insights that paint a picture of the impact it has on women over the age of 18 in Puerto Rico. In order to achieve this objective, the study relies on a strong theoretical framework that is made up of Foucauldian discourse analysis, necropolitics, and sexual scripts theories. As pornography continues to become more easily and readily accessible, children are at higher risk of viewing it. This work aims to investigate the destructive impact of premature exposure to pornography, and the negative perceptions of sex and decision-making towards it as a result. The operational definition used defines “early exposure” as prepubescents (children of elementary-grade school ages) viewing explicit material through involuntary and accidental initial engagement, and “decision-making” as moral agency and choice of response and action. In this paper, we use the pseudonym ABC Pharmaceuticals (ABC) to refer to the company under examination, GlaxoSmithKline (GSK). ABC Pharmaceutical (ABC) must assess, adapt, and change to ensure regulatory compliance, maintain and attract employees, and remain competitive. ABC has a history of erroneous practices and is at risk internally with employees, and externally with regulators and the public sectors. The authors applied J.P. Kotter’s (1996) 8-step and the ADKAR (Hiatt, 2006) models for change management. J.P.
