The rate of weight reduction during obesity treatment declines over time and eventually reaches a weight plateau. We investigated factors associated with time to weight plateau (TTWP) in tirzepatide-treated participants with obesity or overweight in a post-hoc analysis of SURMOUNT-1 and SURMOUNT-4 trials. Participants adherent to tirzepatide treatment and achieving ≥5% weight loss by primary endpoint (week 72 SURMOUNT-1; week 88 SURMOUNT-4) were included. Weight plateau was defined as a weight change < 5% over a 12-week interval and all subsequent 12-week intervals. TTWP was time from randomization to the start of the first 12-week interval. Association between baseline characteristics and TTWP was assessed. Overall, 1438 participants in SURMOUNT-1 and 259 in SURMOUNT-4 were included. Across BMI categories (overweight, class I, II, and III), median TTWP in SURMOUNT-1 was 24.3, 26.0, 36.1, and 36.1 weeks, respectively (p < .05, class II and III vs. overweight). By week 72, 90.2%, 88.9%, 87.6%, and 87.8% of participants in SURMOUNT-1 had reached a weight plateau across respective BMI categories [Correction added on 22 January 2025, after first online publication: The "72%" has been changed to "72" in this version.]. Higher doses of tirzepatide (10/15 mg), younger age, and female sex were more likely to reach a weight plateau later. Results in SURMOUNT-4 were similar. In this post-hoc analysis, most participants reached a weight plateau by week 72. Higher doses of tirzepatide, younger age, and female sex were associated with a longer TTWP. Further research into modifiers of weight reduction phases with tirzepatide may inform treatment decisions for its use in chronic weight management. Clinical Trial Registration: ClinicalTrials.gov, identifiers NCT04184622 (SURMOUNT-1) and NCT04660643 (SURMOUNT-4), available at http://www.clinicaltrials.gov/.

Expanding The Clinical Spectrum Of Porokeratosis Ptychotropica, Brooke R Bartley, Dylan R Maldonado, Steven Mays, Ronald P Rapini Jun 2025

Expanding The Clinical Spectrum Of Porokeratosis Ptychotropica, Brooke R Bartley, Dylan R Maldonado, Steven Mays, Ronald P Rapini

Faculty, Staff and Student Publications

Porokeratosis ptychotropica (PP) is a rare variant of porokeratosis, typically presenting as pink to brown hyperkeratotic plaques in the gluteal cleft and buttocks, defined histologically by multiple cornoid lamellae. Diagnosis of PP is challenging due to its clinical overlap with other dermatologic conditions and inconsistent terminology in the literature. This case report describes a 47-year-old woman exhibiting clinical and pathological features of PP but with a more diffuse presentation than previously documented. The case emphasizes the diagnostic complexities of PP, particularly in atypical presentations, and highlights the critical role of histological examination in distinguishing it from other porokeratoses.

Background: Alcohol, tobacco, and cannabis are the most commonly used substances among young populations, often leading to polysubstance use, which correlates with problematic consumption and adverse health outcomes. The contextualized reinforcer pathology (CRP) model explains addictive behaviors through several constructs, including the following: drug demand, delay discounting (DD), and substance-free and substance-related reinforcement. Most previous studies have focused on a single CRP construct and the couse of only two substances. This study aimed to characterize three subgroups of young adults reporting past month use of alcohol only, alcohol + tobacco or cannabis, and alcohol + tobacco + cannabis, in terms …

An Enzyme-Linked Immunosorbent Assay (Elisa)-Based Activity Assay For Amp-Activated Protein Kinase (Ampk), Trezze P Nguyen, Shangze Lyu, Yang Liu

Faculty, Staff and Student Publications

AMP-activated protein kinase (AMPK) is the master regulator of cellular and organismal energy homeostasis, playing an essential role in modulating metabolism and other cellular processes. Substantial efforts have been made to develop pharmacological modulators of AMPK activity due to their therapeutic potential against various diseases. Measuring AMPK activity in vitro is a fundamental step for testing AMPK activators and inhibitors. Here, we report an enzyme-linked immunosorbent assay (ELISA)-based AMPK activity assay with simple steps and high sensitivity. This assay offers a robust, in-house alternative to the traditional radioactive methods and other approaches that rely on specialized reagents or commercial kits.

Research outputs 2022 to 2026

Circulating tumour DNA (ctDNA) is a promising biomarker for personalised oncology. However, its clinical utility is limited by detection sensitivity, particularly in early-stage disease. T-Oligo Primed Polymerase Chain Reaction (TOP-PCR) is a commercial amplification approach utilising an efficient “half-adapter” ligation design and a single-primer-based PCR strategy. This study evaluated the clinical value and application of cell-free DNA (cfDNA) pre-amplification. cfDNA amplification with TOP-PCR preserved DNA size profiles and resulted in a 22 bp size increase due to the half-adaptor ligation. Gene target amplification rates varied, showing lower efficiency for the GC-rich TERT promoter amplicon and higher efficiency for the BRAF …

Paradoxical Leadership As Facilitator In The Idea Journey: The Role Of Regulatory Focus, Irene E. De Pater, Shenghui Wang, Ming Yi, Eric F. Rietzschel

Research outputs 2022 to 2026

Drawing upon theory and research on innovation, conservation of resources, and paradoxical leadership, we examine relationships between three stages of the innovative process (i.e., idea generation, idea dissemination, and idea implementation) and the role of paradoxical leadership as a facilitator of the transitions between these stages. In addition, we examine the motivational mechanisms (promotion and prevention focus) that can explain the relevance of paradoxical leadership in the innovative process. Results of a four-wave study show that paradoxical leadership, through employee promotion focus, strengthens the relationship between idea generation and idea dissemination, whereas paradoxical leadership, through employee prevention focus, strengthens the …

A Predictive Model For The Shear Capacity Of Ultra-High-Performance Concrete Deep Beams Reinforced With Fibers Using A Hybrid Ann-Anfis Algorithm, Hossein Mirzaaghabeik, Nuha S. Mashaan, Sanjay Kumar Shukla

Research outputs 2022 to 2026

Ultra-high-performance concrete (UHPC) has attracted considerable attention from both the construction industry and researchers due to its outstanding durability and exceptional mechanical properties, particularly its high compressive strength. Several factors influence the shear capacity of UHPC deep beams, including compressive strength, the shear span-to-depth ratio (λ), fiber content (FC), vertical web reinforcement (ρsv), horizontal web reinforcement (ρsh), and longitudinal web reinforcement (ρs). Considering these factors, this research proposes a novel hybrid algorithm that combines an adaptive neuro-fuzzy inference system (ANFIS) with an artificial neural network (ANN) to predict the shear capacity of UHPC deep beams. To achieve this, ANN and …

Stress Reactivity And Sociocultural Learning: More Stress-Reactive Individuals Are Quicker At Learning Sociocultural Norms From Experiential Feedback, Shilpa Madan, Krishna Savani, Pranjal H. Mehta, Desiree Y. Phua, Ying-Yi Hong, Michael W. Morris

Research Collection Lee Kong Chian School Of Business

When interacting with others in unfamiliar sociocultural settings, people need to learn the norms guiding appropriate behavior. The present research investigates an individual difference that helps this kind of learning: stress reactivity. Interactions in an unfamiliar sociocultural setting are stressful, particularly when the actor fails to follow its rules. Although stress is typically considered a liability, more stress-reactive individuals may be more motivated to improve, and, thus, quicker to learn these rules. Consistent with this idea, a pilot study found that people genetically inclined to stress reactivity, as computed by a genetic profile score across 59 single nucleotide polymorphisms on …

Harnessing Redox-Active Molecules In Alkylammonium Halide-Based Eutectic Solvents For Redox Flow Batteries, Zeru Syum, Jesse Wainright, Desiree Mae Prado, Burcu Gurkan, Palani Sabhapathy, Clemens Burda, Matthew Vasil, Robert Savinell

Faculty Scholarship

Redox flow batteries (RFBs) are promising for large-scale energy storage, however, advancements in performance and cost-effectiveness are critical factors for adoption. Here we report on alkylammonium halide based eutectic solvents (ESs) and found that a ESs comprising of diethylammonium chloride or bromide, in ethylene glycol demonstrated exceptional stability and a wide electrochemical stability window, making it a promising candidate for RFB applications. The electrochemical stability and redox behavior of the alkylammonium halide-based ESs are significantly influenced by hydrogen-bonding interactions, modulated by the alkyl chain length of the cation and the nature of the anion. A redox-active eutectic electrolyte containing 0.45 …

Enabling Automatic Solar Pv Array Identification Using Big Satellite Imagery, Qi Li, Keyang Yu, Carson Snow, Dong Chen

Computer Science Faculty Research and Publications

Recently, there has been a growing interest in automatically collecting distributed solar photovoltaic (PV) installation information in smart grid systems, including the quantity and locations of solar PV deployments, as well as their profiling information across a given geospatial region. Most recent approaches are still suffering low detection accuracy due to insufficient sample and principal feature learning when building their models and also separation of rooftop object segmentation and identification during their detection processes. In addition, they cannot report accurate multi-deployment results. To address these problems, we design a new system-SolarDetector+, which can automatically and accurately detect and profile distributed …

Emulating Empire: Washington’S Preoccupation With British Counterinsurgency In Malaya And U.S. Commitment To Vietnam, Wen-Qing Ngoei

Research Collection College of Integrative Studies

This essay examines how Washington’s deep preoccupation with the British-Malayan counterinsurgency tactics–tactics that routed the guerilla fighters of the Malayan Communist Party–shaped US commitment to Vietnam. This US preoccupation, stoked by Britain’s (and Malaya’s) promotion of their counterinsurgency knowledge to US leaders, also influenced the lessons that US analysts and thinkers absorbed from the United States’ defeat in Indochina. Most studies of British-Malayan counterinsurgency and its connection to the Vietnam War fixate upon debating the relevance of the former to the latter and how this bears on US failures in Indochina. However, this essay focuses on the understudied fervor with …

Testing For Broad Alternatives In Stratified Contingency Tables, Nan Mi Jun 2025

Testing For Broad Alternatives In Stratified Contingency Tables, Nan Mi

Dissertations

In medical and social sciences fields, data are measured in terms of discrete categories. The primary question of interest involves the relationship between a set of factors and a set of response variables under studies. Moreover, the distribution of the response variables may be influenced by another set of variables called confounders. The data from such studies are summarized in 3-way tables. The hypothesis we are interested in can be expressed in terms of "no partial association" between the sub-populations and the response levels.

The methods for testing the association or independence in a 2x2 contingency table have been developed, …

Runtime Backdoor Detection For Federated Learning Via Representational Dissimilarity Analysis, Xiyue Zhang, Xiaoyong Xue, Xiaoning Du, Xiaofei Xie, Yang Liu, Meng Sun

Research Collection School Of Computing and Information Systems

Federated learning (FL), as a powerful learning paradigm, trains a shared model by aggregating model updates from distributed clients. However, the decoupling of model learning from local data makes FL highly vulnerable to backdoor attacks, where a single compromised client can poison the shared model. While recent progress has been made in backdoor detection, existing methods face challenges with detection accuracy and runtime effectiveness, particularly when dealing with complex model architectures. In this work, we propose a novel approach to detecting malicious clients in an accurate, stable, and efficient manner. Neural Radiance Field (NeRF) and 3D Gaussian Splatting (3DGS) have noticeably advanced photo-realistic novel view synthesis using images from densely spaced camera viewpoints. However, these methods struggle in few-shot scenarios due to limited supervision. In this paper, we present NexusGS, a 3DGS-based approach that enhances novel view synthesis from sparse-view images by directly embedding depth information into point clouds, without relying on complex manual regularizations. Exploiting the inherent epipolar geometry of 3DGS, our method introduces a novel point cloud densification strategy that initializes 3DGS with a dense point cloud, reducing randomness in point placement while preventing over-smoothing and overfitting. Specifically, …
