Development Of Myelin In Wolfram Syndrome, Olga Reine Neyman May 2023

Development Of Myelin In Wolfram Syndrome, Olga Reine Neyman

Arts & Sciences Graduate Student Theses and Dissertations

Wolfram syndrome is a rare, autosomal recessive disease traditionally characterized by juvenile onset insulin dependent diabetes, optic atrophy, deafness, and neurodegeneration often beginning in childhood and adolescence. The disease is caused by mutations in WFS1, which encodes for the protein wolframin and is associated with endoplasmic (ER) stress-mediated apoptosis. ER stress-related dysfunction may inhibit production of myelin during neurodevelopment in Wolfram syndrome, as active and developing oligodendrocytes are more vulnerable to ER stress than mature ones. It is known that white matter tracts are compromised in the disease, but many questions related to the vulnerability of myelin and its impact …

Mapping Brain Development And Decoding Brain Activity With Diffuse Optical Tomography, Kalyan Tripathy May 2023

Mapping Brain Development And Decoding Brain Activity With Diffuse Optical Tomography, Kalyan Tripathy

Arts & Sciences Graduate Student Theses and Dissertations

Functional neuroimaging has been used to map brain function as well as decode information from brain activity. However, applications like studying early brain development or enabling augmentative communication in patients with severe motor disabilities have been constrained by extant imaging modalities, which can be challenging to use in young children and entail major tradeoffs between logistics and image quality. Diffuse optical tomography (DOT) is an emerging method combining logistical advantages of optical imaging with enhanced image quality. Here, we developed one of the world’s largest DOT systems for high-performance optical brain imaging in children. From visual cortex activity in adults, …

Leveraging Protein Dynamics To Drug Filovirus Protein-Nucleic Acid Interactions Using Simulations And Experiments, Matthew Alberto Cruz May 2023

Leveraging Protein Dynamics To Drug Filovirus Protein-Nucleic Acid Interactions Using Simulations And Experiments, Matthew Alberto Cruz

Arts & Sciences Graduate Student Theses and Dissertations

Drug discovery targeting protein nucleic acid (PNI) and protein-protein interactions (PPI) remains a difficult task. How to identify druggable conformations and discover compounds binding to these conformations is an unsolved problem. Here, I describe how we apply computational and experimental approaches to probe the conformational landscape of a key immune antagonist protein from the Zaire ebolavirus. Viral Protein 35 (VP35) binds to the viral pathogen associated molecular pattern, double stranded RNA (dsRNA), and blocks activation of the interferon (IFN) response. Long time scale molecular dynamics simulations reveal that VP35 adopts an alternative conformation which opens a pocket absent in experimental …

Understanding Thermotolerance In Photosynthetic Cells: From Green Algae To Land Plants, Erin Marie Mattoon May 2023

Understanding Thermotolerance In Photosynthetic Cells: From Green Algae To Land Plants, Erin Marie Mattoon

Arts & Sciences Graduate Student Theses and Dissertations

Global temperatures are increasing, causing damage to crop plants, algae, and reducing food and biofuel production. To meet increasing demand for food and energy, we must engineer thermotolerant crops and algae that will thrive in the increasingly warm environment. However, there are many things we do not understand about how photosynthetic cells cope with high temperatures. Primarily, we do not fully understand which genes are required for thermotolerance and how different intensities and durations of high temperature periods may change this list. In this thesis, I employed high-throughput, quantitative, and genomic approaches including transcriptomics, proteomics, and genome-wide pooled mutant screens …

Spike-Timing-Dependent Plasticity Alters Sensory Network Connectivity, Adalee Lube May 2023

Spike-Timing-Dependent Plasticity Alters Sensory Network Connectivity, Adalee Lube

Arts & Sciences Graduate Student Theses and Dissertations

A fundamental question in neuroscience is: how does a sensory system optimize detection of behaviorally relevant stimuli, when those stimuli and the sensory environment are constantly changing? Spike-timing-dependent plasticity (STDP), in which synapse strength changes based on the relative timing of pre- and post-synaptic spiking, has been implicated in changes in neuronal connectivity thought to underlie learning and memory. Synaptic strength changes caused by STDP have been shown in optic tectum, visual cortex, hippocampus, and other brain regions in vitro across many organisms like fish, frogs, and mice. Although it is possible that STDP mechanisms underlie changes in sensory neuron …

The Effect Of Oxytocin Receptor Genetic Variants On Oxytocin Response, Manasi Malik May 2023

The Effect Of Oxytocin Receptor Genetic Variants On Oxytocin Response, Manasi Malik

Arts & Sciences Graduate Student Theses and Dissertations

Oxytocin is a nonapeptide hormone that modulates social behavior, mediates the lactation reflex, and induces and strengthens uterine contractions. Each year, oxytocin is administered to about half of laboring patients for induction and augmentation, and to almost all patients for prevention of post-partum hemorrhage – a total of four million patients per year in the United States alone. However, response to oxytocin varies widely between individuals, and complications including uterine hyperstimulation, uterine atony, required Cesarean section, and postpartum hemorrhage can arise from either excessive or inadequate oxytocin response. To avoid these adverse events, it is critically important to identify individuals …

Zebrafish Swim Capacity, Gait, And Posture During Innate Spinal Cord Repair, Nicholas Oliver Jensen May 2023

Zebrafish Swim Capacity, Gait, And Posture During Innate Spinal Cord Repair, Nicholas Oliver Jensen

Arts & Sciences Graduate Student Theses and Dissertations

Adult zebrafish are capable of anatomical and functional recovery following severe spinal cord injury. Significant advances for understanding the cellular and molecular mechanisms of innate spinal cord repair are best supported by accurate assessment of swim function. This study introduces neuromuscular-associated measurements derived from gait and posture, compares new and classical functional measurements longitudinally, and provides a comprehensive correlative analysis of various functional and cellular regeneration outputs. It also explores trajectories of functional regeneration through tensor decomposition and demonstrates that swim function at 2 weeks post-injury can be used to predict 8-week regeneration outcomes. By establishing swim quality metrics which …

Solo Dance In Greek And Roman Comedy, Marleigh Anderson May 2023

Solo Dance In Greek And Roman Comedy, Marleigh Anderson

Arts & Sciences Graduate Student Theses and Dissertations

In the first full-length study focusing exclusively on comic solo dance, I argue for the presence and significance of solo dance in both Greek and Roman comedy. My dissertation aims to answer two driving questions: where does solo dance (broadly defined as dance by actors) occur in extant Greek and Roman comedy, and how does such dancing contribute to the plays? The introduction lays out my methodology for approaching these questions. My study then moves from what can be known with certainty (definite solo dance scenes and references to dance) to what requires more speculation (likely dance scenes). Chapter one, …

Heterozygous mutations in the gene encoding RagD GTPase were shown to cause a novel autosomal dominant condition characterized by kidney tubulopathy and cardiomyopathy. We previously demonstrated that RagD, and its paralogue RagC, mediate a non-canonical mTORC1 signaling pathway that inhibits the activity of TFEB and TFE3, transcription factors of the MiT/TFE family and master regulators of lysosomal biogenesis and autophagy. Here we show that RagD mutations causing kidney tubulopathy and cardiomyopathy are "auto- activating", even in the absence of Folliculin, the GAP responsible for RagC/D activation, and cause constitutive phosphorylation of TFEB and TFE3 by mTORC1, without affecting the phosphorylation …

Glutamatergic Cerebellar Neurons Differentially Contribute To The Acquisition Of Motor And Social Behaviors, Meike E Van Der Heijden, Alejandro G Rey Hipolito, Linda H Kim, Dominic J Kizek, Ross M Perez, Tao Lin, Roy V Sillitoe

Duncan NRI Faculty and Staff Publications

Insults to the developing cerebellum can cause motor, language, and social deficits. Here, we investigate whether developmental insults to different cerebellar neurons constrain the ability to acquire cerebellar-dependent behaviors. We perturb cerebellar cortical or nuclei neuron function by eliminating glutamatergic neurotransmission during development, and then we measure motor and social behaviors in early postnatal and adult mice. Altering cortical and nuclei neurons impacts postnatal motor control and social vocalizations. Normalizing neurotransmission in cortical neurons but not nuclei neurons restores social behaviors while the motor deficits remain impaired in adults. In contrast, manipulating only a subset of nuclei neurons leaves social …

Despite the prevalence of pericytes in the microvasculature of the heart, their role during ischemia-induced remodeling remains unclear. We used multiple lineage-tracing mouse models and found that pericytes migrated to the injury site and expressed profibrotic genes, coinciding with increased vessel leakage after myocardial infarction (MI). Single-cell RNA-Seq of cardiac pericytes at various time points after MI revealed the temporally regulated induction of genes related to vascular permeability, extracellular matrix production, basement membrane degradation, and TGF-β signaling. Deleting TGF-β receptor 1 in chondroitin sulfate proteoglycan 4-expressing (Cspg4-expressing) cells reduced fibrosis following MI, leading to a transient improvement in the cardiac …

The Body Knows: Embodiment And Whiteness A Rhetorical Analysis Of White Bodies Engaged In Antiracism Work, Otsego County, New York, Kerry Lynn Mess May 2023

The Body Knows: Embodiment And Whiteness A Rhetorical Analysis Of White Bodies Engaged In Antiracism Work, Otsego County, New York, Kerry Lynn Mess

Theses - ALL

Multiculturalism, diversity, inclusion, equity, antiracism, belonging… The humanities, social sciences, and rhetorical scholarship have all slowly given attention to questions of race and racism. Situated at the intersection of somatic psychology and rhetorical studies, this thesis uses a somatic theory lens to explore how symbolic systems, specifically language and bodies, are intertwined to understand the embodied or material effects of racism we have not yet accounted for. I borrow Karma Chávez’s language of “abstract” and “actual” (2018) to observe the rhetorical dynamics, patterns and strategies, of white bodies accounting for themselves and apply Resmaa Menakem and Douglas Robinson’s somatic theories …

Theaters Of The Intimate: The Feminine Culture Of Self-Care On Tiktok, Rayana Weller May 2023

Theaters Of The Intimate: The Feminine Culture Of Self-Care On Tiktok, Rayana Weller

Theses - ALL

In this thesis, I study the feminine culture of self-care on TikTok as it exists in prominent digital genres: skincare and soft living. These two genres represent the aspiration toward a soft perfection of both the physical self and the space around it. Situated within the theories of the female complaint (Berlant, 2008) and the paradigm of perfection (McRobbie, 2015), I argue that feminine culture on TikTok reframes the demanding and unending labor required to approximate standards of feminine perfection as a pleasing and satisfying experience of self-care. Self-care rhetoric on TikTok disguises the discipline required to pursue cultural definitions …

This thesis explores the concept of utopia across different cultures, analyzing the ideal lands in Eastern and Western societies. The author categorizes utopian ideals into two types: Thomas More-style utopias, which aim for social and political perfection, and Arcadian ideals, which prioritize individual inner peace and escape from society. The author compares the ways of entering, the environments, the lifestyles, and the consequences of leaving mentioned in these stories to identify the visual language involved. These visual elements of ideal lands provide a solid foundation for the author's visual thesis, a video game that allows players to build their own …

Associations Between Parental Separation In Adolescence And Romantic Relationship Quality In Emerging Adulthood, Chloe Ludden May 2023

Associations Between Parental Separation In Adolescence And Romantic Relationship Quality In Emerging Adulthood, Chloe Ludden

Theses - ALL

Abstract Parental separation can impact multiple aspects of an individual’s life, including romantic relationship satisfaction (Amato, 2010). Albert Bandura’s social learning theory suggests that children model their behavior based on their parents’ example, which means those whose parents separate may adopt similar relationship practices that lead to their eventual separation with their romantic partner– this could contribute to the pattern of divorce being more common in those whose parents’ divorced (Amato & Deboer, 2001; Bandura, 1977; Ross & Mirowsky, 1999). It is important to interrupt this pattern to prevent the cycle of negative impacts of parental divorce on children being …

Macroproblems Require Microsolutions: The Case For Microgrids In The U.S. Energy Infrastructure, David Howie May 2023

Macroproblems Require Microsolutions: The Case For Microgrids In The U.S. Energy Infrastructure, David Howie

Student Theses 2015-Present

This paper explores the shortcomings of the modern United States Power Grid in the face of climate change and worsening climate conditions, and seeks to explore how alternative styles of power grids could be viable replacements for current designs. The power grid is one of the most fundamental aspects of modern life, without it there would be no reliable energy supply to power devices and machinery. Oftentimes when energy generation is discussed in the United States, it only focuses on the source of generation for the energy, not the transmission of the energy. The lack of complete discussion around this …
