We present an ocean‑basin‑scale dataset that includes tail fluke photographic identification (photo‑ID) and encounter data for most living individual humpback whales (Megaptera novaeangliae) in the North Pacific Ocean. The dataset was built through a broad collaboration combining 39 separate curated photo‑ID catalogs, supplemented with community science data. Data from throughout the North Pacific were aggregated into 13 regions, including six breeding regions, six feeding regions, and one migratory corridor. All images were compared with minimal pre‑processing using a recently developed image recognition algorithm based on machine learning through artificial intelligence; this system is capable of rapidly detecting matches between individuals …
Exploring Strain Variation And Bacteriophage Predation In The Gut Microbiome Of Ciona Robusta, Celine Grace F. Atkinson
Exploring Strain Variation And Bacteriophage Predation In The Gut Microbiome Of Ciona Robusta, Celine Grace F. Atkinson
USF Tampa Graduate Theses and Dissertations
Current microbiome studies have shown that the maintenance of homeostasis betweenmicrobial populations (e.g. bacteria, viruses) and the host immune system (e.g. innate immune molecules) is necessary for balancing health and disease outcomes within the host. These studies most often utilize vertebrate models; however, research in this field can benefit from diverse model systems that facilitate our ability to conduct experiments to identify phylogenically conserved rules influencing homeostasis in the gut of animals. The Dishaw has developed the use of a filter-feeding marine invertebrate chordate, Ciona robusta, to model such fundamental interactions[1]–[6]. While most biological diversity and functional contribution within microbiomes …
Utilization Of Detonation Nanodiamonds: Nanocarrier For Gene Therapy In Non-Small Cell Lung Cancer, Allan E. Gutierrez
Utilization Of Detonation Nanodiamonds: Nanocarrier For Gene Therapy In Non-Small Cell Lung Cancer, Allan E. Gutierrez
USF Tampa Graduate Theses and Dissertations
Lung cancer is one of the most prevalent and high mortality cancers in the world. Two different categories of lung cancer are non-small cell lung cancer which makes up 85% of cases, and small cell lung cancer which makes up the remaining 15% of cases. A significant risk factor that can lead to lung cancer is cigarette smoking (CS) however non-smoking cases i.e., secondhand CS, exposure to certain toxins and family history are on the rise. Both the main types of lung cancer have different system of stages, different ranges of treatment, and consequently low survival rates. Unfortunately, detection and …

BACKGROUND: ERK5 (extracellular signal-regulated kinase 5) is a dual kinase transcription factor containing an N-terminal kinase domain and a C-terminal transcriptional activation domain. Many ERK5 kinase inhibitors have been developed and tested to treat cancer and inflammatory diseases. However, recent data have raised questions about the role of the catalytic activity of ERK5 in proliferation and inflammation. We aimed to investigate how ERK5 reprograms myeloid cells to the proinflammatory senescent phenotype, subsequently leading to atherosclerosis.
METHODS: A ERK5 S496A (dephosphorylation mimic) knock in (KI) mouse model was generated using CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/clustered regularly interspaced short palindromic …
Variation In Salamander Life History And Community Composition Across An Urban Gradient In Atlanta, Georgia, Usa, Leah Rittenburg
Variation In Salamander Life History And Community Composition Across An Urban Gradient In Atlanta, Georgia, Usa, Leah Rittenburg
Master of Science in Integrative Biology Theses
Urbanization threatens to alter stream systems in watersheds in the eastern United States. These highly disturbed systems may result in many local extirpations of sensitive salamander species. One group of organisms that often persists in urban streams is the Eurycea bislineata (Two-lined Salamander) species complex. Many aspects of Two-lined Salamander life history are linked to environmental factors, particularly stream temperature, prey abundance, and stream hydrology. Urbanization threatens to alter these environmental factors, potentially influencing Two-lined Salamander life history. At 14 sites spanning an urban gradient in Atlanta, GA, I investigated the variation in life history and phenology in the Southern …

First Shasta Ground Sloth (Nothrotheriops; Xenarthra) From The Eastern Great Basin, Nevada, Jim I. Mead, Sandra L. Swift, H. Gregory Mcdonald, Steven D. Emslie
Western North American Naturalist
We present the first record of the Shasta ground sloth (Nothrotheriops shastensis) for the eastern Great Basin. The single right upper second molariform was recovered from the back dirt pile of archaeological test pits excavated during the 1960s and 1970s in Smith Creek Cave, northern Snake Range, White Pine County, Nevada. The precise age of the specimen is indeterminate, although all sediment layers in the area produced radiocarbon dates older than 11,000 cal BP; thus, the specimen is considered of the Rancholabrean Land Mammal Age. All previous accounts of this sloth in the Great Basin are from its …

Anaerobic Digestion Of Brewery Waste Including Spent Yeast And Hops, Dhanashree Rawalgaonkar
USF Tampa Graduate Theses and Dissertations
Florida is home to more than 300 craft breweries, that brew over a million barrels of beer annually. Brewing is an energy intensive process and produces large quantities of high strength waste including spent grains, yeast, hops, and high strength wastewater. Brewery wastewater, and spent yeast have high Chemical Oxygen Demand (COD) concentrations between 5000 mg/L – 10,000 mg/L, and 200,000 mg/L – 300,000 mg/L respectively. Brewery effluent wastewater is sent to Publicly Owned Treatment Works (POTWs) which implement high surcharges based on the strength of brewery effluent (COD concentration). While some of the spent yeast and hops can be …

Outdated United States' Online Copyright Infringement Practices: What We Can Learn From The International Community, Ian Carstens
San Diego International Law Journal
Current United States practices and regulations regarding what is known as “the Server Test,” the DMCA, and online piracy need to be studied and brought into the realities of the modern world. With new technologies and uses being invented for the internet every day, the U.S. cannot afford to drag its heels any longer or it risks their statutory and case law becoming dangerously underequipped to deal with daily life. This paper proposes that the United States government listen to its own departments, the wishes of its citizens, and the examples being broadcasted internationally to form a new system to …
Leveraging Protein Dynamics To Drug Filovirus Protein-Nucleic Acid Interactions Using Simulations And Experiments, Matthew Alberto Cruz
Leveraging Protein Dynamics To Drug Filovirus Protein-Nucleic Acid Interactions Using Simulations And Experiments, Matthew Alberto Cruz
Arts & Sciences Graduate Student Theses and Dissertations
Drug discovery targeting protein nucleic acid (PNI) and protein-protein interactions (PPI) remains a difficult task. How to identify druggable conformations and discover compounds binding to these conformations is an unsolved problem. Here, I describe how we apply computational and experimental approaches to probe the conformational landscape of a key immune antagonist protein from the Zaire ebolavirus. Viral Protein 35 (VP35) binds to the viral pathogen associated molecular pattern, double stranded RNA (dsRNA), and blocks activation of the interferon (IFN) response. Long time scale molecular dynamics simulations reveal that VP35 adopts an alternative conformation which opens a pocket absent in experimental …

Mesenchymal stromal cells (MSCs) are a key component of the bone marrow (BM) niche, providing essential support required for the maintenance of hematopoietic stem cells. To advance our understanding of physiological functions of p53 and Mdm2 in BM-MSCs, we developed traceable conditional mouse models targeting Mdm2 and/or Trp53 in vivo. We demonstrate that Mdm2 is essential for the emergence, maintenance, and hematopoietic support of BM-MSCs. Mdm2 haploinsufficiency in BM-MSCs resulted in genotoxic stress-associated thrombocytopenia, suggesting a functional role for Mdm2 in hematopoiesis. In a syngeneic mouse model of acute myeloid leukemia (AML), Trp53 deletion in BM-MSCs improved survival, and protected …

Gastrulation begins when the epiblast forms the primitive streak or becomes definitive ectoderm. During this lineage bifurcation, the DNA dioxygenase TET1 has bipartite functions in transcriptional activation and repression, but the mechanisms remain unclear. By converting mouse embryonic stem cells (ESCs) into neuroprogenitors, we defined how Tet1-/- cells switch from neuroectoderm fate to form mesoderm and endoderm. We identified the Wnt repressor Tcf7l1 as a TET1 target that suppresses Wnt/β-catenin and Nodal signalling. ESCs expressing catalytic dead TET1 retain neural potential but activate Nodal and subsequently Wnt/β-catenin pathways to generate also mesoderm and endoderm. At CpG-poor distal enhancers, TET1 maintains …

The Pact Act As Indicum Of The Due Process Validity Of The Marketplace Fairness Act, Eric S. Smith
The Pact Act As Indicum Of The Due Process Validity Of The Marketplace Fairness Act, Eric S. Smith
Florida Tax Review
If passed into law, the Marketplace Fairness Act would impose a federal duty on out-of-state sellers to collect a state-defined and state-benefitting use tax. This unique exercise of federal power implicates due process. The Prevent All Cigarette Trafficking Act of 2009 (PACT Act) represents the only other instance in which Congress has similarly compelled state law compliance. Two circuit courts of appeals have found the PACT Act vulnerable on concerns of due process. This Article relies on the PACT Act analogue to simulate how the Marketplace Fairness Act would fare under similar scrutiny. To this end and in this uncommon …
Tax Compliance As A Wicked System, J. T. Manhire
Tax Compliance As A Wicked System, J. T. Manhire
Florida Tax Review
This Article proposes a new typology and framework for tax compliance systems. Traditionally-competing approaches such as deterrence theory, behaviorist theory, and game theoretic models taken together suggest that tax compliance is perhaps a new type of system—a “wicked system”—that is only partially comprehensible by understanding the traditional theories alone. If correct, previously competing theories become simply different limiting cases of the same underlying “wicked system.” The Article concludes with a discussion of the framework’s limitations and presents initial solutions and challenges for future work.

Evaluating The Post-Mining Land Uses Of Former Mine Sites For Sustainable Purposes In South Africa, Sphiwe E Mhlongo
Journal of Sustainable Mining
This paper uses a case study of an abandoned magnesite mine in the Limpopo Province of South Africa to find ways of identifying post-mining land used from the current uses of the abandoned mine sites or features. The approach used involved carrying out a field characterization of the mine site and documentation of the current uses of the features of the abandoned mine site. The technique used to identify the internal and external factors of the land uses involved analyzing their Strength, Weakness, Opportunities, and Threats. The Analytic Hierarchy Process (AHP) technique was used for further ranking of the land …

Diabetic retinopathy (DR) is a leading cause of blindness in working-age adults and remains an important public health issue worldwide. Here we demonstrate that the expression of stimulator of interferon genes (STING) is increased in patients with DR and animal models of diabetic eye disease. STING has been previously shown to regulate cell senescence and inflammation, key contributors to the development and progression of DR. To investigate the mechanism whereby STING contributes to the pathogenesis of DR, diabetes was induced in STING-KO mice and STINGGT (loss-of-function mutation) mice, and molecular alterations and pathological changes in the retina were characterized. We …

Homeostatic plasticity represents a set of mechanisms that are thought to recover some aspect of neural function. One such mechanism called AMPAergic scaling was thought to be a likely candidate to homeostatically control spiking activity. However, recent findings have forced us to reconsider this idea as several studies suggest AMPAergic scaling is not directly triggered by changes in spiking. Moreover, studies examining homeostatic perturbations in vivo have suggested that GABAergic synapses may be more critical in terms of spiking homeostasis. Here we show results that GABAergic scaling can act to homeostatically control spiking levels. We find that increased or decreased …

Purpose: The Rio Grande Valley in South Texas comprises 5% percent of Texas's population yet 17%of Texas's COVID-19 deaths. We aimed to address underlying mistrust and systemic racism in our Hispanic community that contributes to health inequities by developing a cultural competence guide for public health messaging.
Methods: We employed a mixed method design (e.g., focus groups, surveys, interviews) to develop and implement a cultural competence guide in an iterative community-informed process. We created a general cultural competence guide, one for the Hispanic community and one for the hard-of-hearing community.
Results: Our cultural competence guides provide an interpretation as to …

Respiration can positively influence cerebrospinal fluid (CSF) flow in the brain, yet its effects on central nervous system (CNS) fluid homeostasis, including waste clearance function via glymphatic and meningeal lymphatic systems, remain unclear. Here, we investigated the effect of supporting respiratory function via continuous positive airway pressure (CPAP) on glymphatic-lymphatic function in spontaneously breathing anesthetized rodents. To do this, we used a systems approach combining engineering, MRI, computational fluid dynamics analysis, and physiological testing. We first designed a nasal CPAP device for use in the rat and demonstrated that it functioned similarly to clinical devices, as evidenced by its ability …

Ecology And Retribution: Blake, Tokarczuk, And Animal Rights, Kristina Isaak Powell
Theses and Dissertations
This thesis explores how Nobel laureate Olga Tokarczuk's 2008 novel, Drive Your Plow Over the Bones of the Dead, engages with William Blake's life and his writings on animal welfare and speaks to current conversations about multispecies justice in the environmental humanities. It argues, first, that in recognizing how this novel's protagonist, Janina, selectively reads Blake to rationalize retributive justice, readers should resist a tendency to mistake this character for Tokarczuk's ideal advocate for environmental ethics. Secondly, it asserts that legal scholars' division between retributive and restorative justice offers valuable framework for approaching both this novel and ongoing debates about …
Concentrated Animal Feeding Operations, Environmental Quality And Public Health, Suraj Ghimire
Concentrated Animal Feeding Operations, Environmental Quality And Public Health, Suraj Ghimire
Economics ETDs
Using dairy farm data, this dissertation investigates the environmental and health implications of concentrated animal feeding operations (CAFOs) in New Mexico. It seeks to answer three primary research questions: (1) the relationship between environmental justice indicators and proximity to dairy farms; (2) the overall health consequences of dairy air pollutants; and (3) the viability of implementing an anaerobic digester (AD) system as a potential solution to externality concerns while maintaining dairy farm economic sustainability.
The first study demonstrates that foreign-born populations and Hispanics disproportionately experience emissions from CAFOs, with living near dairy farms or in areas with elevated ammonia levels …
The Manhattant: Identification, Natural History, And Nutritional Ecology Of A Recently Introduced Pest In New York City, Lasius Emarginatus (Hymenoptera: Formicidae), Samantha Kennett
Master of Science in Integrative Biology Theses
New York City is one of the oldest and largest cities in North America, and it is a center for global trade and shipping. As such, it has also been the site of introduction for some of North America’s most damaging invasive pests, from chestnut blight to the Asian long-horned beetle. Despite these cautionary examples, there has been no formal research on a newly introduced ant species that was tentatively identified as a European ant species, Lasius cf. emarginatus. Since its discovery in 2011, L. cf. emarginatus has become among the most common urban ant species in New York …
Potassium Ion Channels As A Molecular Target To Reduce Virus Infection And Mortality Of Honey Bee Colonies, Christopher J. Fellows, Michael Simone‑Finstrom, Troy D. Anderson, Daniel R. Swale
Potassium Ion Channels As A Molecular Target To Reduce Virus Infection And Mortality Of Honey Bee Colonies, Christopher J. Fellows, Michael Simone‑Finstrom, Troy D. Anderson, Daniel R. Swale

Declines in managed honey bee populations are multifactorial but closely associated with reduced virus immunocompetence and thus, mechanisms to enhance immune function are likely to reduce viral infection rates and increase colony viability. However, gaps in knowledge regarding physiological mechanisms or ‘druggable’ target sites to enhance bee immunocompetence has prevented therapeutics development to reduce virus infection. Our data bridge this knowledge gap by identifying ATP-sensitive inward rectifier potassium ( KATP) channels as a pharmacologically tractable target for reducing virus-mediated mortality and viral replication in bees, as well as increasing an aspect of colony-level immunity. Bees infected with Israeli …

Colonial documents preserve information that allows us to know the local Andean history of the Viceroyalty of New Granada. These manuscripts reveal forms of violence that shaped the subjectivities of the time and the resistance of oppressed women. This dissertation examines the effects of slavery and the response of three enslaved women to that colonial violence. This analysis seeks to better understand and make visible how the intersection between racism and patriarchy impacted the lives of three racialized women in the colonial context.
This dissertation focuses on the experiences, struggles, and resistance of three women present in the manuscripts consigned …
Examination Of Microplastics And Organic Contaminants In Tampa Bay’S Eastern Oyster (Crassostrea Virginica) Through Visual And Chemical Analysis, Andrea Murray
USF Tampa Graduate Theses and Dissertations
Microplastic pollution is an increasingly alarming concern with widespread global distribution in aquatic environments. Filter feeding organisms such as bivalves, have a heightened risk of microplastic ingestion through free floating particulates in the water. This can lead to bioaccumulation moving up through trophic levels to apex predators and humans. Spatial and temporal differences in microplastic abundance were evaluated in the Eastern Oyster, Crassostrea virginica, at six sites within Tampa Bay. Oyster tissue was digested using 30% hydrogen peroxide (H2O2) and microplastics were quantified using Nile Red dye and fluorescent particle excitement. A total of 3025 microplastics were found throughout six …
Stable Isotope Analysis Of Doryteuthis (Amerigo) Pealeii Eye Lenses To Determine Migratory Patterns In The Eastern Gulf Of Mexico Using Statoliths For Age Determination, Hannah M. Schwaiger
Stable Isotope Analysis Of Doryteuthis (Amerigo) Pealeii Eye Lenses To Determine Migratory Patterns In The Eastern Gulf Of Mexico Using Statoliths For Age Determination, Hannah M. Schwaiger
USF Tampa Graduate Theses and Dissertations
Doryteuthis (Amerigo) pealeii is a common fisheries squid that occurs in the Gulf of Mexico (GoM) and along the east coast of the United States. These squid are an important link in the food web, linking higher and lower trophic positions (Madsen et al., 2007), and they are also used for human consumption. The migration patterns of D. pealeii populations in the northwestern North Atlantic have been thoroughly studied when compared to those in the GoM. This research aims to combine statolith aging and stable isotope analysis to enhance the understanding of D. pealeii migration patterns throughout their short, sub-annual …
