Rate Of Tau Propagation Is A Heritable Disease Trait In Genetically Diverse Mouse Strains., Lindsay A Welikovitch, Simon Dujardin, Amy R Dunn, Analiese R Fernandes, Anita Khasnavis, Lori B Chibnik, Catherine Kaczorowski, Bradley T Hyman

Faculty Research 2023

The speed and scope of cognitive deterioration in Alzheimer's disease is highly associated with the advancement of tau neurofibrillary lesions across brain networks. We tested whether the rate of tau propagation is a heritable disease trait in a large, well-characterized cohort of genetically divergent mouse strains. Using an AAV-based model system, P301L-mutant human tau (hTau) was introduced into the entorhinal cortex of mice derived from 18 distinct lines. The extent of tau propagation was measured by distinguishing hTau-producing cells from neurons that were recipients of tau transfer. Heritability calculation revealed that 43% of the variability in tau spread was due …

Tax Noncompliance: The Role Of Tax Morale In Smokers’ Behavior, James Prieger Feb 2023

Tax Noncompliance: The Role Of Tax Morale In Smokers’ Behavior, James Prieger

School of Public Policy Working Papers

Measuring and predicting compliance with tax obligations is important but challenging. Survey data from California smokers are investigated to show that several forms of tax avoidance and evasion were common. About 43% of smokers avoided taxes by purchasing cigarettes outside the state in the previous year, 15% admit to evading taxes through cross-border purchases, and 26% bought likely or certainly untaxed cigarettes in the state in the past month. Attitudinal factors involving tax morale contributed more toward explaining the variance in compliance rates than demographic or law-and-economics factors. The implications for policy are discussed.

A Scoping Review Of Transgender Policies In The 15 Most Commonly Played Uk Professional Sports., Michael Mclarnon, Jane Thornton, Gail Knudson, Nigel Jones, Danny Glover, Andrew Murray, Michael Cummings, Neil Heron

Family Medicine Publications

INTRODUCTION: There has been much debate recently on the participation of transgender and gender-diverse (TGD) athletes in sport, particularly in relation to fairness, safety and inclusion. The 2021 IOC Framework on Fairness, Inclusion and Non-discrimination acknowledges the central role that eligibility criteria play in ensuring fairness, particularly in the female category, and states that athletes should not be excluded solely on the basis of their TGD identity.

AIMS: To identify policies that address TGD athlete participation in the 15 major United Kingdom (UK) sporting organisations and to summarise the evidence for each of these policies.

METHODS: A scoping review of …

Acetylcholinesterase Activity Staining In Freshwater Planarians, Christina Rabeler, Taixi Gong , '24, Danielle Ireland, O. Cochet-Escartin, Eva-Maria S. Collins

Biology Faculty Works

The serine hydrolase acetylcholinesterase (AChE) is an important neuronal enzyme which catalyzes the hydrolysis of the neurotransmitter acetylcholine and other choline esters. The breakdown of acetylcholine by AChE terminates synaptic transmission and regulates neuromuscular communication. AChE inhibition is a common mode of action of various insecticides, such as carbamates and organophosphorus pesticides. Freshwater planarians, especially the species Dugesia japonica, have been shown to possess AChE activity and to be a suitable alternative model for studying the effects of pesticides in vivo. AChE activity can be quantified in homogenates using the Ellman assay. However, this biochemical assay requires specialized equipment …

A Data-Driven Optimization Method For Coarse-Graining Gene Regulatory Networks., Cristian Caranica, Mingyang Lu

Faculty Research 2023

One major challenge in systems biology is to understand how various genes in a gene regulatory network (GRN) collectively perform their functions and control network dynamics. This task becomes extremely hard to tackle in the case of large networks with hundreds of genes and edges, many of which have redundant regulatory roles and functions. The existing methods for model reduction usually require the detailed mathematical description of dynamical systems and their corresponding kinetic parameters, which are often not available. Here, we present a data-driven method for coarse-graining large GRNs, named SacoGraci, using ensemble-based mathematical modeling, dimensionality reduction, and gene circuit …

Was Kaposi’S Sarcoma-Associated Herpesvirus Introduced Into China Via The Ancient Silk Road? An Evolutionary Perspective, Zhenqiu Liu, Qiwen Fang, Jialu Zuo, Veenu Minhas, Charles Wood, Na He, Tiejun Zhang

Nebraska Center for Virology: Faculty Publications

Kaposi’s sarcoma-associated herpesvirus (KSHV) has become widely dispersed worldwide since it was first reported in 1994, but the seroprevalence of KSHV varies geographically. KSHV is relatively ubiquitous in Mediterranean areas and the Xinjiang Uygur Autonomous Region, China. The origin of KSHV has long been puzzling. In the present study, we collected and analysed 154 KSHV ORF-K1 sequences obtained from samples originating from Xinjiang, Italy, Greece, Iran and southern Siberia using Bayesian evolutionary analysis in BEAST to test the hypothesis that KSHV was introduced into Xinjiang via the ancient Silk Road. According to the phylogenetic analysis, 72 sequences were subtype A …

Chemical and Biochemical Engineering Faculty Research & Creative Works

Pulmonary arterial hypertension (PAH) features pathogenic and abnormal endothelial cells (ECs), and one potential origin is clonal selection. We studied the role of p53 and toll-like receptor 3 (TLR3) in clonal expansion and pulmonary hypertension (PH) via regulation of bone morphogenetic protein (BMPR2) signaling. ECs of PAH patients had reduced p53 expression. EC-specific p53 knockout exaggerated PH, and clonal expansion reduced p53 and TLR3 expression in rat lung CD117+ ECs. Reduced p53 degradation (Nutlin 3a) abolished clonal EC expansion, induced TLR3 and BMPR2, and ameliorated PH. Polyinosinic/polycytidylic acid [Poly(I:C)] increased BMPR2 signaling in ECs via enhanced binding of interferon regulatory …

Background

Transcatheter aortic valve replacement (TAVR) is a relatively novel minimally invasive procedure for the treatment of symptomatic patients with severe aortic stenosis. Although it has been proven effective in improving mortality and quality of life, TAVR is associated with serious complications, such as acute kidney injury (AKI).

Summary

TAVR-associated AKI is likely due to several factors such as sustained hypotension, transapical approach, volume of contrast use, and baseline low GFR. This narrative review aims to present an overview of the latest literature and evidence regarding the definition of TAVR-associated AKI, its risk factors, and its impact on morbidity and …

Welcome Home: A Policy Briefing Book For Housing Opportunity In St. Louis County, Molly W. Metzger

Center for Social Development Research

“Equality of opportunity” is a beautiful American ideal, but it stands in stark relief to the history of racist policies in the United States—including policies in the St. Louis region. Edited by Molly Metzger, this briefing book aims to broaden opportunity by offering ideas for housing policy change in the majority-white, inner-ring suburbs of St. Louis. The policy works in this collection were written by masters of social work students at Washington University’s Brown School as part of a social-policy course taught by Metzger in partnership with Mayor Laura Arnold of Webster Groves, Missouri, Webster Groves’ Alliance for Interracial Dignity, …

Stress is associated with contextual memory deficits, which may mediate avoidance of trauma-associated contexts in posttraumatic stress disorder. These deficits may emerge from impaired pattern separation, the independent representation of similar experiences by the dentate gyrus-Cornu Ammonis 3 (DG-CA3) circuit of the dorsal hippocampus, which allows for appropriate behavioral responses to specific environmental stimuli. Neurogenesis in the DG is controlled by mitochondrial reactive oxygen species (ROS) production, and may contribute to pattern separation. In Experiment 1, we performed RNA sequencing of the dorsal hippocampus 16 days after stress in rats that either develop conditioned place avoidance to a predator urine-associated …

Efficacy Of A Combined Food-Response Inhibition And Attention Training For Weight Loss, Eric Stice, Sonja Yokum, Timothy D. Nelson, Elliot Berkman, Harm Veling, Natalia Lawrence

Department of Psychology: Faculty Publications

This Current Opinion in Behavioral Sciences article reviews trials that evaluated an obesity treatment that combines response-inhibition training with high-calorie foods and training designed to reduce attention for high-calorie foods. Two randomized controlled trials suggest that food-response inhibition and attention training produced significant body-fat loss, along with a reduction in valuation of, and reward-region response to, high-calorie foods. However, these effects did not emerge in a third trial, potentially because this trial used more heterogeneous food images, which reduced inhibition learning and attentional learning. Collectively, results suggest that food-response inhibition and attention training can devalue high-calorie foods and result in …
