TP53 mutations are found in 10% to 15% of myeloid neoplasms and are associated with a dismal prognosis. Although hypomethylating agents (HMAs), such as decitabine, are active in TP53-mutated myeloid neoplasms (TP53-MN), mutation clearance is rarely complete and nearly all patients relapse. Molecular determinants of response to HMAs in TP53-MN are poorly understood. Here, we show that decitabine induces replicative stress with decreased replication fork progression, induction of single-strand DNA breaks, and activation of the ataxia telangiectasia mutated-Rad3-related (ATR) pathway. Resolution of decitabine-induced replication stress is impaired in TP53-mutated acute myeloid leukemia (AML) cells, representing a potential therapeutic vulnerability. Indeed, …

Background/aim: Tumor budding (TB) and poorly differentiated clusters (PDCs) are histopathological parameters that have been associated with poor prognosis in various malignancies, particularly colorectal carcinoma. Although numerous studies have demonstrated an association between the microcystic, elongated, and fragmented (MELF) pattern in endometrial carcinomas and lymphovascular invasion as well as lymph node metastasis, the literature regarding TB and PDCs in this context remains limited. This study aimed to investigate the potential associations of the MELF pattern, TB, and PDCs with overall survival, progression-free survival, and clinicopathological parameters in cases of endometrioid endometrial carcinoma (EEC).

Acute Compartment Syndrome (ACS) is a limb- and life-threatening complication of lower leg fractures. Rapid recognition and fasciotomy can prevent irreversible ischemic injury, but ACS is often under-recognized. Vascular injury may increase the risk of ACS by impairing perfusion and exacerbating swelling. This study aimed to determine the incidence, risk factors, and outcomes of ACS in patients with lower leg fractures, with particular emphasis on the impact of vascular injury. This report provides a comprehensive analysis of South Carolina’s public school teacher workforce in 2024–25. Aligned with SC TEACHER’s mission, it offers data to support decision-making at both state and district levels. Using teacher-level information for 55,634 educators, this report describes key workforce characteristics and trends of classroom teachers (PK–12), special education teachers, and retired educators who have returned to the classroom.
In addition to exploring broad workforce patterns, the report highlights age profiles across different facets of the teacher workforce. Understanding the distribution of early-, mid-, and late-career educators is essential for anticipating retirement trends, identifying support needs for …

This study analyzes the spatiotemporal evolution and near-future trajectory of land surface temperature (LST) in Bizerte, a rapidly urbanizing Tunisian coastal governorate. Using multi-temporal Landsat imagery (2009–2024) processed in Google Earth Engine, we quantify bi-seasonal warming and project thermal conditions to 2029. Over 2009–2024, mean summer LST rose by 2.8 °C (to 41.7 °C) and winter LST by 2.5 °C (to 18.9 °C), with urban and bare-soil areas warming most intensely (+5.3 °C). Utah State University has developed a high-performance “green” hybrid propulsion technology based on the unique electrical breakdown properties of 3D-printed acrylonitrile butadiene styrene. Using 3D-printed ABS as fuel, typical startup sequences require approximately 5–15 joules and, once started, the system can be sequentially fired with no additional energy inputs required. The number of possible ignitions is limited only by the amount of fuel. The most technologically mature version uses gaseous oxygen (GOX) as oxidizer and 3D-printed ABS as fuel. While GOX is mass-efficient, it lacks volumetric efficiency unless highly pressurized. Nytrox, a blend of GOX and nitrous oxide, improves propellant …

Accurate quantification of atmospheric carbon dioxide (CO₂) fluxes in wetland ecosystems is essential for understanding their role in both regional and global carbon dynamics, particularly in the context of climate change. This study examines how Bangladeshi freelancers use digital platform work to cope with precarious local labor markets and how this strategy both mitigates and reproduces insecurity. In this form of work, freelancers compete for short-term, skill-based remote jobs posted by global clients. Drawing on 30 in-depth interviews and virtual ethnography, this study challenges dominant Western understandings of the precariat that assume workers fall from stable employment into insecure work. Instead, the findings show that many freelancers enter platform labor strategically as a way out of already unstable and limited local opportunities. Infestation of potato by the potato tuber moth (Phthorimaea operculella) varies markedly among cultivars, yet the chemical and molecular mechanisms underlying this variation remain poorly understood. Here, we combine field surveys, chemical ecology, functional genomics, and structural modeling to reveal how cultivar-specific volatile profiles shape moth oviposition behavior and pest outcomes. Field and laboratory assays identified marked differences in adult attraction, oviposition, and larval damage among potato cultivars, which correlated with distinct blends of emitted volatile organic compounds (VOCs). Background: Sarcopenia is a progressive skeletal muscle disorder increasingly seen in individuals with diabetes, leading to functional decline and poor quality of life. Despite Kerala's high diabetes burden, sarcopenia remains underrecognized. Background: Gingivitis is a common inflammatory disease of the periodontal tissues characterized by immune responses triggered by microbial biofilm. The inflammatory process involves the release of several cytokines and chemokines, including interleukin-1 beta (IL-1β ) and chemokine CXCL8, which play crucial roles in regulating leukocyte recruitment and inflammatory signaling within the gingival tissues. Proteome and transcriptome data combined can help assess the relevance of non-coding germline variants. Here, we combine germline Structural Variants (SVs) with mass spectrometry-based proteomics on tumors from 1637 cancer patients spanning various tumor tissues of origin to determine the extent SV breakpoint patterns involve differential protein expression of nearby genes. Rare and singleton SVs disrupting protein expression of known cancer susceptibility genes collectively involve 6% of patients. About 24% of the hundreds of genes with SV-associated non-coding cis-regulatory alterations at the mRNA level are similarly associated at the protein level. Background: Oscillatory shear stress (OSS), resulting from disturbed blood flow, is implicated in atherosclerotic plaque formation by incompletely understood mechanisms. This study aims to elucidate the involvement of death-associated protein kinase (DAPK) 2 in OSS-induced endothelial cell (EC) activation and atherosclerosis.
Methods: Publicly available resources, including genome-wide microarray, RNA sequencing, and single-cell RNA sequencing, were utilized to identify key OSS-sensitive regulatory factors. Background and objectives: Hypothalamic hamartomas (HH) are rare brain lesions associated with epilepsy and numerous comorbidities. Worldwide treatment is varied. There is a paucity of high-quality evidence to guide treatment. This study aimed to establish expert consensus on the evaluation and management of HH.
Methods: A modified Delphi survey was designed by the Medical Advisory Board of Hope for Hypothalamic Hamartomas and was conducted among 17 International League Against Epilepsy level II epilepsy surgery centers. To improve traffic safety, communities first need to know where serious crashes are actually happening. High-Injury Networks are designed to identify these locations, but they are usually built using only police-reported crash data. This research asks: Are we missing crashes—and injuries—by relying on police data alone? The research team first demonstrates that data from Emergency Medical Services and those from official databases differ substantially, and neither data set captures the full extent of collisions in a community. This doctoral capstone experience (DCE) was completed at Corewell Health Grand Rapids Hospitals Hand Therapy Clinic and focused on two ACOTE concentration areas including advanced clinical practice skills and education. The experience combined hands-on clinical practice in a specialized outpatient hand therapy setting with the development of high-quality client education materials and home exercise programs (HEPs). Objectives were created to address identified needs through the creation of client education handouts, development of diagnosis-specific Medbridge HEP templates, and creation of training resources to improve therapist efficiency with Medbridge use. Lung cancer remains the leading cause of cancer-related mortality worldwide, largely due to late-stage diagnosis. Low-dose computed tomography (LDCT) screening has demonstrated significant mortality reduction in high-risk populations; however, its widespread implementation is limited by high false-positive rates, inter-reader variability, and substantial workflow burden. Artificial intelligence (AI) has emerged as a promising adjunct to address these challenges by enhancing diagnostic consistency, efficiency, and risk stratification in LDCT-based screening. APOBEC3 enzymes are key effectors of antiviral immunity that introduce mutations into viral genomes. We show that viral R-loops serve as preferred substrates for APOBEC3-mediated mutagenesis during herpes simplex virus type 1 (HSV-1) infection. APOBEC3 enzymes are recruited to viral R-loops, generating clustered C-to-T mutations in genes critical for viral assembly. Importantly, R-loops alone are not mutagenic, and APOBEC3 enzymes do not edit HSV-1 without an R-loop; mutagenesis occurs when R-loops and APOBEC3 enzymes interact. These findings identify endogenous R-loops formed during the HSV-1 life cycle as focal vulnerabilities and highlight R-loop-APOBEC3 coupling as a mechanism for antiviral mutagenesis.

Forest fire and climate change are reshaping the snow hydrology of the Pacific Northwest of the United States, yet the combined effects of these disturbances on snow accumulation, storage, and melt timing remain poorly quantified in these Mediterranean mountain watersheds. This study integrates field-based snow and radiation observations with physically based modelling to evaluate how postfire forest structure, postfire snow albedo decay, and future climate change influence snow-water storage in the Breitenbush watershed of the western Oregon Cascades. This research aimed to identify the most common forms of bullying experienced by eighth-grade students, according to data from the TIMSS 2023, and those with the greatest impact on academic achievement, in Saudi Arabia, Japan, and Finland. The sample included all students who participated in the TIMSS 2023 study, totaling 15,667 students. The descriptive method, employing both causal-comparative and correlational approaches, was used to achieve the study’s objectives. The results showed that the most common form of bullying in Saudi Arabia, Japan, and Finland was “listening to hurtful things about one’s appearance,” with rates of 35.5%, 12.1%, and 24%, respectively. …

Drawing on Goffman’s concept of persona as a strategic and situational performance, the study examines written responses elicited through guided workplace scenarios designed to simulate high-stakes professional interaction on WhatsApp. The study adopted a qualitative, interpretive, discourse-pragmatic research design. The sample of the study included 30 women and 10 men employed in academic or administrative roles at the University of Bisha, and data were collected through semi‑structured, DCT‑style guided written prompts tailored to WhatsApp communication. The objectives of this research are to determine the effect of drawing based on story- telling toward language development and social development of children age 5–6 years.
