A Tale Of Two Genomes: The Complex Interplay Between The Mitochondrial And The Nuclear Genomes, Abhilesh S. Dhawanjewar Jun 2022

A Tale Of Two Genomes: The Complex Interplay Between The Mitochondrial And The Nuclear Genomes, Abhilesh S. Dhawanjewar

School of Biological Sciences: Dissertations, Theses, and Student Research

Mitochondria, the product of an ancient endosymbiotic event are pivotal to eukaryotic cells by synthesizing the majority of the cell’s ATP output. However, modern- day mitochondria are completely dependent on more than one thousand nuclear-encoded products for their function and the maintenance of their genomes. The fundamentally different ways in which the mitochondrial (mtDNA) and the nuclear (nucDNA) genomes are replicated and inherited lead to captivating coevolutionary dynamics between them. The aims of this dissertation are to investigate the coevolutionary dynamics between the mitochondrial and nuclear genomes at three distinct biological scales. At the organismal level, we use a Drosophila …

Discovering Novel Polyextremotolerant Fungi And Determining Their Ecological Role Within The Biological Soil Crust Consortium, Erin C. Carr Jun 2022

Discovering Novel Polyextremotolerant Fungi And Determining Their Ecological Role Within The Biological Soil Crust Consortium, Erin C. Carr

School of Biological Sciences: Dissertations, Theses, and Student Research

The ecological niche of polyextremotolerant fungi within oligotrophic ecosystems such as biological soil crusts has not yet been determined. These fungi persist in locations where nutrients are depleted while simultaneously surrounded by autotrophic microbes such as algae and cyanobacteria. Yet it has not been shown that they are engaging in any exchange of nutrients the way lichens do. However, there is seemingly no other way for these fungi to obtain vital nutrients, such as carbon or nitrogen, other than from these microbes. Here we have isolated polyextremotolerant fungi from cold desert biological soil crusts which are a microbial biofilm that …

Helping-Seeking Tendencies And Subjective Well-Being: A Cross-Cultural Comparison Of The United States And Japan, Verity Yu Qing Lua, Nadyannam N. Majeed, Andree Hartanto, Angela K. Y. Leung Jun 2022

Helping-Seeking Tendencies And Subjective Well-Being: A Cross-Cultural Comparison Of The United States And Japan, Verity Yu Qing Lua, Nadyannam N. Majeed, Andree Hartanto, Angela K. Y. Leung

Research Collection School of Social Sciences

Help-seeking is commonly conceived as an instrumental behavior that improves people’s subjective well-being. However, most findings supporting a positive association between help-seeking and subjective well-being are observed in independence-preferring countries. Drawing from research demonstrating that the pathways to subjective well-being are culturally divergent, we posit that help-seeking tendencies may be detrimental to subjective well-being for members in interdependence-preferring countries where norms for preserving relational harmony and face concerns are prevalent. This study tested the moderating role of country in the relationship between help-seeking tendencies and subjective well-being using data from 5,068 American and Japanese participants. Results revealed that although help-seeking …

Obstetrics and Gynaecology, East Africa

We urgently need answers to basic epidemiological questions regarding SARS-CoV-2 infection in pregnant and postpartum women and its effect on their newborns. While many national registries, health facilities, and research groups are collecting relevant data, we need a collaborative and methodologically rigorous approach to better combine these data and address knowledge gaps, especially those related to rare outcomes. We propose that using a sequential, prospective meta-analysis (PMA) is the best approach to generate data for policy- and practice-oriented guidelines. As the pandemic evolves, additional studies identified retrospectively by the steering committee or through living systematic reviews will be invited to …

Integrated Multilayer Omics Reveals The Genomic, Proteomic, And Metabolic Influences Of Histidyl Dipeptides On The Heart, Keqiang Yan, Zhanlong Mei, Jingjing Zhao, Md Aminul Islam Prodhan, Detlef Obal, Kartik Katragadda, Benjamin Doelling, David Hoetker, Dheeraj Kumar Posa

Internal Medicine, East Africa

Background: Histidyl dipeptides such as carnosine are present in a micromolar to millimolar range in mammalian hearts. These dipeptides facilitate glycolysis by proton buffering. They form conjugates with reactive aldehydes, such as acrolein, and attenuate myocardial ischemia-reperfusion injury. Although these dipeptides exhibit multifunctional properties, a composite understanding of their role in the myocardium is lacking.

Methods and Results: To identify histidyl dipeptide-mediated responses in the heart, we used an integrated triomics approach, which involved genome-wide RNA sequencing, global proteomics, and unbiased metabolomics to identify the effects of cardiospecific transgenic overexpression of the carnosine synthesizing enzyme, carnosine synthase (Carns), in …

BACKGROUND AND AIMS: Risk factor cutoffs are derived from associations with clinical cardiovascular disease (CVD), but how these risk factors associate with preserved cardiovascular health into old age is not well studied. We investigated midlife determinants of healthy versus nonhealthy cardiovascular aging in the Atherosclerosis Risk in Communities (ARIC) study.

METHODS: ARIC participants were categorized by cardiovascular status in older age (mean age 75.8 ± 5.3 years, range 66-90): healthy, subclinical disease (assessed by biomarkers and left ventricular function), clinical CVD (coronary heart disease, stroke, or heart failure), or prior death. We examined associations of midlife (mean age 52.1 ± …

guide the discussion.

Tribal, state, and city authorities are currently pursuing or have settled product liability, public nuisance, and other common law and statutory tort claims brought against Monsanto[1] for PCB contamination.[2] “Sovereign-led” litigation melds traditional plaintiff common law tort litigation with sovereign-led environmental suits and is an emerging trend in environmental law.[3] Tort claims against the manufacturers of contaminants ubiquitous in the environment give sovereigns a new angle for pursuing damages separate from a traditional statutory environmental claim under federal or state regulatory schemes, such as the Comprehensive Environmental Response, Compensation, and Liability Act (CERCLA)

College of Business Theses and Dissertations

April 2020, COVID-19 (COVID) changed the world and how society functions, transforming the means in which we communicate. Face-to-face interactions were abruptly and immediately replaced with technology mediums. The increase in technology use changed our interpersonal communication habits, placing greater reliance on new and existing electronic methods. Although new forms continue to evolve, the predominate electronic communication method utilized is email. This research uses grounded theory on stress and technology acceptance, and further assesses perceived email volume changes to analyze the relationship of these constructs to job satisfaction during COVID. The data was collected from 147 employees of a mid-sized …

Black & Boring, Daryhl Gregory Covington

College of Liberal Arts & Social Sciences Theses and Dissertations

This collection of poetry and flash fiction was written when I was 24 years old. It was inspired heavily by the two years I spent living in Southgate — a blue collar suburb of Detroit — before moving to Chicago to complete my second year of grad school at DePaul. Most of my time in Southgate was uneventful. I spent those two years playing basketball, writing raps, taking pictures, watching indie movies with Aaron, smoking weed, skateboarding, and walking through different parks in the area. My life was more mundane and repetitive than it had ever been, but I’d never …

The Role And Immunogenicity Of Cbfa2t3-Glis2 In Pediatric Acute Megakaryoblastic Leukemia, Elizabeth A. Garfinkle Jun 2022

The Role And Immunogenicity Of Cbfa2t3-Glis2 In Pediatric Acute Megakaryoblastic Leukemia, Elizabeth A. Garfinkle

Theses and Dissertations (ETD)

CBFA2T3-GLIS2 is the most prevalent fusion oncogene in pediatric acute megakaryoblastic leukemia in patients without Down syndrome (non-DS-AMKL) and is associated with an event free survival of only 8% even with high intensity chemotherapy and stem cell transplant in first remission. A cryptic inversion event on chromosome 16 joins the three nervy homology regions (NHR) of CBFA2T3 to the five zinc fingers of GLIS2. This configuration enables the encoded chimeric transcription factor to bind GLIS consensus sequences throughout the genome and recruit transcriptional activators and repressors to alter gene expression and enhance self-renewal capability. Few cooperating mutations have been identified …

Identification Of Therapeutic Targets For Th17 Cell-Mediated Diseases, Jin Wang Jun 2022

Identification Of Therapeutic Targets For Th17 Cell-Mediated Diseases, Jin Wang

Theses and Dissertations (ETD)

Hypersensitivity and autoimmune diseases are complex synergistic effect of environmental, genetic and other factors. T helper 17 (Th17) cells play a critical role in the immune pathology. Th17 cells are a group of specialized CD4+ T cells with potential pro-inflammatory function which are different from other T cells such as T helper 1 (Th1) cells or T helper 2 (Th2) cells. Th17 cell development and differentiation processes are strictly regulated through complex pathways. Th17 cells are characterized by secreting cytokines such as IL-17 and IL-22. Studies have shown that Th17 cells and IL-17 have been implicated in the pathogenesis of …

Host-Pathogen Interactions During Leptospirosis In Male C3h/Hej Mice, Advait Bhaskar Shetty Jun 2022

Host-Pathogen Interactions During Leptospirosis In Male C3h/Hej Mice, Advait Bhaskar Shetty

Theses and Dissertations (ETD)

Leptospirosis is a zoonotic disease transmitted from reservoir hosts like rodents to other animals and humans, who are incidental hosts. This infection is prevalent in the areas of Southeast Asia, Africa and South America. Inactivated Leptospira serovars vaccines are available in endemic areas but they have certain limitations such as serovar specificity and short-term protection. Antibiotics are effective treatment strategies useful at least during early stages of infection. Early diagnosis of Leptospirosis is difficult due to lack of accessible laboratory and point of care tests and due to clinical similarities with other diseases. Thus, it is important to understand host-Leptospira …

Characterizing T Cell Features Of Atypical Lymphocytes For The Optimization Of Adoptive Immunotherapies, Taylor L. Wilson Jun 2022

Characterizing T Cell Features Of Atypical Lymphocytes For The Optimization Of Adoptive Immunotherapies, Taylor L. Wilson

Theses and Dissertations (ETD)

T lymphocytes with atypical antigen targets or mechanisms or antigen recognition have been implemented into therapeutic approaches for the treatment of disease with auspicious results. Two such populations are regulatory T cells (Tregs) and CAR (chimeric antigen receptor-modified) T cells. The functional role of Tregs in the immune system is non-canonical, in that they prevent hyper-inflammation and resolve immunopathology by constraining conventional immune cell effector functions and producing mediators that promote tissue repair. Similarly, the targets of CAR T cells are also self-derived but are recognized through an engineered receptor designed to bypass the conventional mechanism of antigen-recognition. Despite their …

Remote Patient Monitoring As A Means To Reduce 30-Day Readmissions In Skilled Patients: An Integrative Review, Sharee Lenae Birkett

Doctoral Dissertations and Projects

Readmissions among skilled patients are a problem nationwide with rising rates despite federal mandates. Per the AHRQ (2021), readmission costs are estimated to average $15, 200 per patient. Interventions are needed to reduce readmissions and improve patient outcomes. Remote patient monitoring (RPM), especially with continuous vital sign monitoring (CVM) is one method that could reduce readmission rates. The purpose of this integrative review is to determine whether RPM could be used to reduce emergency department and 30-day readmission visits for Medicare part A skilled nursing patients. Utilizing the Whittemore and Knafl (2005) framework, 29 articles were reviewed with resulting themes …

The Scorpion Mud turtle, Kinosternon scorpioides, is a widely distributed, semiaquatic species known to engage in regular terrestrial movement and terrestrial estivation. We studied terrestrial behavior in this species in Palo Verde National Park, northwestern Costa Rica. We determined terrestrial activity patterns by searching for active individuals during transects along 3.4 km of trails. We determined the distance that turtles traveled between the wetlands and estivation sites by fitting 10 terrestrially active individuals with a thread-bobbin tracking device attached to the rear of the carapace. We identified sex and marked and measured every turtle found active. We accumulated 92 observations …

Surgical Treatment Of Multiple Deforming Xanthomas Of The Foot And Ankle With Complete Excision And Rotational Skin Flaps: A Case Report, Qanita Ali, Rachelle Randall, Lawrence M. Fallat

Articles

Xanthomas are cholesterol rich nodular dermal lesions characterized by lipid-laden macrophages that accumulate in skin and tendons.1 These lesions are frequently associated with hyperlipidemia disorders characterized by impaired cholesterol metabolism and accelerated atherosclerotic plaque development.2 Until recently, xanthomas were considered mere cosmetic lesions, however extensive evidence has shown that the presence of xanthomas signify increased cardiovascular risk and reduced average lifespan.3 Treatment centers around lipid lowering agents and rarely are these masses large enough to require surgical resection, thus there is very little literature discussing proper surgical excision.4, 5 We present a rare case of a 65 year old female …
