Department of Pharmacology, Physiology, and Cancer Biology Faculty Papers

Histone methyltransferase NSD2 (MMSET) overexpression in multiple myeloma (MM) patients plays an important role in the development of this disease subtype. Through the expansion of transcriptional activating H3K36me2 and the suppression of repressive H3K27me3 marks, NSD2 activates an aberrant set of genes that contribute to myeloma growth, adhesive and invasive activities. NSD2 transcriptional activity also depends on its non-catalytic domains, which facilitate its recruitment to chromatin through histone binding. In this study, using NMR, ITC and molecular dynamics simulations, we show that the tandem PHD domain of NSD2 (PHDVC5HCHNSD2) is a combinatorial reader of unmodified histone H3K4 and tri-methylated H3K27 …

Johann Georg Hamann And The Liberal Project, Jesse D. Ansley Jan 2025

Johann Georg Hamann And The Liberal Project, Jesse D. Ansley

LSU Doctoral Dissertations

This dissertation examines the political thought of Johann Georg Hamann, his relationship to liberalism, and the importance to modern concerns for consensus formation and intercultural dialogue. This contributes to the line of literature sparked by Isaiah Berlin debating the political legacy of Hamann and its relation to the development of modern politics. Berlin labels Hamann an anti-rationalist whose current developed into the ideology of the Nazi party. My work supports the countervailing arguments that Hamann’s work is importantly different from the Romantics he inspired and even more so from the Nazis. By examining two complete works and a number of …

There are no evidence-based management guidelines for the management of factor X deficiency due to limited literature. Detailed history, clinical examination and specialized tests (coagulation profile, mixing studies, factor assay) are required for the diagnosis of factor X deficiency. In this case report, we are discussing the case of a 1-month-old, 12-day-old female child who presented with the complaint of spontaneous bleeding from the nose since 1 day with no other bleeding manifestations from …

Sialylation Of Tnfr1 By St6gal1 Modulates Receptor Dynamics And Determines Cell Fate, Jihye Hwang Jan 2025

Sialylation Of Tnfr1 By St6gal1 Modulates Receptor Dynamics And Determines Cell Fate, Jihye Hwang

All ETDs from UAB

Abnormal glycosylation has long been identified as a key feature of cancer. Aberrant glycan structures on tumor cells arise from the dysregulation of select glycosyltransferases. For example, sialyltransferase ST6GAL1 is notable for its high expression in various cancers, including breast, colon, ovarian, and pancreatic. ST6GAL1 introduces α2-6-linkage sialylation to N-glycans on proteins, endowing cancer cells with stem cell-like properties. This modification contributes to resistance to apoptosis mediated by death receptors like TNFR1. Despite its significance in cancer, the regulatory mechanisms controlling ST6GAL1 expression remain largely unexplored. Additionally, it is unclear whether the apoptosis-resistant effect of sialylation is caused by an …

Characterizing The Molecular Complexities Of Iga1 By Serum Proteomics In The Context Of Iga Nephropathy, Mary A. Cunningham Jan 2025

Characterizing The Molecular Complexities Of Iga1 By Serum Proteomics In The Context Of Iga Nephropathy, Mary A. Cunningham

All ETDs from UAB

Immunoglobulin A1 (IgA1) is the second most abundantly produced immunoglobulin in circulation per day and plays a key role in human immunity. Two molecular complexities of IgA1 make accurately studying the protein challenging. The first is that IgA1 exists in three main molecular forms in the serum of humans: monomeric (mIgA1, 80-90% of serum IgA1), polymeric (pIgA1, 10-20% of serum IgA1), and IgA1-containing immune complexes (IgA1-IC, <0.5% of serum IgA1). The second complexity is that the hinge-region of IgA1 is extensively O-glycosylated and exhibits a high level of variation in its O-glycans, particularly in the degree of sialylation. In context of the autoimmune disease IgA nephropathy (IgAN), patients have elevated levels of IgA1 that has reduced levels of galactose (Gal) in the IgA1 hinge-region (Gal-deficient IgA1). This Gal-deficient IgA1 forms immune complexes with IgG autoantibodies (IgA1-IC), deposits in the glomeruli, and induces kidney injury. While it is established that these large IgA1-IC (>700 kDa) formed in the circulation are pathogenic, there remains a gap in knowledge about other blood proteins that may be associating with IgA1 in complexes. It is also necessary to understand the native …

Neural Mechanisms Of Proactive And Reactive Cognitive Control In Humans, Anas Khan Jan 2025

Neural Mechanisms Of Proactive And Reactive Cognitive Control In Humans, Anas Khan

All ETDs from UAB

Cognitive control enables flexible, goal-directed behavior by dynamically adjusting our actions in response to changing task demands. This dissertation investigates the neural mechanisms underlying proactive and reactive cognitive control, particularly focusing on the dorsolateral prefrontal cortex (dlPFC) and the dorsal anterior cingulate cortex (dACC) during tasks that require response inhibition and conflict processing. Using intracranial electroencephalography (iEEG) recordings from human participants, this research examines the distinct roles of oscillatory activity in these regions in both short-term reactive adjustments and long-term proactive control strategies. Chapter 2 explores the role of low-frequency oscillations in the mid-rostral dlPFC during motor inhibition, revealing that …

Development Of A Lifestyle Wellness Assessment For Obesity Treatment-Seeking Adults, Katie M. Ellison

All ETDs from UAB

Obesity is strongly associated with, and effective at, preventing and managing cardiometabolic disease but proponents of Health at Every Size® approaches challenge the importance of weight loss for improving health. While some patients want to lose weight, there is evidence that others desire to focus on improving broader aspects of health and wellness such as metabolic, fitness, and psychosocial health. Clinicians may benefit from a comprehensive understanding of the multifactorial benefits of obesity treatments to effectively capture overall patient wellness. However, existing wellness assessments are often either not comprehensive, unreliable, or subject to time, space, or cost constraints in clinical …

A Biomechanical Comparison Of Varying Peg Angles On Glenoid Implant Stability For Anatomic Total Shoulder Arthroplasty, Erica Nicole Schilling

All ETDs from UAB

Anatomic total shoulder arthroplasty (ATSA) is a surgical procedure in which both the humeral head and the glenoid cavity are replaced with implants resembling the natural anatomy of the shoulder. Several complications may occur after an ATSA, but the most common is glenoid loosening, which makes up about 37.7% of these complications. The overall structure of a glenoid implant is a concave oval plate commonly manufactured from ultra-high molecular weight polyethylene (UHMWPE) with various stabilizing structures, including pegs, protruding from the back of the implant. For this study, the focus was on Catalyst OrthoScience’s pegged glenoid implants and how the …

Changes in epigenetic processes such as histone acetylation are proposed as key events influencing cancer cell function and the initiation and progression of pediatric brain tumors. Valproic acid (VPA) is an antiepileptic drug that acts partially by inhibiting histone deacetylases (HDACs) and could be repurposed as an epigenetic anticancer therapy. Here, we show that VPA reduced medulloblastoma (MB) cell viability and led to cell cycle arrest. These effects were accompanied by enhanced H3K9 histone acetylation (H3K9ac) and decreased expression of the MYC oncogene. VPA impaired the expansion of MB neurospheres enriched in stemness markers and reduced MYC while increasing TP53 …

Spontaneous preterm birth (sPTB) poses significant challenges, affecting neonatal health and neurodevelopmental outcomes worldwide. The specific effects of placental trophoblasts on the pathological development of sPTB subtypes-preterm premature rupture of fetal membranes (pPROM) and spontaneous preterm labor (sPTL)-are not fully understood, making it crucial to uncover these impacts for the development of effective therapeutic strategies. Using single-nucleus RNA sequencing, we investigated transcriptomic and cellular differences at the maternal-fetal interface in pPROM and sPTL placentas. Our findings revealed distinct trophoblast compositions with pPROM characterized predominantly by extravillous trophoblasts (EVTs), while sPTL showed an abundance of syncytiotrophoblasts (STBs). Through cell differentiation and …

Two-way dual language (TWDL) programs have traditionally served white and non-Black Latinx students, with limited attention to other cultures and races. This limitation can result in the erasure of Afro-Latinx culture from bilingual programs. This study explores the ways Black characters are represented in bilingual picturebooks, which can serve to perpetuate or combat anti-blackness within bilingual education. We completed a critical content analysis of Spanish-English bilingual picturebooks that include Black or Afro-Latinx characters, employing LatCrit and BlackCrit theoretical frameworks to explore how Black characters are positioned. Findings show a significant dearth of bilingual books that include Black or Afro-Latinx characters. …

Method For Minimizing Connector Stress When Installing Field-Replaceable Units Across Multiple Boards Simultaneously, Anonymous Jan 2025

Method For Minimizing Connector Stress When Installing Field-Replaceable Units Across Multiple Boards Simultaneously, Anonymous

Defensive Publications Series

This disclosure addresses the challenge of aligning multiple PCBs—such as a backplane and multiple switch cards—with a single Field Replaceable Unit (FRU) during installation. Traditionally, fixed mounting of all PCBs can cause misalignment and introduce harmful shear forces on connector pins, leading to deformation, signal degradation, or permanent damage. By fixing the backplane and allowing the FRU to self-align within its rail-mounted tolerances, the system ensures proper initial positioning. Additionally, each switch card is mounted on elastic neoprene grommets, enabling the card to “float” and adjust its position in any direction. This innovative approach accommodates manufacturing tolerances, reduces stress on …
