Small-conductance Ca2+-activated potassium channels subtype 2 (KCa2.2, also called SK2) are operated exclusively by a Ca2+-calmodulin gating mechanism. Heterozygous genetic mutations of KCa2.2 channels have been associated with autosomal dominant neurodevelopmental disorders including cerebellar ataxia and tremor in humans and rodents. Taking advantage of these pathogenic mutations, we performed structure-function studies of the rat KCa2.2 channel. No measurable current was detected from HEK293 cells heterologously expressing these pathogenic KCa2.2 mutants. When co-expressed with the KCa2.2_WT channel, mutations of the pore-lining amino acid residues (I360M, Y362C, G363S …

We use large eddy simulations to investigate the puff and droplet dynamics from violent expiratory events such as coughs and sneezes in the first few seconds following an ejection. For each of the eleven simulations considered, over 60,000 droplets are ejected and individually tracked using the point-particle Euler–Lagrange approach. We test the sensitivity of the puff and droplet dynamics to various parameters including the ejection volume, momentum, and orientation. We also explore the effect of the mouth shape on the aforementioned dynamics by considering elliptical and circular inlet cross-sections. The results from the simulations compare favorably with a recent theoretical …

BACKGROUND: Coronavirus disease 2019 (COVID-19) is associated with provoked thrombo-inflammatory responses. Early in the COVID-19 pandemic this was thought to contribute to hypercoagulability and multi-organ system complications in infected patients. Limited studies have evaluated the impact of therapeutic anti-coagulation therapy (AC) in alleviating these risks in COVID-19 positive patients. Our study aimed to investigate whether long-term therapeutic AC can decrease the risk of multi-organ system complications (MOSC) including stroke, limb ischemia, gastrointestinal (GI) bleeding, in-hospital and intensive care unit death in COVID-19 positive patients hospitalized during the early phase of the pandemic in the United States.
METHODS: A retrospective analysis …

Recent preclinical and clinical studies have highlighted the improved outcomes of combination radiotherapy and immunotherapy. Concurrently, the development of high-Z metallic nanoparticles as radiation dose enhancers has been explored to widen the therapeutic window of radiotherapy and potentially enhance immune activation. In this study, we evaluate folate-modified hafnium-based metal-organic frameworks (HfMOF-PEG-FA) in combination with imiquimod, a TLR7 agonist, as a well-defined interferon regulatory factor (IRF) stimulator for local antitumor immunotherapy. The enhancement of radiation dose deposition by HfMOF-PEG-FA and subsequent generation of reactive oxygen species (ROS) deregulates cell proliferation and increases apoptosis. HfMOF-PEG-FA loaded with imiquimod (HfMOF-PEG-FA@IMQ) increases DNA double-strand …

Blastoid B-cell neoplasms mainly include B-lymphoblastic leukemia/lymphoma (B-ALL), blastoid mantle cell lymphoma, and high-grade B-cell lymphoma with blastoid morphologic features (blastoid HGBL). Distinguishing blastoid HGBL from B-ALL can be challenging and we previously developed six-point flow cytometry-focused and three-point immunohistochemistry-focused scoring systems to aid in differential diagnosis. However, the six-point scoring system was derived from bone marrow cases and occasional cases may have a misleading score using either system. In this study, we assessed 121 cases of blastoid-HGBL (37 BM and 84 extramedullary) to validate the six-point scoring system in all tissue types and to further compare the two scoring …

Extracellular Matrix (Ecm) And Fibrosis In Adipose Tissue: Overview And Perspectives, Kai Sun, Xin Li, Philipp E Scherer
Faculty, Staff and Student Publications
Fibrosis in adipose tissue is a major driver of obesity-related metabolic dysregulation. It is characterized by an overaccumulation of extracellular matrix (ECM) during unhealthy expansion of adipose tissue in response to over nutrition. In obese adipose-depots, hypoxia stimulates multiple pro-fibrotic signaling pathways in different cell populations, thereby inducing the overproduction of the ECM components, including collagens, noncollagenous proteins, and additional enzymatic components of ECM synthesis. As a consequence, local fibrosis develops. The result of fibrosis-induced mechanical stress not only triggers cell necrosis and inflammation locally in adipose tissue but also leads to system-wide lipotoxicity and insulin resistance. A better understanding …

Prior theoretical and empirical studies have linked the first 3 years of children's life with later life outcomes. One primary explanation is the critical role these experiences play in children's early brain development (including their early language and cognitive abilities) and subsequent schooling achievement. The current study is a registered report with two complementary research objectives: (1) examine to what extent stimulating and responsive interactions with mothers and nonparental caregivers during the first 3 years of life show additive or synergistic associations with key socioeconomic outcomes in adulthood (i.e., educational attainment, salary, and employment status), and (2) examine to what …

Suicidal and self-injurious incidents in correctional settings deplete the institutional and healthcare resources, create disorder and stress for staff and other inmates. Traditional statistical analyses provide some guidance, but they can only be applied to structured data that are often difficult to collect and their recommendations are often expensive to act upon. This study aims to extract information from medical and mental health progress notes using AI algorithms to make actionable predictions of suicidal and self-injurious events to improve the efficiency of triage for health care services and prevent suicidal and injurious events from happening at California's Orange County Jails. …

Background: Previous studies have indicated that lower lung function is related to a higher risk of venous thromboembolism (VTE). However, causal inferences may be affected by confounders, coheritability or reverse causality. We aimed to explore the causal association between lung function and VTE. Methods: Summary data from public genome-wide association studies (GWAS) for lung function and VTE were obtained from published meta-analysis studies and the FinnGen consortium, respectively. Independent genetic variables significantly related to exposure were filtered as proxy instruments. We adopted linkage disequilibrium score regression (LDSC) and two-sample Mendelian randomization (MR) analyses to infer the genetic backgrounds and causal …

Bayesian Nonparametric Modelling Of Conditional Multidimensional Dependence Structures, R Barone, L Dalla Valle
School of Engineering, Computing and Mathematics
In recent years, conditional copulas, that allow dependence between variables to vary according to the values of one or more covariates, have attracted increasing attention. However, the literature mainly focused on the bivariate case, since the constraints on the multivariate copulas correlation matrices would make the specifications of covariates arduous. In high dimension, vine copulas offer greater flexibility compared to multivariate copulas, since they are constructed using bivariate copulas as building blocks. We present a novel inferential approach for multivariate distributions, which combines the flexibility of vine constructions with the advantages of Bayesian nonparametrics, not requiring the specification of parametric …

Estimating The Effects Of The Ada Amendments Act On The Hiring And Termination Of Individuals With Disabilities, Using New Disability Categorizations, Patrick Button, Philip Armour, Simon Hollands
Upjohn Institute Working Papers
Disability discrimination laws are often used to potentially increase employment for individuals with disabilities. However, legal theory and empirical economics research do not provide conclusive answers as to how expansions in disability discrimination laws affect economic outcomes, namely hiring rates, for individuals with disabilities. We estimate the effect of the ADA Amendments Act (ADAAA) on employment transitions: hirings and terminations for individuals with disabilities relative to those without disabilities. To calculate employment transitions, we use data from the Survey of Income and Program Participation (SIPP). We also use the SIPP to develop additional measures and categorizations of disability based on …

Leadership is a dynamic process with a far-reaching impact. Leadership is the process of guiding a group of individuals towards the achievement of a common goal. Healthcare leaderships looks to guide staff towards the common goal of positive patient outcomes. The literature has demonstrated that staff outcomes related to the health of their work environment can impact patient outcomes. A contributor to the health of the work environment is the relational tone of the unit; this tone is partly shaped by the skill and philosophy of the leader. Relational dynamics can influence several processes, including psychological safety which is a …
