INTRODUCTION: Proteinuria is a modifiable risk factor for chronic kidney disease (CKD) progression in children. Finerenone, a selective, non-steroidal, mineralocorticoid receptor antagonist (MRA) has been approved to treat adults with CKD associated with type 2 diabetes mellitus (T2DM) following results from the phase III clinical trials FIDELIO-DKD (NCT02540993) and FIGARO-DKD (NCT02545049). In a pre-specified pooled analysis of both studies (N = 13,026), finerenone was shown to have an acceptable safety profile and was efficacious in decreasing the risk of adverse kidney and cardiovascular outcomes and of proteinuria.

OBJECTIVE: FIONA and the associated open-label extension (OLE) study aim to demonstrate that …

Tools that assess school supports for highly mobile, military-connected students are lacking. This study describes the development and preliminary validation of the Inventory of School Supports-Parent Report (ISS-PR). Participants were 433 parents (74% female; 62.5% White, 12% Black, 6.5% Asian, 5.5% Pacific Islander, 4% Native American, and 9.5% bi/multiracial; 19% Latinx) of students (grades 3–5) from families with an active-duty military parent. Parents completed the ISS-PR and rated: (a) how welcoming schools were toward military-connected families; (b) parent-teacher relationship quality; and (c) satisfaction with their child’s school. We created three proportional index composite scores: a 26-item school supports score, a …

Farber Institute for Neuroscience Faculty Papers

Small-molecule modulators of diverse voltage-gated K+ (Kv) channels may help treat a wide range of neurological disorders. However, developing effective modulators requires understanding of their mechanism of action. We apply an orthogonal approach to elucidate the mechanism of action of an imidazolidinedione derivative (AUT5), a highly selective positive allosteric modulator of Kv3.1 and Kv3.2 channels. AUT5 modulation involves positive cooperativity and preferential stabilization of the open state. The cryo-EM structure of the Kv3.1/AUT5 complex at a resolution of 2.5 Å reveals four equivalent AUT5 binding sites at the extracellular inter-subunit interface between the voltage-sensing and pore domains of the …

Objectives: The Global Aging & Geriatric Experiments in Bipolar Disorder Database (GAGE-BD) project pools archival datasets on older age bipolar disorder (OABD). An initial Wave 1 (W1; n = 1369) analysis found both manic and depressive symptoms reduced among older patients. To replicate this finding, we gathered an independent Wave 2 (W2; n = 1232, mean ± standard deviation age 47.2 ± 13.5, 65% women, 49% aged over 50) dataset. Design/Methods: Using mixed models with random effects for cohort, we examined associations between BD symptoms, somatic burden and age and the contribution of these to functioning in W2 and the …

Building Community Through Learning Traditional Irish Music, Laura Olson Mar 2024

Building Community Through Learning Traditional Irish Music, Laura Olson

Music ETDs

This thesis foregrounds traditional Irish music and the role it plays in building community. I demonstrate how the genre is defined and transmitted plays an important role in how inclusive the genre can be and where changes need to be made to be more welcoming to new individuals. Playing or singing in musical groups has been shown to combat loneliness but there has been little focus on specific music genres, especially those that are instrumental in nature. This thesis shows that instrumental music can be another tool to combat loneliness and promote overall health and wellbeing using the vehicle of …

Flowering Of Spring-Planted Greens, Elizabeth Maynard, Marian M. Rodriguez-Soto, Rebecca Koetz

Midwest Vegetable Trial Reports

Greens for salad or cooking are a significant part of the spring crop mix sold at Indiana farmers' markets. The harvest period for many spring-planted greens is shortened because they flower in response to environmental conditions. This paper reports on flowering response of two varieties of arugula, collards, mizuna, two types of mustard, and shungiku, planted in mid-April through May in northern Indiana.

Stare Decisis And Remedy, Melissa Murray Mar 2024

Stare Decisis And Remedy, Melissa Murray

Duke Law Journal

Much ink has been spilled on the Roberts Court’s approach to stare decisis and precedent. Such commentary is hardly surprising. In just the last five years, the Court has overruled extant precedents on issues that range from abortion and jury convictions to property rights and public unions. It has also substantially narrowed and limited existing precedents, curbing the reach of earlier decisions in ways that disrupt and distort the jurisprudential landscape.

Some view the Court’s uneven approach to precedent as ideologically determined. As these critics maintain, the Court adheres to precedents that are consistent with the views of its six-member …

The Return Of Three-Judge Constitutional Courts, Matt Queen Mar 2024

The Return Of Three-Judge Constitutional Courts, Matt Queen

Duke Law Journal

State courts wield the authority to elevate state constitutional protections above those afforded by the U.S. Constitution. That power is great—so great that some legislatures have intervened in constitutional adjudication, purportedly to undermine forum shopping and check a single judge’s influence. Accordingly, North Carolina and Tennessee require that three-judge trial courts hear constitutional challenges to state laws. These courts echo twentieth-century congressional efforts to trim federal courts’ equitable jurisdiction. They also present new and familiar drawbacks spawned by their federal ancestors.

This Note examines these new constitutional courts through several lenses: their historical context, political development, advantages, and drawbacks. Although …

Publications

INTRODUCTION: Many people, often older adults, living in long-term care homes (OA-LTCH) became socially isolated during the COVID-19 pandemic due to variable restrictions on in-person visits and challenges associated with using technology for social connectivity. Health providers were key to supporting these OA by providing additional care and facilitating their connections with family using technology such as smartphones and iPads. It is important to learn from these experiences to move forwards from the COVID-19 pandemic with evidence-informed strategies that will better position health providers to foster social engagement for OA-LTCH across a range of contextual situations. OBJECTIVE: This exploratory qualitative …

An Investigation Of The Validity Of A Framework Constructed To Predict And Facilitate Upward-Stability, Timothy Streett Mar 2024

An Investigation Of The Validity Of A Framework Constructed To Predict And Facilitate Upward-Stability, Timothy Streett

Graduate Theses, Dissertations, and Capstones

This study is an investigation of the validity of the theoretical assumptions of a programming model which posits that it is not income but rather a collection of personality and dispositional constructs which provide the foundation of an economically stable life, and that empowering those in poverty to improve their economic situation must begin with these constructs or will be a failed effort. The author provides a review of significant literature supporting the idea that these constructs are associated with better outcomes in many aspects of life. The author then provides original investigative research utilizing a self-administered, 40 item agree/disagree …

Spinocerebellar ataxia type 1 (SCA1) is a fatal neurodegenerative disease caused by an expanded polyglutamine tract in the widely expressed ataxin-1 (ATXN1) protein. To elucidate anatomical regions and cell types that underlie mutant ATXN1-induced disease phenotypes, we developed a floxed conditional knockin mouse (f-ATXN1146Q/2Q) with mouse Atxn1 coding exons replaced by human ATXN1 exons encoding 146 glutamines. f-ATXN1146Q/2Q mice manifested SCA1-like phenotypes including motor and cognitive deficits, wasting, and decreased survival. Central nervous system (CNS) contributions to disease were revealed using f-ATXN1146Q/2Q;Nestin-Cre mice, which showed improved rotarod, open field, and Barnes maze performance by 6-12 weeks of age. In contrast, …

Somatostatin (SST)-producing pancreatic delta-cells play an important role in maintaining the balance of insulin and glucagon secretion within the islets. This study aimed to generate a human embryonic stem cell (hESC) line with a SST-P2A-mCherry reporter using CRISPR/Cas9 system. The SST-P2A-mCherry reporter cell line was shown to maintain typical pluripotent characteristics and able to be induced into SST-producing pancreatic delta-cells. The generation of the cell line would provide useful platform for the characterization of stem cell-derived delta-cells, discovery of delta-cell surface markers and investigation of paracrine mechanisms, which will ultimately promote the drug discovery and cell therapy of diabetes mellitus.

Comparative Assessment Of Gold Nanoparticle-Antibody Conjugates With Two Differently Shaped Particles For Multimodal Colorimetric Lateral Flow Assay, Vinayak Sharma, Bilal Javed, Furong Tian

Articles

Novel detection strategies that exploit the unique properties of gold nanoparticles (AuNPs) hold great potential for the advancement of point-of-care (POC) diagnostics, such as lateral flow and dipstick immunoassay [1–3]. Owing to its biocompatibility and convenient surface modifications, gold nanoparticles with enhanced sensitivity reduce the sample-to-answer times to less than 5 min (Figure 1). Methods like covalent conjugation and electrostatic adsorption are explored for antibody immobilization to develop a gold nanoparticle-based biosensor for fast and sensitive localized surface plasmon resonance (LSPR) for both basic research and clinical diagnostics. The mounting of antibodies on the gold nanoparticle surface requires selective binding …

ATP synthase inhibitory factor subunit 1 (IF1) is an inhibitory subunit of mitochondrial ATP synthase, playing a crucial role in regulating mitochondrial respiration and energetics. It is well-established that IF1 interacts with the F1 sector of ATP synthase to inhibit the reversal rotation and, thus, ATP hydrolysis. Recent evidence supports that IF1 also inhibits forward rotation or the ATP synthesis activity. Adding to the complexity, IF1 may also facilitate mitophagy and cristae formation. The implications of these complex actions of IF1 for cellular function remain obscure. In the present study, we found that IF1 expression was markedly upregulated in hypoxic …

The Past As A Colonialist Resource, Deepa Das Acevedo

Duke Law Journal

Originalism’s critics have failed to block its rise. For many jurists and legal scholars, the question is no longer whether to espouse originalism but how to espouse it. This Article argues that critics have ceded too much ground by focusing on discrediting originalism as either bad history or shoddy linguistics. To disrupt the cycle of endless “methodological” refinements and effectively address originalism’s continued popularity, critics must do two things: identify a better disciplinary analogue for originalist interpretation and advance an argument that moves beyond methods.

Anthropology can assist with both tasks. Both anthropological analysis and originalist interpretation are premised on …

Glioblastoma, the most common primary brain tumor, has a 6.8% survival rate 5 years post diagnosis. Our team developed an oncolytic adenovirus with an OX-40L expression cassette named Delta-24-RGDOX. While studies have revealed the interaction between the gut microbiota and immunotherapy agents, there are no studies linking the gut microbiota with viroimmunotherapy efficacy. We hypothesize that gut bacterial signatures will be associated with oncolytic viral therapy efficacy. To test this hypothesis, we evaluated the changes in gut microbiota in two mouse cohorts: (1) GSC-005 glioblastoma-bearing mice treated orally with indoximod, an immunotherapeutic agent, or with Delta-24-RGDOX by intratumoral injection and …
