Background: Posttraumatic stress disorder (PTSD) is a mental disorder that may occur in the aftermath of severe psychological trauma. Epigenetic changes in the brain may play a critical role in understanding the neurobiology of PTSD by linking environmental traumatic stress exposure to lasting alterations in gene expression that shape neuronal function.
Methods: We examined 1,065,750 DNA methylation (DNAm) sites from 171 donors including neurotypical controls and PTSD and major depressive disorder (MDD) cases across 6 regions implicated in the fear circuitry of the brain. Historic and recent elevation data for the glaciers in the conterminous US were differenced to estimate glacier mass and volume change.
Historic glacier elevations were obtained from the USGS National Elevation Dataset (https://doi.org/10.15760/geology-data.04), a digitized product from the USGS 1:24,000 paper topographic maps that included the glacier extent (Gesch, 2007). Recent glacier elevations were derived from aerial lidar and from satellite radar. The lidar data were largely obtained from the USGS 3D Elevation Program (Stoker and Miller, 2022; US Geological Survey, 2024), and filtered by acquisition date using only those data collected between mid-summer and early autumn. Background: The first international clinical standards of care for the gender-diverse population were formulated in the late 1970s. In the last 15 years, multiple subspecialty societies within the United States have developed clinical care guidelines for those with gender dysphoria, including gender-affirming hormone therapy (GAHT) and surgical treatments. To date, there are no pathology- and laboratory medicine-specific recommendations in the United States for the gender-diverse population.

Total energy expenditure (TEE) is commonly modelled as the sum of resting energy expenditure (REE), activity energy expenditure (AEE), and diet-induced thermogenesis (DIT). This additive model has recently been challenged by the constrained energy balance model, which proposes that reductions in mass-adjusted REE compensate to some extent for increases in AEE, rendering mass-adjusted TEE effectively invariant. We tested these competing models using two complementary approaches. Background: Necator americanus glutathione S-transferase-1 (Na-GST-1) performs a crucial enzymatic step in hookworm blood feeding and is a leading target for vaccine development. Phase 1 trials in Brazil, Gabon, and the USA of recombinant Na-GST-1 adsorbed on Alhydrogel (Na-GST-1/Al), co-administered with or without AP 10-701 (Toll-like receptor [TLR] 4 agonist) or CpG 10104 (TLR9 agonist), showed the vaccine was well tolerated, safe, and immunogenic. Background: Accurate segmentation of brain metastases (BM) is essential for diagnosis, stereotactic radiosurgery planning, and longitudinal assessment. However, manual contouring is time-intensive, limiting clinical scalability, and exhibits substantial inter-observer variability. This variability complicates objective assessment of automated segmentation methods and challenges interpretation of model performance.
Purpose: To address these limitations, we developed TUM-SAM, a hybrid foundation-model framework for fully automated BM segmentation, and introduced a bias-controlled, blinded multi-rater evaluation paradigm to determine whether AI-based BM segmentation has reached expert-level performance and whether AI-generated contours are preferred by human experts under unbiased assessment.

Purpose: The NEW DAY (ClinicalTrials.gov identifier, NCT04469595) study assessed the efficacy and safety of the fluocinolone acetonide (FAc; 0.19 mg) intravitreal implant as baseline therapy in diabetic macular edema (DME).
Design: Prospective, randomized, single-masked, active-controlled, multicenter, 18-month, phase 4 study.
Participants: Adults with type 1 or 2 diabetes and center-involving DME confirmed by central subfield thickness (CST).

Caspase-2 has positive and negative effects on tumor growth in different cancer models. The mechanisms determining these dual outcomes of caspase-2 activation are unclear underscoring the need to assess caspase-2 function in different cell types. We report that high caspase-2 expression is associated with lower survival in patients with colon cancer and increased tumor size in HCT116 colon cancer xenografts. We investigated the interplay between caspase-2 and p53 in regulating the proliferation of these cells. While neither of these proteins had much of an effect on cell cycle arrest on their own, deficiency in both caspase-2 and p53 led to …

Biallelic pathogenic variants in PNPT1 cause combined oxidative phosphorylation deficiency 13 (COXPD13) (MIM #614932), linking mitochondrial dysfunction to type I interferon (IFN) activation through cytosolic leakage of mitochondrial double‐stranded RNA (mt‐dsRNA). This mechanism connects mitochondrial disease to interferonopathies such as Aicardi–Goutières syndrome (AGS). We describe a 7‐month‐old female infant with compound heterozygous PNPT1 variants presenting with severe hypotonia, feeding difficulties necessitating gastrostomy, dystonia, and elevated serum lactate. Brain magnetic resonance imaging (MRI) demonstrated marked cerebellar, brainstem, and basal ganglia atrophy, with a lactate peak on MR spectroscopy (consistent with an inverted doublet). Exposure-based cognitive behavioral therapy (CBT) is the frontline treatment for pediatric obsessive-compulsive disorder (OCD), but not all youth fully respond to this treatment. While multiple factors may influence CBT response, homework adherence in CBT is a modifiable target that can improve treatment outcomes. This report examines the relationship between homework adherence and clinical outcomes in a large sample of youth with OCD who received exposure-based CBT. Here, 137 youth with OCD between 7 and 17 years old (M = 12.42, SD = 2.88) participated in a randomized controlled trial of exposure-based CBT. Changes in leisure consumption are causing transformations in the relationships between young people and brands. A reflection of this is found in video games, whose predominant user profile is that of young people between 18 and 30 years old. They invest more and more time in digital universes: they play, socialize and even buy, and income from the video game sector has increased year after year in Spain and in the world, surpassing those generated jointly by cinema and music. The study aimed to examine the mediating role of rumination in the relationship between the dimensions of alexithymia and body image among first-year students at Sultan Qaboos University in the Sultanate of Oman. The descriptive method was employed, and the sample consisted of 310 male and female first-year students at Sultan Qaboos University, who were selected using a convenience sampling method. The study instruments included three scales: the Body Image Scale, the Toronto Alexithymia Scale, and the Rumination Scale. Passive emitter localization is an important capability for wireless sensing, interference detection, and spectrum awareness. Classical timing-based localization methods estimate emitter position from differences in signal arrival time across distributed sensor platforms, but their performance depends on precise receiver synchronization and favorable signal bandwidth characteristics. This thesis investigates subspace-decomposition-based spatial parameter estimation as an alternative approach in which directional information is extracted from coherent antenna-array measurements. This work develops the theoretical foundation and signal-processing framework for angle-of-arrival estimation. The purpose of this mixed-methods study is to examine the competence of special education teachers in rural East Texas, focusing on their experiences, perceptions, and professional development needs related to behavior management and inclusive education. Quantitative data were gathered through questionnaires given to special education teachers and elaborated upon through focus group interviews, where themes emerged. An explanatory sequential mixed-methods design was used to develop a better understanding of teachers’ preparedness and the problems involved in the implementation of inclusive practices. The findings may inform more effective teacher training, policy-making, and support systems in rural educational settings by highlighting professional …

BACKGROUND: Meningiomas exhibit clinical heterogeneity. Radiotherapy (RT) remains the only adjuvant therapy, but tumor-control is variable, and biomarkers are limited. NRG/RTOG-0539 is the first prospective phase 2 trial to stratify meningioma patients for adjuvant RT based on clinical risk. Here, we apply modern molecular tools to this cohort and identify correlates of RT response.
METHODS: Tumor tissue from 100 RTOG-0539 patients was profiled using DNA methylation arrays, RNA sequencing, and whole-exome sequencing. OBJECTIVE: To evaluate if a single-subject study (S3) design, utilizing paired transcriptome samples from the same patient (eg, "sepsis" vs "recovered"), can replicate transcriptomic signatures from small case-control studies, addressing challenges in patient accrual for rare or sub-stratified diseases.
METHODS: We generated a sepsis gene signature (SGS) comprising 300 differentially expressed genes (DEGs; FDR < 5%) from a human sepsis case-control cohort using general linear models (GLMs). Reproducibility of SGS was assessed through three approaches applied to sub-sampled independent datasets: single-subject analyses (N-of-1-MixEnrich), anticipated to perform better; conventional paired-sample GLM analyses; and a traditional case-control GLM analysis.
RESULTS: SGS reproducibility in GLM analyses was inconsistent at smaller cohort sizes (∼80% reproducibility; n = 5) but stabilized at cohort sizes >6. Remarkably, the single-subject-study approach consistently reproduced SGS in each of the 18 subjects individually (100% reproducibility; n = 1).

BACKGROUND: Laser interstitial thermal therapy (LITT) is a minimally invasive treatment for glioblastoma that increases blood-brain barrier (BBB) permeability. However, the mechanisms and spatiotemporal features of this effect remain unclear. It is also unknown whether LITT promotes release of circulating tumor DNA (ctDNA).
METHODS: Using our previously developed LITT mouse model, we employed single-cell RNA-sequencing (scRNA-seq) to investigate gene expression changes in endothelial cells following LITT in the naive mouse brain. A system is proposed herein through which a set of techniques can be utilized to create a coordinated, latency-aware allocation policy for Dynamic Sub-band Operation (DSO). DSO can be used to create multiple parallel contention domains. For example, an access point (AP) can use fine-grained per-sub-band measurements, including overlapping basic service set (OBSS) frame ratio, jitter variance, airtime fairness, clear channel assessment (CCA), and retry metrics, to compute a condition score per sub-band. A closed-loop GPU kernel optimization technique collects hardware performance counters to form a normalized counter-profile vector and classifies the kernel into a bottleneck class. Candidate structured transforms are selected using probabilistic ranking based on stored successes and failures for transforms conditioned on bottleneck class and hardware. After each transform, the kernel is re-profiled (in addition to benchmarking), re-classified, and a bottleneck shift is detected when the bottleneck class changes. Upon a shift, remaining transforms are re-ranked for the new bottleneck class. A directed acyclic graph represents proxy metrics across layers, including a ground-truth metric and downstream proxies. For each adjacent layer pair, a drift state is computed using sliding-window Spearman correlation, a trend estimate, and volatility, with adaptive window sizing via change-point detection.
