The extremes of lightning, specifically lightning with long-lasting continuing currents and lightning with extremely high peak currents, are investigated from both ground- and space-based observations. Additionally, the potential solar influence on lightning is investigated on large spatial scales.

Continuing currents may occur following the impulsive flow of current during a cloud-to-ground (CG) return stroke and are usually low amplitude (from a few amperes to a few kiloamperes) and long duration (several to hundreds of milliseconds). Remotely estimating their duration from the electromagnetic fields measured by existing ground-based lightning locating systems (LLSs) is not possible, but some space-based lightning detection systems …

In recent years, social media-driven misinformation and disinformation have helped erode public trust in democratic institutions. This thesis examines how constant exposure to false online content correlates with declining confidence in democracy and evaluates the roles of platforms, influencers, and mechanisms. Using a convergent mixed-methods design, this thesis pairs a quantitative survey with a qualitative analysis of three disinformation case studies: the 2020 U.S. election and the “Stop the Steal” movement, the Rohingya crisis in Myanmar crisis, and Russian denial of wartime atrocities in their invasion against Ukraine. Ensuring dimensional precision in parts with complex overhangs is a significant concern in Metal Additive Manufacturing (MAM), as undetected geometric deviations can compromise functionality and reliability. This research introduces a Conditional Generative Adversarial Network (cGAN), specifically the Pix2Pix framework, to predict layer-wise geometric deviations in Laser Powder Bed Fusion (LPBF) printed parts with overhang geometries using paired 2D CAD slices and corresponding X-ray Computed Tomography (XCT) based ground truth images. A key innovation is using RGB color-coded CAD slices to encode overhang angle information, enhancing feature distinction and prediction accuracy compared to non-color-coded inputs. Research reveals that women parolees or probationers encounter both challenges and support systems as they navigate their reintegration into the community. More specifically, most women returning from prison and those on probation are released into a society of stigma where access to housing, employment, education, transportation, and healthcare services is either severely limited or nonexistent. Background: Investigation of circulating plasma proteins may reveal insights into molecular pathways that contribute to the development of heart failure (HF) among people with chronic kidney disease.

Objectives: The authors aimed to identify circulating kidney function-related plasma proteins that are associated with the risk of incident HF.

Methods: The authors studied participants of the ongoing longitudinal ARIC (Atherosclerosis Risk In Communities) study. Relative plasma concentrations of 4,697 unique proteins were measured with the use of an aptamer assay (Somalogic). Purpose: FLASH radiation therapy (FLASH-RT) is an emerging modality that delivers radiation at ultra-high dose rates (UHDR) and has shown consistent normal tissue sparing while preserving tumor control compared to conventional RT—a phenomenon termed the FLASH effect. Despite promising results, the mechanisms behind the FLASH effect remain unclear. Many hypotheses point to radiolytic interactions, but experimental validation is limited. This work aims to develop a robust experimental platform to evaluate the relationship between radiolytic species production and physical beam parameters relevant to FLASH-RT, alongside dosimetry tools for reliable UHDR beam characterization.

Breast cancer is a highly heterogeneous disease characterized by diverse genetic, transcriptional, and epigenetic landscapes. Understanding how intra-tumoral heterogeneity (ITH) influences therapeutic response and drives tumor evolution remains a central challenge in cancer biology. Here, we leverage single-cell multi-omics technologies to dissect the cell states and gene expression programs underlying breast cancer heterogeneity, with a focus on triple-negative breast cancer (TNBC) and estrogen receptor-positive (ER+) breast cancer.

Prion diseases are fatal neurodegenerative disorders caused by the misfolding of the normal prion protein (PrPC) into its infectious form (PrPSc). While the zoonotic potential of chronic wasting disease (CWD) remains uncertain, the presence of prions in food products raises public health concerns. Additionally, therapeutic strategies for prion diseases remain limited. Among these interactions, synthetic lethality, where simultaneous loss of two genes is lethal but loss of either alone is tolerated, offers a powerful framework for selectively targeting tumor-specific dependencies. Huntington’s disease (HD) is a devastating neurodegenerative disorder caused by an expanded CAG trinucleotide repeat in the Huntingtin (HTT) gene, resulting in a mutant huntingtin (mHTT) protein with an elongated polyglutamine tract. While the genetic basis of HD is well established, the mechanisms underlying mHTT toxicity and its impact on cellular function remain incompletely understood. Recent discoveries implicate Huntingtin-associated protein 40 (HAP40) as a critical regulator of HTT stability and intracellular function. Li-Fraumeni syndrome (LFS) is a heritable disorder caused by germline mutations in the TP53 gene that result in an increased risk of cancer. The TP53 tumor suppressor gene regulates cell division and prevents the accumulation of cells that may become cancerous. LFS patients are susceptible to various types of cancers, including osteosarcoma, soft tissue sarcoma, acute leukemia, and adrenal cortical tumors, as well as breast and brain cancer. Recent evidence suggests that systemic factors, particularly the gut microbiota and its metabolites, play a significant role in shaping brain immune responses, including microglial activation. However, the mechanisms linking gut dysbiosis to microglial dysfunction in AD are poorly understood. In this dissertation, I explore the posthuman play literacies of multilingual makerspaces with four-year-olds, emphasizing the need for such a study while situating my arguments within the existing literature and outlining my methodological approach. This study examines how middle school mathematics teachers conceptualize and enact culturally responsive mathematics teaching (CRMT) within a district-supported professional learning initiative. Guided by the CRMT framework developed by Aguirre and Zavala (2013), the research explores how instructional decision-making intersects with equity-focused pedagogy.

Exploring The Effects Of Remote Work On Leadership Pipelines: With A Focus On Professional Women And Minorities, Mark S. Andrekovich

I analyzed data from five U.S.-based companies to understand how remote work models affect leadership advancement. Using Role Congruity, Status Characteristics, and Intersectionality theories, I analyzed five hypotheses about the impact of remote work on leadership pipelines. Two of the five hypotheses were supported, revealing bias patterns rather than issues related to remote work. For instance, white employees had greater access to remote work opportunities than non-white employees, who had fewer promotions despite similar performance scores. Contrary to expectations, I found that remote work does not inherently affect promotional advancements. Via a post hoc analysis, I found that hybrid professionals …

We consider a nonlinear transport problem to model the chromatography process of high-capacity multimodal membranes. Robust and efficient algorithms that simulate these bioseparation processes are critical to developing therapeutics for various chronic illnesses and infectious diseases. However, much of the current methodology focuses on stabilization and linearization techniques, often implementing low-order time-discretizations and linearized adsorption, resulting in inefficiencies and inaccuracies in the numerical solution. This Dissertation in Practice utilized improvement science methods to increase teacher self-efficacy in foundational reading practices and to address the oral reading fluency (ORF) challenges of elementary students who consistently score below proficiency. Rooted in Bandura’s theory of self-efficacy and the Active View of Reading, the study aimed to build teacher confidence in delivering foundational reading instruction while improving student ORF rates. A Plan-Do-Study-Act (PDSA) cycle was implemented with second-grade teachers and targeted the following aim: increase ORF by 12 words correct per minute (wcpm) for students receiving Tier 3 intervention over an eight-week cycle. Online optimization (OO) is an iterative process of decision making under uncertainty. At every step, a decision is made before the outcome of this decision is known. For the online optimization model, the objective function is unknown at the time the decision is being made.
