Induced Reduction Of N-Glycosylation Leads To Heart Failure, Anthony M. Young Jan 2025

Induced Reduction Of N-Glycosylation Leads To Heart Failure, Anthony M. Young

Browse all Theses and Dissertations

Cardiovascular disease is the leading cause of death and contributes to the increasing global prevalence of heart failure (HF). N-glycosylation is a common co-/post-translational modification where branching oligosaccharides are bound to the extracellular domain of membrane proteins. This creates a diverse range of glycan structures and requires the coordination of hundreds of regulated genes. Inherited mutations in glycan synthesis frequently present with cardiomyopathies leading to HF with reduced ejection fraction (HFrEF). Additionally, gene expression studies of HFrEF patients have shown altered expression of glycosylation-related genes, including alpha-1,3-mannosyl-glycoproten 2-beta-N acetlyglucosaminyltransferase (Mgat1). This gene encodes N-acetylglucosaminyl transferase 1 (GlcNAcT1) which is required …

The Impact Of Ahr And Hs1.2 Enhancer Genetic Variations On Igh Expression And Antibody Production In Human B Cells, Mili Santosh Bhakta-Yadav

Browse all Theses and Dissertations

Antibody production is an essential component of the immune response against pathogens. The immunoglobulin heavy chain (IgH) gene codes for the heavy chain of antibodies. The IgH constant regions Cμ, Cδ, Cγ1-4, Cα1-2, and Cε encode the five major classes of antibodies, i.e., IgM, IgD, IgG1-4, IgA1-2, and IgE, respectively. The transcription of the IgH gene and class switch from IgM to other isotypes is regulated by two 3’ IgH regulatory regions (3’IgHRRs), each of which is a cluster of three enhancer regions (hs3, hs1.2 and hs4). The genetic variations in the hs1.2 enhancer have been identified; a ~53 bp …

Characterizing The Microvesicle Particles From Keratinocytes Stimulated With Multiple Stressors, Muslima Begum

Browse all Theses and Dissertations

Many types of environmental and physical stressors act upon the skin and can initiate inflammation, systemic effects, and even damage to internal organs. Recent studies have identified that these harmful events are driven by the release of subcellular microvesicle particles (MVPs), an inflammatory process promoted by the glycerophosphocholines-derived lipid mediator, Platelet-activating Factor (PAF). These harmful effects are driven by the release of MVPs, a process promoted by the inflammatory lipid mediator, PAF. We have previously demonstrated that exposure of human HaCaT keratinocyte and mouse skin to stressors such as thermal burn injury, ultraviolet B (UVB) radiation, and topical photodynamic therapy …

Wrongful Birth After Dobbs And The Limits Of Tort Law In Areas Of Dissensus, E. Travis Ramey Jan 2025

Wrongful Birth After Dobbs And The Limits Of Tort Law In Areas Of Dissensus, E. Travis Ramey

Articles

As the abortion debate has raged in this country, a secondary debate over wrongful birth has accompanied it. Wrongful birth is a medical-malpractice-like claim brought by parents who allege that a medical provider negligently failed to provide accurate information about the fetus and that had they received accurate information they would have terminated the pregnancy. Courts rejected the tort theory until Roe v. Wade was decided. Post-Roe, courts were heavily divided over whether to recognize the claim and what damages were recoverable. Now, after Dobbs v. Jackson Women's Health Organization, the continued viability of wrongful birth is in question, and …

Promoting Financial Empowerment Via 401(K) Plan Domestic Abuse Victim Distributions, Samantha Prince

Faculty Scholarship

Domestic violence is sadly and shockingly all too prevalent in the United States. According to the U.S. Centers for Disease Control, more than one in four women and one in seven men in this country are subject to domestic abuse “affecting an estimated 10 million people every year.”

Finances and financial abuse play a significant role in 99% of domestic abuse cases. “[L]acking financial knowledge or resources is the number one indicator of whether a domestic violence victim will stay, leave, or return to an abusive relationship.”When abusers have control over financial assets, victims are monetarily paralyzed and have little …

Taking From The Most Vulnerable: Legal Implications Of Seizing Federal Benefits From Foster Kids, Lucy Johnston-Walsh Jan 2025

Taking From The Most Vulnerable: Legal Implications Of Seizing Federal Benefits From Foster Kids, Lucy Johnston-Walsh

Faculty Scholarship

Foster youth in some states are paying the cost of their own room and board when state and local government agencies seize a child's federal social security benefits to cover the costs of care. Other states, however, have passed laws forbidding such a practice, allowing youth to save these funds for their own financial needs. Federal social security benefits are an individual's own property and should not be taken without adequate notice, nor without an opportunity to challenge the taking. Tet too many youth who are involved in the foster care system are unaware they are even receiving benefits and …

How Can We Go Far Together In Healthcare Education And Training?, Nader Nadershahi Jan 2025

How Can We Go Far Together In Healthcare Education And Training?, Nader Nadershahi

Journal of the American College of Dentists

“If you want to go fast, go alone. If you want to go far, go together.” This African proverb highlights the importance of collaboration and cooperation to achieve long-term success and impact. This sentiment applies to many aspects of life, and it certainly applies to our reflection on the importance of breaking down the silos that have developed in healthcare. Our healthcare system has been siloed in all aspects, including delivery models, payment systems, education, and training.

A Constitutional False Claims Act, Benjamin Mcmichael, Mackenzi Barrett, W. Kip Viscusi Jan 2025

A Constitutional False Claims Act, Benjamin Mcmichael, Mackenzi Barrett, W. Kip Viscusi

Articles

The False Claims Act (FCA) represents one of the most important sources, if not the most important source, of liability in the healthcare system and other industries that routinely provide goods and services to the federal government. Originally designed to police fraud during the Civil War, the FCA has become a general statute to enforce many other complex legal schemes. Because failure to comply with complicated statutes and regulations can lead to a reimbursement claim being defined as false under the FCA, the FCA serves as a blunt instrument to cudgel those who fail to comply with the minutiae of …

Unfairness, Reconstructed, Luke Herrine Jan 2025

Unfairness, Reconstructed, Luke Herrine

Articles

A paradigm shift is afoot at major federal consumer protection agencies. For four decades, a bipartisan bloc of bureaucrats has seen the purpose of consumer protection as promoting informed consumer choice or "consumer sovereignty." The idea was that informed consumers in competitive markets would protect themselves by choosing among sellers. Ensuring access to information would then shore up markets' self-correcting tendencies without requiring moral judgment. In the past few years, by contrast, regulators have prioritized sector-wide regulation, enforcement sweeps, and strategic cases against market leaders. They have justified their actions not ( exclusively) in terms of informed choice or efficiency …

Categorizing Clinical Response To Mepolizumab For Patients With Chronic Rhinosinusitis With Nasal Polyps: Synapse Trial, Claire Hopkins, Larry Borish, Jared Silver, Peter Howarth, Robert Chan, Priya Dsilva, Joseph K. Han

Department of Otolaryngology (ENT) Faculty Publications

[Introduction] Chronic rhinosinusitis with nasal polyps (CRSwNP) can substantially affect quality of life (QoL), and is often challenging to manage.¹ First-line treatment for the management of CRSwNP involves intranasal corticosteroids, with short courses of systemic corticosteroids (SCS) for exacerbations, and sinus surgery for uncontrolled disease.² Biologics targeting type 2 inflammation (eg, dupilumab, omalizumab, and mepolizumab) have become a treatment option for refractory CRSwNP. With the advent of disease-modifying treatments such as biologics, control, defined as patients reporting no/low-symptom levels, has increasingly become the treatment goal for CRSwNP.³ However, the optimal use of biologics for symptom control in individual patients remains …

Acute Exacerbations Of Chronic Rhinosinusitis, Robert M. Frederick, Kent Lam, Joseph K. Han Jan 2025

Acute Exacerbations Of Chronic Rhinosinusitis, Robert M. Frederick, Kent Lam, Joseph K. Han

Department of Otolaryngology (ENT) Faculty Publications

Purpose of Review

We aim to highlight recent advancements on the evolving chronic rhinosinusitis (CRS) phenotype: acute exacerbations of chronic rhinosinusitis (AECRS). We focused on studies that expanded the current understanding of its pathophysiology, patient characteristics, and disease burden.

Recent findings

Defining AECRS has been a topic of discussion for many years. A recent regulatory definition of AECRS in the literature incorporates a > 3 day requirement of worsened symptoms and an escalation of treatment. It is important not to rely on patient-reported rescue medication frequency as it was recently demonstrated these are only obtained for 1/3 of reported AECRS episodes. …

Causal Discovery On The Effect Of Antipsychotic Drugs On Delirium Patients In The Icu Using Large Observational Ehr Dataset, Riddhiman Adib, Md. Osman Gani, Sheikh Iqbal Ahamed, Mohammad Adibuzzaman

Computer Science Faculty Research and Publications

Delirium occurs in about 80% of cases in the Intensive Care Unit (ICU) and is associated with an extended hospital stay, increased mortality, and other complications. Delirium lacks biomarker-based diagnosis and is frequently treated with antipsychotic drugs (APD), despite numerous studies debating its efficacy. Since randomized controlled trials (RCT) are expensive and time-consuming, we approach the research question of estimating the efficacy and safety outcomes of APD in treating delirium through retrospective cohort analysis. We employed the Causal inference framework to explore the underlying causal model for Delirium patient cohort. We focus on building a structural causal model for delirium …

Using A Dimension Reduction Approach To Parse Heterogeneity In Dietary Restriction, Amanda Makara Jan 2025

Using A Dimension Reduction Approach To Parse Heterogeneity In Dietary Restriction, Amanda Makara

Theses and Dissertations

Dietary restriction, the problematic reduction of caloric or macronutrient intake, is a critical eating behavior occurring transdiagnostically across eating disorders and subclinical disordered eating. The definition of dietary restriction is highly heterogeneous, and restriction is often conflated with other distinct constructs, such as dietary restraint. The current practices related to the definition and measurement of dietary restriction have notable flaws, negatively impacting the ability to identify and establish risk, diagnosis, and treatment of eating disorders. There is also limited evidence that commonly-used dietary restriction measures can validly predict food choice and objective intake. These issues raise questions about whether dietary …

How South Dakota Can Criminalize Child Sexual Grooming, Jackson Marnette Jan 2025

How South Dakota Can Criminalize Child Sexual Grooming, Jackson Marnette

South Dakota Law Review

In 2023, the South Dakota Legislature proposed and rejected Senate Bill 94, which would have criminalized child sexual grooming. South Dakota, like most states, lacks a law that effectively deters potential child sexual abusers from initiating the child sexual abuse process. Other states’ grooming laws have been challenged on constitutional grounds with differing results. This article considers the benefits of grooming laws, the pros and cons associated with other states’ passed legislation, and any issues with Senate Bill 94’s language. The article concludes with a legislative proposal based on child sexual grooming academic theory.
