Immobilization of large biomacromolecules is often required for analytical quantification and physicochemical characterization. However, immobilization can alter the structure and size of the particles being studied. Here, two exosomes (derived from HEK-293 and MDA-MB-231 cells) and three viral particles (Suid herpesvirus 1 (SuHV), xenotropic murine leukemia virus (XmuLV), and porcine parvovirus (PPV)) were immobilized to different covalent chemistries to understand how surface chemistry influences particle deformation during immobilization. The surface chemistries explored were: (i) NHS (N-hydroxysulfosuccinimide) and EDC (1-ethyl-3-(3-(dimethylamino)propyl) carbodiimide hydrochloride), and (ii) poly l-lysine (PLL) and glutaraldehyde (GA). Introduction
It is unclear why people of Mexican ancestry who smoke report relatively high rates of discussing quitting-related topics, which predicts cessation attempts.Methods
Using Ecological Momentary Assessment (EMA), adults from Mexico (n=40) and with Mexican heritage living in the US (n=52) who smoked daily were sent texts/emails each morning with cessation tips and cessation benefits over 15 days. At baseline, participants reported their personal network characteristics across up to eight different alters. Each evening of the 15 days, participants reported forgoing any cigarettes they normally would smoke (yes/no) and having had any quitting-related conversations (yes/no). Advances in computing and machine learning (ML) methods have led to a rapid rise in artificial intelligence (AI) research and applications in many fields. AI research benefitted from advances in computation hardware, collection and distribution of large data sets, and proliferation of software techniques. AI techniques include ML for provable results, deep learning for data exploration, reinforcement learning for control, and active learning for adaptive systems. Likewise, AI algorithms can handle large amounts of data, construct unknown representations, and provide a direct link between data and classification for decision making. Background
We examined fentanyl use and outcomes among people who inject drugs (PWID) in the urban Pacific Northwest (PNW).
Methods We conducted secondary analysis of data from the 2022 National HIV Behavioral Surveillance PWID cycle from Portland, OR and Seattle, WA. We calculated proportions of any fentanyl, injection fentanyl, non-injection fentanyl, and street pill use. Next, we compared characteristics, behaviors, and outcomes by any fentanyl use. Finally, we estimated adjusted relative risk (aRR) of overdose outcomes associated with fentanyl use and injecting less associated with type of street pill use.

Retinal ganglion cells (RGCs) form the only connection between the eye and the brain via their axons in the optic nerve. In diseases such as glaucoma, RGCs undergo irreversible neurodegeneration leading to vision loss. RGCs consist of multiple types differing in morphology, gene expression, and signaling properties. Moreover, these types exhibit differential survival after axon injury. What cellular properties vary between RGC types that could underlie their heterogeneous response to neurodegeneration? Given that energy metabolism is tightly optimized for the homeostatic and active functions of different neurons, physiologically distinct RGC types likely exhibit divergent energetic characteristics. Background: An emerging feature in modern biomedical research is collecting and analyzing numerous variables. In the presence of many potential covariates, inference becomes challenging requiring both distinguishing a set of covariates truly associated with an outcome and estimating their corresponding regression coefficients consistently. Traditional statistical inference typically focuses on estimating coefficients assuming a pre-specified set of covariates. Further, advanced machine/statistical learning methods performing both selection and estimation predominantly focus on outcome prediction rather than association inference.

Reared In Disaster: The Long-Run Harm From Childhood Storm Exposure, Lucas Kaplan

I estimate how exposure to natural disasters in childhood shapes earnings and schooling later in life, using newly assembled county-date disaster records dating back to 1951 linked to residential histories in the NLSY79. I compare adult outcomes for children who are born in the same county but due to disaster timing experience different amounts of exposure, finding that severe and repeat disasters reduce earnings. I show that school-age disaster exposure drives long-run harm: exposure between ages 5 and 10 reduces educational attainment, while exposure between ages 10 and 15 reduces adult earnings. The development of therapies that can be applied in human health care to promote longer and healthier lives is one of the main goals of aging research. However, many of the interventions currently available have serious adverse effects that limit their translational potential. Thus, new strategies are urgently needed to promote healthy aging in humans. Here, we reveal a functional interaction between the insulin signaling pathway and mTOR pathway that promotes healthy aging in C. elegans via lysosomal tubulation. Their respective transcription factors, DAF-16/FOXO and HLH-30/TFEB cooperate, to drive the formation of tubular lysosomes (TLs) under various contexts, which contributes …

BACKGROUND: This review assessed the cost-effectiveness and implementation strategies of hypertension management by non-physician healthcare workers (NPHCWs) in low- and middle-income countries (LMICs).
METHODS: A systematic search (inception-May 2024) included adults ≥18 years managed by NPHCWs LMICs, following Preferred Reporting Items for Systematic Reviews and Meta-analyses (PRISMA) guidelines. Economic evaluations were assessed using Drummond's checklist and ROBINS-I.
RESULTS: Seven studies (2002-2022) conducted across eight countries enrolled 96-10,000 participants and included randomized, modeling, observational, and quasi-experimental designs. NPHCWs included pharmacists, community and village health workers, and nurses. Patients' mean age ranged 58-71 years, with 57-82% female. Outcomes assessed included cost per …

An early event after lung injury is extracellular matrix (ECM) remodeling and the formation of a provisional matrix. While megakaryocytes and platelets (Mgk/plt) play important roles in hemostasis, their impact on the matrix in lung injury is not fully understood. Using lung intravital microscopy, 3D large area scanning with multiphoton confocal hybrid imaging, and label-free quantitative proteomics, the matricellular protein thrombospondin-1 (TSP1) arising from Mgk/plt protects the lung from alveolar injury. Mgk/plt cell-specific Thbs1 knockout (cKO) mice show increased alveolar barrier disruption with exaggerated neutrophil-mediated injury. BACKGROUND: Executive functioning (EF) difficulties are common in autistic children and may aggregate in families. Longitudinal studies examining EF development in autistic children and their siblings are limited. In the current study, we characterized EF development from later infancy through school-age in autistic and non-autistic high likelihood (HL; with an older autistic sibling) and low likelihood (LL; without older autistic sibling) children. METHODS: Participants were 83 autistic and 235 non-autistic HL children, and 132 LL children from the Infant Brain Imaging Study. PURPOSE: Partial nephrectomy is increasingly favored for small renal masses due to its renal function-preserving benefits. This study compared contemporary utilization patterns, perioperative outcomes, and hospital costs of robotic-assisted (RAPN), laparoscopic (LAPN), and open partial nephrectomy (OPN) among U.S. patients with renal cancer.

Objectives: In 2022, the FDA issued a drug safety communication based on case studies that transmucosal buprenorphine, a medication for opioid use disorder (MOUD), may contribute to dental disease. We sought to assess longitudinal dental care utilization patterns among patients with opioid use disorder (OUD) in the 18 months before and 36 months after MOUD initiation.

Organisms commonly respond to stressors in their environment by increasing the secretion of corticosterone (CORT). While increased CORT is acknowledged as a response to predation risk by organisms inhabiting a landscape of fear, new research raises the potential for changes in CORT as a response to the risk of infection by organisms in their environment. An increase in CORT of a healthy, uninfected individual in response to their perception of the risk of infection in their environment could change their susceptibility to infection, highlighting a significant role for CORT to alter host–pathogen dynamics. BACKGROUND AND OBJECTIVES: The Arms Race Control Score (ARCS) was developed to provide a quantitative assessment of publications among residency applicants. International medical graduates (IMGs) often have higher numbers of publications compared to U.S medical graduates (US MDs). However, there is limited knowledge of publication quality and effort among IMGs and US MDs. Our study analyzes publication productivity among IMGs and US MDs using the ARCS.
METHODS: Applicants were selected from ACGME-accredited training programs that enrolled both IMG and US MD residents over the 2018-2024 match cycle. Programs without IMG residents in a given year were excluded from the analysis. …

For over thirty years, patients have been visiting the Arkansas Epilepsy Program for diagnosis and treatment for seizures and seizure-like episodes. As part of their clinical evaluation, patients often undergo ambulatory EEG (electroencephalogram) monitoring. This routine process produces valuable data for treating the patient. In this study, over 2000 ambulatory EEG reports from 1998 to 2016 were reviewed. A large database of seizures was created from the reports, with information on 407 patients, 1611 EEG-confirmed seizures, and 1726 patient-reported seizures (IRB Protocol #17-093). Introduction: Effective intraoperative feedback is a cornerstone of surgical training, but remains inconsistently delivered. Surgeons who never received structured feedback during their own training lack the tools to deliver it effectively. At our institution, low faculty engagement scores on an Accreditation Council for Graduate Medical Education (ACGME) survey prompted a residency-wide needs assessment, which revealed that most attendings provide feedback perceived by residents as ineffective. To address this, we developed a simulation-based faculty development program grounded in the “Debriefing with Good Judgment” model.
