Regenerative Medicine For Tendon/Ligament Injuries: De Novo Equine Tendon/Ligament Neotissue Generation And Application, Takashi Taguchi

Tendon and ligament injuries are debilitating conditions across species. Poor regenerative capacities of these tissues limit restoration of original functions. The first study evaluated the effect of cellular administration on tendon/ligament injuries in horses using meta-analysis. The cellular administration was effective in restoring ultrasonographic echogenicity and increasing vascularity during early phase of healing. Additionally, it improved microstructural organization of healed tissue in terms of cellularity and fiber alignment. However, the study did not support its use for increasing rate of return to performance, expression/deposition of tendon-specific genes/proteins, or mechanical properties.

Due to advances in high-throughput parallel sequencing, researchers have conducted novel studies exploring relationships between microbiome compositions and different aspects of organism health. Some of these studies have shown that the gut microbiome of rodent models has effects on organism health and behavior and that infection with pathogens and the composition of the skin microbiome are linked to changes in gut microbiome composition. While previous studies have shown how vector microbiota impact vector behavior and pathogen transmission, the effect vectors have on reservoir species microbiomes has been a less prominent focus. This thesis investigates the struggle with work-life balance among veteran secondary teachers in public North Mississippi schools. Primary research questions include a) How do veteran teachers describe the struggle with work-life balance, b) What coping strategies over their career seem helpful?, and c)What changes could be made to the educational system to better support public school teachers in their pursuit of work-life balance? At what level should those changes occur? Qualitative data was collected by interviewing five teachers from middle and high schools in the North Mississippi region and analyzed to understand emergent themes from the interviews. “The Queer, Lonely, Intense, Inner Lives Of Their Children”: Psychoanalysis, Mysticism, And Mabel Dodge Luhan’S Narrative Approach To The Story Of Her Childhood, Lauren M. Franken

This thesis explores Mabel Dodge Luhan’s narrative approach to writing Background (1933), the first of her four published volumes of autobiography titled Intimate Memories. In the first section I lay the groundwork for this analysis with a brief examination of Background’s publication history. The succeeding two sections offer a historical framework for understanding late nineteenth and early twentieth-century American conceptualizations of childhood, Freudian psychoanalysis, and mysticism. Considering the various lenses through which Luhan analyzed her childhood memories provides a more complex awareness of her narrative approach. In parallel with the genetic and epigenetic changes that accumulate in tumor cells, chronic tumor-promoting inflammation establishes a local microenvironment that fosters the development of malignancy. While knowledge of the specific factors that distinguish tumor-promoting from non-tumor-promoting inflammation remains inchoate, nevertheless, as highlighted in this series on the 'Hallmarks of Cancer', it is clear that tumor-promoting inflammation is essential to neoplasia and metastatic progression making identification of specific factors critical. Studies of immunometabolism and inflamometabolism have revealed a role for the tryptophan catabolizing enzyme IDO1 as a core element in tumor-promoting inflammation. Adipose tissue is a complex organ whose functions go beyond being an energy reservoir to sustain proper body energy homeostasis. Functioning as an endocrine organ, the adipose tissue has an active role in the body's metabolic balance regulation through several secreted factors generally termed as adipokines. Thus, adipose tissue dysregulation in chronic kidney disease (CKD) can have a deep impact in the pathophysiology of diseases associated with metabolic dysregulation including metabolic syndrome, insulin resistance (IR), atherosclerosis, and even cachexia. “But first, we must believe.” So concludes The Antiracist Constitution, where Brandon Hasbrouck confronts an uneasy question: In the quest for racial justice, is the Constitution friend or foe? Even the casual observer knows that constitutional law is no friend to racial justice. In the nineteenth century, Plessy v. Ferguson blessed Jim Crow. In the twentieth century, Washington v. Davis insulated practices that reproduce Jim Crow. Now in the twenty-first century, pending affirmative action litigation invites the Supreme Court to outlaw efforts to remedy Jim Crow.

This thesis joins a vibrant conversation on the importance of storytelling in an age of climate change through an analysis of Jeff VanderMeer’s Annihilation, a strange and prophetic novel whose environments and characters are confronted with significant ecological devastation and transformation. It explores the ways in which VanderMeer opens liminal spaces between the human and nonhuman through his usage of the New Weird genre, uncanny and abcanny imagery, and monstrous characters.

In 1983 Benjamin Libet and colleagues published a paper apparently challenging the view that the conscious intention to move precedes the brain’s preparation for movement. The experiment initiated debates about the nature of intention, the neurophysiology of movement, and philosophical and legal understanding of free will and moral responsibility. Here we review the concept of “conscious intention” and attempts to measure its timing. Scalp electroencephalographic activity prior to movement, the Bereitschaftspotential, clearly begins prior to the reported onset of conscious intent. Based on previous results showing a pivotal role of endogenous interleukin-10 (IL-10) in the recovery from cisplatin-induced peripheral neuropathy, the present experiments were carried out to determine whether this cytokine plays any role in the recovery from cisplatin-induced fatigue in male mice. Fatigue was measured by decreased voluntary wheel running in mice trained to run in a wheel in response to cisplatin. Mice were treated with a monoclonal neutralizing antibody (IL-10na) administered intranasally during the recovery period to neutralize endogenous IL-10. In the first experiment, mice were treated with cisplatin (2.83 mg/kg/day) for five days and IL-10na (12 μg/day for …

Somatic mutations in nonmalignant tissues accumulate with age and injury, but whether these mutations are adaptive on the cellular or organismal levels is unclear. To interrogate genes in human metabolic disease, we performed lineage tracing in mice harboring somatic mosaicism subjected to nonalcoholic steatohepatitis (NASH). Proof-of-concept studies with mosaic loss of Mboat7, a membrane lipid acyltransferase, showed that increased steatosis accelerated clonal disappearance. Next, we induced pooled mosaicism in 63 known NASH genes, allowing us to trace mutant clones side by side. Human sapoviruses (HuSaVs), like human noroviruses (HuNoV), belong to the Caliciviridae family and cause acute gastroenteritis in humans. Since their discovery in 1976, numerous attempts to grow HuSaVs in vitro were unsuccessful until 2020, when these viruses were reported to replicate in a duodenal cancer cell-derived line. Physiological cellular models allowing viral replication are essential to investigate HuSaV biology and replication mechanisms such as genetic susceptibility, restriction factors, and immune responses to infection. In this study, we demonstrate replication of two HuSaV strains in human intestinal enteroids (HIEs) known to support the replication of HuNoV and other human enteric viruses. …

Chronic liver disease is a major public health burden worldwide1. Although different aetiologies and mechanisms of liver injury exist, progression of chronic liver disease follows a common pathway of liver inflammation, injury and fibrosis2. Here we examined the association between clonal haematopoiesis of indeterminate potential (CHIP) and chronic liver disease in 214,563 individuals from 4 independent cohorts with whole-exome sequencing data (Framingham Heart Study, Atherosclerosis Risk in Communities Study, UK Biobank and Mass General Brigham Biobank). ‘Sex’ is often used to describe a suite of phenotypic and genotypic traits of an organism related to reproduction. However, not all these traits are necessarily linked, and the rhetorical collapse of variation into a single term elides much of the complexity inherent in reproductive phenotypes. We conducted a meta-analysis of scientific literature to investigate how terminology related sexual phenotypes changes over time. We find that the conflation of gender and sex has increased, and there is a mammalian bias in this conflation. We highlight how a more inclusive and expansive framework for sex can clarify studies of sexual diversity.

This multidisciplinary study examines the liquid dairy and alternative milk markets through four different lenses, exploring the intersection of consumer sentiment, industry financial performance, nutritional science, and environmental implication using metrics compiled from industry financial and market research, consumer sentiment surveys, and empirical scientific research. Through our analysis we realized that our results were rife with contradictions. Consumers often pick non-dairy alternatives for health or environmental reasons, but this choice is often rooted in misinformation. Road salt pollution in the Great Lakes region has been steadily increasing in aquatic systems due to urban sprawl and road safety concerns. Excessive road salt (measured as Cl- mg/L) impacts water quality adjacent to roadside ecosystems by increasing electro- conductivity and altering soil solution osmotic gradients, thus rendering habitats inhospitable for native plants and animals. To date, few mitigation strategies exist to manage salinity in freshwater streams, lakes, and wetlands. Homing, a behavior demonstrated by the threespine stickleback allows an organism to return to their home site when displaced. We predicted that male stickleback fish would home well because they guard their young in nests and have high incentive to return. Here we marked and displaced nesting male stickleback over a variety of distances to see whether homing ability declines with distance displaced. Our results show that stickleback can home, but there was no significant relationship between the distance and return rate. However, some nests were taken over by new males after displacement, meaning that even if the experimental fish …

African American women have the highest burden of cardiovascular disease compared to women of other racial and ethnic groups. The American Heart Association (AHA) developed the Life’s Essential 8 (LE8) to promote ideal cardiovascular health. No known studies have examined adherence to the AHA’s Life’s Essential 8 in young African American women. Based on AHA’s definitions of ideal cardiovascular health, we operationalized the LE8 strategies into a quantitative score and assessed young African American women's adherence.
