Characterizing Marr Homologs That Mediate Bacterial Fitness And Survival, George Chukwuemeka Nwokocha
Characterizing Marr Homologs That Mediate Bacterial Fitness And Survival, George Chukwuemeka Nwokocha
LSU Doctoral Dissertations
Plant disease outbreaks are on the rise, threatening food security in many parts of the globe. To address these enormous challenges, new intervention strategies like bio-controls are being used as an important sustainable alternative to mitigate the negative effects of pathogens and increase the quality and productivity of agricultural crops. MarR family transcriptional regulators have been studied for their involvement in regulating bacterial virulence genes that help colonize plant hosts and therefore could serve as targets for biocontrol. The transcriptional regulator PecS, a member of the MarR family, is encoded by select bacterial pathogens and we show here that PecS …

The Abolitionist Spectator: Resistant Readings Of Punishment, Rehabilitation, And Reform At The Louisiana State Penitentiary, Kathryn Marie Morris
LSU Doctoral Dissertations
For the majority of the twenty-first century, Louisiana has been the global leader in rates of incarceration. Despite its prevalence, many people encounter prisons and punishment only through representations in movies, television, and the news, remaining distant from the actual processes of punishment and prisons built in remote areas. The Louisiana State Penitentiary at Angola, however, is unique in the number of opportunities it presents for the public to enter the prison for large-scale events like the famous Angola Prison Rodeo, select drama club performances, or to visit the Angola Prison Museum. These opportunities are often read cynically, as moments …
The Regulation Of Autoimmunity Genes In Pancreatic Β-Cells By Nf-Κb, Thomas Malone Martin
The Regulation Of Autoimmunity Genes In Pancreatic Β-Cells By Nf-Κb, Thomas Malone Martin
LSU Doctoral Dissertations
Type 1 diabetes (T1D) is an autoimmune disease where the immune system targets the pancreatic islet beta-cells, which are responsible for insulin production and secretion, leading to a state of insulin insufficiency. Many of the molecular details of disease onset and progression remain poorly understood, including key aspects of cytokine signaling. Therefore, we have examined some of the consequences of inflammatory cytokine signaling as a critical module relevant to T1D. Using interleukin-1 (IL-1) signaling as a well-defined paradigm for inflammation, we investigated key components of this signaling pathway in pancreatic beta-cells. We identified ICAM-1, one of the proteins expressed in …

Development, Optimization, And In Vitro/In Vivo Evaluation Of Azelaic Acid Transethosomal Gel For Antidermatophyte Activity, Ali Nasr, Noha Badawi, Yasmine Tartor, Nader Sobhy, Ali Nasr
Pharmacy
Treatment of dermatophytosis is quite challenging. This work aims to investigate the antidermatophyte action of Azelaic acid (AzA) and evaluate its efficacy upon entrapment into transethosomes (TEs) and incorporation into a gel to enhance its application. Optimization of formulation variables of TEs was carried out after preparation using the thin film hydration technique. The antidermatophyte activity of AzA-TEs was first evaluated in vitro. In addition, two guinea pig infection models with Trichophyton (T.) mentagrophytes and Microsporum (M.) canis were established for the in vivo assessment. The optimized formula showed a mean particle size of 219.8 ± 4.7 nm and a …
Role Of Macrophages In Regression Of Myocardial Fibrosis Following Alleviation Of Left Ventricular Pressure Overload, Lily Neff
MUSC Theses and Dissertations
Antecedent conditions that affect the heart, such as aortic stenosis (AS), can develop into left ventricular pressure overload (LVPO). LVPO is associated with increased myocardial interstitial fibrosis, specifically fibrillar collagen, that leads to increased myocardial stiffness. Patients undergoing surgical aortic valve replacement (SAVR) to correct AS demonstrate significant but incomplete regression of fibrosis. Although current therapies normalize hemodynamic load, treatments that regress collagen content within the myocardium remain a critical need. To elucidate cellular mechanisms of this persistent fibrosis in LVPO, we utilized a transverse aortic constriction (TAC) and removal of the constriction (unTAC) murine model. Outputs were assessed for: …

Engineered Production Of Bioactive Polyphenolic O-Glycosides, Jie Ren, Caleb Don Barton, Jixun Zhan
Biological Engineering Faculty Publications
Polyphenolic compounds (such as quercetin and resveratrol) possess potential medicinal values due to their various bioactivities, but poor water solubility hinders their health benefits to humankind. Glycosylation is a well-known post-modification method to biosynthesize natural product glycosides with improved hydrophilicity. Glycosylation has profound effects on decreasing toxicity, increasing bioavailability and stability, together with changing bioactivity of polyphenolic compounds. Therefore, polyphenolic glycosides can be used as food additives, therapeutics, and nutraceuticals. Engineered biosynthesis provides an environmentally friendly and cost-effective approach to generate polyphenolic glycosides through the use of various glycosyltransferases (GTs) and sugar biosynthetic enzymes. GTs transfer the sugar moieties from …

Introduction: Intravesical therapy (IVT), including Bacillus Calmette-Guérin (BCG), is the standard of care for high grade (HG) non-muscle invasive bladder cancer (NMIBC). Despite the use of IVT, many patients recur after treatment. The bladder microbiome and its role in disease processes has recently risen to prominence. We aim to characterize changes that occur in the bladder microbiome over the course of intravesical therapy and assess whether these changes correlate with outcomes in patients with NMIBC.
Methods: Patients with NMIBC undergoing induction BCG or intravesical therapy were prospectively enrolled from January 2019 to March 2020. Patients with clinical T2 or greater …

Viruses are the most numerically abundant biological entities on Earth. As ubiquitous replicators of molecular information and agents of community change, viruses have potent effects on the life on Earth, and may play a critical role in human spaceflight, for life-detection missions to other planetary bodies and planetary protection. However, major knowledge gaps constrain our understanding of the Earth’s virosphere: (1) the role viruses play in biogeochemical cycles, (2) the origin(s) of viruses and (3) the involvement of viruses in the evolution, distribution and persistence of life. As viruses are the only replicators that span all known types of nucleic …

Conjugation is used by bacteria to propagate antimicrobial resistance (AMR) in the environment. Central to this process are widespread conjugative F-pili that establish the connection between donor and recipient cells, thereby facilitating the spread of IncF plasmids among enteropathogenic bacteria. Here, we show that the F-pilus is highly flexible but robust at the same time, properties that increase its resistance to thermochemical and mechanical stresses. By a combination of biophysical and molecular dynamics methods, we establish that the presence of phosphatidylglycerol molecules in the F-pilus contributes to the structural stability of the polymer. Moreover, this structural stability is important for …

Does SPH Curricula Promote ‘Health Equity’, Reproduce Injustice, or both?
The social justice movements of recent years (preceded by [generations of] insurmountable suffering) have facilitated a collective recognition of the systemic effects of racism and epistemic violence. Despite the ambitious and well-intentioned vision of “health equity” as defined by epidemiologic scholarship - progress is slow and injustices prevail.
Students, scholars, and researchers of ‘Public Health’ are uniquely positioned to imagine and create innovative ways of understanding and addressing the harmful inequities and injustices perpetuated by white settler colonialism. I argue that Academic institutions delivering Public Health education are uniquely positioned …
Adaptive Plasticity Of Coloration In Response To Environmental Change, Karissa Coffield
Adaptive Plasticity Of Coloration In Response To Environmental Change, Karissa Coffield
Scholars Week
When rapid environmental changes occur, different selective forces can create phenotypic trade-offs in which a trait can provide fitness benefits or costs under different environmental conditions. Amphibians are particularly vulnerable to environmental change, and previous research has revealed that some species will plastically respond to variation in temperature and ultra-violet radiation (UVR) by altering their coloration. Divergent selection on coloration may change with elevation and climate induced shifts in temperature because high temperatures are likely to result in lighter color morphs but as elevation increases, UVR exposure increases leading to the prediction that darker color morphs will be more common. …

Intersections Between Precarious Housing And Residential Tenancy Law: A Review Of A Complex Exile And Recent Legal Scholarship On Residential Tenancies, Anna Lund
Journal of Law and Social Policy
This review essay challenges siloed thinking about housing precarity by bringing a sociological account of emergency shelters in Ottawa, Canada–Erin Dej’s book A Complex Exile–into conversation with recent scholarship from Canadian academics on residential tenancy law.3 One intuition underlying this essay is that we need to think about these disparate legal regimes as comprising a bigger system of housing law. Bringing these areas of law into conversation with one another allows us to identify common themes and these may inform statutory reform initiatives, changes to practice, and advocacy on related social issues. Promising innovations in one area of housing law …

The Colonialism Of Eviction, Sarah Buhler, Patricia Barkaskas
Journal of Law and Social Policy
Very little scholarship has specifically considered the phenomenon of eviction as a colonial process, or examined the role of the eviction legal system in reproducing colonial structures and relations. Our aim in this article is to address this gap and thereby extend to the eviction legal system context the work of scholars who have theorized the colonialism of the criminal justice, child welfare, and carceral systems. This article begins with an overview of current issues relating to urban Indigenous housing in Canada. The next section introduces Indigenous concepts of home and homelessness, which are distinct from dominant understandings. We then …

After deadly landmines are removed and booby-traps and improvised explosive devices are cleared, wheat fields are now ready for harvesting, children can run to school on a path, families can return to their partially destroyed homes, and elephants are able to migrate through grasslands. Elsewhere, man-portable air defense systems (MANPADS) and small arms and light weapons were secured or destroyed to prevent terrorists from acquiring them and attacking civilians. These are just some of the successes the U.S. Conventional Weapons Destruction Program achieves day after day, year after year, one step at a time. In fiscal year 2022, the United …

Intra-Arterial Delivery Of Radiopharmaceuticals In Oncology: Current Trends And The Future Of Alpha-Particle Therapeutics, Nathan Kauffman, James Morrison, Kevin O'Brien, Jinda Fan, Kurt R. Zinn
Diagnostic Radiology Articles
A paradigm shift is underway in cancer diagnosis and therapy using radioactivity-based agents called radiopharmaceuticals. In the new strategy, diagnostic imaging measures the tumor uptake of radioactive agent "X" in a patient's specific cancer, and if uptake metrics are realized, the patient can be selected for therapy with radioactive agent "Y". The X and Y represent different radioisotopes that are optimized for each application. X-Y pairs are known as radiotheranostics, with the currently approved route of therapy being intravenous administration. The field is now evaluating the potential of intra-arterial dosing of radiotheranostics. In this manner, a higher initial concentration can …

DPP8/9 inhibition induces either pyroptotic or apoptotic cell death in hematological malignancies. We previously reported that treatment with the DPP8/9 inhibitor 1G244 resulted in apoptotic cell death in myeloma, and our current study further evaluates the mechanism of action of 1G244 in different blood cancer cell lines. Specifically, 1G244 inhibited DPP9 to induce GSDMD-mediated-pyroptosis at low concentrations and inhibited DPP8 to cause caspase-3-mediated-apoptosis at high concentrations. HCK expression is necessary to induce susceptibility to pyroptosis but does not participate in the induction of apoptosis. To further characterize this DPP8-dependent broad-spectrum apoptosis induction effect, we evaluated the potential antineoplastic role for …

BACKGROUND: Molecular characterization of late-onset Alzheimer's disease (LOAD), the leading cause of age-related dementia, has revealed transcripts, proteins, and pathway alterations associated with disease. Assessing these postmortem signatures of LOAD in experimental model systems can further elucidate their relevance to disease origins and progression. Model organisms engineered with human genetic factors further link these signatures to disease-associated variants, especially when studies are designed to leverage homology across species. Here we assess differential gene splicing patterns in aging mouse models carrying humanized APOE4 and/or the Trem2*R47H variant on a C57BL/6J background. We performed a differential expression of gene (DEG) and differential …

Methodological Precision Of In Situ And In Vitro Algal Density Measurements In The Model Cnidarian, Exaiptasia Diaphana, Lauren R. Bolzan, Alison M. Roark
Open Access Fund Publications
In cnidarian symbiosis research, studying algal uptake, maintenance, and expulsion typically requires quantification of algal density in host tissue. Multiple methods are used to measure algal density including in vitro cell counts of holobiont homogenate and in situ cell counts of tentacle clippings. The relative precision of both types of measurement has not previously been reported for the model cnidarian Exaiptasia diaphana in the fully symbiotic state. The objective of this study was to evaluate the precision of in vitro and in situ algal density measurement protocols using light, fluorescent, and confocal microscopy and an automated cell counter. In situ …
