Spatial transcriptomics (ST) enables systematic profiling of whole-transcriptome gene expression in tissues while preserving spatial context. Recent advances in sequencing- and imaging-based ST technologies have ushered in the era of microscopic-resolution ST (μST), allowing transcriptome mapping at cellular and even subcellular scales with unprecedented precision. Despite these advances, μST faces substantial challenges, including sparse transcript discovery per submicron (or micron)-sized spatial units and data fragmentation across platforms, hindering integration and analysis. There is also a growing demand for scalable, segmentation-free, and universally applicable analysis methods, as well as strategies for 3D mapping, multi-omics integration, and artificial intelligence (AI)-driven spatial analysis. …

This longitudinal study examined whether specific cognitive patterns during pregnancy predict postpartum obsessive-compulsive (OC) symptoms. A diverse sample of 256 women was assessed at 20 weeks of pregnancy (Time 1), 6 weeks postpartum (Time 2; n = 233), and 6 months postpartum (Time 3; n = 231). At each point, participants completed interview and/or self-report measures of OC symptoms, psychological distress, and obsessive beliefs (i.e., cognitive patterns related to OC symptoms). Postpartum OC symptoms were common, reported by 87.1 % at Time 2 and 74.5 % at Time 3. Stronger prenatal obsessive beliefs during pregnancy predicted greater postpartum OC symptom …

Background: Lifestyle interventions for weight loss might exacerbate age-related bone loss and osteoporosis. However, there is limited knowledge about their effects on bone quality. We examined whether lifestyle intervention can preserve or enhance bone quality, despite reductions in bone mineral density.
Methods: The Lifestyle Intervention to Improve Bone Quality (LIMB-Q) study was a randomised controlled trial conducted at Baylor College of Medicine and the Michael E DeBakey VA Medical Center (Houston, TX, USA) including older adults (aged 65-85 years) with obesity (BMI ≥30 kg/m2). GeSn (germanium tin) alloys are potentially well suited for near-mid infrared space optoelectronic applications. Alloys of GeSn have similar properties to group III-V and mercury-cadmium-telluride semiconductors and are compatible with cost-effective complementary metal oxide semiconductor (CMOS) manufacturing technology. Recent progress in non-equilibrium remote plasma-enhanced chemical vapor deposition (RPECVD) has enabled the crystalline growth of GeSn with Sn concentrations of up to 10% without Sn surface segregation. Several experimental studies in previous literature report CVD- or molecular beam epitaxy (MBE)-grown GeSn alloys achieving a direct bandgap with 6%-9% Sn content. A comprehensive postmortem examination is an essential component of a work-up after stillbirth. Its findings can support accurate counseling of parents about causes and risk of recurrence. It also supports providers' decisions about most appropriate testing and management plans for future pregnancy to prevent recurrence. Informing parents about fetal autopsy and obtaining their consent is challenging, and conducting a fetal autopsy requires expertise that is, not universally available. Newer non-invasive or minimally invasive methods such as postmortem MRI and targeted biopsies can replace or supplement autopsies, but one must recognize that expertise in these methods is likewise not broadly available. …

Background: Molecular aberrations have been incorporated into tumour classification guidelines of meningioma. TERT-promoter (TERTp) mutation is associated with worse prognosis and is designated a WHO grade 3 biomarker. However, it remains unclear whether TERTp mutation is context-dependent, with other co-occurring genetic alterations potentially driving its association with prognosis. We sought to characterise the role of TERTp mutation in meningioma and guide TERTp sequencing.

As the U.S. Air Force confronts growing complexity in system acquisition, the implementation of digital models in system design and logistics process management allows the incorporation of digital tools and the possibility for automation of portions of logistics processes. This thesis investigates where these technologies can be most effectively integrated within the Air Force Life Cycle Management Center logistics enterprise (AFLCMC). Advanced aerospace systems require structural materials that can perform reliably in high-temperature environments for long durations. The performance of standard polymer matrix composites (PMCs) in these conditions is often limited by their susceptibility to time-dependent deformation, such as creep. Fuel represents over 70% of the logistical resupply demand within the Department of Defense, with the United States Air Force accounting for most of that consumption. While prior fuel efficiency efforts have primarily focused on technical upgrades, this research explores how behavioral interventions can influence pilot decision-making and operational energy outcomes. Using the Theory of Planned Behavior (TPB) as a guiding framework, this study investigates the effects of targeted behavioral strategies on pilots’ attitudes, subjective norms, perceived behavioral control, intentions, and actual fuel-efficient behaviors. Hypersonic vehicle design requires understanding complex aerodynamic phenomena across the full flight regime. This study presents a novel MF surrogate modeling methodology that enables the prediction the full field response across a vehicle’s surface. A Space-Filling Curve (SFC) is used to convert unstructured data into 1D vectors. The a Convolutional Autoencoder is used with transfer learning to reduce the dimensionality of the data. An Emulator-Embedded Neural Network (E2NN) combines multi-fidelity data for fast, accurate predictions. A benchmark analytical example and hypersonic application are used to evaluate the methodology. CubeSats have an inherent limitation on volume due to their small form factor. Naturally, this extends to the capacity of on-board ΔV, thus limiting the maneuverability of the satellites. This limited maneuverability makes it extremely important to be intentional when planning out mission trajectories. The cislunar region subjects these trajectories to the gravitational forces of both the Earth and the Moon, making it all the more necessary to deliberately plan out maneuvers. This work seeks to answer the unstudied questions “Does a layer of vegetation impact the prompt radiation energy deposition in the soil from an atmospheric nuclear detonation?” and “If yes, by how much?” A new methodology was developed to repeatably automate and streamline a process of generating a three-dimensional geometry for subsequent radiation transport simulations and statistical analysis. The methodology streamlined three diverse applications: a three-dimensional mesh generator, a radiation transport simulation code, and a sensitivity analysis & uncertainty quantification software. We investigate the spin state of the electron-positron system positronium to determine experimental criteria that can identify a positronium anyon. Anyons have been posited to be the basis for a truly fault-tolerant quantum computer, but they have not been indisputably observed in experimental systems. Through the analysis of the fully relativistic symmetry group of fermions and bosons in three dimensions, we investigate the symmetry of its subgroups to determine that positronium in two dimensions behaves as a scalar with respect to rotations. Background and objectives: Acute kidney injury (AKI) poses a serious clinical challenge, particularly in high-risk environments, due to its association with increased morbidity and mortality. Traditional diagnostic markers, such as serum creatinine, often detect AKI only after significant kidney damage has occurred, limiting opportunities for early intervention. Neutrophil gelatinase-associated lipocalin (NGAL) has emerged as a promising early biomarker due to its rapid upregulation following kidney ischemia. NGAL supports renal recovery by reducing toxicity and promoting tubular regeneration via heme oxygenase-1 activity. Background: Anhedonia is a core symptom of major depressive disorder (MDD) that may result from aberrant lateral habenula hyperactivity. Targeting G-protein coupled receptor 139 (GPR139) may improve anhedonia by modulating lateral habenula activity. NBI-1065846 is an investigational GPR139 agonist that improved anhedonia, anxiety, and depression in rodent models.
Methods: TERPSIS was a phase 2, proof-of-concept clinical study. Adults with MDD experiencing a major depressive episode with anhedonia were randomized 1:1 to NBI-1065846 or placebo for 8 weeks. The importance of collective motion in atomic nuclei has been well-established for many decades. Even before the advent of the nuclear shell model and its much-needed answers to surprising empirical phenomena, collectivity and surface deformation have played a vital role in broadening our knowledge. Reconciling the collective and single-particle camps into a unified description required tremendous strides in the application of group theory and advances in high-performance supercomputing, and thanks to these efforts the symmetry-adapted no-core shell model, or SA-NCSM, has developed into a reliable model for the study of deformed light and medium-mass nuclei. The Coalition for Adolescent Girls (CAG), hosted by the Population Council, in collaboration with Akili Dada, convened a high-level donor roundtable on June 4, 2025. Against the backdrop of funding freezes, policy shifts, and widening global crises, this convening brought together funders, grassroots organizations, adolescent and youth leaders, and global experts, to reflect on emerging trends, lived experiences, and actions needed urgently to ensure adolescent girls remain at the center of global development priorities. This knowledge brief synthesizes key insights from the roundtable discussion. A new form of “autocratic legalism” (Scheppele) that relies on law rather than guns raises a serious challenge to democratic constitutionalism. While authors tend to focus on the “illiberal” challenge, this article is concerned with its jurisprudential dimension: what exactly is wrong with autocratic legalism? That is, to what extent does legalism, understood as the formalistic and one-sided instrumentalization of law, represent a hollowed-out, defective, and potentially harmful theory of law? Introduction: Monitoring fluid balance (FB) is important yet challenging in the pediatric intensive care unit (PICU). Currently used, conventional methods of fluid assessment are either not feasible or unreliable. Also, there is a lack of composite scores or clinical algorithms that use different parameters of fluid status. We aim to develop a fluid assessment scoring tool (FAST) to clinically assess the fluid status of children in the PICU.

Introduction: Obesity presents a significant health risk for the aging population. Research shows that weight loss and regular exercise can greatly improve the functional status of older adults who are obese. However, weight loss may also result in a decrease in bone mass. To properly assess changes in fracture risk due to lifestyle interventions, a direct biomechanical evaluation of bone strength and fracture risk at metabolically active sites is essential.

Vocal cord paralysis (VCP) presents with unique challenges when managing airways, particularly during endotracheal intubation. This literature review explores the effects of unilateral and bilateral VCP on intubation technique, first-pass success, extubation safety, and perioperative airway planning. While video laryngoscopy (VL) is becoming increasingly favored for its superior airway visualization, it does not always confirm successful placement in patients with VCP. Complications such as airway edema, aspiration risk, and cuff misplacements are increased in this patient population, and they may be more vulnerable to post-extubation complications, especially without the use of steroid prophylaxis or intensive post-operative planning. The dual rAAVrh8-HEXA and rAAVrh8-HEXB vector can restore central nervous system hexosaminidase (Hex) enzyme activity, decrease GM2 levels in cerebrospinal fluid and rescue phenotypic consequences of GM2 gangliosidosis, Tay-Sachs and Sandhoff diseases in animal models following simultaneous bi-thalamic (BiT) injections. Following up on an n = 2 expanded access trial, we initiated a phase 1/2, single-dose, dose-escalation of combined BiT, intra-cisterna magna and intrathecal infusion in children with Tay-Sachs and Sandhoff diseases (six infantile, three juvenile). Efficient lymph flow is ensured by lymphatic valves (LVs). The mechanisms that regulate LV development are incompletely understood. Here, we show that the deletion of the GPCR sphingosine 1-phosphate receptor-1 (S1PR1) from lymphatic endothelial cells (LECs) results in fewer LVs. Interestingly, LVs that remained in the terminal ileum-draining lymphatic vessels were specifically dysfunctional. Furthermore, tertiary lymphoid organs (TLOs) formed in the terminal ileum of the mutant mice. TLOs in this location are associated with ileitis in humans and mice. However, mice lacking S1PR1 did not develop obvious characteristics of ileitis. Mechanistically, S1PR1 regulates shear stress signaling and the expression of …

OBJECTIVES: Machine learning (ML) is an emerging discipline centered around complex pattern matching and large data-based prediction modeling and can improve precision medicine healthcare. Cochlear implants (CI) are highly effective, however, outcomes vary widely, and accurately predicting speech perception performance outcomes between patients remains a challenge. This study aims to evaluate the ability of ML to predict speech perception performance among CI recipients at 6-month post-implantation using only preoperative variables on one of the largest CI datasets to date, with an emphasis placed on identification of poor performers.

In contemporary digital environments, exponential cyber threat growth has made cryptographic key generation a critical security challenge. Traditional Pseudo-Random Number Generators (PRNGs) and existing chaos-based methods often exhibit insufficient entropy, limited randomness quality, and inadequate resistance to statistical attacks. Current implementations frequently produce suboptimal entropy values and fail to meet modern cryptographic security standards and rigorous randomness testing protocols. Federated learning (FL) enables decentralized, privacy-preserving machine learning by training models across distributed data without sharing raw patient information. However, most FL frameworks focus on unimodal data and overlook critical challenges in multimodal healthcare settings, such as privacy risks, fairness disparities, and reduced model interpretability. We present PRIFLEX (Privacy-Resilient Integration Framework for Learning Exchange), a novel FL framework designed for secure integration of structured and unstructured medical data. The proposed work aims to develop a DDoS attack detection model that targets web servers by selecting the appropriate classifier based on several criteria and enhancing the accuracy. The detection system has been constructed using machine-learning algorithms to train and test the CIC-DDoS2019 dataset, and it is integrated with a stacked classifier to enhance accuracy. The classifiers depend on multiple criteria. For that, we employed the BWM to calculate the criteria weights and VIKOR to rank the classifiers, which are part of the MCDM methods.
